本発明は、単離された人工抗原提示細胞(aAPC)を包含し、前記aAPCはレンチウイルスベクター(LV)を使用して形質導入されたK562細胞を含んでなり、前記LVは最低1種の免疫刺激リガンドおよび最低1種の共刺激リガンドをコードする核酸を含んでなり、かつ、さらに、前記aAPCは前記刺激リガンドおよび前記共刺激リガンドを発現しそして前記aAPCと接触させたT細胞を刺激しかつ増殖させ得る。
本発明は、レンチウイルスベクターを使用して、多数のT細胞刺激および共刺激リガンド、ならびにそれらに対する抗体、ならびに、とりわけ抗原、サイトカインを安定に発現するaAPCを効率的に製造し得るという驚くべき発見に関する。本発明はまた、製造される新規aAPC、ならびに所望のT細胞を増殖させ、特定のT細胞サブセットを活性化しかつ/若しくは増殖させ、特定のT細胞サブセットの増殖を促進し得る刺激分子、共刺激分子およびそれらの組合せを同定するためのそれらの使用方法、ならびに該新規aAPCを使用するT細胞の増殖および刺激に関する多数の治療的用途にも関する。
本明細書で使用されるところの以下の用語のそれぞれは、この節でそれと関連した意味するところを有する。
本発明は、MHCクラスI若しくはクラスII分子を発現せずかつ遺伝子操作技術に反応しないと以前は考えられていたヒト赤白血病細胞株K562が、限定されるものでないが、刺激リガンド、共刺激リガンド、抗原(例えば腫瘍、ウイルスなど)、サイトカインなどを挙げることができる多数の分子を発現するように、レンチウイルスベクターを使用して容易に形質導入し得るという驚くべき発見に関する。
本発明は、単離された人工抗原提示細胞(aAPC)を包含し、該細胞はレンチウイルスベクター(LV)を使用して形質導入されたK562細胞を含んでなる。さらに、該LVは最低1種の免疫刺激および共刺激リガンドをコードする。本明細書に開示されるデータは、K562細胞に形質導入された約9種の異なる分子をコードする約9種の核酸が、長期培養において安定かつ高度に発現されたことを示す一方、これがこれらの細胞中に導入され得る分子の数若しくは種類の制限であることを示唆するものは何も存在しない。代わりに、いかなる分子若しくはリガンドも、刺激、共刺激、サイトカイン、抗原、Fcγ受容体などであろうと、これらの細胞中に導入して本発明のaAPCを生じさせ得る。

本発明は、既知の共刺激分子を発現するT細胞の増殖の特異的誘導方法を包含する。該方法は、増殖されるべきであるT細胞を、その共刺激分子と特異的に結合するリガンドをコードするレンチウイルスベクターを含んでなるaAPCと接触させることを含んでなる。本明細書の別の場所に示されるとおり、とりわけT細胞表面上で発現されるコグネイトの共刺激分子を特異的に結合する共刺激リガンドを含んでなるK562に基づくaAPCとT細胞を接触させることは、T細胞を刺激し、そして、多数の特異的T細胞が容易に産生され得るようなT細胞増殖を誘導する。aAPCは、特定の共刺激分子を発現するT細胞のみが該aAPCにより増殖されるために、「特異的に」T細胞を増殖させる。従って、発現されるべきT細胞が、そのいくつか若しくは大部分が共刺激分子を発現しない細胞の混合物中に存在する場合は、目的のT細胞のみを誘導して増殖させかつ細胞数を大きくすることができる。T細胞は、当該技術分野で既知かつ/若しくは本明細書の別の場所に記述されるもののような多様な細胞分離および精製技術を使用してさらに精製し得る。
本発明は、本発明のaAPC、多様なタンパク質をコードする核酸、T細胞の表面上の共刺激分子に特異的に結合する抗体、および/または本発明の抗体、抗原若しくはサイトカインをコードする核酸、アプリケーター、ならびに本発明の方法を実施するためのキットの使用を記述する説明資料を含んでなる多様なキットを包含する。例示的キットを下述するとは言え、他の有用なキットの内容は本開示に照らして当業者に明らかであろう。これらのキットのそれぞれが本発明内に包含される。
実施例
CD4およびCD8 T細胞の固有の増殖要件は異なることが示されている(Deethsら、1997、Eur.J.Immunol.27:598−608;Lauxら、2000、Clin.Immunol.96:187−197;Fouldsら、2002、J.Immunol.168:1528−1532)。CD8細胞の長期増殖のためのCD28に加えて多様な共刺激分子の発現の遺伝子操作を可能にするように、細胞に基づくaAPCを設計した。該培養系は、共刺激シグナルが、CD28により提供されるものに加えて最適なCD8細胞の増殖に必要とされるという事実に基づいた。ヒト赤白血病CML細胞株K562(Lozzioら、1975、Blood 45:321−334)は、同種応答を促進するとみられるHLAタンパク質を発現しないため、この細胞株を細胞性aAPCの足場構造として使用した。しかしながら、K562はICAM(CD54)およびLFA−3(CD58)を発現し、それらの双方はT細胞との相互作用を促進する(図1)。