Claims (31)

  1. 妊娠女性由来の生物試料中の胎児DNAを検出する方法であって、
    (a)メチル化DNA及び非メチル化DNAを別個に修飾する薬物で該試料を処置し;及び
    (b)該試料中のRASSF1A、APC、CASP8、RARB、SCGB3A1、DAB21P、PTPN6、THY1、TMEFF2又はPYCARDのDNA配列を検出し、ここで、該DNA配列の存在が試料中の胎児DNAの存在を指示し、該DNA配列の不存在が試料中の胎児DNAの不存在を指示する
    工程を含む前記方法。
  2. 前記試料が全血、血漿、血清、尿、及び唾液からなる群から選択される、請求項1に記載の方法。
  3. 前記薬物が非メチル化DNAを消化するが、メチル化DNAを消化しない、請求項1に記載の方法。
  4. 前記薬物がメチル化感受性酵素である、請求項に記載の方法。
  5. 前記メチル化感受性酵素がメチル化感受性制限酵素である、請求項に記載の方法。
  6. 前記メチル化感受性制限酵素がHpaII又はBstUIである、請求項に記載の方法。
  7. 前記薬物が亜硫酸水素塩を含む、請求項1に記載の方法。
  8. 工程(b)が増幅プロセスを含む、請求項1に記載の方法。
  9. 前記増幅プロセスがポリメラーゼ連鎖反応(PCR)である、請求項に記載の方法。
  10. 前記PCRがリアルタイムPCRである、請求項に記載の方法。
  11. 工程(b)がDNA配列の量を測定する、請求項1に記載の方法。
  12. 工程(b)が試料中の胎児DNAの存在を指示し、さらに、
    (c)第2試料中の第2胎児DNA配列の量を測定し、ここで、第2試料が薬物で処理される前の工程(a)の試料と同一であり、第2配列がRASSF1A、APC、CASP8、RARB、SCGB3A1、DAB21P、PTPN6、THY1、TMEFF2又はPYCARDでなく;及び
    (d)該2配列の量と標準対照とを比較し、ここで、対照からの第2配列の量の増加が妊娠に関連した状態の存在又はそれを発症する危険性の増加を指示する
    工程を含む、請求項1に記載の方法。
  13. 前記工程(c)の第2試料が、メチル化DNA及び非メチル化DNAを別個に修飾するいずれの薬物でも処理されていない、請求項12に記載の方法。
  14. 前記工程(c)の第2試料が、第2胎児DNA配列の量が測定される前に、薬物で処理される請求項12に記載の方法。
  15. 前記工程(c)の第2試料が、第2胎児DNA配列の量が測定される前に、メチル化DNA及び非メチル化DNAを別個に修飾する第2の異なった薬物で処理される、請求項12に記載の方法。
  16. 妊娠に関連した状態が、子癇前症、早期陣痛又は子宮内発育遅延(IUGR)である、請求項12に記載の方法。
  17. 工程(b)が試料中の胎児DNAの存在を指示し、さらに、
    (c)第2試料中の第2胎児DNAを検出し、ここで、第2試料が薬物で処理される前の工程(a)の試料と同一であり、第2配列がRhD血液型の遺伝子、ABO血液型の遺伝子、RhC血液型の遺伝子、RhE血液型の遺伝子、HLA型の遺伝子、Y染色体上に位置した遺伝子、又は所定の突然変異を含む遺伝子であり、第2配列の存在が胎児ゲノム中にRhD血液型、ABO血液型、RhC血液型、RhE血液型、HLA型、Y染色体又は突然変異の存在を指示する
    工程を含む、請求項1に記載の方法。
  18. 工程(c)の第2試料がメチル化DNA及び非メチル化DNAを別個に修飾するいずれの薬物でも処理されていない、請求項17に記載の方法。
  19. 工程(c)が増幅プロセスである、請求項17に記載の方法。
  20. 前記増幅プロセスがポリメラーゼ連鎖反応(PCR)である、請求項19に記載の方法。
  21. PCRがリアルタイムPCRである、請求項20に記載の方法。
  22. 妊娠女性における妊娠に関連した状態を検出する方法であって、
    (a)メチル化DNA及び非メチル化DNAを別個に修飾する薬物で女性から得た生物試料を処理し;
    (b)該試料中のRASSF1A、CASP8、RARB、SCGB3A1、DAB21P、PTPN6、THY1、TMEFF2又はPYCARDのDNA配列の量を検出し;及び
    (c)該DNA配列の量と標準対照とを比較し、ここで、対照からの増加が妊娠に関連した状態の存在又はそれを発症する危険性の増加を指示する
    工程を含む前記方法。
  23. 前記薬物が非メチル化DNAを消化するが、メチル化DNAを消化しない、請求項22に記載の方法。
  24. 前記薬物がメチル化感受性酵素である、請求項23に記載の方法。
  25. 前記メチル化感受性酵素がメチル化感受性制限酵素である、請求項24に記載の方法。
  26. 前記メチル化感受性制限酵素がHpaII又はBstUIである、請求項25に記載の方法。
  27. 前記薬物が亜硫酸水素塩を含む、請求項22に記載の方法。
  28. 工程(b)が増幅プロセスを含む、請求項22に記載の方法。
  29. 前記増幅プロセスがポリメラーゼ連鎖反応(PCR)である、請求項28に記載の方法。
  30. 前記PCRがリアルタイムPCRである、請求項29に記載の方法。
  31. 妊娠に関連した状態が、子癇前症、早期陣痛又は子宮内発育遅延(IUGR)である、請求項22に記載の方法。
