Description
本発明は、サンプル中の分析物の検出のための側流式迅速視覚化試験に関する。
側流式試験デバイスは、当該分野において公知であり、サンプル(例えば、体液)中の目的の分析物の存在を検出するための臨床診断において使用され得る。しかし、このようなデバイスは、代表的に、比較的大きなサンプル容量および多くの量の結合体を必要としている。これらはまた、試験結果が読み出され得るまで長い待ち時間がかかっている。
1つの局面において、サンプル中の目的の分析物の検出のために、免疫診断試験片が適用される。いくつかの実施形態において、分析物は、抗ヒト免疫不全ウイルス(HIV)抗体または抗C型肝炎ウイルス(HCV)抗体のような抗体である。他の実施形態において、分析物は、ヒト絨毛性性腺刺激ホルモン(hCG)のような抗原である。
診断デバイスおよび診断方法が、サンプルにおける分析物の存在を迅速に決定するために、提供される。サンプル中の抗体または抗原の検出のための免疫診断デバイスに関して一般的に記載されるが、当業者は、本発明が、目的とする任意の分析物を検出するために容易に改変され得ることを、認識する。
他のように規定されない限り、本明細書中で使用されるすべての技術用語および科学用語は、本発明が属する分野の当業者によって一般的に理解されるのと同じ意味を有する。本明細書中に記載されるものと類似するかまたは等価である任意の方法、デバイス、および材料が、本発明の実施において使用され得る。
本明細書中に開示される診断デバイスは、好ましくは、図1〜3に示されるように、試験カード1を含む。図1は、プラスチックハウジング10を備える試験カード1を示し、一緒にパチンと閉じられた上側部分15および下側部分18を有する。プラスチックハウジング10の上側部分15内に2つの開口部(サンプルウインドウ20および緩衝液ウインドウ30)が存在する。サンプルウインドウ20は、プラスチックハウジング10内に含まれる膜のサンプル適用領域へのサンプルの適用を可能にする。サンプル適用領域は、好ましくは、試験領域140を含む。コントロール領域150は、好ましくは、膜の試験領域140の下流に位置する。緩衝液ウインドウ30によって、結合体供給源領域50への緩衝液の適用が可能になる。膜は、以下に詳細に記載される試験片の一部である。
試験片は、少なくとも1つの試験領域を有する試験膜を備える。目的の分析物に特異的な分析物結合剤は、試験領域に結合されるかまたはそれ以外で固定される。試験領域の少なくとも一部は、目的の分析物がサンプル中に存在する場合、試験の完了時に可視化される。特定の実施形態において、試験領域は、固定化抗原を含み、そしてこの試験は、生物学的流体のようなサンプル中の特定の抗体の同定を可能にする。別の実施形態において、試験領域は、結合された抗体を含み、この試験は、サンプル中の特定の抗原の同定を可能にする。
目的の分析物の存在について試験されるサンプルは、試験領域と接触するように、試験膜のサンプル適用領域に適用される。好ましい実施形態において、サンプルは、膜に適用され、結合体が試験領域に接触する前に、試験領域に接触し得る。従って、サンプル中の目的の分析物は、存在する場合、結合体の結合の前に、試験領域内の分析結合剤に結合する機会を有する。
ヒト免疫不全ウイルスに対する抗体の検出のための側流式迅速視覚化試験を調製した。この試験片の配置は、本質的に図1に図示されたとおりである。
血清または血漿を、適切な静脈穿刺技術を使用して、HIVについて試験すべき患者から得た全血サンプルから調製した。サンプルを、2℃〜8℃にて保存して24時間以内に使用するか、または−20℃にて凍結して2週間以内に使用した。凍結サンプルを、使用前に融解した。
C型肝炎ウイルスの診断のための試験片を、実施例1においてのように調製した。しかし、コア(0.3mg/ml)と、NS3(0.4mg/ml)と、NS4(0.1mg/ml)とを含む、C型肝炎抗原混合物(HIV抗原ではない)を、上記膜上の試験線中に噴霧して、試験領域を作製した。合計約0.4μgのHCV抗原混合物を、試験線に適用した。1mg/mlになるように希釈したウサギIgGを、コントロール線に噴霧した。
