Description
−カルボキシヘプタデカノイル)−des(B30)ヒトインスリン及びB29−N−(ω−カルボキシヘプタデカノイル)ヒトインスリンである。
(a)プレプロインスリン、プレプロインスリンアナログ又はプレプロインスリン誘導体は、Ser172Alaブタトリプシンで切断されるものとし、
(b)この得られる切断産物は、分離され、そして
(aa)この得られる切断産物のうちの1つが、インスリンアナログ若しくはインスリン誘導体である場合、このインスリンアナログ若しくはインスリン誘導体が得られることになり、又は
(bb)インスリン、インスリンアナログ若しくはインスリン誘導体の前駆体であるこれらの切断産物はさらにプロセシングされ、そしてこのようなさらなるプロセシングから得られるインスリン、インスリンアナログ若しくはインスリン誘導体が分離されそして得られることになり;
ここでインスリンは、好ましくはヒトインスリンであり;そしてインスリンアナログは、LysB28ProB29ヒトインスリン、B28Aspヒトインスリン、B28位のプロリンが、Asp、Lys、Leu、Val又はAlaで置換されており、そしてここでB29位のLysが、Proで置換され得るヒトインスリン;AlaB26ヒトインスリン;des(B28−B30)ヒトインスリン; des(B27)ヒトインスリン及びdes(B30)ヒトインスリンを含む群より選択され、そしてインスリン誘導体は、B29−N−ミリストイル−des(B30)ヒトインスリン、B29−N−パルミトイル−des(B30)ヒトインスリン、B29−N−ミリストイルヒトインスリン、B29−N−パルミトイルヒトインスリン、B28−N−ミリストイルLysB28ProB29ヒトインスリン、B28−N−パルミトイル−LysB28ProB29ヒトインスリン、B30−N−ミリストイル−ThrB29LysB30ヒトインスリン、B30−N−パルミトイル−ThrB29LysB30ヒトインスリン、B29−N−(N−パルミトイル−Y−グルタミル)−des(B30)ヒトインスリン、B29−N−(N−リトコリル−Y−グルタミル)−des(B30)ヒトインスリン、B29−N−(ω−カルボキシヘプタデカノイル)−des(B30)ヒトインスリン及びB29−N−(ω−カルボキシヘプタデカノイル)ヒトインスリンを含む群より選択されるものとする。
本発明のさらなる実施態様において、インスリン、インスリンアナログ又はインスリン誘導体の前駆体であるこれらの切断産物の上記のさらなるプロセシングは、インスリングラルギンを除くカルボキシペプチダーゼBでこの産物を切断することを含むものである。
本発明の別の実施態様は、上記のDNAを含むベクターである。
(a)請求項16に記載のベクターを提供すること、
(b)このベクターで微生物宿主株を形質転換すること、
(c)この形質転換された微生物宿主株を、栄養分を含む増殖培地で培養し、これによってこの微生物宿主株が、Ser172Alaブタトリプシン又はSer172Alaブタトリプシノーゲンを発現すること、

を含むものであり、特にここで、微生物宿主株は、Hansenula、Pichia、Candida及びTorulopsis種を含む群より選択されるメチロトローフ酵母株であり;好ましくはここで、微生物宿主株は、Pichia pastoris、Hansenula polymorpha、Candida boidinii及びTorulopsis glabrataを含む群より選択されるものとする。
トリプシン活性が定量化されると、本明細書では「単位」(U)を適用する。トリプシン及びその変異体のタンパク質分解活性は、基質としてChromozym TRY、Chromozym TH及びChromozym PL(Roche Diagnostics GmbH)を使用した測光アッセイを用いて定量化される。与えられた調製物の「タンパク質分解比活性」又は「比活性」は、調製物中のタンパク質1mg当たりの単位数として定義され、実施例9に記載される方法によって測定される。