K562細胞を使用することの他の利点は、限定されるものでないが、照射したK562細胞が臨床の設定に導入され得るという事実を挙げることができる。これらの細胞はマイコプラスマを含まず、血清を含まない培地中で増殖し、そしてナチュラルキラー(NK)細胞により容易に死滅されるからである。こうしたaAPCを産生するためにK562を使用することの望ましさにもかかわらず、K562細胞は形質導入するのが有名に困難であった(Kahlら、2004、J.Virol.78:1421)。驚くべきことに、本明細書に開示されるデータは、K562細胞が、無数の外因性核酸で連続してかつ/若しくは同時にのいずれでも形質導入されて多数の分子を発現し、それにより所望の表現型をもつaAPCのライブラリーを得ることができることを初めて示す。こうしたLVに基づく形質導入は、連続してかつ/若しくは同時に実施され、そしてMoFloを使用して所望の表現型を示す細胞がクローン化される。さらに、該データは、最適のプロモーターを選択し得、そして、プロモーターの競合を評価し得かつ必要若しくは所望の場合は排除し得ることを示す。本発明の方法を使用して製造されるaAPCのライブラリーをその後、限定されるものでないがNOD/SCIDマウスモデルを挙げることができる、技術に認識されたモデルを使用して、生物学的機能についてin vivoで評価する。
K562細胞に遺伝子を導入するための以前に記述されたトランスフェクションに基づくアプローチの制限を回避するため、一連の高力価レンチウイルスベクターを、本明細書の別の場所に開示されるとおり使用して、K562細胞中に多数の共刺激リガンドおよびMHC分子を安定に導入した。これは、多様なaAPCの系統的かつ迅速な産生を見込み、また、HIV特異的T細胞に対する最適の増殖およびエフェクター機能を生じる共刺激分子の組合せの決定を見込む。本明細書に開示されるデータはこのアプローチを示す。
K562細胞は、末期の急性転化の慢性骨髄性白血病を伴う患者から単離した(Lozzioら、1975、Blood、45:321−334)。K562はMHC分子若しくはT細胞共刺激リガンドを発現しないDC前駆細胞を表しうるが、しかし、限定されるものでないがサイトカイン産生、接着分子発現およびマクロ飲作用を挙げることができる、DCを効果的なAPCにする多くの他の属性を保持する。単球細胞株U−937は効果的なaAPCとして機能することが不可能であったため、これらの属性はK562細胞に独特でありうる。従って、K562細胞は、その上に所望のMHC分子および共刺激リガンドを導入してCD様aAPCを樹立し得る理想的な足場構造を表す。こうしたaAPCは、高レベルのMHC発現、多数の共刺激リガンド、およびT細胞とのサイトカインのクロストークに携わる能力を包含するDCの全部の利点を有する。K562に基づくaAPCはまた、それらの制限された寿命、複製能力の欠如、および明確に定義されていない成熟要件のようなDCの欠点も欠く(Leeら、2002、Vaccine 20:A8−A22)。
aAPCを製造するためのK562細胞の形質導入
本明細書に開示されるデータは、K562細胞がDCの強力なT細胞刺激能力を良好に模倣することを可能にする、共刺激リガンドのレンチウイルスに媒介される導入を介するK562 aAPCの創製を示す。
本発明は、治療的使用のためのT細胞のin vivo治療的ワクチン接種およびex vivo増殖のためのLVで工作したK562 aAPCを包含する。抗原、サイトカインおよび/若しくは共刺激分子を、同一若しくは別個のプロモーター/制御配列の制御下にK562細胞に形質導入し得る。さらに、腫瘍細胞抗原をコードする核酸を細胞中に形質導入し得るか、若しくは、細胞が適切なエピトープをプロセシングしかつMHCタンパク質の情況で提示するような細胞に別の方法で抗原を負荷し得る。これは、K562細胞が、必要とされる特異的抗原若しくはエピトープを最初に同定若しくは単離する必要性を伴わずに抗原をプロセシングかつ提示する能力を有することが、本明細書の別の場所で示されているからである。従って、細胞抽出物(腫瘍細胞の最低1種の膜成分を含んでなる)をK562に基づくaAPCに負荷し得、そして、関連する抗原をプロセシングかつ提示する該細胞の天然の能力を活用する。カスタマイズしたaAPCを本明細書に示す一方、これらは具体的説明の目的上のみであり、そして本発明は表3に示されるこれらの態様に制限されない。これは、本明細書の別の場所に提供される教示を備えた当業者により認識されるであろうとおり、無数の分子を事実上無制限の組合せでaAPCにより形質導入しかつ発現させ得るからである。
