Description
また、前記培養物の着色度が明度L*値で10以上であるので、上記β−グルカンを高濃度に含有し、且つその見た目によって製品の商品性に影響を与えないβ−グルカン含有組成物を提供することができる。
また、アウレオバシジウム属(Aureobasidium sp.)に属する微生物が、アウレオバシジウム プルランス M−2(Aureobasidium pullulans M-2)(FERM BP-10014)であるので、入手が容易であり菌学的性質のよく知られた安全性の高い微生物を原材料にすることができる。
本発明のβ−グルカン含有組成物の製造方法によれば、アウレオバシジウム属(Aureobasidium sp.)に属する微生物が増殖中に菌体外に産出するβ−グルカンを高濃度に含有し、しかも着色度合の低いβ−グルカン含有組成物を効率よく製造することができる。
また、β―グルカンを生産する培養中の培養液pHは4.5〜6.0の範囲にあり、特に培養後半にはpHが5.0未満となるので、雑菌汚染による品質劣化を受けにくいばかりでなく、培養終了後に必要な加熱殺菌が容易になる。
また、アウレオバシジウム属(Aureobasidium sp.)に属する微生物が、アウレオバシジウム プルランス M−2(Aureobasidium pullulans M-2)(FERM BP-10014)であるので、入手が容易であり菌学的性質のよく知られた安全性の高い微生物を原材料にすることができる。
本発明のβ−グルカン含有組成物を経口的に摂取する場合、その1日当りの摂取量に特に制限はないが、β−グルカン換算で0.5mg〜10mg/Kg/日であることが好ましい。
(アウレオバシジウム プルランス M−2(Aureobasidium pullulans M-2)の作出)
親株であるアウレオバシジウム プルランスM−1(Aureobasidium pullulans M-1、独立行政法人産業技術総合研究所特許生物寄託センター寄託番号FERM BP-08615)を、PDA (Potato Dextrose Agar:Difco 社製)スラントで7日間培養し、得られた菌体をPBS 10mlに懸濁し、1,000 CFU/ml程度の溶液とした。その菌体懸濁液0.2mlをPDA (Potato Dextrose Agar:Difco 社製)プレートに塗布し、その後のプレート上の菌体に対して0、5、10分間とそれぞれ照射時間を変えてUV照射処理を行った。25℃で 4日間静置培養してコロニー形成を確認し、更にプレートを4℃で3日間培養して菌体を厚膜胞子化させた。これらのプレートから白色コロニーを選択し複数の変異株を単離した。白色コロニーの出現頻度は1/1200程度であった。
(β−グルカン含有組成物の製造)
アウレオバシジウム プルランス M−2(Aureobasidium pullulans M-2)を、ショ糖2.0%、米糠0.2%、アスコルビン酸ナトリウム0.1%、を含む2リットルの液体栄養培地(pH5.2〜5.4)に植菌し、3リットルタンク中0.5v/v/mの通気攪拌条件で、環境温度25℃において増殖定常期に至るまで(植菌から120時間)培養した。得られた培養物は薄い乳白色を呈し、アウレオバシジウム プルランスの通常有する濃緑色の色調を欠いていた。
(色素蓄積性)
アウレオバシジウム プルランス M−2(Aureobasidium pullulans M-2)の色素蓄積性を、親株であるアウレオバシジウム プルランス M−1(Aureobasidium pullulans M-1)の色素蓄積性と比較するために、両者を実施例2の培養条件で増殖定常期に至るまで通気攪拌培養し、その培養時間中、植菌から72時間後及び264時間後にそれぞれ培養物の一部を採取した。採取した培養物の色成分を定性分析するために、「分光測色計JS555」(商品名、株式会社カラーテクノシステム製)を用いて分析した。表2には、その分析結果をCIE(国際照明委員会)Labカラー表色系によって数値化した測定値を示す。
(β−グルカン蓄積性)
アウレオバシジウム プルランス M−2(Aureobasidium pullulans M-2)のβ−グルカン蓄積性を、親株であるアウレオバシジウム プルランス M−1(Aureobasidium pullulans M-1)と比較した。そのために、両者を実施例2の培養条件で増殖定常期に至るまで通気攪拌培養し、その培養時間中、植菌から48時間後から264時間後に至るまで、経時的に培養物を採取して、培養物100g中のβ−グルカン量を上記実施例2における方法で測定した。図3には、その結果を示す。