「ベクター」は、本発明のDNAフラグメントを含み得る、すなわち保有及び維持し得るDNAとして定義され、例えばファージ及びプラスミドが挙げられる。これらの用語は、遺伝子工学分野の当業者に理解されている。用語「発現カセット」は、プロモーター及びターミネーターに作動可能に連結した、プレタンパク質をコードするヌクレオチド配列を表している。発現カセットを含むベクターに関しては、用語「ベクター」及び「発現ベクター」は、同義語として使用される。
「形質転換」は、DNAを生物、すなわち宿主生物に導入して、その結果このDNAが、染色体外エレメントとして、又は染色体への組込みのいずれかによって複製可能なことを意味している。
核酸又はそのその相補体の少なくとも一部分が直接翻訳されて、ペプチド若しくはタンパク質のアミノ酸配列をもたらし得るか、又はこの単離核酸が、単独若しくは発現ベクターの一部として使用されて、インビトロ、原核生物宿主細胞内若しくは真核生物宿主細胞内でこのペプチド若しくはタンパク質を発現し得るときに、ヌクレオチド配列は、ペプチド又はタンパク質を「コードしている」。
「プロモーター」は、転写を刺激する調節ヌクレオチド配列である。これらの用語は、遺伝子工学分野の当業者に理解されている。プロモーターと同様に、「プロモーターエレメント」は、転写を刺激するが、より長いプロモーター配列のサブフラグメントを構成される。
「シグナルペプチド」は、分泌タンパク質のプレタンパク質形態又はプレプロタンパク質形態で存在しているアミノ酸の切断可能なシグナル配列である。細胞膜を横切って輸送される、すなわち「分泌される」タンパク質は、代表的には、疎水性アミノ酸が豊富なN末端配列を有しており、代表的には、およそ15〜30アミノ酸長である。膜を通過するプロセスの間のある時に、このシグナル配列は、シグナルペプチダーゼによって切断される(Alberts,B.、Johnson,A.、Lewis,J.、Raff,M.、Roberts,K.、Walter,P.(編)、Molecular Biology of the Cell、第4版、2002、Garland Science Publishing)。シグナルペプチドの多くの供給源は、当業者に周知であり、そして例えば、Saccharomyces cerevisiae由来のα−因子シグナルペプチドのアミノ酸配列などが挙げられ得る。別の例は、配列番号5の1〜16位のネイティブブタ膵臓トリプシノーゲンプレタンパク質のブタシグナルペプチドである。一般に、本質的にどのような分泌タンパク質のプレタンパク質のN末端でも、本発明の使用に好適なシグナルペプチドの可能性のある供給源なのである。シグナルペプチドはまた、二分性(bipartite)であり得るものであり、プレタンパク質を第一の細胞区画及び第二の細胞区画に導く2つのシグナルペプチドを含んでいる。二分性のシグナルペプチドは、分泌経路の経過の間に、段階的に切り離される。従って特定の例は、Saccharomyces cerevisiae由来のα−因子のプレプロペプチドである(Watersら、J.Biol.Chem.263(1988)6209〜14)。
5,032,516、US 5,122,465、US 5,135,868、US 5,166,329、WO 00/56903)。グルコースが存在しないとき、Pichia pastorisは、炭素源としてメタノールを用いており、これは同時に、メチロトローフ生物の特徴である。配列番号7で与えられるアルコールオキシダーゼ(AOX1)プロモーターは、アルコールオキシダーゼの発現を制御しており、これは、メタノール代謝の最初の工程を触媒している。代表的には、メタノール誘導性細胞の可溶性タンパク質全体の30%が、アルコールオキシダーゼである。いくつかのPichia発現ベクターは、AOX1プロモーターを保有し、そしてメタノールを使用して、所望の異種タンパク質の高レベル発現を誘導している。発現構築物はまた、Pichia pastorisゲノムに組み込まれて、形質転換され、そして遺伝的に安定な宿主を生成する。
増殖培地へのブタトリプシン変異体の分泌は、増殖培地中に拡散する細胞質外空間に成熟組換えタンパク質を導くことである。液体培養液中で増殖した形質転換されたメチロトローフ酵母は、組換えブタ膵臓トリプシン変異体を液体増殖培地、すなわち液体培養基に分泌する。これにより、例えば濾過技術を用いて、酵母バイオマスと組換えタンパク質とを極めて効率的に分離し得る。結果として、この供給源から精製された組換えブタトリプシン変異体は、他の酵素活性、例えばリボヌクレアーゼ活性又は他の(非トリプシン)プロテアーゼ活性と極めて効率的に分けられる。
