本发明涉及两性离子化合物及其制备,其可以通过短链二烷基氨基烷基酰胺的羧烷基化得到。
背景技术
很早就已经知道通过脂肪酸的二甲基氨基丙基酰胺的羧甲基化得到的甘氨酸盐化合物。它们以名称“酰氨基丙基甜菜碱”广泛地作为表面活性剂(参见例如,Parfümerie und Kosmetik,1996,77(4),244-248)。
这类物质的已知化合物例如衍生自椰子油脂肪酸、氢化椰子油脂肪酸或椰子油脂肪酸馏分(例如椰油酰氨基丙基甜菜碱)。它们广泛地例如用作化妆制剂(例如,淋浴凝胶、洗发水、液体肥皂)和餐具洗涤剂中的辅助表面活性剂。
已知的酰氨基丙基甜菜碱的特征是它们的表面活性行为,即降低水性体系的表面张力的性质。这种性质是分子结构引起的,即,一方面,由疏水的、长链脂肪酸基团引起,另一方面,由亲水的、两性离子二甲基甘氨酸铵基团引起。目前的脂肪酸基团尤其包括天然脂肪中的那些,尤其是椰子油脂肪或氢化椰子油脂肪,即,饱和及不饱和的C8-C18脂肪酸。己酸(C6)的酰氨基丙基甜菜碱也已被公开。
许多两性离子物质,例如,胺氧化物、脯氨酸、ectoin或三甲基甘氨酸,被称作cosmotropes,即它们加强了水性体系的结构或秩序。这些物质中的许多物质已知是渗透溶质(osmolytes),即它们控制细胞的水含量,从而具有稳定作用。
许多表面活性剂的不利影响是刺激皮肤和眼睛。虽然酰氨基丙基二甲基甘氨酸盐的刺激低于例如烷基硫酸盐或烷基醚硫酸盐,但是它们也不是完全没有可能的刺激效果。
发明内容
因此,本发明的目的是提供新的化合物,尽管其具有两性离子的、亲水性的头部基团,但是没有现有技术中表面活性剂那样的细胞破坏作用,因而没有表面活性结构,因此有利地不刺激皮肤和眼睛。更有利地,可以使用允许大规模生产的方法制备这些两性离子化合物。
出人意料地已经发现,与已知的上述基于长链脂肪酸的酰氨基丙基二甲基甘氨酸盐相比,根据本发明的化合物没有细胞破坏作用,并且是非刺激性的,这大概因为缺乏表面活性性质。
特别出人意料地已经发现,本发明的化合物在皮肤模型中产生抗炎效果。
因此,本发明提供了如权利要求1~9所述的两性离子化合物,它们如权利要求10-12所述用作各种治疗剂,以及权利要求13所述的其制备方法。
本发明的化合物的进一步优点在于,它们比常规使用的化合物更低的分子量,因而可以实现较高浓度的离子和/或两性离子结构。
本发明的方法的一个优点在于,可以制备高浓度的本发明化合物,而没有凝胶形成,从而没有粘度问题。

在本发明式I的化合物的更优选实施方案中,m=1和X=H。特别优选的化合物是其中n=3,m=1,R1=R2=CH3,Y=CH2和X=H的式I的实施方案。
本发明还提供由活性量的至少一种本发明的化合物构成的药物,以及本发明的化合物作为药物的用途。
如实施例3.1~3.4所示,本发明的化合物对重组人表皮同时具有细胞保护作用和抗炎作用。因此本发明提供用作治疗活性成分的本发明的化合物。
由于细胞保护和抗凋亡作用特别是可以在皮肤细胞上得到极好的证实,因此,本发明的化合物的优选用途是用于治疗皮肤疾病,特别优选用于治疗炎症性皮肤疾病。

本发明的药物可以本领域技术人员已知的任何给药途径给予,例如,鼻内、静脉内、肌肉内、经皮和局部,在皮肤疾病的情况下,优选经皮和局部给药。皮肤疾病的治疗可以常规方式进行,例如借助于绷带、膏药、敷布、可复水的冻干物,含有本发明化合物的膏药或粘贴或凝胶,但是本发明的化合物也可以脂质体复合物或金颗粒复合物的形式在病变皮肤区经皮和局部给药。其它可以采用的经皮给药的方式包括例如以下形式:溶液、乳液、霜剂、药膏、泡沫、气雾剂、凝胶基质、作为可复水的冻干物、膏药或粘贴的绵球、、滴剂或洗剂。
根据本发明的式I的化合物例如可以使用下述本发明的方法制备。

