Description
本出願は、2005年12月5日に提出された「POLYGLUTAMATE−AMINO ACID AND METHODS」と題された米国仮特許出願番号60/742,291;2006年1月10日に提出された「POLYGLUTAMATE−ASPARTATE−TAXANES」と題された米国仮特許出願番号60/757,917;および2006年4月10日に提出された「POLYGLUTAMATE−ASPARATATE−MRI CHELATES」と題された米国仮特許出願番号60/790,735に対して優先権を主張する。これらは全て、それらの全体が引用により本明細書中に組み入れられる。
本発明は、概して、ペンダント官能基を有している生体適合性の水溶性ポリマー、およびそれらを作製する方法に関し、特に、様々な薬物、生体分子、および造影剤の送達用途に有用であるポリグルタミン酸アミノ酸結合体に関する。
薬物、生体分子、および造影剤の送達は様々な系が使用されている。例えば、そのような系としては、カプセル、リポソーム、マイクロ粒子、ナノ粒子、およびポリマーが挙げられる。
いくつかの実施形態においては、本明細書中に開示される化合物(例えば、ポリマー結合体および/またはそれを含む薬剤)のプロドラッグ、代謝物、立体異性体、水和物、溶媒和物、多形体、および薬学的に許容される塩が提供される。
化合物または薬学的組成物は、任意の適切な手段によって患者に投与することができる。投与方法の限定ではない例としては、中でも、以下が挙げられる:活性化合物を生存している組織と接触させるための当業者によって適切であると考えられる、(a)経口経路からの投与(この投与には、カプセル剤、錠剤、顆粒剤、噴霧剤、シロップ剤、または他のそのような形態での投与が含まれる);(b)経口以外の経路による投与(例えば、直腸、膣、尿道内、眼内、鼻腔内、または耳介内)(この投与には、水性懸濁剤、油状調製物などとして、または液滴、噴霧剤、坐剤、軟膏、膏薬などとしての投与が含まれる);(c)注射(皮下、腹腔内、静脈内、筋肉内、皮内、眼窩内、莢膜内、髄腔内、胸骨内など)(注入ポンプによる送達が含まれる);(d)腎臓または心臓の領域への直接の注射によるような局所的な投与(例えば、デポー移植物による);さらには、(e)局所投与。
様々な分子量(多角光散乱(MALS)に基づいて41,400(PGA(97k))、17,600(PGA(44k))、16,000(PGA(32k))、および10,900(PGA(21k))ダルトンの平均分子量)を有しているポリ−L−グルタミン酸ナトリウム塩;1,3−ジシクロヘキシルカルボジイミド(DCC);N−(3−ジメチルアミノプロピル)−N’−エチルカルボジイミド塩酸塩(EDC);ヒドロキシベンゾトリアゾール(HOBt);ピリジン;4−ジメチルアミノピリジン(DMAP);N,N’−ジメチルホルムアミド(DMF);酢酸ガドリニウム;クロロホルム;および重炭酸ナトリウムは、Sigma−Aldrich Chemical社から購入した。ポリ−L−グルタミン酸を、2Nの塩酸溶液を使用してポリ−L−グルタミン酸に変換した。トロフルオロ酢酸(TFA)は、Bioscienceから購入した。Omniscan(商標)(ガドジアミド)はGE healthcareから購入した。

ポリ−(γ−L−アスパルチル−グルタミン)を、以下のように、図1の一般的な反応スキームにしたがって調製した:
ポリ−(γ−アスパルチル−グルタミン)−ポリ−L−グルタミン酸を、以下のように、図2に示す一般的な反応スキームにしたがって調製した。
ポリ−(γ−L−アスパルチル−グルタミン)を、以下のように、図3に示した一般的な反応スキームにしたがって調製した。
様々な平均分子量(MALS検出器を用いるHeleosシステムに基づき、19,800および9,400ダルトン)を有している出発ポリグルタミン酸ナトリウム塩からのポリ−(γ−L−アスパルチル−グルタミン)の合成を、実施例3の手順を使用して行い、得られたポリマーであるポリ−(γ−L−アスパルチル−グルタミン)の平均分子量を測定し、そしてそれぞれ、39,700および17,700ダルトンであることを見出した。