他のaAPCを、限定されるものでないが養子免疫療法および遺伝子治療を挙げることができるex vivoの使用のため製造し得る。こうしたex vivo使用のためのaAPCのカスタマイズしたバージョンのいくつかは、とりわけ下の表4に開示される構築物である。すなわち、被験体から単離したT細胞を、これら若しくは無数の他のaAPCを使用してin vitroで刺激かつ増殖させ得、その後、該T細胞を被験体に導入してそれによりそれに対する養子免疫療法を提供し得る。加えて、増殖させたT細胞を、遺伝子工作前若しくはその非存在下での該T細胞中での該タンパク質の発現に比較して、発現されなかったか若しくはより低レベルで発現された外因性タンパク質を発現するように遺伝子的に工作し得る。従って、本発明は、それの必要な被験体の自己移植に使用されるT細胞を増殖させるために本発明のaAPCを使用するex vivoの細胞に基づく養子免疫療法および遺伝子治療の双方を提供する。表4は単にこうした細胞/遺伝子治療に使用し得るaAPCのいくつかの具体的に説明する例を示すが、しかし、本発明は、本発明のこれらの例示的な「在庫があり入手可能な」aAPCに制限されない。
本明細書に開示されるデータは、CD4 T細胞の長期増殖が、外因性サイトカインの存在下でK32/CD3/28およびK32/86CD3 aAPCを使用して得られたが、しかしU937に基づくaAPCは有効でなかったことを示す(図5)。さらに、該データは、K562に基づくaAPC(例えばK32/CD3/28、K32/86/CD3)が、ビーズおよび細胞双方にCD3およびCD28を負荷する場合にCD4 T細胞の長期増殖をより効果的にビーズに基づくaAPC(CD3/28被覆ビーズ)を媒介することを示す。これらの結果は、ビーズに基づく系を使用して可能でない、検出可能な「クロストーク」がK562に基づくaAPCとT細胞の間に存在することを示す。図5に具体的に説明されるとおり、全部の腫瘍細胞株が人工APCとしてはたらく能力を有するわけではなく、また、該データは、予期されなかった、強力なAPCとしてはたらくK562細胞の能力をさらに示す。これらの驚くべき結果は、K562に基づくaAPCを使用して可能である、従来技術の方法を上回る有意の改良を裏付ける。
簡潔には、50,000の照射したKT32/4−1BBL/CD86を抗CD3 Abで被覆し、そして健康なドナーからの100,000の新たに単離したCD8 T細胞と混合した。10〜12日ごとに、CD8 T細胞を、新たに照射したKT32/4−1BBL/CD86 aAPCで再刺激した。細胞を廃棄しなかった場合に蓄積していたとみられる細胞の総数を、培養日数に対する総細胞数の半対数プロットとして描く(図9)。図9でまた具体的に説明されるとおり、CD32および4−1BBLで形質導入したaAPC(K32/4−1BBL)により増殖させたポリクローナルCD8 T細胞は、43日後に18,600倍増殖した。
抗原特異的CD8 T細胞の自己移入によるHIV特異的T細胞応答を増強するための試みは、HIV感染の長期抑制をもたらしていない(Tanら、1999、Blood 93:1506;Koenigら、1995、Nature Med.1:330−336;Brodieら、1999、Nature Med.5:34(欠落)、Riddellら、1996、Nature Med.2:216−223;Liebermanら、1997、Blood 90:2196−2206)。これらの細胞のin vivoで生存することの不能は、臨床上意味のある方法で抗HIV応答を測定するためのいかなる試みも不可能にした。若干の症例において、これらの細胞の早期の死亡は選択可能なマーカーの免疫認識として容易に説明された(Riddellら、1996、Nature Med.2:216−223)。他の症例においては、注入に際してのT細胞の死の理由はより少なく明確であった。これらの早期の研究は、HIV特異的ペプチドを末梢単核血液細胞(PMBC)若しくはリンパ芽球腫細胞株(LCL)に負荷すること、限界希釈を実施してクローンを単離すること、ならびに、高濃度の外因性IL−2および共刺激の非存在下でトリガーするTCRを使用してT細胞をex vivoで数か月間増殖させて1×109までのHIV特異的T細胞を生じさせることというわずかに異なる変形物を使用した。いずれかの特定の論理により拘束されることを願わないが、もしかしたら高濃度のIL−2への依存性と結合された長期のex vivo培養が、それらの宿主に一旦戻し注入されればこれらの細胞でのアポトーシスの開始につながったのかもしれない。
HLA−A2ドナーを、該集団中でのこの対立遺伝子の高頻度のため使用する。最初は、ウイルス表現型をHLA−A2ドナーの患者選択基準として使用しない。ナイーブなCD8 T細胞が活性化後にそれらの細胞表面上に少量のCD4を発現して、それらをHIV感染に感受性にすることが示されている(Yangら、1998、J.Exp.Med.187:1139−1144;Kitchenら、1998、J.Virol.72:9054−9060;Flamandら、1998、Proc.Natl.Acad.Sci.U.S.A 95:3111−3116;Imalchら、2001、J.Virol.75:11555−11564)。しかしながら、ナイーブなCD8 T細胞のみがこの可塑性を有するようであり、そして使用されたPol特異的T細胞は本質的にメモリーT細胞である。従って、いずれかの特定の論理に拘束されることを願わず、これらの細胞は感染せず、そしてex vivo増殖に際して感染した状態になることができない。
