本明細書に示される18F−補欠分子族は、標的分子とインビボでの生物学的標的(例えば細胞または組織)の間の相互作用に影響することなしに改変されうる誘導体化基を含むものであれば、実質的にあらゆる標的分子に結合されうる。
わずか2個のアミノ酸残基、好ましくは2から10個の残基を有するペプチドが用いられてもよく、また他の部位に結合されてもよい。標的可能な構築物はまた、当該ペプチドのインビボでの安定性を向上させるため、骨格構造に非天然アミノ酸、例えばD−アミノ酸を含みうる。別の実施態様では、非天然アミノ酸またはペプトイドから構築されるような、他の骨格構造が用いられうる。
いくつかの実施態様において、本明細書に記載の放射性トレーサー組成物に用いられる標的分子は抗体である。本明細書で用いられる用語「抗体」は、抗体全体、およびいずれかの抗原結合性フラグメント(すなわち、「抗原−結合部位」)またはそれらの一本鎖を含みうる。例えば、「抗体」は自然発生的および非自然発生的抗体;モノクローナルおよびポリクローナル抗体;キメラおよびヒト化抗体;ヒトおよび非ヒト抗体;二重特異性抗体;全合成抗体;および1本鎖抗体のいずれをもを指しうる。本明細書で用いられる用語「抗原」は、タンパク質、ペプチドまたはハプテンなどの、いずれかの天然または合成免疫原性物質をいう。
いくつかの実施態様において、本明細書に記載の放射性トレーサー組成物に用いられる標的分子はFBSタンパク質である。一般に、FBSタンパク質分子は本質的に急速な血液クリアランス速度を示すため、非関連組織からのプローブのバックグラウンドシグナルに必要な時間を最小化することによって、18Fイメージング技術の使用に有利でありうる。急速にクリアランスされるプローブによって、プローブが注入されるのと同じ日に高コントラスト画像を得ることが可能になり、特に重要なことには、対象に対する全体の放射線被曝を軽減させることができる。
本明細書に記載の18F−標識プローブに結合する、インビボでの標的分子の例は、悪性疾患、心血管疾患、感染性疾患、炎症性疾患、自己免疫性疾患、または神経性疾患などの様々な疾患または病状に関連するものである。
本明細書に記載の方法によって検出可能な感染を引き起こす病原性細菌のいくつかの例としては、クラミジア、リケッチア細菌、マイコバクテリア、ブドウ球菌、レンサ球菌、肺炎球菌、髄膜炎菌および淋菌、クレブシエラ菌、プロテウス菌、セラチア菌、シュードモナス菌、レジオネラ菌、ジフテリア菌、サルモネラ菌、桿菌、コレラ菌、破傷風菌、ボツリヌス菌、炭疽菌、ペスト菌、レプトスピラ、およびライム病細菌が挙げられる。
本明細書に記載のタンパク質標的分子の標的分子への結合は、平衡定数(例えば、解離、KD)から、および速度定数(例えば、結合速度定数、konおよび解離速度定数、koff)から評価されうる。koffが十分に小さい、またはkonが十分に大きい場合には、大きなKD値は許容されうるが、タンパク質標的分子は一般にKDが500nM、100nM、10nM、1nM、500pM、200pM、または100pM未満で標的分子に結合する。
薬学的に許容可能な担体と共に製剤化される、本明細書に記載の18F−標識標的薬剤のうちの1つ、またはそれらの組み合わせを含む、組成物、例えば医薬組成物がさらに示される。このような組成物は、本明細書に記載の薬剤のうちの1つ、またはそれらの組み合わせ(例えば、2つまたはそれ以上)を含みうる。例えば、本明細書に記載の医薬組成物は、18F−標識標的薬剤および薬物の組み合わせを含みうる。