ポリ−(γ−L−アスパルチル−グルタミン)を、以下のように、図4に示した一般的な反応スキームにしたがって調製した。
様々な平均分子量(MALS検出器を用いるHeleosシステムに基づき、49,000、19,450および10,900)を有しているポリ−L−グルタミン酸ナトリウム塩からのポリ−(γ−L−グルタミル−グルタミン)の合成を、実施例4の手順を使用して行った。それらのポリ(γ−L−グルタミル−グルタミン)ポリマーの平均分子量を測定し、そしてそれぞれ、110,800、37,400、および19,800ダルトンであることを見出した。
ポリ−(γ−L−グルタミル−グルタミン)−ポリ−L−グルタミン酸を、以下のように、図5に示した一般的な反応スキームにしたがって調製した。
PGA−97−A−Texas Redと本明細書中で呼ばれるポリマー結合体を、以下のように、図6に示した一般的な反応スキームにしたがって調製した。
PGA−97−A−DTPAと本明細書中で呼ばれるポリマー結合体を、以下のように、図7に示した一般的な反応スキームにしたがって調製した。
PGA−97−A−DTPA−Gd(III)と本明細書中で呼ばれるポリマー結合体を、以下のように、図8に示した一般的な反応スキームにしたがって調製した。
PGA−97−A−10と本明細書中で呼ばれるポリマー結合体を、以下のように、図9に示した一般的な反応スキームにしたがって調製した。
PGA−97−A−20と本明細書中で呼ばれるポリマー結合体を、以下のように、図9に示した一般的な反応スキームにしたがって調製した。
それぞれ、39,700および17,700ダルトンの平均分子量を有しているポリ−(γ−L−アスパルチル−グルタミン)ポリマーからのPGA−−44−A−20およびPGA−−21−A−20と本明細書中で呼ばれるポリマー結合体の合成を、実施例10の手順を使用して行った。ポリマー中のパクリタキセルの含有量を、UV分光分析によって、20重量%と決定した。
39,700ダルトンの平均分子量を有しているポリ−(γ−L−アスパルチル−グルタミン)からのPGA−−44−A−19と本明細書中で呼ばれるポリマー結合体の合成を、パクリタキセルだけを添加する代わりにパクリタキセルと3H−パクリタキセルとの混合物を添加するよう変更して、実施例10の手順を使用して行った。ポリマー中のパクリタキセルの含有量を、UV分光分析によって19重量%と決定した。
PGA−−97−G−20と本明細書中で呼ばれるポリマー結合体を、以下のように、図10に示した一般的な反応スキームにしたがって調製した。

(細胞培養および調製):
B16F0細胞をATCC(CRL−6322、ATCC アメリカンタイプカルチャーコレクション,Rockville,MD)から購入し、これを10%のウシ胎児血清および100単位/mLのペニシリンを含むダルベッコ改変イーグル培地(DMEM)で増殖させた。細胞は、5%のCO2環境下37℃で増殖させた。培養培地を除去し、廃棄した。細胞を、ダルベッコリン酸緩衝溶液(Dulbecco Phosphate Buffer Solution(DPBS))ですすぎ、トリプシン−エチレンジアミン四酢酸(EDTA)溶液(0.5ml)を加え、そして細胞を倒立顕微鏡下で観察して、これらが分散していることを確認した。完全な増殖培地(6.0から8.0ml)を加え、そして細胞をゆっくりとピペッティングすることによって吸引した。適切なアリコート中の細胞懸濁液を新しい培養プレートに移した。細胞を、5%のCO2中37℃で24時間増殖させ、その後、さらに実験を行った。
(インビトロでの細胞による取り込み実験)
PGA−97−A−Texas RedおよびTexas Red色素(TR)をDPBS中に別々に溶解させた。色素を含む両方の溶液を、1.0μMから10μMの最終濃度で細胞に添加した。化合物を含む細胞を37℃で8〜24時間インキュベートし、その後、細胞をDPBSで3回洗浄した。処理した細胞を、OLYMPUS蛍光顕微鏡下で試験し、そして励起および放射波長を、それぞれ、591nmおよび612nmで測定した。結果は、細胞はPGA−97−A−Texas Redに由来するTexas red色素を取り込んだが、Texas Redのみに由来する色素は取り込まなかったことを示している。