可以在步骤A中使用的羧酸均是一元、二元或多元羧酸,或满足对式II限定的条件的它们的混合物。为制备其中m=2的式I的二两性离子的化合物,在步骤A中使用的羧酸优选是草酸(HOOC-COOH)、丙醇二酸(HOOC-CH2(OH)-COOH)、苹果酸(HOOC-CH2(OH)-CH2-COOH)和酒石酸(HOOC-CH2(OH)-CH2(OH)-COOH),特别优选丙二酸(HOOC-CH2-COOH)。在步骤A中使用的制备本发明优选m=1的式I物质的优选羧酸是乳酸、丙酸和乙醇酸,特别优选甲酸(HCOOH)。
可以使用的胺组分是满足式III条件的任何适当的胺类化合物。优选地,使用3-(二乙基氨基)丙胺、2-(二乙基氨基)乙胺或2-(二甲基氨基)乙胺。特别优选的胺组分是二甲基氨基丙胺(DMAPA)。
优选地,根据本发明方法的步骤A中,式II的酸组分与式III的胺组分在90℃~220℃的温度下反应,特别优选在约180℃的温度下,得到式IV的酰氨基胺。根据本发明方法的步骤A特别优选使用适合的催化剂进行。适合使用的催化剂优选是强碱性催化剂,例如,醇盐,特别优选乙醇钠、乙醇钾、甲醇钠和甲醇钾。
反应中形成的水可以从产物中除去。优选在反应条件下蒸馏掉水,因而从产物混合物中除去。特别是在低于约130℃的温度下,有利的是使用低于大气压的压力,以提高通过蒸馏除水的速率。
下面的反应方案显示步骤A的一种可能的反应过程:
由于步骤A中形成的盐混合物在反应开始时是固体,因此,在本发明的方法中式II的酸组分优选加到最初加入的式III的胺组分中,这种顺序与现有技术相反。
在式IV的短链酰氨胺制备中,应该考虑式III的胺和式II的酸之间的盐形成过程中与现有技术已知的长链脂肪酸的酰氨基胺相比大大增加的放热,这是由于低分子量和由此的较高定量浓度造成的。在这方面,在本发明方法的优选实施方案中,可以使用在步骤A中特定适用的工艺参数,使得羧酸组分缓慢加到胺组分中,这样在加入过程中反应混合物的温度不超过130℃,优选不超过100℃。在较高的温度时,形成的水可能除去大量胺组分,这对所用组分的化学计量学有负面影响。为保持指定的温度范围,优选进行逆流冷却,以实现经济有用的定量比。
步骤B可以在适合的溶剂存在下进行,溶剂的量确保反应混合物在工艺中的任何时间点均具有可搅拌性和可抽吸性。优选地,反应在水作为溶剂存在下进行。步骤B优选在约70~100℃的温度下进行。作为副产物制得的卤化物Z可以从反应液中除去,或留在其中。如果要除去卤化物,那么,例如,可以用适合的溶剂沉淀或渗析。优选的沉淀溶剂是乙醇。
在本发明方法的优选实施方案中,卤化物Z留在溶液中。
可以使用的单卤代羧酸或式V酸根的单卤代羧酸盐均可以是酸根满足式V的条件的卤代羧酸。特别优选的式V的单卤代羧酸盐是单氯乙酸盐。
如步骤A中所述,在步骤B中,也应该考虑由短链酰氨基胺组分的低摩尔质量所造成的与现有技术的方法相比大大增加的放热。因此,在本发明方法的步骤B中,反应优选进行如下:在将卤代羧酸组分加到酰氨基胺组分过程中或完全加入之后,直到反应放热衰减时,反应温度保持在最高约70℃,如果需要进行逆流冷却。随后的反应优选在略低于溶剂沸点下进行,如果使用水作溶剂,优选使用的温度范围为95-99℃。
下面的反应方案显示步骤B的一种可能的反应过程。
式IV的酰氨基胺优选在溶剂中反应得到式I的相应的化合物。酰氨基胺的优选使用浓度为3~75%,优选5~50%。该步骤中形成的式I化合物的溶液可以进行进一步的浓缩或脱盐步骤,或不进行,例如为制备化妆制剂。在本发明方法的更优选实施方案中,通过使用适合溶剂使卤化物组分沉淀或通过渗析使本发明的式I的化合物脱盐,或者通过蒸馏所有或部分溶剂进行浓缩。优选地,不经过进一步处理步骤使用步骤B后形成的溶液。
在本发明方法的优选实施方案中,在步骤B中与卤代羧酸组分反应之前,除去步骤A中过量使用的胺II。例如,可以通过减压蒸馏进行。