本明細書に記載の18F−標識標的薬剤は、様々なインビボイメージングの応用(例えば、組織または全身のイメージング)において有用である。いくつかの実施態様において、18F−標識標的薬剤は、例えば標的発現性腫瘍などの標的−陽性細胞または組織をイメージングするために用いられうる。例えば、標識された18F−標識標的薬剤は、対象の組織に18F−標識標的薬剤が取り込まれるのに十分な分量において、対象に投与される。対象は次いで、PETなどのイメージングシステムを用いて、18F放射性核種に必要な時間にわたってイメージングされる。標的物質を発現している細胞または組織に結合した18F−標識標的薬剤が、次いでイメージングシステムによって検出される。
本明細書に記載の18F−放射性標識標的組成物の生成のためのキットおよび使用のための説明書もまた示される。キットには、一般に、所定量の試薬と説明書のパッケージ化された組み合わせおよびキットの内容物の意図される用途を表すラベルが含まれる。用語ラベルには、キットに組み込まれたまたはキットに含まれる、またはそうでなければ、キットに付随する、あらゆる文書または記録されたものが含まれる。
18F−標識標的薬剤を用いたイメージング方法が本明細書において示される。18F−放射性標識タンパク質ベースのPETプローブなどの陽電子放出断層撮影(PET)トレーサーは、インビトロおよびインビボでの画像診査および診断の応用において使用するための、現在利用可能なPET技術と共に用いられうる。PETスキャンによる18Fイメージングのためのイメージング技術および装置は、当技術分野において周知であり(例えば、米国特許第6,358,489号;6,953,567号;Page et al.,Nuclear Medicine and Biology,21:911−919,1994;Choi et al.,Cancer Research 55:5323−5329,1995;Zalutsky et al.,J.Nuclear Med.,33:575−582,1992を参照されたい)、このような周知のPETイメージング技術または装置のいずれかが用いられうる。

2−(2−(2−(2−アジドエトキシ)エトキシ)エトキシ)エチル 4−メチルベンゼンスルホネートの合成
((オキシビス(エタン−2,1−ジイル))ビス(オキシ))ビス(エタン−2,1−ジイル)ビス(4−メチルベンゼンスルホネート)(5g、9.95mmol)およびアジ化ナトリウム(0.647g、9.95mmol)の混合物を、エタノール(50mL)に溶解し、反応液を90℃で17時間にわたり還流した。溶媒を部分真空によって留去し、次いで40グラムのシリカカートリッジに加え、フラッシュクロマトグラフィー(IscoCombiFlash−ヘキサン中の10%酢酸エチルからヘキサン中の90%酢酸エチルまでの、45分間の直線状勾配によって溶出)を用いて精製した。回収した画分をTLCによって確認し、合わせて、2−(2−(2−(2−アジドエトキシ)エトキシ)エトキシ)エチル 4−メチルベンゼンスルホネートを無色の油状物として得た。2−(2−(2−(2−アジドエトキシ)エトキシ)エトキシ)エチル 4−メチルベンゼンスルホネート生成物の反応特性によって、当該物質はさらなる特性評価をすることなく、そのままで用いられた。
3−(2−(2−(2−(2−アジドエトキシ)エトキシ)エトキシ)エトキシ)−2−フルオロピリジンの合成

3−(2−(2−(2−(2−アジドエトキシ)エトキシ)エトキシ)エトキシ)−N,N,N−トリメチルピリジン−2−アミニウム、トリフルオロメタンスルホン酸塩を用いた、[18F]−3−(2−(2−(2−(2−アジドエトキシ)エトキシ)エトキシ)エトキシ)−2−フルオロピリジンの合成
