ペプチドマスクのスクリーニング方法
一般に、望ましいマスキング表現型を有するペプチドマスクおよびペプチドマスクのスクリーニング方法は、(機能性タンパク質を結合するメンバを同定するための)ポジティブ・スクリーニング・ステップおよび(機能性タンパク質を結合しないメンバを同定するための)ネガティブ・スクリーニング・ステップによって達成される。ネガティブ・スクリーニング・ステップは、ペプチドマスクの非存在下で機能性タンパク質を結合するメンバを集団から消耗させることによって達成することができる。本明細書に記載するライブラリスクリーニング方法は、最初にネガティブスクリーニングを行って機能性タンパク質を結合しない候補を選択して、次にポジティブスクリーニング(すなわち候補ペプチドマスクを提示する複製可能な生物学的実体のライブラリを機能性タンパク質に曝露して、機能性タンパク質を結合するメンバを選択する)を行うことによって開始できることに注目すべきである。

本明細書で使用する場合、「標識」、「検出可能な標識」および「検出可能な部分」は互換的に使用されて、これに限定されるわけではないが、放射性同位元素、蛍光剤、化学発光剤、発色団、酵素、酵素基質、酵素補因子、酵素阻害剤、発色団、染料、金属イオン、金属ゾル、リガンド(たとえばビオチン、アビジン、ストレプトアビジンまたはハプテン)などを含む検出可能な分子を指す。「蛍光剤」という用語は、検出可能な範囲の蛍光を呈することが可能な物質またはその部分(portion)を指す。標識としての使用に好適な例示的な検出可能な部分は、親和性タグおよび蛍光タンパク質を含む。
興味のあるプロタンパク質の分離および回収を提供するいずれの好適な方法も利用され得る。たとえば興味のあるプロタンパク質を提示する細胞は、FACS、免疫クロマトグラフィーによって、または検出可能な標識が磁性である場合には、磁気分離によって分離され得る。分離の結果として、集団は、望ましい特徴を呈する、たとえば切断後に結合パートナーへの結合を呈するまたは切断の非存在下で結合パートナーへの結合の低下を有するもしくは検出可能な結合を有さない細胞が濃縮される。
本明細書に記載するプロタンパク質は、提供された切断可能なリンカー−タンパク質の併用による好適な被験体の処置方法での使用のために選択することができる。プロタンパク質の例示的な非制限的な使用は、C型肝炎、および新脈管形成のためである。たとえば(たとえば上記のような)病態に罹患している患者に、治療的有効量のプロタンパク質を投与することができる。
本明細書で提供される組成物およびプロタンパク質は、治療薬および診断薬を含む多様な目的に有用であることができる。
肝臓病態の処置におけるインターフェロンシグナル伝達経路を調節するプロタンパク質の使用
プロタンパク質がインターフェロンシグナル伝達を調節する機能性タンパク質を含有する場合、たとえば機能性タンパク質がIFN−αであるとき、プロタンパク質はC型肝炎ウイルス感染および肝癌(たとえば肝細胞癌に)などの病態の処置に用途が見出される。
癌阻害プロタンパク質は、複数の種類の腫瘍の処置に用途が見出される。
本明細書に記載する組成物の治療能力は、改変機能性タンパク質のより高い生体内分布生体内分布および生物学的利用能を可能にする。本明細書に記載する組成物は、改善された生物学的利用能を有するタンパク質治療薬を提供し、該機能性タンパク質治療薬のその結合パートナーへの結合の親和性は、罹患組織と比較したときに健常組織において低い。ペプチドマスクにカップリングされた機能性タンパク質を含む薬学的組成物は、健常組織よりも罹患組織において、その結合パートナーに対するより高い親和性を提示することができる。好ましい実施形態において、罹患組織での親和性は、健常組織での親和性よりも5〜10,000,000倍高い。例示的な実施形態において、罹患組織における親和性は、健常組織における親和性よりも約10,000倍高い。