細胞による取り込みをまた、共焦点顕微鏡(Olympus FV1000)によっても確認した。細胞の核をHoechst 33342で5〜20分間染色し、DPBSで2〜3回洗浄し、そしてレーザースキャン共焦点顕微鏡下で観察した。Hoechst 33342の励起および放射波長を、それぞれ、405nmおよび461nmで測定した。Texas Red(TR)を543nmのレーザーで励起させ、そして、37℃の5%のCO2環境下615nmで検出した。結果は、暴露の24時間後に、PGA−97−A−Texas Redに由来するTexas red色素がB16F0細胞によって取り込まれたことを示している。PGA−97−A−Texas Redに由来するTexas red色素は細胞質で見られ、そして核からは排除されていた。
(同系腫瘍モデル)
動物:Nu/nuマウス、雌、6〜8週齢(22〜25g)。孤立性腫瘍を、2×105のマウス黒色腫細胞(B16F0)を右腿の皮下に注射することによって生じさせた。5〜7日後に、腫瘍が約500mm3に達すると、PGA−97−A−Texas RedまたはTexas Red色素を腫瘍に対して静脈内に注射した。
(PGA−97−A−Texas RedまたはTRの投与およびクリオスタット切片)
PGA−97−A−Texas RedおよびTexas RedをDPBS中に別々に溶解させ、動物に投与する前に0.2μlのフィルターを通して濾過した。100μlのPGA−97−A−Texas Red(2.5%のTR負荷)または0.1〜10mMのTexas Redを、実施例19の同系腫瘍モデルを使用して腫瘍に静脈内注射した。腫瘍を最適な切開温度で切り取り、包埋し、そして液体窒素中で凍結させた。クリオスタット切片(6〜15μm)を作製し、そして氷上で10〜30分間、0.03Mのスクロースを含む4%のパラホルムアルデヒドで固定した。切片をDPBSで2回洗浄し、Hoechst 33342(1μg/ml)で10分間染色し、そしてDPBSで再び洗浄した。その後、切片を、蛍光封入剤(DakoCytomation)でマウントし、カバーガラスで覆った。腫瘍のクリオスタット切片を、レーザースキャン共焦点顕微鏡下で観察した。画像は、PGA−97−A−Texas Redに由来するTexas red色素が、PGA−97−A−Texas Redの静脈内投与の24時間後にインビボで腫瘍細胞中に蓄積したが、Texas Red色素のみの場合には蓄積しなかったことを示していた。
(インビトロでの細胞傷害性MTT試験)
パクリタキセルを含む本明細書中に記載したポリマー結合体を、いくつかの異なる薬物濃度でB16F0黒色腫細胞の増殖に対するそれらの効果について評価した。細胞傷害性MTTアッセイを、Monksら、JNCI 1991,83.757−766(その全体が引用により本明細書中に組み入れられる)に報告されているように行った。PGA−44−A−20を、実施例10aと同様に、MALS検出器を用いるHeleosシステムに基づいて39,700ダルトンの平均分子量を有しているポリ(γ−L−アスパルチル−グルタミン)から調製し、そしてポリマー中のパクリタキセルの重量百分率は20重量%であった。PGA−97−A−20を、実施例10と同様に、MALS検出器を用いるHeleosシステムに基づいて99,400ダルトンの平均分子量を有しているポリ(γ−L−アスパルチル−グルタミン)から調製し、そしてポリマー中のパクリタキセルの重量百分率は20重量%であった。PGA(97k)−PTX−20を本実施例の対照ポリマーとして使用し、これを、MALS検出器を用いるHeleosシステムに基づいて49,000ダルトンの平均分子量を有しているポリ−L−グルタミン酸から先の文献の手順にしたがって調製し、ポリマー中のパクリタキセルの重量百分率は20重量%であった(Liら、「Complete Regression of Well−established tumors using novel water soluble poly(L−glutamic acid)−paclitaxel conjugate.」Cancer Research 1998,58,2404−2409を参照のこと)。結果を図13に示す。黒色腫細胞の生存性は、図13に示したように、薬物濃度が増大するに伴い低下した。これらの結果は、PGA−44−A−20およびPGA−97−A−20が有効な抗ガン剤であることを示している。