[18F]−フルオライドの水溶液(2.0ml,33.3GBq/900mCi)を、ペンシルベニア州ウエスト・ポイント、P.E.T.Net(登録商標)Pharmaceuticalsから購入し、直接Sep−Pak light QMAに移した[Sep−Pak light QMAカートリッジは、使用前に、5mlの0.5Mの炭酸水素カリウム、5mlの脱イオン水、および5mlのMeCNの順で前処理を行った]。移行が終了すると、炭酸カリウム(15mg/ml;0.1ml)、次いで、炭酸カリウム(30mg/ml、0.1ml)、4,7,13,16,21,24−ヘキサオキサ−1,10−ジアザビシクロ[8.8.8]ヘキサコサン(15mg、0.04mmol)および1.2mlのMeCNの混合物を逐次添加することによって、[18F]フルオライド水溶液がQMA Sep−Pakから溶出した。溶媒を90℃の窒素の緩やかな気流の下でおよび真空で留去した。共沸乾燥を、1mlのアセトニトリルによって2回繰り返し、無水K.2.2.2/K[18F]F複合体を得た。3−(2−(2−(2−(2−アジドエトキシ)エトキシ)エトキシ)エトキシ)−N,N,N−トリメチルピリジン−2−アミニウム、トリフルオロメタンスルホン酸塩(2mg、5.6μmol)を、500マイクロリットルのDMSOに溶解し、乾燥クリプタンドに加えた。この溶液を120℃で10分間加熱した。その後、粗製反応混合物を3mlのDI水で希釈した。次いで、粗製反応混合物の全量を、逆相HPLCに移し、ロードし、以下の条件で精製した:HPLCカラム:Luna C18 250×10 溶媒A:0.1%TFAを含むDI水;溶媒B:0.1%TFAを含むアセトニトリル、流速4.6ml/分、32%のBの定組成メソッドを用い、UVは280nmにおいてモニターされた。[18F]−3−(2−(2−(2−(2−アジドエトキシ)エトキシ)エトキシ)エトキシ)−2−フルオロピリジンを、クロマトグラムの24分のピークにおいて単離し、2分間回収した。この生成物を10mlのDI水を含む100mlのフラスコに回収し、全ての内容物をWatersのSep−Pak Vac tC18 6cc 1g sep packに移した。6.1GBq/164mCiの[18F]−3−(2−(2−(2−(2−アジドエトキシ)エトキシ)エトキシ)エトキシ)−2−フルオロピリジンをこの反応液から単離した。これは3mlのエタノールによってsep−pakから回収され、この溶液を98C熱源、窒素の安定な気流、および真空によって15分間、当該バイアルにフィルムのみが残るまで蒸発させた。最終生成物を100%の1×PBS緩衝液に再溶解し、37℃で1時間以上は当溶媒に安定である。
「クリックケミストリー」を用いた18F−放射性標識タンパク質の製造
本例において、図1および5に説明されるように、[18F]−3−(2−(2−(2−(2−アジドエトキシ)エトキシ)エトキシ)エトキシ)−2−フルオロピリジンは、標的タンパク質を放射性標識するために用いられた。
900mCiの18Fの18O水(3ml)の放射能(IBA Molecularから購入)を、4,7,13,16,21,24−ヘキサオキサ−1,10−ジアザビシクロ[8.8.8]ヘキサコサン(2.8mg、7.44μmol)および炭酸カリウム(1.7mg、0.012mmol)を含むマイクロバイアル(no QMA)に直接移した。追加の2.0mlのアセトニトリルをこの粗製反応混合物に加え、全ての混合物を共沸によって乾燥させた。98℃のオイルバスを用いて溶液を蒸発させることによって、並びにN2の穏やかな気流を加えることおよび部分真空によって、乾燥を完了させた。溶液の体積は約2mlに減少した。追加の2mlのアセトニトリルを加え、このプロセスを40分にわたり3回繰り返した。液体の体積が0.3ml未満まで減少した時、0.7mlの分取量のアセトニトリルを加え、溶液をさらに共沸蒸留することで体積を〜0.1mlにまで減少させた。追加の0.9mlのアセトニトリルを加え、このプロセスを白色の固形物が生成するまで行った。このプロセスには〜55分要した。最後の手順の間は、溶液が乾燥する前にバイアルをオイルバスから外し、バイアル中の残留物を20分間室温で、完全真空(窒素気流なし)下に設置した。[18F]−FPPEGAクリプタンド混合物の移行および乾燥に要した合計時間は65分であった。