7例のHLA−A2乳癌患者を、PolペプチドILKEPVHGV(配列番号3)に対し、対照治療群としてhTERTペプチドワクチンに対し、ワクチン接種した(Vonderheide、2004、Clin.Cancer Res.10:828−839)。ワクチン接種したHLA−A2乳癌患者からの細胞を使用して、HIV陰性宿主からのPol特異的細胞を増殖させる。これらの患者からの凍結PMBCを使用してPol特異的T細胞を精製する。これらのPol特異的T細胞の頻度は、HIVに感染したドナーから期待される頻度より低く(およそ0.1%)、従って、より少数(しかしなお十分な数)のこれらの細胞が、実験を開始するために得られる。Pol特異的T細胞患者は、Polペプチドでワクチン接種していたいずれの患者からも得ることができる。
HIV感染患者からの大量のT細胞を増殖させる能力を、HIV特異的T細胞を増殖させるこれらのaAPCの能力を特徴付けする前に検査する。これは、特定の1T細胞サブセットが1種のaAPCにより他者を上回って優先的に増殖されるかどうかを決定するための増殖率の測定を見込む。加えて、IFN−γ分泌およびパーフォリン発現と結合された四量体染色の分析を使用して、特定の1aAPCが、flu、CMV、EBV若しくはHIV特異的CD8 T細胞を優先的に増殖しかつ/若しくはそれらに向上されたエフェクター機能を与えるかどうかを決定する。
治療レベルのHIV特異的CD8 T細胞を生成させるため、T細胞を約10,000と約1,000,000倍の間(約13〜20回の集団倍加)まで増殖させる(Riddellら、1995、Annu.Rev.Immunol.13:545−586)。HIV特異的CD8 T細胞がex vivo培養物中で過ごす時間の量と、これらの細胞がHIV感染患者に提供する潜在的な臨床上の利益の間に逆相関が存在する。従って、HIV感染患者からのCD8 T細胞を最も迅速に増殖させるaAPCを決定し、そして本明細書に開示される方法で使用する。
ex vivo増殖後のT細胞の健康状態は、in vivoで機能するそれらの能力の優れた予測因子である。これらのaAPCの重要な細胞生存遺伝子Bcl−xLを誘導する能力もまた測定する。増殖過程の間の培養物中のアポトーシス細胞の比率を使用して、細胞に基づくaAPCのいずれかが増殖されたT細胞に特定の生存の利点を与えるかどうかを決定する。加えて、ex vivo増殖後の細胞のテロメア長を測定して、特定の1aAPCが、それが増殖させる細胞の複製能力の保存においてより効果的であるかどうかを決定する。各染色体の端に、テロメアと呼ばれる多数のTTAGGGヌクレオチド反復配列が存在する。細胞が分割する度ごとにそれは損失そのテロメアの一部分。大部分の細胞は、染色体の端の該DNA反復配列のコピーを修復し得る酵素テロメラーゼを発現しないため、細胞がその遠隔測定(telemetric)長さの決定的な量を一旦喪失してしまえばそれは分割するその能力を喪失すると考えられている。テロメア長は、細胞が何回複製したかを計るため、および推論によりその将来の複製能力を評価するための一方法として、当該技術分野で使用されている(Palmerら、1997、J.Exp.Med.185:1381−1386;Wengら、1997、J.Immunol.158:3215−3220)。しかしながら、Tリンパ球はテロメラーゼ活性を誘導し得る数種の細胞型の1つであり(Wengら、1996、J.Exp.Med.183:2471−2479)、そして従って異なる方法を使用して増殖させたT細胞間の相対的差違は、T細胞の有糸分裂事象の回数ならびにテロメラーゼが誘導された程度の双方を反映する。最も高い複製能力を有する細胞を注入することは、養子移入したHIV特異的T細胞がHIV感染を長期間制御することを確実にするために最も重要である。
サイトカインは重要なエフェクター分子であり、そしてT細胞分化への洞察を提供する。HIVに感染した個体由来のCD8 T細胞からの以下のサイトカインすなわちIL−2(ex vivo増殖のための重要なT細胞増殖因子であり、そして、IL−2を誘導する細胞の能力はその長期増殖能力と良好に相関する);IL−4(TH2分化のマーカー);およびIL−10(制御性T細胞の増殖の代理となりうる免疫抑制性サイトカイン)を誘導するaAPCのそれぞれの能力を定量する。HIV感染患者には、低レベルのIL−10を産生するようにT細胞を誘導するaAPCが好ましい。他のサイトカインは、限定されるものでないがTGF−β(IL−10と同一の論理的根拠のため);IFN−γ(TH1分化のマーカーかつ重要なエフェクターサイトカイン);およびTNFα(重要なエフェクターサイトカイン)を挙げることができる。