れたメチロトローフ酵母株に対して、Zeocin(登録商標)に対する耐性レベルのさらなる増大がこれ以上得られないか、又は選択培地中のZeocin(登録商標)濃度のさらなる増大がこれ以上は不可能になるまで、形質転換を反復し、そしてなおより耐性の形質転換体の選択を反復することがさらに有利である。
固定相:Nucleosil 120−5 C18、Macherey&Nagel、250×4mm;移動相A:45mMリン酸ナトリウム緩衝液(pH2.5)、315mM NaCl、25%(v/v)アセトニトリル;移動相B:45mMリン酸ナトリウム緩衝液(pH2.5)、55mM NaCl、65%(v/v)アセトニトリル;勾配:30分以内で6%相B〜10%相Bの直線。
実施例1:組換えヒト野生型トリプシン及びセリン190アラニンヒトトリプシン変異体を用いた、プレプロインスリングラルギンの切断
これらの実験を、8℃及びpH値8.3(緩衝化溶液)で行い、そして50mLスケールまでで行った。
PPI溶液を、適切なサーモスタット反応容器に満たし、そして酵素調製物を添加することにより反応を開始させた。一定の時間間隔後にサンプルを採取した。1N又は2N
HCl溶液によってサンプル溶液を酸性化することにより、酵素反応を直ちに停止させた。それぞれの産物の濃度を、HPLCによって測定した。
一般に、Sambrook,Fritsch&Maniatis、Molecular Cloning、A Laboratory Manual、第3版、CSHL Press、2001に記載されるような分子生物学の標準的方法を適用した。以下に説明される方法は、「部位特異的突然変異誘発」としてまた公知の極めて一般的な方法の具体的な適用である。
突然変異を、ポリメラーゼ連鎖反応(PCR)を用いて部位特異的様式で生成した。所望のコドン、すなわち塩基トリプレットを突然変異させるために、ブタ膵臓トリプシノーゲンをコードする合成ヌクレオチド配列の変異体部分を表す相補的な一本鎖DNAオリゴヌクレオチド対を設計及び合成した。この一本鎖DNAオリゴヌクレオチドは、突然変異されるトリプレット配列を除いては、配列番号2で与えられる配列と同一又は相補的であった。代表的には、DNAオリゴヌクレオチドは、およそ20〜45のヌクレオチド長を有し、突然変異されるトリプレット配列又はその相補体は、それを含むDNAオリゴヌクレオチドの中央部分に位置し、そして両側がおよそ10〜12ヌクレオチド隣接していた。このDNAオリゴヌクレオチドを、「野生型」組換えブタ膵臓トリプシノーゲンDNA(配列番号2)とのDNAオリゴヌクレオチドのハイブリダイゼーションが、中央のミスマッチとのハイブリッドをもたらすが、このミスマッチの側面のインタクトな塩基対合(各DNAオリゴヌクレオチドの5’末端及び3’末端を含む)とはハイブリッドを生じないように設計した。
野生型成熟ブタ膵臓トリプシンタンパク質のアミノ酸置換による変異体をコードするヌクレオチド配列を、いくつかのPCRに基づいた工程によって合成した。
得られる2つの中間増幅産物を、アガロースゲル電気泳動によって分析し、所望のフラグメントを切り出し、そして「QIAquick Gel Extraction Kit」(Qiagen、カタログ番号28704)を用いて、アガロースブロックからDNAを単離した。
続いて、突然変異した全長DNAフラグメントを、「PCRクローニングキット−平滑末端」(Roche Diagnostics GmbH、Mannheim;カタログ番号1 939 645)を用いてクローニングベクターに挿入した。このDNAフラグメントを、制限酵素分析及び配列決定によって確認した。次いで、確認したDNAフラグメントを、Xho I及びNot Iでの切断によって切り出し、そして同じ制限酵素で切断したPichia pastoris発現ベクターに挿入した(実施例3及び実施例5を参照のこと)。