抗PD−L1アドネクチンイメージング剤を用いた、PD−L1−陽性腫瘍のPD−L1−陰性腫瘍とのインビボでの識別

カニクイサルにおけるインビボイメージング
カニクイサルにおいて行われた場合、18F−放射性標識E01イメージング剤はまた同様の結果を示した。これらの実験において、実施例9に記載されるように合成された18F−E01抗PD−L1は、カニクイサルにおいて高コントラスト画像が得られるかを試験された。本明細書に記載の抗PD−L1アドネクチンは、カニクイサルPD−L1に対して高い親和性を維持する(しかし、齧歯類PD−L1に対しては低い親和性を持つ)。さらに、カニクイサルは、マウスモデルにおいて見られるようなPD−L1(+)腫瘍を有していないため、イメージング性能は、内因性PD−L1発現(バックグラウンドが低いため、PD−L1(+)組織の高感度検出が可能になる)に関連する画像において測定される、バックグラウンド量に基づいて第一に評価される。これらの実験において、得られたPET画像におけるバックグラウンド量は極めて低く、顕著な放射性トレーサーの蓄積は主に腎臓、脾臓、および膀胱に見られた。
[18F]−A02抗PD−L1アドネクチンを用いたインビトロオートラジオグラフィー
ヒトの肺腫瘍組織をOCTに設置し、2−メチルブタン中で2−5分間、冷凍するまで冷却した。使用までサンプルを−80℃の冷凍庫に保存した。ヒトの異種移植片組織もまた、当該アッセイにおいてインキュベートした。両側性の異種移植片を有するマウスは、4×106のPD−L1(+)L2987細胞および1.5×106のPD−L1(−)HT−29t細胞を、nu/nuマウスの両側に皮下投与することで生成した。異種移植腫瘍が適切な大きさ(およそ200−300mm3)に到達した後に、マウスを2%イソフルランによって麻酔し、頸椎脱臼によって犠死させた。新たな腫瘍組織を切除し、OCTに浸し、2−メチルブタン中で2−5分間、冷凍するまで冷却した。次いで組織をfoil/ZIPLOC(登録商標)袋に包み、使用まで−80℃で保存した。全ての組織(ヒトの肺腫瘍および異種移植片)について、5μmの厚さの切片(2切片/スライドを回収した)を低温保持装置を用いて切り取り、顕微鏡のガラススライド上で解凍し、およそ30分間空気乾燥させた。
市販のGE TRACERlab FX2 N合成ユニットを用いた、放射性合成のための一般的な方法による、[18F]−3−(2−(2−(2−(2−アジドエトキシ)エトキシ)エトキシ)エトキシ)−2−フルオロピリジンの自動合成
手順:
非カセット型のGE TRACERlab FX2 N合成モジュールを用いて、[18F]−3−(2−(2−(2−(2−アジドエトキシ)エトキシ)エトキシ)エトキシ)−2−フルオロピリジンの自動合成を行った。合成ユニットの配置は表1および図6にまとめられる。[18F]−フルオライド水溶液(2.0ml、29.6GBq/800mCi)が、Sep−Pak light QMAに移された[Sep−Pak light QMAカートリッジは使用前に、5mlの0.5Mの炭酸水素カリウム、5mlの脱イオン水、および5mlのアセトニトリルの順で前処理を行った]。この移動が完了すると、反応容器に溶出混合液を加えることによって(「V1」から)、[18F]フルオライド水溶液をQMA Sep−Pakから溶出させた。溶媒を窒素の穏やかな気流および真空下で留去した。前駆体の溶液を(「V3」から)乾燥クリプタンド残留物に加え、この反応混合物を120℃で10分間加熱した。次いで、4mlの蒸留水を(「V4」から)反応容器中の粗製反応混合物に加え、混合物を、ローディングの終わりを制御する液体センサーを介して、セミ−分取HPLCの5mlサンプル注入ループに移した。