(薬物動態実験のための動物および腫瘍モデル)
ヌードマウス(6〜7週齢、体重25〜30グラム、雌)を、Charles River Lab(Willington,MA)から購入した、B16F0細胞株はATCC(CRL−6322、ATCC アメリカンタイプカルチャーコレクション,Rockville,MD)から購入した。B16F0細胞を10%のウシ胎児血清、2μMのグルタミン、1mMの必須ではないアミノ酸、1mMのピルビン酸ナトリウム、100U/mLのペニシリン、および100μg/mlのストレプトマイシンを補充したDMEMで培養した。組織培養物から採取したB16F0細胞をカウントし、5×106/mLの濃度で再度懸濁した。TB注射器を使用して、0.4mL(全部で2×106細胞)を、それぞれのマウスに皮下注射によって投与した。1匹の動物あたり4つの腫瘍を、右肩、左肩、右臀部、および左臀部に接種した。
実施例23によるマウスの集団全体について平均の腫瘍の容積が200〜300mm3(6〜8mmの直径)に達した時点で、腫瘍を有しているそれぞれのマウスに、3H−Taxol(対照)またはPGA−44−A−19の1回のIVボーラス注射を尾静脈から投与した。
実施例23によるマウスの集団の全てについて、平均の腫瘍容積が200〜300mm3(6〜8mmの直径)に達した時点で、腫瘍を有しているそれぞれのマウス(ヌードnu/nuマウス)に、3H−Taxol(対照)またはPGA−44−A−19の1回のIVボーラス注射を、尾静脈から投与した。
(インビボでの効力の実験のための動物および腫瘍モデル)
ヌードマウス(6〜8週齢、体重21〜25グラム、雄)を、Charles River Lab(Willington,MA)から購入した。B16−F0−EGFP安定細胞を、10%のウシ血清、100U/mLのペニシリン、および100μg/mlのストレプトマイシンを補充したDMEMで増殖させた細胞培養物中で維持した。細胞を、接種の48時間前に分け、その結果、これらを採取時に対数増殖期になるようにした。細胞を、トリプシン−EDTAを使用して組織培養物から採取し、そして生存している細胞の数を、トリパンブルーの存在下で血球計においてカウントすることによって決定した。細胞を、血清を含まないDMEM培地中に5×106/mlの濃度で再度懸濁した。腫瘍細胞懸濁液を、それぞれの肩およびそれぞれの臀部の上に5×106/mlの濃度で1ccのインシュリン注射器を使用して、0.1mlの腫瘍細胞懸濁液を注射することによって接種した(4部位/マウス)。
実施例11a〜11cにしたがって作成したポリマーの最大耐量(MTD)での体重減少の毒性を測定した。MTDは、本明細書中では、2週間以内に最大15%の体重の減少を生じる用量と定義する。PGA−21−G−20およびPGA−32−G−20を、それぞれ、実施例11cおよび11bに開示したように、MALS検出器を用いるHeleosシステムに基づいて、それぞれ、19,800および37,400ダルトンの平均分子量を有している出発ポリ(γ−L−アスパルチル−グルタミン)ポリマーから調製し、そしてそれぞれのポリマー中のパクリタキセルの重量百分率は20重量%であった。PGA−21−G−20およびPGA−32−G−20を、50mg/mLで生理食塩水中に溶解させた。この実施例についての対照抗ガン剤はAbraxaneであり、これは、抗ガン剤としてFDAによって承認されている。生理食塩水もまた、抗腫瘍薬を含まないネガティブ対照として使用した。