Ser172Ala:配列番号2の528〜531位に見出される塩基トリプレット「TCT」を、「GCT」で置換した。この目的のために、第一のPCRにおいて、DNAオリゴヌクレオチド「5’−Try−Ser172Ala」(配列番号10)を、「3’−トリプシン」と組み合わせてプライマーとして使用し、そして第二のPCRにおいて、「3’−Try−Ser172Ala」(配列番号11)を、「5’−トリプシン」と組み合わせてプライマーとして使用した。続いて、全長産物を生成するために、単離された中間フラグメントを第三のPCRに使用した。
PCRフラグメントから生成された変異体組換えブタ膵臓トリプシノーゲンをコードするDNAフラグメント(実施例2を参照のこと)を、EcoRI及びXbaII(Roche Diagnostics GmbH)で切り出した。このフラグメントを、「QIAquick Gel Extraction Kit」を用いて、製造業者の指示書に従って単離した。
このフラグメントを、pPICZαAベクターに連結し、それによって、変異体組換えブタ膵臓トリプシノーゲンをコードするヌクレオチド配列を、Saccharomyces cerevisiae由来のα−因子シグナルペプチドをコードするヌクレオチド配列に融合させた。連結反応の前に、ベクターをEcoRI及びXbaIで同様に切断し、そして単離した。
このクローニング方法により、Saccharomyces cerevisiae由来のα−因子シグナルペプチドをコードするヌクレオチド配列の後に、直接及びインフレームで、変異体組換えブタ膵臓トリプシノーゲンをコードするヌクレオチド配列を挿入した。
配列番号6で与えられる組換えプレプロタンパク質をコードするヌクレオチド配列を、例えば、Pichia pastorisにおいては、メタノールにより誘導性のP.pastoris AOX−1プロモーター(配列番号7)の制御下に置いた。
用いた宿主株は、Pichia pastorisX−33、GS115、KM71H及びSMD1168(Invitrogen)であった。好ましい株は、X−33及びKM71Hであった。宿主生物のゲノムに発現構築物を安定に組込むことが形質転換の目的であった。最初に、YPD培地(YPD=酵母ペプトンデキストロース;Invitrogen)5mlを、P.pastorisコロニーで接種し、そして30℃で一晩、シェーカーで前培養した。形質転換コンピテントな細胞を調製するために、前培養物100μlを新鮮なYPD培地200mlに接種材料として添加し、そしてOD600nmが1.3と1.5との間に達するまで増殖させた。細胞を、1,500×gで5分間遠心分離し、そして氷冷(0℃)滅菌水200mlに再懸濁した。細胞を再び1,500×gで5分間遠心分離し、そして氷冷滅菌水100mlに再懸濁した。細胞をもう一度1,500×gで5分間遠心分離し、そして氷冷1Mソルビトール(ICN)10mlに再懸濁した。このように調製された細胞を氷上に置き、そして直ちに形質転換に使用した。
形質転換に使用されるpPICZαA由来のpTrySer172Ala発現ベクターを、SacI制限エンドヌクレアーゼ(Roche Diagnostics GmbH)を用いて線状化し、沈殿させ、そして水に再懸濁した。「Gene Pulser II(登録商標)」(BioRad)を用いたエレクトロポレーションによって形質転換を達成した。形質転換の設定のために、1Mソルビトール溶液中のP.pastoris細胞80μlを、線状化発現ベクターDNA1μgと穏やかに混合し、そして氷冷キュベットに移し、次いで氷上に5分間保持した。その後、キュベットをGene Pulserに移した。エレクトロポレーションパラメータは、1kV、1kΩ及び25μFであった。エレクトロポレーションの後、1Mソルビトール溶液1mlを、細胞懸濁液に添加し、その後100μg/ml Zeocin(登録商標)(Invitrogen)を含むYPDSプレート(YPDS=酵母ペプトンデキストロースソルビトール;Invitrogen)にプレーティングし、細胞懸濁液100〜150μlを、1つのプレートに広げた。YPDSプレートを、30℃で2〜4日間インキュベートした。酵母クローンを、グリッド入りの最小デキストロースプレートに移した。これらのプレートからコロニーを選び、そして別々に滅菌水に再懸濁した。