混合物をセミ−分取HPLCカラム(Luna C18(2).250x10mm、Phenomenex)にロードした。0.1%トリフルオロ酢酸水溶液中の35%アセトニトリルの混合液を、毎分4.6mlの速度でカラムに通した。生成物をこのHPLCカラムから、15mlの蒸留水を含む希釈フラスコに回収し、全ての内容物を、tC18 1グラムの固相抽出カートリッジに移した。3mlのエタノールによって、このカートリッジから352mCi(13GBq)の[18F]−3−(2−(2−(2−(2−アジドエトキシ)エトキシ)エトキシ)エトキシ)−2−フルオロピリジンが溶出され(「V14」から)、アルキンを含む適切な生体分子との「クリック」アジド−アルキン反応を利用した18F標識生物学的製剤の生成に用いられうる。
市販のIBA Synthera合成ユニットを用いた、放射性合成のための一般的な方法による、[18F]−3−(2−(2−(2−(2−アジドエトキシ)エトキシ)エトキシ)エトキシ)−2−フルオロピリジンの自動合成
手順:

を含む、請求項20〜55のいずれか1つに記載の18F−放射性標識タンパク質ベースのプローブの画像を得る方法。 - 18F−放射性標識タンパク質ベースのプローブの分布の画像が、疾患の存在または非存在を示す、請求項56記載の方法。
- 対象における疾患の存在を診断する方法であって、
(a)疾患の存在に関係する標的分子に結合する、請求項20〜55のいずれか1つに記載の18F−放射性標識タンパク質ベースのプローブを、それを必要とする対象に投与すること;および
(b)当該18F−放射性標識タンパク質ベースのプローブの存在または非存在を検出するために、対象の少なくとも1部分の放射線画像を得ること;
を含み、前記18F−放射性標識タンパク質ベースのプローブの存在および位置がバックグラウンドに対して疾患の存在および位置を示す方法。 - 対象における疾患の進行をモニターする方法であって、
(a)疾患の存在に関係する標的分子に結合する、請求項20〜55のいずれか1つに記載の18F−放射性標識タンパク質ベースのプローブを最初の時点において、それを必要とする対象に投与すること、および疾患細胞または組織の量を決定するために、対象の少なくとも1部分の画像を得ること;および

を含み、疾患細胞または組織における放射性放射の量および分布が疾患細胞または組織の定量的な測定基準である方法。 - 疾患が固形癌、造血癌、血液癌、自己免疫性疾患、神経変性疾患、心血管疾患、および病原性感染から成る群から選択される、請求項58〜60のいずれか1つに記載の方法。
- 標的タンパク質を発現する組織または細胞の定量画像を得る方法であって、当該標的タンパク質に結合する、請求項20〜55のいずれか1つに記載の18F−放射性標識タンパク質ベースのプローブに、当該細胞または組織が接触すること、および陽電子放出断層撮影(PET)を用いて、当該標的タンパク質が発現している組織の検出または定量を行うことを含む方法。
- 疾患を治療する薬剤のスクリーニング方法であって、以下のステップ:
(a)疾患に関連する標的タンパク質を発現する細胞が、候補薬剤の存在下または非存在下において当該標的タンパク質と結合する、請求項20〜55のいずれか1つに記載の18F−放射性標識タンパク質ベースのプローブと接触すること;および
(b)当該候補薬剤の存在下または非存在において、陽電子放出断層撮影(PET)を用いて当該細胞のイメージングを行うこと
を含み、当該候補薬剤の存在において放射性放射量の減少が、当該薬剤が当該標的タンパク質に結合していることを示す方法。 - 18F−放射性標識タンパク質ベースのプローブがリガンドを含む、請求項56〜63のいずれか1つに記載の方法。
