Abstract
La presente invención se refiere a sistemas, métodos y aparatos que pueden determinar y utilizar perfiles de metilación de diversos tejidos y muestras. Se proporcionan ejemplos. Un perfil de metilación se puede deducir para el tejido fetal/de tumor con base en una comparación de la metilación del plasma (u otra muestra con el ADN libre de células) con un perfil de metilación de la madre/paciente. Un perfil de metilación se puede determinar para el tejido fetal/de tumor utilizando alelos específicos de tejido para identificar el ADN del feto/tumor cuando la muestra tiene una mezcla de ADN. Un perfil de metilación se puede utilizar para determinar las variaciones de número de copias en el genoma de un feto/tumor. Los marcadores de metilación de un feto se han identificado a través de diversas técnicas. El perfil de metilación se puede determinar mediante la determinación de un parámetro de tamaño de una distribución de tamaño de fragmentos de ADN, donde los valores de referencia para el parámetro de tamaño se pueden utilizar para determinar los niveles de metilación. Además, un nivel de metilación se puede utilizar para determinar un nivel de cáncer.
