Description
cell)」とは、脂肪組織から由来する幹細胞であって、脂肪組織は、多量の組織採取が容易であることから、幹細胞の収穫に良い条件を有し、脂肪由来幹細胞は、培養時に安定的な成長と増殖を示し、分化を誘導したときに多様な細胞への分化が可能である。
1)幹細胞を増殖させる段階と、
2)前記増殖した幹細胞を骨細胞へ分化させる段階と、
3)骨細胞へ分化する幹細胞からエキソソームを抽出及び精製する段階と、を含む方法を用いることができる。
増殖した幹細胞からエキソソームを抽出するために、ヒト脂肪由来幹細胞(passage 3−7)を、10%ウシ胎児血清(fetal bovine serum;FBS)及び1%ペニシリン/ストレプトマイシン(penicillin/streptomycin)が含まれている通常の培養培地(Dulbecco Modified Eagle Medium high glucose;DMEM)で培養した後、無血清、無抗生剤、無フェノールレッド(phenol red−free)のDMEM培地に変えて、24時間維持して得た細胞培養上澄液を回収した。これより、増殖したヒト脂肪由来幹細胞からエキソソームを抽出した。
本発明の一実施例において、エキソソームは、ヒト脂肪由来幹細胞が骨細胞へ分化する過程で抽出した。
前記実施例1及び実施例2から抽出したエキソソームを透過電子顕微鏡(transmission electron microscope)を用いて確認したエキソソームの構造及び模様と、ナノ粒子分析機(nanoparticle tracking analysis)を用いて確認したエキソソームの大きさ分布と、を確認した。その結果は、図2に示した。
本発明の実施例1及び実施例2で抽出したエキソソーム1×108パーチクル/mLをhuman osteoprotegerin ELISA Kitを用いてエキソソーム内のOPG発現を定量分析した。この際、比較対照群として、骨髓由来幹細胞(bone−marrow mesenchymal stem cells)から抽出したエキソソーム(BM−MSC−EXO)を用いた。
エキソソームを用いて骨再生及び骨密度増進を確認するために、増殖した幹細胞からのエキソソーム(ASC−EXO)及び骨細胞へ分化中の幹細胞から抽出されたエキソソーム(Osteo−EXO)を、骨粗鬆症が誘発されたマウス(卵巣摘出モデル)に1×108パーチクル/mLのパーティクル濃度で2週間6回静脈に投与した。陰性対照群としては、PBSを2週間6回静脈に投与した。陽性対照群としては、脂肪由来幹細胞(ASC)を5×105個ずつ2回静脈に投与し、開腹手術のみを行ったsham群も共に分析した。
Claims (5)
- 増殖した脂肪組織由来幹細胞から抽出されたエキソソームを有効成分として含む、骨粗鬆症の予防または治療用の薬学的組成物。
- 前記脂肪組織由来幹細胞が、ヒトまたは哺乳動物由来幹細胞である、請求項1に記載の骨粗鬆症の予防または治療用の薬学的組成物。
- 増殖した脂肪組織由来幹細胞から抽出されたエキソソームを有効成分として含む、骨粗鬆症の予防または治療用の注射剤。
- 前記注射剤は、エキソソームが1×106〜1×1011パーチクル/mLの濃度で含まれるものである、請求項3に記載の骨粗鬆症の予防または治療用の注射剤。
- 増殖した脂肪組織由来幹細胞から抽出されたエキソソームを有効成分として含む、骨粗鬆症の予防または改善用の健康機能食品。