Immunomicsにより開発された四量体/細胞内IFN−γおよびパーフォリン染色アッセイ(製造元の説明書による)は、表現型分析と結合された抗原特異的T細胞の検出および機能アッセイ双方を可能にする。この流れに基づく(flow based)方法は、現在利用可能な抗原特異的T細胞機能の最も正確なアッセイである。このアッセイを使用して、ex vivo増殖がHIV特異的T細胞の総数および機能にどのように影響を及ぼすかを決定する。
これらのaAPCが大量のHIV−1に感染したT細胞をどのように増殖するかを評価するため、各aAPCに照射し、抗CD3 Abで被覆し、そしてHIV−1感染患者からの精製したCD8 T細胞と1:2のaAPC対T細胞比で混合した。細胞増殖の初期速度を比較するため、細胞を3Hチミジン取り込みよりはむしろCFSE染色にかけて、各aAPCが全T細胞の増殖をどのくらい良好に誘導したかを決定した。CFSE染色ははるかにより定量的な終点を提供しかつ増殖させた細胞の同時の表現型分類を可能にするからである。HIVに感染した個体からのおよそ2千万の精製したCD8 T細胞を3μMのCFSEと8分間混合し、徹底的に洗浄して未結合のCFSEを除去し、そしてaAPCで刺激する。刺激後毎日、細胞のアリコートを各培養物から取り出し、そしてフローサイトメトリーにより分析する。CFSE染色は細胞を高度に蛍光性にする。細胞分裂に際して蛍光は半分になり、そして従って細胞が多く分裂するほどそれはより少なく蛍光性になる。T細胞増殖を誘導する各aAPCの能力を、1回、2回、3回、など分裂した細胞の数を測定することにより定量する。特定の時間点で最も多数の細胞分裂を誘導するaAPCは、HIVに感染した個体からのCD8 T細胞の最も強力な増殖体(expander)と考えられる(Wellsら、1997、Clin.Invest.100:3173−3183)。
アポトーシス率およびテロメア長
アネキシンV/To−Pro(Molecular Probes、オレゴン州ユージーン)染色を各再刺激前に実施して、増殖速度の差違がアポトーシスを受けている細胞の数の差違を反映するかどうか決定する。このアッセイの実験の詳細はMausら(2002、Nature Biotechnol.20:143−148)に詳細に記述されている。最少量のアポトーシスに至る培養条件が望ましい。
サイトカイン産生およびBcl−xL発現レベルを検討するため、およそ百万の細胞から各刺激24時間後にRNAを単離し、そして限定されるものでないがIL−2、IL−4、IL−10、IFN−γ、TNF−αおよびBcl−xLの相対的発現を検査するため定量的RT−PCRにかける。これらの確立したアッセイの実験の詳細はMausら(2002、Nature Biotechnol.20:143−148)、Thomasら(2002、Clin.Immunol.105:259−272)およびParryら(2003、J.Immunol.171:166−174)に見出し得る。TGF−α mRNAレベルと分泌されたサイトカインの間の多くの矛盾が示され(Assoianら、1987、Proc.Natl.Acad.Sci.USA 84:6020−6024)、従ってTGF−α産生はELISAにより測定する。
四量体およびELISPOT分析
共通のリコール抗原およびHIVを認識する増殖されたCD8 T細胞の能力を比較する。増殖前に、精製したCD8 T細胞を、以下、とりわけHLA−A2四量体GLCTLVAML(配列番号4)(EBV BMLF)、NLVPMVATV(配列番号5)(CMV p65)、SLYNTVATL(配列番号6)(HIV gag p17)、ILKEPVHGV(配列番号3)(HIV RT pol)、GILGFVFTL(配列番号1)(Fluマトリックス)およびLLFGYPVYV(配列番号7)(HTLV Tax)を使用して染色し、そしてこれらの四量体の頻度をex vivo増殖前に測定する。次に、HIVに感染したドナーからの大量のCD8 T細胞を、本明細書に開示されるaAPCを使用して刺激し、そして、本明細書に記述される方法を使用して該細胞を増殖させる。各再刺激前に、増殖させたT細胞を四量体染色にかけて、EBV、CMVおよびHIV特異的T細胞の相対頻度が特定の一aAPCによる刺激により変えられたかどうか決定する。
本明細書に開示される方法は、ポリクローナルT細胞増殖の環境内でこれらの細胞の増殖および機能を検査することにより、HIV特異的CD8 T細胞の増殖においてどのaAPCが最良であるかへの重要な洞察を提供する。しかしながら、恒常性の機構がCD4 T細胞の喪失について補正しかつHIV非特異的CD8 T細胞に全体的な異常がないように思われる際に、全CD8 T細胞の数が大部分のHIV感染患者で増大しているため、HIVに感染した個体からのex vivoで増殖させたポリクローナルT細胞の翻訳値は低い(Gandhiら、2002、Annu.Rev.Med.53:149−172)。従って、自己養子移入によりHIV特異的CD8 T細胞応答を向上させるため、HIV特異的T細胞のみを増殖させかつ長期間HIV感染を除外し得るエフェクター機能をそれらに与える系が望ましい。