本開示のプロタンパク質はたとえば、治療的有効量の興味のある活性化可能のマスクされたタンパク質および担体である薬学的に許容され得る賦形剤(薬学的に許容され得る担体とも呼ばれる)を含有する薬学的組成物に組み入れることができる。多くの薬学的に許容され得る賦形剤は当分野で公知であり、一般に投与経路、処置される病態、および当分野で十分に理解されている他のこのような変数に従って選択される。薬学的に許容され得る賦形剤は、たとえばA.Gennaro(2000)“Remington:The Science and Practice of Pharmacy,”20th edition,Lippincott,Williams,& Wilkins;Pharmaceutical Dosage Forms and Drug Delivery Systems(1999)H.C.Anselら、eds.,7th ed.,Lippincott,Williams,& Wilkins;およびHandbook of Pharmaceutical Excipients(2000)A.H.Kibbeら、eds.,3rd ed.Amer.Pharmaceutical Assoc.を含む多様な刊行物に詳細に記載されている。薬学的組成物は、他の構成要素、たとえばpH調整剤および緩衝剤、等張化剤、安定剤、湿潤剤なども含むことができる。いくつかの実施形態において、ナノ粒子またはリポソームは、プロタンパク質を含む薬学的組成物を担持する。
プロタンパク質は、診断方法および/または撮像方法でも使用できる。たとえば、酵素的に切断可能なリンカーを有するプロタンパク質を使用して、切断可能なリンカーの切断が可能である酵素の存在または非存在を検出することができる。このようなプロタンパク質は診断に使用することができ、該診断は、所与の宿主生物の所与の組織における活性化プロタンパク質の蓄積の測定を通じた、興味のある結合パートナーの存在を伴う酵素活性のインビボ検出(たとえば定性的または定量的)を含む。
ペプチドライブラリのスクリーニングおよびIFN−αに特異的なペプチドマスクの同定
インターフェロン−α(IFN−α)のペプチドマスクを同定するために、ペプチドライブラリをスクリーニングした。IFN−αを使用してランダム15×ペプチドライブラリをスクリーニングした。ここでXは、総多様性は5×1010である、いずれかのアミノ酸である。スクリーニングは、最初のラウンドのMACS(磁気活性化細胞ソーティング)と、続いての4ラウンドのFACS(蛍光活性化細胞ソーティング)から成った。最初のMACSおよび3ラウンドのFACSは、500nMの濃度のビオチン化IFN−αによって行った。MACSの場合、約1×1011個の細胞の結合をスクリーニングして、3.4×107個の細胞を収集した。ニュートラアビジン−PEを最初のFACSラウンドの蛍光プローブとして使用した。FACS選択の4回目のラウンドは、500nMダイライト標識IFN−α(ダイライト−IFN−α)を用いて行った。FACSソーティングの3回目および4回目のラウンドを図2に、ダイライト−IFN−αで標識して示す。
プロ−IFN−αの構築および発現
PhoA制御の下でのインターフェロン−αの構築:ヒトインターフェロン−α遺伝子をOpen Biosystemsから購入した。IFN−αをファージミド(Phagmid)X(PhoA駆動細菌発現ベクター)中に以下の方式でクローニングした。IFN−αはプライマーCX0573およびCX0566を使用して増幅した。PhoAプロモータをファージミド(Phagmid)Xから、プライマーCX0571およびCX0572を使用して増幅した。これらの2つの重複生成物を1つのポリメラーゼ連鎖反応で合せて、プライマーCX0581およびCX0572を使用して増幅した。HindIIIおよびEcoRI制限部位を使用して、最終生成物をファージミド(Phagmid)X中にクローニングした。