注射した薬物の実際の量は、それぞれの動物の体重から決定した。薬物の最初の用量は、マウスの集団全体の平均の腫瘍の大きさが約15から約50mm3(腫瘍の大きさは、式(w2×l)/2から概算し、式中、「l」は、ミリメートルで測定した腫瘍の最長直径であり、「w」は最長直径に対して垂直な直径である)に達した時点で、マウスに投与した。マウスには、2用量の薬物を連続する2日間に、麻酔をせずに尾静脈への注射によって投与した。ストック溶液は注射を行う日に新しく調製した。薬物のストック溶液を1ccの注射器に抜き取り、静脈内に注射した。マウスを最も近い0.1g単位まで体重を測定した。ヌードnu/nuマウスには、100mg/kgのパクリタキセル等量の用量のAbraxaneと比較して、175mg/kgの用量のPGA−21−G−20と150mg/kgの用量のPGA−32−G−20との両方の高用量を注射した。それぞれの薬物で処置した場合の体重の変化(%)を別々に観察し、経時的に(日)記録した。結果を図17に示す。PGA−21−G−20は、はるかに高い用量でもほとんど体重の減少を示さなかった。PGA−32−G−20は、はるかに高い用量でも、Abraxaneと同等の体重の減少しか示さなかった。これらの結果は、抗ガン剤と結合させた本発明の好ましいポリマーはマウスに対する毒性が低いことを示している。

ポリマー結合体を、様々な分子量のポリマーの選択に関してパクリタキセルが放出される速度を決定するために試験した。PGA−21−G−20、PGA−32−G−20、PGA−97−G−20と、PGA(97k)−PTX−20の対照とを2mg/mLの濃度でリン酸緩衝液中に入れ、放出速度を測定した。ポリマー−パクリタキセル結合体の溶液を37℃でインキュベートした。50μlのアリコートを様々な時点で取って、凍結させた。その後、全てのアリコートをLC−MSによって分析した。HPLCプロフィール上の放出された薬物のピークの累積面積を測定した。放出されたパクリタキセルの量を検量線から計算した。結果を図23に示し、これは、ポリマー結合体の分子量が大きくなるに伴い、放出されるパクリタキセルの百分率が減少することを示している。これらの結果は、パクリタキセルの放出速度を、ポリマーの様々な分子量を選択することによって制御することができることを示している。

ヌードマウス(6〜7週齢、体重25〜30グラム、雌)を、Charles River Lab(Willington,MA)から購入した、B16F0細胞株はATCC(CRL−6322、ATCC アメリカンタイプカルチャーコレクション,Rockville,MD)から購入した。B16F0細胞を10%のウシ胎児血清、2μMのグルタミン、1mMの必須ではないアミノ酸、1mMのピルビン酸ナトリウム、100U/mLのペニシリン、および100μg/mlのストレプトマイシンを補充したDMEMで培養した。組織培養物から採取したB16F0細胞をカウントし、5×106/mLの濃度で再度懸濁した。TB注射器を使用して、0.4mL(全部で2×106細胞)を、それぞれのマウスに皮下注射によって投与した。1匹の動物あたり4つの腫瘍を、右肩、左肩、右臀部、および左臀部に接種した。
様々な薬物結合ポリマーを、経時的に血漿中のパクリタキセル濃度を決定するために、Taxolの対照に対して試験した。実施例32によるマウスの集団全体について平均の腫瘍容積が200〜300mm3(6〜8mmの直径)に達した時点で、腫瘍を有しているそれぞれのマウスに、3H−Taxol(対照)、PGA−21−A−19、PGA−32−A−19、PGA−97−A−24の1回のIVボーラス注射を尾静脈から投与した。
様々な薬物結合ポリマーを、経時的に腫瘍中のパクリタキセル濃度を決定するために、Taxolの対照に対して試験した。実施例32によるマウスの集団全体について平均の腫瘍容積が200〜300mm3(6〜8mmの直径)に達した時点で、腫瘍を有しているそれぞれのマウスに、3H−Taxol(対照)、PGA−21−A−19、PGA−32−A−19、PGA−97−A−24の1回のIVボーラス注射を尾静脈から投与した。
