Claims (28)
- (a) 外因性TGFβスーパーファミリー増殖因子の存在下で多能性ヒト細胞を培養するステップ、
(b) ステップ(a)により得られた細胞を、外因性FGF−ファミリー増殖因子の存在下で培養するステップ、及び
(c) ステップ(b)により得られた細胞をレチノイドの存在下培養し、多能性PDX1−陽性内胚葉細胞を産生するステップ、
を含む、PDX1陽性内胚葉細胞を産生する方法。 - 前記TGFβスーパーファミリー増殖因子がアクチビンAを含む、請求項1に記載の方法。
- 前記FGF−ファミリー増殖因子がFGF−7を含む、請求項1又は請求項2に記載の方法。
- 前記レチノイドがレチノイン酸(RA)である、請求項1〜3の何れか1項に記載の方法。
- RAが0.01μMから約50μMの範囲の濃度で供給される、請求項4に記載の方法。
- RAが0.1μMから約10μMの範囲の濃度で供給される、請求項4に記載の方法。
- 外因性TGFβスーパーファミリー増殖因子が、ステップ(b)に先立ち、取り除かれる、請求項1〜6の何れか1項に記載の方法。
- 前記細胞集団の少なくとも25%がPDX1を発現する、請求項1〜7の何れか1項に記載の方法。
- 前記PDX1陽性内胚葉細胞中において定量的ポリメラーゼ連鎖反応により検出されたとき、PDX1の発現が、アルファ−フェトプロテイン(AFP)、SOX7、SOX1、ZIC1および、NFMから成る群から選択されるマーカーの発現より大きい、前記請求
項1〜8の何れか1項に記載の方法。 - PDX1の発現が免疫細胞化学により検知される、請求項1〜9の何れか1項に記載の方法。
- ヒト細胞を含むin vitroの細胞培養物であって、
前記ヒト細胞の少なくとも10%が
HNF6、SOX9及びPROX1から成る群から選択される1種のマーカーを発現し、膵臓−十二指腸ホメオボックス因子−1(PDX1)タンパクを発現する、内胚葉細胞である細胞培養物。 - 前記ヒト細胞の少なくとも25%がHNF6、SOX9及びPROX1から成る群から選択される1種のマーカーを発現し、膵臓−十二指腸ホメオボックス因子−1(PDX1)タンパクを発現する内胚葉細胞である、請求項11に記載の細胞培養物。
- 前記内胚葉細胞がHNF6、SOX9及びPROX1マーカーを発現する、請求項11又は請求項12に記載の細胞培養物。
- 前記内胚葉細胞が実質的にNKX6.1を発現しない、請求項11〜13の何れか1項に記載の細胞培養物。
- 更にレチノイドを含む、請求項11〜14の何れか1項に記載の細胞培養物。
- 前記レチノイドがレチノイン酸(RA)を含む請求項15に記載の細胞培養物。
- 前記ヒト細胞が非組み換え体である請求項11〜16の何れか1項に記載の細胞培養物。
- 前記細胞培養物が臓側内胚葉細胞、壁側内胚葉細胞、及び神経細胞からなる群から選択される細胞を実質的に含まない、請求項11〜17の何れか1項に記載の細胞培養物。
- 前記内胚葉細胞が前腸から派生した細胞に分化する能力を更に持つ多能性細胞である請求項11〜18の何れか1項に記載の細胞培養物。
- ヒト細胞を含むin vitroでの細胞培養物であって、前記ヒト細胞の少なくとも10%が、SOX17、HNF1β、およびFOXA1からなる群から選択されるマーカーを発現する、PDX1−陰性前腸内胚葉細胞である細胞培養物。
- 前記ヒト細胞の少なくとも25%が、SOX17、HNF1βおよびFOXA1からなる群から選択されるマーカーを発現する、PDX1陰性前腸内胚葉細胞である請求項20に記載の細胞培養物。
- 前記PDX1陰性前腸内胚葉細胞がSOX17、HNF1β、およびFOXA1マーカーを発現する、請求項20又は21に記載の細胞培養物。
- 前記PDX1陰性前腸内胚葉細胞が実質的にCERを発現しない、請求項20〜22の何れか1項に記載の細胞培養物。
- 更にFGFファミリー増殖因子を含む請求項20〜23の何れか1項に記載の細胞培養物。
- 前記FGFファミリー増殖因子がFGF−7を含む、請求項24に記載の細胞培養物。
- 前記ヒト細胞が非組み換え体である請求項20〜25の何れか1項に記載の細胞培養物。
- 前記細胞培養物が実質的に、壁側内胚葉細胞、臓側内胚葉細胞、神経細胞からなる群から選択される細胞を含まない、請求項20〜26の何れか1項に記載の細胞培養物。
- 前記PDX1陰性前腸内胚葉細胞が、前腸から派生した細胞に分化する能力を更に持つ多能性細胞である、請求項20〜27の何れか1項に記載の細胞培養物。