平均して700,000のPol特異的T細胞をHLA−A2陽性のHIVに感染した個体から、また、およそ100,000のPol特異的T細胞をワクチン接種した癌患者から単離する。抗原特異的増殖は約1,500のT細胞を使用して達成し得る(図3)。培養条件を同等にするため、培養は、96ウェルプレート中合計100μlの培地中で、7,500のPol特異的T細胞および15,000の本明細書の別の場所に記述されるaAPCを0.5ng/mlの抗CD3 Abと混合することにより開始する(T細胞対APC比を逆転させることが非常に少ない細胞を増殖させる場合に重要であることが観察されている)。これらの細胞を刺激する抗CD3およびK562にプロセシングかつ提示される抗原の能力を比較するため、そのaAPCもまたPol−GFP発現ベクターで形質導入した理想的な一組の培養物を増殖させる。従って、各ドナーの細胞について10個の培養物が利用可能である。新たに照射したaAPCを、2T細胞ごとに対し推定される1aAPCの比で、増殖するポリクローナルT細胞に10〜12日ごとに添加する。存在する細胞の数の正確な量がコールターカウンターを使用して計算され得る点まで該集団が一旦増殖したら、各集団の増殖速度を追跡する。T細胞増殖の前および後にHIVおよび癌患者から単離したPol特異的T細胞のCD28/CD27表現型を比較する。
サイトカイン産生、Bcl−xL発現、アポトーシス率およびテロメア長の評価
これらの研究は、存在する数百万の細胞を使用すること(およそ30日)から分析が構成されることを除き、本明細書の別の場所に開示される方法を使用して実施する。にもかかわらず、これらの研究は、ポリクローナルT細胞を使用して本明細書の別の場所に開示される結果を確認するはずである。
抗原特異的T細胞を分離して増殖させることの一利点は、複数の死滅および他の機能アッセイを非常に定量的様式で実施し得ることである。第一の試験は四量体/細胞内IFN−γおよびパーフォリン発現アッセイである。本明細書の別の場所に開示されるとおり、ex vivoで増殖させたCD8 T細胞を、自己CD8を枯渇したPBMCと混合する。CD8 T細胞の大部分は四量体陽性であるため、Polペプチドとの接触に際して最高レベルのIFN−γおよびパーフォリンを産生するT細胞をどのaAPCが生じさせるかに関する定量的データが得られる。加えて、四量体細胞の数は制限しないため、CCR7、CD27、CD28、CD62L、CD45 RO、CD45 RAおよびCD57を使用するこれらの細胞の完全な表現型分析(Brenchleyら、2003、Blood 101:2711−2720)を実施し、それによりエフェクター機能(1種若しくは複数)をT細胞表現型と相互に関連づける。
Nefエピトープを認識した単一のCD8クローンの注入は、このエピトープを発現しなかったウイルスの選択につながり(Koenigら、1995、Nature Med.1:330−336)、HIVエスケープミュータントを予防するために複数の特異性をもつT細胞が必要とされることを示す。ある患者からの特定の1ウイルスの複数のエピトープを認識するT細胞を生成させる潜在的に強力な一方法は、細胞に基づくaAPC取り込みを有し、そして天然のAPCの抗原に類似の抗原を提示させることである。MHCを発現するK562に基づくaAPCに、患者の化学的に不活性化されたウイルスを負荷すること、および自己T細胞に混合することにより、患者特異的抗HIV T細胞を増殖させ得る。HIV特異的T細胞がHIV特異的抗原に遭遇するこの最適な状況を提供することにより、免疫優性および潜在(cryptic)双方の抗原を認識するT細胞を増殖させ得、従ってHIV感染を制御するためのより大きな潜在能力をもたらす。さらに、共有されるHLAクラスI対立遺伝子をもつ、感染していない健康な人からのT細胞を、レシピエントの化学的に不活性化したウイルスでex vivoでワクチン接種し得、増殖させ得、そしてHIV感染患者に注入し得る。これは、進行した疾患を伴いかつ制限されたT細胞レパートリーをもつ個体にとって強力な潜在的処置の一選択肢を表す。
K562細胞を、(i)抗CD3および抗CD28抗体の外因性の負荷を可能にするためのヒトFcγ受容体CD32、ならびに、細胞の別個の集団を、抗CD3、抗CD28および他の受容体の高親和性負荷を可能にするヒトFcγ受容体CD64で形質導入した、ならびに(ii)ヒト4−1BBリガンドで安定にコトランスフェクトした。CD137としてもまた知られる4−1BBは、CD8+ T細胞の生存を促進するTNF受容体ファミリーの1メンバーである(Hurtadoら、1997、J.Immunol.158:2600−2609;Takahashiら、1999、J.Immunol.162:5037−5040;Tranら、1995、J.Immunol.155:1000−1009)。