(動物および腫瘍モデル)
ヌードマウス(6〜7週齢、体重25〜30グラム、雄)を、Charles River Lab(Willington,MA)から購入した。B16細胞株はATCC(CRL−6322、ATCC アメリカンタイプカルチャーコレクション,Rockville,MD)から購入した。B16細胞を、10%のウシ胎児血清、2μMのグルタミン、1mMの必須ではないアミノ酸、1mMのピルビン酸ナトリウム、100U/mLのペニシリン、および100μg/mlのストレプトマイシンを補充したDMEMで培養した。組織培養物から採取したB16細胞をカウントし、5×106/mLの濃度で再度懸濁した。TB注射器を使用して、0.2mL(全部で1×106細胞)を、それぞれのマウスに皮下注射によって投与した。1匹の動物あたり1つの腫瘍を、右臀部に接種した。腫瘍の接種部位は、それが成長した場合に腫瘍を測定することを容易にするために、接種前に毛をそり落とした。
(腫瘍の蓄積についての磁気共鳴影像法)
ニーコイル(knee coil)のpre−contrastおよびpost−contrastを使用して、GE 3T MRスキャナーでマウスの画像を獲得した。撮影パラメーターは以下であった:TE:minful、TR=250ms、FOV:8および24スライス/スラブ、および1.0mmのコロナスライス(coronal slice)厚。PGA−97−A−DTPA−Gd(III)を、実施例7〜8と同様に、MALS検出器を用いるHeleosシステムに基づいて、99,400ダルトンの平均分子量を有しているポリ(γ−L−アスパルチル−グルタミン)から調製した。この実施例の対照材料は、Omniscan−Gd(III)−(DTPA−BMA(0.1mmolのGd(III)/kg)であった。PGA−97−A−DTPA−Gd(III)の注射用量は0.1mmol Gd(III)/kgであった。Omniscan(商標)の注射用量は0.1mmol Gd(III)/kgであった。2つの化合物を麻酔したマウスに尾静脈から注射し、画像を、注射前、および造影剤の注射後6分から4時間で獲得した。MRIの結果を図26に示す。図26に示すように、腫瘍組織に蓄積したPGA−97−A−DTPA−Gd(III)キレートの量は、低分子Omniscan−Gd(III)よりも多かった。これらの結果は、PGA−97−A−DTPA−Gd(III)キレートが高い特異性と保持を有していることを示している。
(ナノ粒子の形成の実験)
様々な溶液(0.2μmのフィルターを通して濾過した)を、示した場合を除き、1mg/mLでポリ−(g−アスパルチル−グルタミン)(分子量は99,400ダルトンであった)に加えた。溶液は全て均質に溶解させた。粒子サイズ、多分散性、およびベースライン指数を、光散乱ZeltaPals(Brookhaven Instrument Corporation)によって測定した。結果を表2にまとめた。MilliQ水は、0.2μmのフィルターを有している処理系によって濾過された水を意味する。
(PGA−97−A−10のナノ粒子の形成)
PGA−97−A−10を様々な濃度で脱イオン水に溶解させた。粒子サイズ、多分散性、およびベースライン指数を、光散乱(ZeltaPals,Brookhaven Instrument Corporation)によって測定した。結果を以下の表3に示す。
薬物結合ポリマーの凍結割断電子顕微鏡による画像を、Nano Analytical Laboratory(San Francisco,CA)によって撮影した。ポリマーは、ポリ−(g−L−アスパルチル−グルタミン)から調製したPGA−44−A−20であって、分子量は39,700ダルトンであり、ポリマー中のパクリタキセルの重量による百分率は20%であった。これを、超音波処理(約5分)後に生理食塩水中に1mg/mLの濃度にした。その後、これをパラフィルムで包み、すぐに会社に送った(輸送にほぼまる1日)。到着すると、これを4℃で保存した。その後、ポリマーを、ナノ粒子が形成するかどうかを決定するために生理食塩水に入れた。電子顕微鏡画像の複製を図27に示す。画像で見ることができるように、ポリマー結合体を水溶液に入れると、本発明の好ましい薬物結合ポリマーのナノ粒子が形成された。