cDNAを増幅した。BamHIおよびEcoRI制限部位を使用して、ファージミドXベクターを含有するSTII中に本生成物をクローニングした。本ベクターを次に、MMP−9基質含有ベクターの構築のためにテンプレートとして使用した。2つの重複PCR生成物を、プライマー対CX0573/CX0612およびCX0611/CX0566を使用して増幅した。これらの2つの生成物をPCR中で合せ、プライマーCX0573およびCX0566によって増幅して、HindIIIおよびEcoRI制限部位を使用してファージミドX中にクローニングした。
プロ−IFN−αマスキングおよびアンマスキングの分析
プロ−IFN−αのマスキングを実証するために、再折畳みタンパク質の47−MMP−IFN−αまたは49−MMP−IFN−αをMMP−9消化緩衝液(50mMトリス、20mM NaCl、2mM CaCl2、100μM ZnCl2、pH6.82)中1:1で希釈して、試料の半分をMMP−9約35ユニットで37℃にて3時間消化した。続いて消化および非消化物質60、40、20、および6.6μLをPBS−T(PBS、0.05% ツイーン、pH7.4)中の2%脱脂粉乳400μLに添加して、記載するようにELISAで分析した:
インターフェロンELISA:組換えインターフェロン受容体1−Fc(IFNR1−Fc)融合タンパク質(R&D Systems)を使用してIFN−α結合を検出した。簡潔には、ELISAプレートに受容体をPBS中5μg/mLの濃度でRTにて1時間吸収させた。次にウェルをPBS−T中2%脱脂粉乳でRTにて1時間遮断した。3つの濃度60、40、20および6.6nMのインターフェロン−αをウェルのPBS−T中2%脱脂粉乳100μLに添加した。ウェルをPBS−Tで3回洗浄して、インターフェロンを、ウェル当りPBS−T中の2%脱脂粉乳100uL中の力価1:2000の抗muFc−HRPコンジュゲート(Fisher)と混合した力価1:1000の抗His6モノクローナル抗体(Invitrogen)によって検出した。ELISAを製造者のプロトコルに従って、TMB(Pierce)100μLによって展開させた(図3)。図3は、MMP−9による処置前後の、2つのプロ−インターフェロン−α分子、プロ−インターフェロン−α−47(表7および8)およびプロ−インターフェロン−α−49CS(表8および9)の結合を示す。図3の最初の4本の棒(細かいチェック柄)は、処置前にプロ−インターフェロン−α−49CSはIFNRAに結合できないが、マスク49CSのMMP−9除去後には、得られたIFN−α(2組目の4本の棒、図、大きいチェック柄)分子がIFNRAに結合することを示す。対照的にマスク47は、プロ−インターフェロン−α−47中に包含されているときに、IFNRAへのIFN−α結合を弱く遮断して(図3、3組目の棒、水平線)、これはMMP9を用いた処置によって回復される(図3、最後の4本の棒、垂直線)。

望ましい活性化特徴(すなわち切断配座にあるときのIFNRA受容体結合と比べた、未切断配座にあるときのそのIFNRA受容体への結合の低下)を有するIFN−αプロタンパク質を同定するために、可変マトリプターゼまたはHCV NS3/4切断可能なリンカーおよびペプチドマスク中の異なる可変アミノ酸配列およびペプチドマスク中のシステインの各種位置を有する候補IFN−αプロタンパク質が生成された。
マスクされた可溶性プラスミンまたはMMP−9活性化可能Notch受容体タンパク質の構築
マスクされたプラスミン−活性化可能可溶性Notch受容体フラグメントおよびマスクされたMMP9−活性化可能可溶性Notch受容体フラグメントを構築するための配列が本実施例で供給される。これらのプロタンパク質は、付着ペプチドマスクのために正常条件下で不活性である。細菌細胞表面提示を使用して、可溶性Notch受容体タンパク質の好適なペプチドマスクを見出す。本実施例において、選択されたペプチドマスクは、酵素媒介活性化の後の標的化結合に適するようになるプロタンパク質構築物を作るための誘因として使用されるプラスミンまたはMMP−9酵素基質のどちらかと併用される。
