化合物2.1〜2.6の治療薬および皮膚保護特性の特徴づけを可能にするために、皮膚モデルにおけるさまざまなインビトロ試験(再構成ヒト表皮、会社:スキンエチック(SkinEthic))を実施した。
細胞培養培地におけるLDHの出現は、細胞の細胞質膜の損傷とともに上皮細胞層の損傷の確実な徴候である。さらに、この酵素の発現が細胞にとって「引き返し限界点(point of no return)」であること、すなわち損傷の不可逆性を示すことが周知である。
硫酸ドデシルナトリウム(SDS)は、細胞膜を攻撃し、かつLDH放出の上昇をもたらすことが周知である。次に記載した実験により、どの程度まで化合物2.1がSDSによる損傷後に細胞を保護しうるかを検査するのが適切である。
皮膚モデルに40分、SDSで損傷を与えた。次いで、試験製剤として、1または4%の化合物2.1を有するO/Wクリームを塗布した。LDH放出を試験製剤の投与後24時間および48時間に測定した。

IL1−αは、炎症反応において体内で中心的な役割を果たす伝達物質(Botenstoff)である。硫酸ドデシルナトリウム(SDS)は、モデル刺激物質として対象試験で使用すると、ヒトにおいて刺激性の接触性皮膚炎を引き起こしうるとともに、とりわけIL−1αの放出を誘発する皮膚刺激性の界面活性剤である。IL−1α濃度の測定は、市販の試験キット(ヒトIL−1αイムノアッセイ、R&Dシステムズ有限責任会社(Systems GmbH)、独国ヴィースバーデン(Wiesbaden))で行われた。
本発明による化合物の炎症抑制効果が香粧品製剤の使用に際しても示されるかどうか検査するのが適切である。そのために皮膚モデルにSDSで損傷を与えた。次いで、試験製剤を投与した。投与後24時間および48時間にIL−1α濃度を測定した。