在带有搅拌器、温度计、回流冷凝器和滴液漏斗的500ml四颈烧瓶中,称量加入91g单氯乙酸钠和191g水,并加热到40℃。加入100g实施例1.1的酰氨基胺,反应混合物在70℃的温度下保持,直到反应热量衰减。然后将混合物加热到98℃。约7h后,剩余酰氨基胺的量低于0.5%。
获得以下组成的382g水溶液:
化合物2.1: 38.1%
氯化钠: 11.9%
水: 50%
外观: 液体,透明
实施例2.2:制备化合物2.2
在带有搅拌器、温度计、回流冷凝器和滴液漏斗的500ml四颈烧瓶中,称量加入70g单氯乙酸钠和170g水,并加热到40℃。加入100g实施例1.2的酰氨基胺。然后将反应混合物加热到70℃,并保持该温度,直到反应热量衰减。然后将混合物加热到98℃。约7h后,剩余酰氨基胺的量低于0.5%。
获得以下组成的340g水溶液:
化合物2.2: 39.7%
氯化钠: 10.3%
水: 50%
外观: 液体,透明
实施例2.3:制备化合物2.3
在带有搅拌器、温度计、回流冷凝器和滴液漏斗的1000ml四颈烧瓶中,称量加入153g单氯乙酸钠和353g水,并加热到40℃。加入200g实施例1.3的酰氨基胺。然后将反应混合物加热到70℃,并保持该温度,直到反应热量衰减。然后将混合物加热到98℃。约7h后,剩余酰氨基胺的量低于0.5%。
获得以下组成的706g水溶液:
化合物2.3: 39.2%
氯化钠: 10.8%
水: 50%
外观: 液体,透明
实施例2.4:制备化合物2.4
在带有搅拌器、温度计、回流冷凝器和滴液漏斗的500ml四颈烧瓶中,称量加入107g单氯乙酸钠和207g水,并加热到40℃。加入100g实施例1.4的酰氨基胺。然后将反应混合物加热到70℃,并保持该温度,直到反应热量衰减。然后将混合物加热到98℃。约7h后,剩余酰氨基胺的量低于0.5%。
获得以下组成的414g水溶液:
化合物2.4: 35.9%
氯化钠: 14.1%
水: 50%

实施例2.5:制备化合物2.5
在带有搅拌器、温度计、回流冷凝器和滴液漏斗的500ml四颈烧瓶中,称量加入85g单氯乙酸钠和185g水,并加热到40℃。加入100g实施例1.5的酰氨基胺。然后将反应混合物加热到70℃,并保持该温度,直到反应热量衰减。然后将混合物加热到98℃。约7h后,剩余酰氨基胺的量低于0.5%。
获得以下组成的370g水溶液:
化合物2.5: 38.5%
氯化钠: 11.5%

实施例2.6:制备化合物2.6
在带有搅拌器、温度计、回流冷凝器和滴液漏斗的1000ml四颈烧瓶中,称量加115g单氯乙酸钠和265g水,并加热到40℃。加入150g实施例1.6的酰氨基胺。然后将反应混合物加热到70℃,并保持该温度,直到反应热量衰减。然后将混合物加热到98℃。约7h后,剩余酰氨基胺的量低于0.5%。
获得以下组成的530g水溶液:
化合物2.6: 38.5%
氯化钠: 11.5%