実施例3に記載されるようなHCV試験カードを、上記のホイルパックから取り出した。このHCV試験カードは、試験領域上にHCV抗原を含んだ。
側流式視覚的高速妊娠試験を、調製する。その試験片の配置は、本質的に図7に示される通りである。
サンプルカップを用いる側流式視覚的高速妊娠試験を、実施例5に記載されるように調製する。
芯を用いる側流式視覚的高速妊娠試験を、実施例5において記載されるように調製する。
Claims (10)
- 液体サンプル中の分析物の存在を測定する免疫診断デバイスであって、前記デバイスは、緩衝液を受け取る緩衝液パッド110;
緩衝液パッド110と流体連絡する結合体パッド120であって、前記結合体パッド120は乾燥結合体を含み、前記結合体は緩衝液によって拡散可能になっており、前記結合体は視覚的に検出可能な標識と分析物に結合可能な結合剤を含む結合体パッド120;
試験領域140を含む試験膜130であって、前記試験領域140は分析物を固定化するための、試験領域に固定化された分析物結合剤を含む、試験膜130;
濾紙200を含む除去可能なフィルターホルダー190であって、前記除去可能なフィルターホルダー190は前記デバイスに結合し、前記濾紙200は前記試験領域140の上に配置される、フィルターホルダー190;
を含み、
ここで、前記結合体パッド120は、前記試験膜130と流体連絡していない第1の状態と、前記試験膜130と流体連絡している第2の状態の間で移動可能であり、前記第2の状態において、前記結合体パッド120と前記試験膜130は接触しており;
前記デバイスは経路を有し、
前記結合体パッド120が前記第1の状態にある時に、前記経路に沿って緩衝液が結合体パッド120に流れ、前記結合体を移動可能にし、前記緩衝液と移動可能な結合体とを含む混合物を形成し、
前記結合体パッド120が前記第2の状態にある時に、前記経路に沿って前記混合物が前記試験膜130および前記試験領域140に流れ、
前記緩衝液は試験膜130に流れ、前記緩衝液が結合していない成分を試験領域140から洗い流す、デバイス。 - 前記分析物がヒト免疫不全ウイルス(HIV)に対する抗体であり、前記分析物結合剤が
、HIV−1に対する抗原およびHIV−2に対する抗原からなる群より選ばれる、請求項1に記載のデバイス。 - 前記視覚的に検出可能な標識がコロイド金である、請求項1に記載のデバイス。

- 液体サンプル中の分析物の存在を測定する免疫診断デバイスであって、前記デバイスは、緩衝液を受け取る緩衝液パッド110;
緩衝液パッド110と流体連絡する結合体パッド120であって、前記結合体パッド120は乾燥結合体を含み、前記結合体は緩衝液によって拡散可能になっており、前記結合体は視覚的に検出可能な標識と分析物に結合可能な結合剤を含む結合体パッド120;
試験領域140を含む試験膜130であって、前記試験領域140は分析物を固定化するための、試験領域に固定化された分析物結合剤を含む、試験膜130;
前記液体サンプルを受け取るサンプルカップ500であって、前記サンプルカップ500は前記デバイスに結合し、前記試験領域140の上に直接配置される、サンプルカップ500;
を含み、
ここで、前記結合体パッド120は、前記試験膜130と流体連絡していない第1の状態と、前記試験膜130と流体連絡している第2の状態の間で移動可能であり、前記第2の状態において、前記結合体パッド120と前記試験膜130は接触しており;
前記デバイスは経路を有し、
前記結合体パッド120が前記第1の状態にある時に、前記経路に沿って緩衝液が結合体パッド120に流れ、前記結合体を移動可能にし、前記緩衝液と移動可能な結合体とを含む混合物を形成し、
前記結合体パッド120が前記第2の状態にある時に、前記経路に沿って前記混合物が前記試験膜130および前記試験領域140に流れ、