4−1BB刺激は、優先的に、CD8 T細胞をin vitroで活性化し、CTL応答をin vivoで増幅し、そして活性化されたCTLの生存を向上させる(Shufordら、1997、J.Exp.Med.186:47−55)。4−1BBはCD8 T細胞の長期のex vivo増殖を促進し得る候補分子である。CD3/28ビーズ、若しくは抗CD3およびCD28 Abで被覆したK32/4−1BBL aAPC(K32/4−1BBL CD3/28)いずれかで刺激したCD8 T細胞の初期増殖速度は同等であった。しかしながら、再刺激に際して、K32/4−1BBL CD3/28 aAPCで活性化したCD8 T細胞のみが増殖し続けた。増殖するこの能力は、細胞生存遺伝子Bcl−xLおよびサイトカインIL−2のアップレギュレーションと相関した。誘導されているこれらの遺伝子の非存在下で、培養物の大きな割合がアネキシンV陽性(アポトーシスの初期の兆候)になった(Mausら、2002、Nature Biotechnol.20:143−148)。細胞に基づくaAPCとT細胞の間の「クロストーク」が観察された(Thomasら、2002、Clin.Immunol.105:259−272)。
天然に存在するCD25+CD4+サプレッサー細胞(Treg)細胞は、自己の構成要素(すなわち免疫学的自己寛容)に対する免疫学的非応答性、および非自己抗原に対する多様な免疫応答の負の制御の確立および維持において能動的部分を演じている。Tregを定義するための少数の信頼できるマーカーが存在するが、しかし、蓄積する証拠が、天然に存在するCD25+CD4+ Tregが、免疫学的自己寛容の維持、および病理学的ならびに生理学的免疫応答の負の制御において決定的に重要な役割を演じていることを示すため、この集団が最も広範に研究されている。表現型上別個の集団としての免疫系におけるそれらの天然の存在は、それらを、免疫学的疾患を処置若しくは予防しかつ病理学的ならびに生理学的免疫応答を制御するための方法を設計するための良好な標的にする。しかしながら、細胞のこの集団を増殖させかつ操作するための方法はあるとしてもほとんど存在しない。
Claims (22)
- 単離された人工抗原提示細胞(aAPC)であって、レンチウイルスベクター(LV)を使用して形質導入されたK562細胞を含んでなり、かつ、前記LVは最低1種の免疫刺激リガンドおよび最低1種の共刺激リガンドをコードする核酸を含んでなり、かつ、さらに、前記aAPCは前記刺激リガンドおよび前記共刺激リガンドを発現しそして前記aAPCと接触されたT細胞を刺激かつ増殖させ得る、上記細胞。
- 前記刺激リガンドが、抗原を負荷された主要組織適合遺伝子複合体クラスI(MHCクラスI)分子、抗CD3抗体、抗CD28抗体および抗CD2抗体よりなる群から選択されるポリペプチドである、請求項1に記載の単離されたaAPC。
- 前記共刺激リガンドが、CD7、B7−1(CD80)、B7−2(CD86)、PD−L1、PD−L2、4−1BBL、OX40L、ICOS−L、ICAM、CD30L、CD40、CD70、CD83、HLA−G、MICA、MICB、HVEM、リンホトキシンβ受容体、ILT3、ILT4、3/TR6、およびB7−H3と特異的に結合するリガンドよりなる群から選択される最低1種の共刺激リガンドである、請求項1に記載の単離されたaAPC。
- 前記共刺激リガンドが、CD27、CD28、4−1BB、OX40、CD30、CD40、PD−1、ICOS、LFA−1、CD2、CD7、LIGHT、NKG2C、B7−H3、BTLA、Tollリガンド受容体、およびCD83と特異的に結合するリガンドよりなる群から選択される共刺激分子の最低1種と特異的に結合する、請求項1に記載の単離されたaAPC。
- 前記共刺激リガンドが、CD27、CD28、4−1BB、OX40、CD30、CD40、PD−1、ICOS、LFA−1、CD2、CD7、LIGHT、NKG2C、B7−H3、Tollリガンド受容体、およびCD83と特異的に結合するリガンドよりなる群から選択される最低1種の分子と特異的に結合する抗体である、請求項1に記載の単離されたaAPC。
- 前記aAPCが、CD32分子およびCD64分子よりなる群から選択されるFcγ受容体をさらに含んでなる、請求項1に記載の単離されたaAPC。
- 前記LVが、腫瘍抗原、ウイルス抗原、細菌抗原、ペプチド−MHC四量体、ペプチド−MHC三量体、ペプチド−MHC二量体、およびペプチド−MHC単量体よりなる群から選択される最低1種の抗原をコードする核酸を含んでなる、請求項1に記載の単離されたaAPC。
- 前記腫瘍抗原が、MAGE−1、MAGE−2、MAGE−3、MART−1、GP100、CEA、HER−2/Neu、PSA、WT−1、MUC−1、MUC−2、MUC−3、MUC−4およびテロメラーゼよりなる群から選択される、請求項7に記載の単離されたaAPC。
- 前記LVが、サイトカインおよびケモカインから選択される最低1種のペプチドをコードする核酸を含んでなる、請求項1に記載の単離されたaAPC。