本发明化合物的有效性实施例
在为了能够表征化合物2.1~2.6的治疗和护肤性能,对皮肤模型(重组人表皮,公司:SkinEthic)进行各种体外试验。
实施例3.1 乳酸脱氢酶释放(LDH释放)
细胞培养基中LDH的出现是对细胞的细胞质膜造成损害、从而损害表皮细胞层的可靠迹象。此外,据了解,这种酶的流出代表细胞的“不归点”,即表明损害的不可逆转性。
使用市售检测试剂盒(LDH检测试剂盒,Roche Diagnostics,Mannheim,Germany)测定LDH的浓度。
间隔24小时将试验制剂施用于皮肤模型两次。图1显示最后施用24h后的LDH释放。
试验制剂3.1:施用本发明化合物的4%强度的水溶液。由于两性离子的、非表面活性化合物中每1%的活性物质包括约0.3%氯化钠,因此还测试相应的氯化钠浓度。
由于施用了两性离子化合物,从细胞中释放的LDH保持不变,或者甚至略低于未经处理的皮肤模型。这意味着本发明的化合物不攻击细胞膜,并且没有细胞破坏作用,而是恰恰相反,甚至协助这些细胞和它们的增殖。
实施例3.2 用SDS损害细胞后的LDH释放:
众所周知的是十二烷基硫酸钠(SDS)攻击细胞膜,并导致LDH释放增加。使用下述实验,目的是要调查化合物2.1从多大程度上可以保护用SDS损害后的细胞。
用SDS损害皮肤模型40分钟。然后施用试验制剂,即含有1%或4%的化合物2.1的O/W型霜剂。施用试验制剂后24和48h测定LDH释放。
试验制剂3.2:
聚甘油基-3甲基葡萄糖二硬脂酸酯 3.0%
硬脂酸甘油酯 2.0%
硬脂醇 1.0%
鲸蜡硬脂基乙基己酸酯(Cetearyl ethylhexanoate) 5.0%
矿物油 14.0%
化合物2.1 1.0/4.0%
水 加至100.0%
图2显示24和48h后LDH的总浓度。
由于SDS造成的损害,如所预期的那样,LDH释放大大增加。当在损害后直接施用试验制剂时,这种增加明显降低。即使在载体的情况下,积极影响也是显而易见的,但是当制剂含有化合物2.1时,再次明显增加。在这方面,只有1%的本发明化合物似乎就是足够的,因为4%在效果方面没有显著增加。
实施例3.3 SDS损害后的IL-1α释放:
IL-1α是一种在人体内的炎症反应中起核心作用的信使。十二烷基硫酸钠(SDS)是在人体中引起刺激性接触性皮炎的皮肤刺激性表面活性剂,是用作主题研究中的刺激模型,并且此外,诱导IL-1α的释放。使用市售检测试剂盒(人类IL-1α免疫分析,R&D Systems GmbH,Wiesbaden,Germany)测定IL-1α浓度。
将试验制剂(短链、两性离子化合物的4%强度的水溶液)施用于皮肤模型。施用24h后,使用0.25%强度的SDS溶液产生损害40分钟。然后再次施用试验制剂。在24h培养时间后,测定释放的细胞因子IL-1α。
由于测试溶液中每1%的活性物质包括约0.3%的NaCl,因此还分析了适当浓缩的氯化钠溶液。
图3显示损害后24hIL-1α浓度的测量值。
所有测试的两性离子、非表面活性化合物均减少炎症标记I1-1α的释放,即可以认为,短链、两性离子化合物具有抗炎性能。
实施例3.4 含有化合物2.1的O/W型霜剂的抗炎效果:
目的是要调查以化妆品制剂施用本发明的化合物时是否也有抗炎效果。为此,用SDS损害皮肤模型。然后施用试验制剂。施用后24和48h,测定I1-1α浓度。
试验制剂:
聚甘油基-3甲基葡萄糖二硬脂酸酯 3.0%
硬脂酸甘油酯 2.0%
硬脂醇 1.0%
鲸蜡硬脂基乙基己酸酯 5.0%
矿物油 14.0%
化合物2.1 1.0/4.0%
水 加至100.0%
图4显示24和48h后总的IL-1α浓度。
如所预期的那样,由于SDS造成的损害,细胞因子IL-1α的形成大大增加。这种增加随着加入化合物2.1的浓度而相当大程度地降低,也就是说,以O/W型乳液施用本发明的化合物时,显著的抗炎效果是显而易见的。
Claims (13)
2.如权利要求1所述的化合物,其特征在于,m=2且X是直接共价键、CH2、CH(OH)、CH2CH(OH)或CH(OH)CH(OH)。
3.如权利要求2所述的化合物,其特征在于,X=CH2。
4.如权利要求1所述的化合物,其特征在于,m=1且X=H。
5.如权利要求1~4中至少一项所述的化合物,其特征在于,n=3。
6.如权利要求1~5中至少一项所述的化合物,其特征在于,R1=R2=CH3。
7.如权利要求1~6中至少一项所述的化合物,其特征在于,Y=CH2。
8.如权利要求1所述的化合物,其特征在于,n=3,m=2,R1=R2=CH3,Y=CH2且X=CH2。
9.如权利要求1所述的化合物,其特征在于,n=3,m=1,R1=R2=CH3,Y=CH2且X=H。
10.如权利要求1~9中至少一项所述的化合物作为治疗用活性成分的用途。
11.如权利要求1~10中至少一项所述的化合物在治疗皮肤疾病中的用途。
12.如权利要求1~11中至少一项所述的化合物在治疗炎症性皮肤疾病中的用途。
13.制备如权利要求1~12中至少一项所述的化合物的方法,其特征在于,包括以下步骤,
A:使式II的羧酸与式III的胺反应,得到式IV的酰氨基胺,
其中
n=1~6,
m=1~4,
R1和R2彼此独立地为相同或不同的具有1~6个碳原子的脂肪族烃基,
X是m-价基团或共价键,
其中,
当m=1时,X是H、乙基、羟甲基、1-羟乙基、2-羟乙基、1-羟丙基、2-羟丙基或3-羟丙基,
当m=2时,X是直接键或未取代或被至少一个OH基团取代的2-价C1-~C5-烃基,
当m>2时,X是未取代或被至少一个OH基团取代的m-价C1-~C5-烃基,
B:将得自A的式IV的酰氨基胺与具有式V的酸根的ω-卤代羧酸或其盐反应,得到式I的化合物,
其中z=卤素,以及
Y是二价烃基。