栄養培地中の糖は、β−グルカン生成時の原料になると考えられ、その糖濃度の差がグルカン生成に及ぼす影響について調べることとした。そのために、アウレオバシジウム プルランス M−2(Aureobasidium pullulans M-2)を、ショ糖濃度の異なる栄養培地(ショ糖濃度;1、2,3または4%)で実施例2の培養条件で増殖定常期に至るまで振とう培養した。植菌から192時間後の培養液の一部を採取し、上記比較例2と同様の方法で、培養物100g中のβ−グルカン含量を測定した。その結果、図4に示すとおり、いずれの栄養培地ショ糖濃度においても十分にβグルカンを生産しており、特に2〜4%の栄養培地でその蓄積量が高かった。
一般に、食品加工工程においてはpH環境を酸性に維持することにより、雑菌汚染による品質劣化を受けにくく、また必要に応じて殺菌処理を行う場合、緩和な条件を適用することができる。そこで、アウレオバシジウム プルランス M−2(Aureobasidium pullulans M-2)のpH環境に関する培養条件を検討するために、植菌時初発pHの異なる培地を用い、実施例2の培養条件で増殖定常期に至るまで振とう培養し、植菌から240時間後の培養液の一部を採取した。上記比較例2と同様の方法で、培養物100g中のβ−グルカン含量を測定し、更に、培養後の培養液のpH、粘度を測定した。それらの結果を表3に示す。また、図5には、アウレオバシジウム プルランス M−2(Aureobasidium pullulans M-2)についてその培養中の培養液のpHを経時的に測定した結果を示す。
(β−グルカン含有組成物を配合した高機能ヨーグルトスタータ)
実施例2で得られたアウレオバシジウム プルランス M−2(Aureobasidium pullulans M-2)の培養物(1000g)を加圧下121℃、15分間殺菌した後、このうち300gをミキサーで高速撹拌(1500rpm、20分間)して低粘度化してから、粉末化基剤としてデキストリン(商品名「サンデック」、三和澱粉工業社製)及び/又は脱脂粉乳を280g添加してミキサーで均一に混合した。これを残りの前記培養物(700g)と混合してから凍結乾燥して粉末化し、β−グルカン含有組成物を得た。これに、デキストリン(商品名「サンデック」、三和澱粉工業社製)及び/又は脱脂粉乳155g、ヨーグルト粉末(ヨーグルトスタータを含む)25g脱脂粉乳等20gを加え、ヨーグルトスタータとした。
(β−グルカン含有組成物を配合した皮膚用保湿剤)
実施例2で得られたアウレオバシジウム プルランス M−2(Aureobasidium pullulans M-2)の培養物から遠心分離により菌体部分を除く溶液とし、加圧下121℃、15分間殺菌した。この溶液30重量部にソルビット2重量部、1.3ブチレングリコール2重量部、ポリエチレングリコール1重量部、ポリオキシエチレンイレイルエーテル2重量部、ポリオキシエチレン0.1重量部、エタノール10重量部、精製水52.8重量部を加え、さらにpH調整剤、香料、防腐剤、酸化防止剤を適量添加することで、全体で100重量部からなる増粘型柔軟化粧水とした。

Claims (7)
- アウレオバシジウム プルランス M−2(Aureobasidium pullulans M−2)(FERM BP-10014)を、液体培地中で該液体培地のpHが4.5〜6.0の状態で培養することにより得られた培養物からなり、β−グルカン含量が0.3質量%以上であり、着色度が明度L* 値で10以上であり、粘度が39.3m.Pas以上であることを特徴とするβ−グルカン含有組成物。
- アウレオバシジウム プルランス M−2(Aureobasidium pullulans M−2)(FERM BP-10014)を、液体培地中で該液体培地のpHが4.5〜6.0の状態で培養することにより、該培養物中にβ−グルカンを0.3質量%以上生成させることを特徴とするβ−グルカン含有組成物の製造方法。
- 前記培養物の着色度が明度L*値で10以上である請求項2記載のβ−グルカン含有組成物の製造方法。
- 得られた培養物にビタミンC及び/又は有機酸を添加してpH4.5未満に調整し、その後加熱殺菌する工程を含む請求項2又は3に記載のβグルカン含有組成物の製造方法。
- 請求項1に記載のβ−グルカン含有組成物を含有することを特徴とする飲食品。
- 請求項1に記載のβ−グルカン含有組成物を含有することを特徴とする皮膚用保湿剤。
- アウレオバシジウム プルランス M−2(Aureobasidium pullulans M-2)(FERM BP-10014)。
