Description
本発明は、経済的かつ効率的に高純度のプロスタグランジン誘導体を製造する方法及びそのための中間体に関する。
プロスタグランジン誘導体のうち、化学式2のトラボプロスト(Travoprost)、ビマトプロスト(Bimatoprost)、ラタノプロスト(Latanoprost)は、眼圧の低下及び毛髪や眉毛の成長促進等、有用な臨床効果によって商業的に広く使用されている。
〔発明が解決しようとする課題〕
本発明者らは、共役付加反応を利用したプロスタグランジンF(PGF)誘導体の合成において、前記問題を解決するために鋭意研究、検討した結果、共役付加反応後に得られるプロスタグランジンE(PGE)誘導体の保護基をまず除去してから、シクロペンタノン環に存在するケトン基を立体選択的に還元させることにより、β−OHがほとんど存在しない高純度のPGF誘導体を効率的かつ経済的に製造することができることを発見し、本発明を完成するに至った。
本発明は、下記化学式1のプロスタグランジンF(PGF)誘導体の製造方法に関し、本発明の製造方法は、
(i)下記化学式5の保護化プロスタグランジンE(PGE)誘導体のヒドロキシ保護基を除去するために、下記化学式6のプロスタグランジンE(PGE)誘導体を得るステップ、及び
(ii)下記化学式6の化合物のシクロペンタノン環に存在するケトン基を立体選択的に還元反応させるステップ、を含む。
Xは、O又はNHを示し、
Yは、α−OH又はジフルオロ、好ましくは、α−OHを示し、
Y’は、α−OPG又はジフルオロ、好ましくは、α−OPGを示し、
Zは、CH2、O又はS、好ましくは、CH2又はOを示し、
Rは、H又はC1−C5のアルキル基、好ましくは、C1−C5のアルキル基を示し、
R’は、C1−C5のアルキル基、C3−C7のシクロアルキル基又はアリール基、好ましくは、C1−C5のハロアルキル基又はハロゲン、より好ましくは、CF3、Cl又はF、最も好ましくは、CF3で置換又は非置換されたフェニル基を示し、
PGは、ヒドロキシ保護基、好ましくはテトラヒドロピラニル、トリメチルシリル、トリエチルシリル又はt−ブチルジメチルシリル、より好ましくは、トリエチルシリルを示す。
前記化学式6のプロスタグランジンE(PGE)誘導体は、前記化学式5の保護化プロスタグランジンE(PGE)誘導体のヒドロキシ保護基を除去して製造する。
化学式1のプロスタグランジンF(PGF)誘導体は、前記化学式6の化合物のシクロペンタノン環に存在するケトン基を立体選択的に還元反応させて製造する。
本発明によれば、高純度のプロスタグランジンF(PGF)誘導体を、共役付加反応を通じて得られたプロスタグランジンE(PGE)誘導体の保護基をまず除去してから、シクロペンタノン環に存在するケトン基を立体選択的に還元させることにより、効率的かつ経済的に製造することができる。特に、還元剤として2,6−ジ−t−ブチル−4−メチルフェノール(di−tert−butyl methyl phenol)及び水素化ジイソブチルアルミニウム(DIBAL)を使用する場合、α−OHのみを立体選択的に製造することができる。
以下、実施例により本発明をより具体的に説明することとするが、これらの実施例は、単に本発明を説明するためのものであって、本発明の範囲がこれら実施例に極限されないということは、当業者にとって自明のことである。
Claims (14)
- (i)下記化学式5の保護化プロスタグランジンE(PGE)誘導体のヒドロキシ保護基を除去して、下記化学式6のプロスタグランジンE(PGE)誘導体を得るステップ;及び
(ii)2,6−ジ−t−ブチル−4−メチルフェノール(2,6−di−tert−butyl−4−methyl phenol)及び水素化ジイソブチルアルミニウム(DIBAL)を還元剤として使用して、下記化学式6の化合物のシクロペンタノン環に存在するケトン基を立体選択的に還元反応させるステップ;
を含む、下記化学式1のプロスタグランジンF(PGF)誘導体の製造方法。
Xは、O又はNHを示し、
Yは、α−OH又はジフルオロを示し、
Y’は、α−OPG又はジフルオロを示し、
Zは、CH2、O又はSを示し、
Rは、H又はC1−C5のアルキル基を示し、
R’は、C1−C5のアルキル基、C3−C7のシクロアルキル基又はアリール基を示し、
PGは、ヒドロキシ保護基を示す。) - ヒドロキシ保護基が、テトラヒドロピラニル、トリメチルシリル、トリエチルシリル又はt−ブチルジメチルシリルであることを特徴とする、請求項1〜4のいずれか一項記載の製造方法。
- ヒドロキシの保護基がトリエチルシリルであることを特徴とする、請求項5記載の製造方法。
- ステップ(i)において、化学式5の保護化プロスタグランジンE(PGE)誘導体のヒドロキシ保護基を酸性条件下で除去することを特徴とする、請求項1〜4のいずれか一項記載の製造方法。
- 酸性条件として、p−トルエンスルホン酸ピリジニウム(pyridinium p−toluenesulfonate:PPTS)を使用することを特徴とする、請求項7記載の製造方法。
- XがOであり、Rがメチルである化学式1のプロスタグランジンF(PGF)誘導体をRNH2と反応させて、XがNHである化学式1のプロスタグランジンF(PGF)誘導体を製造することを特徴とする、請求項1〜4のいずれか一項記載の製造方法。
- 化学式1のプロスタグランジンF(PGF)誘導体を、炭化水素とアルコールの混合溶媒又はジクロロメタンとアルコールの混合溶媒でHPLCして精製するステップ、をさらに含むことを特徴とする、請求項1〜4のいずれか一項記載の製造方法。
- 炭化水素とアルコールの混合溶媒が、n−ヘキサンと無水エタノールの混合溶媒又はn−ヘプタンと無水エタノールの混合溶媒であることを特徴とする、請求項12記載の製造方法。
- ジクロロメタンとアルコールの混合溶媒が、ジクロロメタンとイソプロパノールの混合溶媒であることを特徴とする、請求項12記載の製造方法。
