Claims (1)
- 【特許請求の範囲】 1. ルリヂサ△6-デサチュラーゼをコードする単離された核酸。 2. SEQ ID NO:4のヌクレオチド配列を含む請求項1記載の単離され た核酸。 3. SEQ ID NO:5のアミノ酸配列をコードする単離された核酸。 4. 請求項1ないし3のいずれか一項に記載の核酸を含むベクター。 5. 単離された核酸の遺伝子産物の発現に作用しうるプロモーターおよび任意 に終結シグナルに使用可能に連結された請求項1ないし3のいずれか一項に記載 の単離された核酸を含む発現ベクター。 6. プロモーターが、△6-デサチュラーゼプロモーター、アナベナカルボキ シラーゼプロモーター、ヘリアンチニンプロモーター、グリシニンプロモーター 、ナピンプロモーター、CaMVの35Sプロモーターもしくはヘリアンチニン 組織特異的プロモーターである、請求項5記載の発現ベクター。 7. プロモーターが、構成性もしくは組織特異的である、請求項5記載の発現 ベクター。 8. 終結シグナルが、シネコシスティス終結シグナル、ノパリンシンターゼ終 結シグナル、もしくは種子終結シグナルである、請求項5記載の発現ベクター。 9. 請求項4ないし8のいずれか一項に記載のベクターを含む細胞。 10. 動物細胞、細菌細胞、植物細胞もしくは真菌細胞である、請求項9記載 の細胞。 11. 請求項1ないし3のいずれか一項に記載の単離された核酸を含むトラン スジェニック生物体。 12. 請求項4ないし8のいずれか一項記載のベクターを含むトランスジェニ ック生物体。 13. 細菌、真菌、植物もしくは動物である、請求項11または12記載のト ランスジェニック生物体。 14. 請求項10記載の植物細胞から再生された植物もしくは該植物の子孫。 15. ヒマワリ、大豆、トウモロコシ、タバコ、ピーナツ、ニンジンもしくは アブラナである、請求項14記載の植物。 16. (a)請求項1ないし3のいずれか一項に記載の単離された核酸で植物 細胞を形質転換し、かつ、 (b)前記植物細胞からγリノレン酸(GLA)含量が増大した植物を再生する ことを含む、GLA含量が増大した植物を産生する方法。 17. (a)請求項4ないし8のいずれか一項に記載のベクターで植物細胞を 形質転換し、かつ、 (b)前記植物細胞からγリノレン酸(GLA)含量が増大した植物を再生する ことを含む、GLA含量が増大した植物を産生する方法。 18. 植物が、ヒマワリ、大豆、トウモロコシ、タバコ、ピーナツ、ニンジン もしくはアブラナである、請求項16または17記載の方法。 19. 請求項1ないし3のいずれか一項記載の単離された核酸で生物体を形質 転換することを含む、γリノレン酸(GLA)が不十分もしくはGLAを欠失し た生物体におけるGLAの産生を誘導する方法。 20. 請求項4ないし8のいずれか一項に記載のベクターで生物体を形質転換 することを含む、γリノレン酸(GLA)が不十分もしくはGLAを欠失した生 物体におけるGLAの産生を誘導する方法。 21. ルリヂサ△6-デサチュラーゼをコードする単離された核酸および△1 2-デサチュラーゼをコードする単離された核酸で生物体を形質転換することを 含む、γリノレン酸(GLA)およびリノール酸(LA)が不十分もしくはGL AおよびLAを欠失した生物体におけるGLAの産生を誘導する方法。 22. △6-デサチュラーゼをコードする単離された核酸が、SEQ ID N O:4のヌクレオチド44〜1390を含む、請求項21記載の方法。 23. 請求項1ないし3のいずれか一項記載の単離された核酸で生物体を形質 転換することを含む、γリノレン酸が不十分もしくはγリノレン酸を欠失した生 物体におけるオクタデカテトラエン酸の産生を誘導する方法。 24. 請求項4ないし8のいずれか一項に記載のベクターで生物体を形質転換 することを含む、γリノレン酸が不十分もしくはγリノレン酸を欠失した生物体 におけるオクタデカテトラエン酸の産生を誘導する方法。 25. 生物体が、細菌、真菌、植物もしくは動物である、請求項23または2 4記載の方法。 26. (a)請求項1ないし3のいずれか一項に記載の単離された核酸で植物 細胞を形質転換し、かつ、 (b)前記形質転換された植物細胞から寒冷耐性が改良された植物を再生するこ とを含む、寒冷耐性が改良された植物を産生する方法。 27. (a)請求項4ないし8のいずれか一項に記載のベクターで植物細胞を 形質転換し、かつ、 (b)前記形質転換された植物細胞から寒冷耐性が改良された植物を再生するこ とを含む、寒冷耐性が改良された植物を産生する方法。 28. 植物が、ヒマワリ、大豆、トウモロコシ、タバコ、ピーナツ、ニンジン もしくはアブラナである、請求項26または27記載の方法。