薬物結合ポリマーの粒子を、様々な薬物濃度での安定性を決定するために試験した。PGA−44−A−20およびPGA−97−A−20を様々な薬物濃度で粒子になるように形成し、粒子サイズを測定した。結果を図28に示す。粒子はナノ粒子サイズの範囲を維持し、そして薬物濃度が高くなるに伴い、さらに安定となった。これらの結果は、安定なナノ粒子を、広い範囲の薬物濃度で形成することができることを示している。
薬物結合ポリマーの粒子を、様々な薬物濃度での安定性を決定するために試験した。PGA−21−G−20およびPGA−32−G−20を様々な薬物濃度で粒子になるように形成し、粒子サイズを測定した。結果を図29に示す。粒子はナノ粒子サイズの範囲を維持し、そして薬物濃度が高くなるに伴い、さらに安定となった。これらの結果はさらに、安定なナノ粒子を、広い範囲の薬物濃度で形成することができることを示している。
Claims (58)
- 式(I)の繰り返し単位と式(II)の繰り返し単位とを含むポリマー結合体であって:
nはそれぞれ独立して、1または2であり;
A1はそれぞれ、酸素はまたNR5であり;
A2はそれぞれ酸素であり;
R1およびR2はそれぞれ独立して、C1〜10アルキル、C6〜20アリール、アンモニウム、アルカリ金属、多座リガンド、保護されている酸素原子を有している多座リガンド前駆体、および薬剤を含む化合物からなる群より選択され;
前記薬剤は、抗ガン剤、標的化剤、光学的造影剤、および磁気共鳴造影剤からなる群より選択され;
R1およびR2の少なくとも一方は、薬剤を含む基であり;
R3およびR4はそれぞれ独立して、水素、アンモニウム、およびアルカリ金属からなる群より選択され;
ポリマー結合体には、ポリマー結合体に対する前記薬剤の質量比に基づいて約1から約50%(重量/重量)の範囲の前記薬剤の量が含まれ;
R5は水素またはC1〜4アルキルであり;そして
薬剤の量、式(I)の繰り返し単位の百分率および式(II)の繰り返し単位の百分率は、実質的に同じ量の薬剤を含む比較可能なポリグルタミン酸結合体の溶解度よりも大きいポリマー結合体の溶解度が得られるように選択され、約22℃の0.9重量%のNaCl水溶液に少なくとも5mg/mLのポリマー結合体が含まれている試験されたポリマー結合体溶液が、比較可能な試験されたポリグルタミン酸結合体溶液の光学的透明度よりも、広いpH範囲にわたって高い光学的透明度を有している場合には、ポリマー結合体の溶解度はより大きい
ポリマー結合体。 - 前記薬剤を含む前記化合物にリンカー基がさらに含まれている、請求項1から2のいずれか1項に記載にポリマー結合体。
- 前記薬剤が光学的造影剤である、請求項1から3のいずれか1項に記載のポリマー結合体。
- 前記光学的造影剤が、アクリジン色素、クマリン色素、ローダミン色素、キサンテン色素、シアニン色素、およびピレン色素からなる群より選択される、請求項4に記載のポリマー結合体。
- 前記薬剤が抗ガン剤である、請求項1から3のいずれか1項に記載のポリマー結合体。
- 前記抗ガン剤が、タキサン、カンプトテシン、およびドキソルビシンからなる群より選択される、請求項6に記載のポリマー結合体。
- 前記タキサンが、パクリタキセルおよびドセタキセルからなる群より選択される、請求項7に記載のポリマー結合体。
- パクリタキセルが、C2’−炭素に結合された酸素原子で、式(I)の繰り返し単位に結合されている、請求項8に記載のポリマー結合体。
- パクリタキセルが、C7−炭素に結合された酸素原子で、式(I)の繰り返し単位に結合されている、請求項8に記載のポリマー結合体。
- 前記薬剤が磁気共鳴造影剤である、請求項1から3のいずれか1項に記載のポリマー結合体。
- 前記磁気共鳴造影剤にGd(III)化合物が含まれる、請求項11に記載のポリマー結合体。
- 前記ポリマーに、式(I)および式(II)の繰り返し単位の総モル数に基づいて、約1モル%から約30モル%の式(I)の繰り返し単位が含まれている、請求項1または3から15のいずれか1項に記載のポリマー結合体。
