Claims (18)
- PDX1陽性内胚葉細胞を作製する方法であって、
外因性TGFβスーパーファミリー増殖因子の存在下で、多分化能ヒト細胞の集団を培養する工程;
該細胞集団から外因性TGFβスーパーファミリー増殖因子を取り除く工程;および
レチノイドの存在下で該細胞集団を連続培養し、多分化能PDX1陽性内胚葉細胞を作製する工程、
を包含する、方法。 - 前記細胞集団を連続培養する工程が、FGFファミリー増殖因子を該細胞集団に供給する工程を包含する、請求項1に記載の方法。
- 前記FGFファミリー増殖因子は、前記レチノイドとほぼ同時に供給される請求項2に記載の方法。
- 前記FGFファミリー増殖因子がFGF−10を含む、請求項2または3に記載の方法。
- 前記細胞集団の少なくとも25%がPDX1を発現する、請求項1〜4のいずれか1項に記載の方法。
- 定量ポリメラーゼ連鎖反応によって決定される場合、前記PDX1陽性内胚葉細胞において、PDX1の発現が、α−フェトプロテイン(AFP)、SOX7、SOX1、ZIC1およびNFMからなる群から選択されるマーカーの発現より高い請求項1〜5のいずれか1項に記載の方法。
- 前記PDX1の発現が、免疫細胞化学により決定される請求項1〜6のいずれか1項に記載の方法。
- 前記レチノイドは、レチノイン酸(RA)である請求項1〜7のいずれか1項に記載の方法。
- RAは、0.01μMから約50μMまでの範囲の濃度で供給される請求項8に記載の方法。
- RAは、0.1μMから約10μMまでの範囲の濃度で供給される請求項8に記載の方法。
- 前記外因性TGFβスーパーファミリー増殖因子がアクチビンAを含む、請求項1〜10のいずれか1項に記載の方法。
- ヒト細胞の少なくとも約10%がPDX1陽性内胚葉細胞である、ヒト細胞を含むインビトロ細胞培養物。
- 前記ヒト細胞の少なくとも約25%が、PDX1陽性内胚葉細胞である請
求項12に記載の細胞培養物。 - レチノイドをさらに含む請求項12または13に記載の細胞培養物。
- 前記レチノイドがレチノイン酸(RA)を含む、請求項14に記載の細胞培養物。