細胞を、30℃で1時間、リチカーゼ(lyticase)(Roche Diagnostics GmbH)17.5単位で消化し、そしてその後、−80℃で10分間凍結させた。PCRによって、それぞれのpPICZαA由来のpTrySer172Ala発現ベクターの発現カセットの存在を確認した。用語「発現カセット」は、AOX1プロモーター及びAOX1ターミネーターに作動可能に連結した変異体組換えブタ膵臓トリプシンプレタンパク質をコードするヌクレオチド配列を表しており、ここでこの発現カセットは、形質転換に使用されるそれぞれのpPICZαA−ベクターに由来している。発現カセットを含むベクターに関しては、用語「ベクター」及び「発現ベクター」は、同義語である。
ポジティブクローン、すなわちゲノムに安定に組込まれた完全な発現カセットの存在に対してポジティブであると試験されたクローンを、変異体組換えブタ膵臓トリプシン発現のさらなる特徴付けに使用した。
さらに、コントロールの形質転換を、オリジナルのpPICZαAベクターを用いてレシピエントのPichia pastorisKM71H株で行った。ポジティブクローンを得て、同様の様式で確認した。
pPICZαA−pTrySer172Ala発現ベクターで形質転換された一連のポジティブクローン(20〜30)を、各々BMGY培地(BMGY=緩衝化グリセロール複合培地;Invitrogen)3ml中で振盪培養として一晩増殖させた。その後、培養物のOD600nm値を、各々pH3のBMMY培地(Invitrogen)10mlを含む振盪フラスコに継代する前に測定した。前培養を接種材料として使用して、各々OD600nmが1になった。この培養物を、シェーカー上30℃に維持した。同時に、ポジティブコントロールクローンを、同じ条件下で培養した。
BMMY(BMMY=緩衝液メタノール複合培地)培地は、変異体組換えブタ膵臓トリプシノーゲンをコードするヌクレオチド配列の転写を制御するAOX−1プロモーターの誘導物質であるメタノール(Mallinckrodt Baker B.V.)を含んだ。
サンプル500μlを、計72時間にわたって24時間間隔で振盪フラスコから採取した。サンプルアリコートを取り出すとき、この培養物にまた、0.5%メタノールを与えた。上清増殖培地のサンプルを、トリプシン酵素活性について試験した。
実施例5に記載されるように得られたサンプルアリコートのOD600nmを最初に測定した。その後、細胞を遠心分離によってペレット化し、そして上清は残しておいた。トリプシン活性を、上清を希釈せずに、そして1:10希釈で測定した。
pPICZαAベクターで形質転換されたコントロールクローンは、培地中でいかなる測定可能なトリプシン活性も導かなかったが、pPICZαA−pTrySer172Ala発現ベクターで形質転換されたPichia株は、増殖培地、すなわち培養基に分泌された組換えブタ膵臓トリプシンのそれぞれの変異体に起因したトリプシン活性を示した。従って、この場合、Saccharomyces cerevisiae由来のα−因子シグナルペプチドを含む組換えプレタンパク質の発現は、タンパク質分解活性を有する活性な酵素の分泌を可能にすると結論付けられ得る。
上清培地中で最も高いトリプシン活性を生じることが見出されたpPICZαA−及びpPICZA−由来のpDNM発現ベクターで形質転換された酵母クローンを、以前と同じ発現ベクターを用いて繰り返しエレクトロポレーションを行った。エレクトロポレーション条件は、YPDSプレートが、Zeocin(登録商標)を高い濃度で、すなわち1,000μg/mlと2,000μg/mlとの間で含むこと以外は実施例4に記載される通りであった。それぞれの発現ベクターの多コピーがゲノムに組込まれた形質転換体を選択するために、抗生物質濃度を増大した。抗生物質に対する耐性が増大した酵母クローンを、グリッド入り最小デキストロースプレートに移した。すでに実施例5に記載される通り、前培養を個々の酵母クローンから行い、そして実施例6に記載される通りに、増殖培地に分泌されたトリプシン酵素活性を測定することによって発現を測定した。トリプシン活性量の増大を生じた個々のクローンを見出した。平均して、それぞれのpPICZαA−pTry−Ser172Ala発現ベクターで繰り返し形質転換されたPichia株の上清で測定されたトリプシン活性は、1回の形質転換しか受けなかったそれぞれの前駆体株と比較して、2倍〜3倍高かった。