- 前記ポリマー結合体に、式(I)および式(II)の繰り返し単位の総モル数に基づいて、約1モル%から約20モル%の式(I)の繰り返し単位が含まれている、請求項1または3から15のいずれか1項に記載のポリマー結合体。
- 前記ポリマー結合体に、式(I)および式(II)の繰り返し単位の総モル数に基づいて、約1モル%から約10モル%の式(I)の繰り返し単位が含まれている、請求項1または3から15のいずれか1項に記載のポリマー結合体。
- 前記ポリマー結合体に、式(I)、(II)、および(III)の繰り返し単位の総モル数に基づいて、約1モル%から約99モル%の式(I)の繰り返し単位が含まれている、請求項2から15のいずれか1項に記載のポリマー結合体。
- 前記ポリマー結合体に、式(I)、(II)、および(III)の繰り返し単位の総モル数に基づいて、約1モル%から約30モル%の式(I)の繰り返し単位が含まれている、請求項2から15のいずれか1項に記載のポリマー結合体。
- 前記ポリマー結合体に、式(I)、(II)、および(III)の繰り返し単位の総モル数に基づいて、約1モル%から約20モル%の式(I)の繰り返し単位が含まれている、請求項2から15のいずれか1項に記載のポリマー結合体。
- 前記ポリマー結合体に、式(I)、(II)、および(III)の繰り返し単位の総モル数に基づいて、約1モル%から約10モル%の式(I)の繰り返し単位が含まれている、請求項2から15のいずれか1項に記載のポリマー結合体。

- 請求項1から33のいずれか1項に記載のポリマー結合体を作製する方法であって、ポリマー反応体を溶媒に溶解または部分的に溶解させて、溶解または部分的に溶解されたポリマー反応体を形成する工程;および前記溶解または部分的に溶解されたポリマー反応体を第2の反応体と反応させる工程を含み、第2の反応体には、多座リガンド、保護されている酸素原子を有している多座リガンド前駆体、および薬剤を含む化合物からなる群より選択される少なくとも1つが含まれる、方法。
- 前記第2の反応体に、ヒドロキシおよびアミンからなる群より選択される置換基が含まれる、請求項34から36のいずれか1項に記載の方法。

- 前記パクリタキセルが、C2’−炭素に結合された酸素原子で、式(I)の繰り返し単位に結合されている、請求項41に記載の方法。
- 前記パクリタキセルが、C7−炭素に結合された酸素原子で、式(I)の繰り返し単位に結合されている、請求項41に記載の方法。
- 前記磁気共鳴造影剤にGd(III)化合物が含まれる、請求項38に記載の方法。
- 溶解または部分的に溶解されたポリマー反応体をカップリング剤の存在下で反応させる工程をさらに含む、請求項34から47のいずれか1項に記載の方法。
- 前記カップリング剤が以下:1−エチル−3−(3−ジメチルアミノプロピル)−カルボジイミド(EDC)、1,3−ジシクロヘキシルカルボジイミド(DCC)、1,1’−カルボニル−ジイミダゾール(CDI)、N,N’−ジスクシンイミジルカルボネート(DSC)、N−[(ジメチルアミノ)−1H−1,2,3−トリアゾロ−[4,5−b]ピリジン−1−イル−メチレン]−N−メチルメタンアミニウムヘキサフルオロホスフェートN−オキサイド(HATU)、2−[(1H−ベンゾトリアゾール−1−イル)−1,1,3,3−テトラメチルアミニウムヘキサフルオロホスフェート(HBTU)、2−[(6−クロロ−1H−ベンゾトリアゾール−1−イル)−1,1,3,3−テトラメチルアミニウムヘキサフルオロホスフェート(HCTU)、ベンゾトリアゾール−1−イル−オキシ−トリス−ピロリジノ−ホスホニウムヘキサフルオロホスフェート、ブロモ−トリス−ピロリジノ−ホスホニウムヘキサフルオロホスフェート、2−[(1H−ベンゾトリアゾール−1−イル)−1,1,3,3−テトラメチルアミニウムテトラフルオロボレート(TBTU)、およびベンゾトリアゾール−1−イル−オキシ−トリス−(ジメチルアミノ)ホスホニウムヘキサフルオロホスフェート(BOP);
