発明の背景
腫瘍細胞は自己由来性でありしたがって細菌およびウィルスのような外因性の病原体のようには免疫原性ではないので、癌治療に免疫学的アプローチを使用することは困難である。その結果、癌の免疫治療の見込みは、免疫系により認識できる腫瘍関連抗原(TAA)の同定に依存する。特に、T細胞媒介応答を誘発する標的抗原は、非常に関心がある。このことは、細胞傷害性Tリンパ球(CTL)が、動物モデル(Kast W. et al (1989) Cell 59:6035; Greenberg P. (1991) Adv. Immunol. 49:281)およびヒト(Boon T. et al. (1994) Annu. Rev. 予め調製した基質をウェルごとに100μL配置する。例えば、1プレートにつき、調製する:9.6mLの滅菌水、0.4mLの25X緩衝剤、0.1mLの溶液A(NBT)および0.1mLの溶液B(BCIP)。室温で30−45分間インキュベートする。滅菌水で洗浄する。乾燥させプラスチックフィルムに移す。Zeiss画像解析機を使用してスポットの数を計数する。それぞれのスポットは、T細胞を分泌するそれぞれのIFN-γに対応する。

結果
ELISPOT分析において陽性であることがテストされた動物は、図1−4に示される。全体的に、この結果により、gp100抗原の結節内投与を受けた動物の2匹のうち2匹(すなわち100%)、およびgp100抗原の皮下投与を受けた動物の4匹のうち2匹(すなわち50%)は陽性の細胞媒介免疫応答を有したことが示される。

ELISA 分析
当該技術において知られる標準の原理を使用してELISAを行った。簡単に言えば、ヒトgp100(“hgp100”;バキュロウィルスで産生される)をコーティング緩衝剤(カルボネート−ビカルボネート、pH9.6)で希釈し、0.5μg/ウェルで96ウェルに加えた。プレートを4℃で一晩配置した。次にプレートを洗浄し、阻止緩衝剤(リン酸緩衝化生理食塩水/0.5% Tween 20/1.0% BSA、pH7.2)を37℃で2時間加えた。その後プレートを洗浄し、血清を希釈緩衝剤で希釈した(リン酸緩衝化生理食塩水/0.5% Tween 20/1.0% BSA、pH7.2)。この研究のために、サル血清を1:800まで希釈し、テストしたそれぞれのサンプルについて”7”つの連続した3倍希釈を行った。ヒト血清対照を、希釈緩衝剤中で1:50に希釈し、”7”つの連続した2倍希釈を行った。それぞれの希釈は、2度行った。プレートを、37℃でさらに2時間インキュベートした。プレートを洗浄し、希釈緩衝剤で1:100に希釈したホースラディッシュペルオキシダーゼ(HRP)−抱合された抗−ヒト二次抗体(すべてヒツジからの抗−ヒトIg(Amersham Life Science, NA933))をウェルに加え、37℃で1時間インキュベートした。プレートを洗浄し、基質緩衝剤(50mM リン酸塩/25mM クエン酸塩、pH7.2)中でH2O2を有するOPD(o-フェニレンジアミンジヒドクロライド)基質をウェルに加えた。速度論ELISAについては、プレートを連続的に(「停止」緩衝剤を使用せず)反復的に(15分間につき2分間の間隔で)読み取った。プレートを450nmで読み取った。

結果
上述の実験の結果は、表8および図5に示される。グループ2は、ALVAC(2)-gp100(mod)の結節内注射を受け、その後修飾されたgp100ペプチド209(2M)および290(9V)により後押しされる;グループ3の動物は、ALVAC(2)構成体の皮下注射を受け、その後ペプチドにより後押しされる;グループ1の動物は、対照として食塩水の結節内注射を受けた。

Claims (12)
- 動物において腫瘍抗原に対する免疫応答を誘発するための組成物であって、単離された腫瘍抗原ペプチドIMDQVPFSY(配列番号1)およびYLEPGPVTV(配列番号2)を含有し、腫瘍抗原IMDQVPFSY(配列番号1)およびYLEPGPVTV(配列番号2)をコードする核酸が動物中のリンパ節に直接投与された後に、前記動物中のリンパ節に直接投与されることを特徴とする組成物。
- 前記核酸が、ウィルス核酸、細菌DNA、プラスミドDNA、裸/遊離DNA、およびRNAから群より選択されることを特徴とする請求項1記載の組成物。
- 前記ウィルス核酸が、アデノウィルス、アルファウィルスおよびポックスウィルス核酸からなる群より選択されることを特徴とする請求項2記載の組成物。
- 前記ポックスウィルス核酸が、アビポックス、オルトポックスおよびスイポックス核酸からなる群より選択されることを特徴とする請求項3記載の組成物。
- 前記ポックスウィルス核酸が、ワクチニア、ニワトリポックス、カナリアポックスおよびブタポックス核酸からなる群より選択されることを特徴とする請求項3記載の組成物。
- 前記ポックスウィルス核酸が、MVA、NYVAC、TROVAC、およびALVAC核酸からなる群より選択されることを特徴とする請求項3記載の組成物。
- 前記核酸が、ベクター中に含まれることを特徴とする請求項1から6いずれか1項記載の組成物。
- 前記ベクターが、組換えウィルスまたは細菌であることを特徴とする請求項7記載の組成物。
- 前記組換えウィルスが、アデノウィルス、アルファウィルスおよびポックスウィルスからなる群より選択されることを特徴とする請求項8記載の組成物。
- 前記ポックスウィルスが、アビポックス、オルトポックスおよびスイポックスからなる群より選択されることを特徴とする請求項9記載の組成物。
- 前記ポックスウィルスが、ワクチニア、ニワトリポックス、カナリアポックスおよびブタポックスからなる群より選択されることを特徴とする請求項9記載の組成物。