完全発酵ブロスを、5〜30mM塩化カルシウム、pH3.5を含む酢酸アンモニウム緩衝液(5〜20mM)で、およそ1:2〜1:4の比で希釈した。トリプシノーゲン変異体を、陽イオン交換体(例えば、Streamline(登録商標)SP、XL)を用いた膨張層クロマトグラフィー(McCornick(1993);EP 0 699 687)により精製した。クロマトグラフィーを、酵母細胞を事前に分離せずに行った。さらなる精製を、充填層カラム(例えば、SP−Sepharose(登録商標)XL、ff)を用いて行った。自己触媒的活性化を、20mM CaCl2の存在下でpHを7〜8に再緩衝化することによって開始させた。pHを2〜4の範囲に戻すことによって、活性化を終結させた。精製トリプシンを、自己タンパク質分解を避けるためにpH1.5〜3で保管した。
トリプシンの活性を、100mM Tris pH8.0、20mM CaCl2中25℃で、Chromozym TRY(Roche Diagnostics GmbH)を用いて測定した。光度測定を、405nmで行った。アルギニン対リジン間の基質特異性を区別するために、Chromozym TH(アルギニン/Roche Diagnostics GmbHを含む)及びChromozym PL(リジン/Roche Diagnostics GmbHを含む)を使用した。
すべての実験を、8℃及びpH値8.3(緩衝化溶液又はNaOHの投与で制御)で行い、そして20mL〜3.5Lスケールで行った。いくつかの実験において、プレプロインスリンをまた、両方の酵素を直接比較するために、ネイティブの組換えブタトリプシンを用いて切断した。
PPI溶液を、適切なサーモスタット反応溶液に満たし、そして酵素調製物を添加することによって反応を開始させた。一定の時間間隔後にサンプルを採取した。1N又は2N HCl溶液によってサンプル溶液を酸性化することにより、酵素反応を直ちに停止させた。それぞれの産物の濃度を、HPLCによって測定した。
表1は、一実験の例示的な結果を示している。
図5は、ネイティブの組換えブタトリプシンを用いたプレプロインスリンの転換を示しており、図6は、S172A変異体トリプシンを用いたPPIの転換を示している。
実施例10の実験条件を用いた。プレプロインスリンをまた、両方の酵素を直接比較するためにネイティブの組換えブタトリプシンを用いて切断した。

- インスリン、インスリンアナログ又はインスリン誘導体の調製方法におけるSer172Alaブタトリプシン変異体の使用であって、Ser172Alaは、配列番号1の172位に対応する位置のセリン残基がアラニン残基で置換されていることを表す、使用。
- インスリン、インスリンアナログ又はインスリン誘導体の調製方法であって、
(a)プレプロインスリン、プレプロインスリンアナログ又はプレプロインスリン誘導体は、Ser172Alaブタトリプシンで切断されるものとし、ここで、Ser172Alaは、配列番号1の172位に対応する位置のセリン残基がアラニン残基で置換されていることを表し、

- Ser172Alaブタトリプシンを産生する方法であって、以下の工程
(a)請求項16に記載のベクターを提供すること、
(b)このベクターで微生物宿主株を形質転換すること、
(c)この形質転換された微生物宿主株を、栄養分を含む増殖培地で培養し、これによってこの微生物宿主株が、Ser172Alaブタトリプシン又はSer172Alaブタトリプシノーゲンを発現すること、
(d)(c)の発現産物が、Ser172Alaブタトリプシノーゲンである場合、成熟Ser172Alaブタトリプシンへ転換すること、並びに
(e)微生物宿主株及び/又は増殖培地からSer172Alaブタトリプシンを精製すること、
を含むものであり、Ser172Alaは、配列番号1の172位に対応する位置のセリン残基がアラニン残基で置換されていることを表す、方法。 - 微生物宿主株が、Hansenula、Pichia、Candida及びTorulopsis種を含む群から選択されるメチロトローフ酵母株である、請求項17に記載の方法。