- 前記サイトカインが、IL−2、IL−4、IL−6、IL−7、IL−10、IL−12、IL−15、IL−21、インターフェロン−α(IFNα)、インターフェロン−β(IFNβ)、インターフェロン−γ(IFNγ)、腫瘍壊死因子−α(TNFα)、腫瘍壊死因子−β(TNFβ)、顆粒球マクロファージコロニー刺激因子(GM−CSF)および顆粒球コロニー刺激因子(GCSF)よりなる群から選択される最低1種のサイトカインである、請求項9に記載の単離されたaAPC。
- 既知の共刺激分子を発現するT細胞の増殖の特異的誘導方法であって、前記T細胞を請求項1に記載のaAPCと接触させることを含んでなり、かつ、さらに、前記共刺激リガンドが前記既知の共刺激分子と特異的に結合してそれにより前記T細胞の増殖を特異的に誘導する、上記方法。
- 既知の共刺激分子を発現するT細胞の増殖の特異的誘導方法であって、前記既知の共刺激分子を発現する最低1種のT細胞を含んでなるT細胞の集団を請求項1に記載のaAPCと接触させることを含んでなり、かつ、前記aAPCは前記既知の共刺激分子と特異的に結合する最低1種の共刺激リガンドを発現し、前記既知の共刺激分子の前記共刺激リガンドとの結合が前記T細胞の増殖を誘導する、上記方法。
- T細胞集団の、サブセットの特異的増殖方法であって、前記サブセットの最低1種のT細胞を含んでなるT細胞の集団を請求項1に記載のaAPCと接触させることを含んでなり、かつ、前記aAPCは、前記サブセットの前記T細胞上の共刺激分子と特異的に結合する最低1種の共刺激リガンドを含んでなり、前記共刺激分子の前記共刺激リガンドとの結合が前記サブセットの前記T細胞の増殖を誘導して、それによりT細胞集団の1サブセットを特異的に増殖させる、上記方法。
- T細胞サブセットの活性化を特異的に誘導する共刺激リガンド若しくはその組合せの同定方法であって、請求項1に記載のaAPCとT細胞の集団を接触させること、および、前記T細胞集団の増殖のレベルを、前記aAPCと接触されないT細胞のそれ以外は同一の集団の増殖のレベルと比較することを含んでなり、かつ、前記aAPCと接触されないT細胞の前記それ以外は同一の集団の増殖のレベルと比較した、前記aAPCと接触させた前記T細胞の増殖のより高いレベルが、前記共刺激リガンドが前記T細胞の活性化を特異的に誘導することの指標である、上記方法。
- 哺乳動物における抗原に対するT細胞応答の誘導方法であって、請求項1に記載のaAPCを前記哺乳動物に投与することを含んでなり、かつ、前記aAPCが前記抗原を負荷されたMHCクラスI分子をさらに含んでなり、前記aAPCが、前記抗原に特異的なT細胞の増殖を誘導して、それにより前記哺乳動物における前記抗原に対するT細胞応答を誘導する、上記方法。
- それの必要な哺乳動物における抗原に対するT細胞応答の誘導方法であって、前記哺乳動物から細胞の集団を得ること(前記集団はT細胞を含んでなる)、細胞の前記集団を請求項1に記載のaAPCと接触させること(前記aAPCは前記抗原を負荷されたMHCクラスI複合体をさらに含んでなり、かつ、それにより、前記細胞を前記aAPCと接触させることが前記抗原に特異的な抗原特異的T細胞の増殖を誘導する)、前記抗原特異的T細胞を細胞の前記集団から単離すること、および前記抗原特異的T細胞を前記哺乳動物に投与して、それにより前記哺乳動物において前記抗原に対するT細胞応答を誘導することを含んでなる、上記方法。
- 制御性T(Treg)細胞の集団の特異的増殖方法であって、前記集団を請求項1に記載のaAPCと接触させることを含んでなり、かつ、前記aAPCは抗CD3抗体および抗CD28抗体を負荷されたFcγ受容体をさらに含んでなり、該方法は、細胞の前記集団をサイトカインと接触させることをさらに含んでなり、前記抗CD3抗体および前記抗CD28抗体の前記Treg細胞との結合が前記Treg細胞の増殖を誘導して、それによりTreg細胞の集団を特異的に増殖させる、上記方法。
- 前記サイトカインがインターロイキン−2である、請求項17に記載の方法。
- 既知の共刺激分子を発現するT細胞の増殖を特異的に誘導するためのキットであって、有効量のaAPCを含んでなり、前記aAPCはレンチウイルスベクター(LV)を使用して形質導入されたK562細胞を含んでなり、かつ、前記LVは前記既知の共刺激分子を特異的に結合する最低1種の共刺激リガンドをコードする核酸を含んでなり、前記既知の共刺激分子の前記共刺激リガンドとの結合が前記T細胞を刺激しかつ増殖させ、前記キットが、アプリケーター、および前記キットの使用のための説明資料をさらに含んでなる、上記キット。
- 既知の刺激分子を発現するT細胞の増殖を特異的に誘導するためのキットであって、有効量のaAPCを含んでなり、かつ、前記aAPCはレンチウイルスベクター(LV)を使用して形質導入されたK562細胞を含んでなり、前記LVは前記既知の刺激分子を特異的に結合する最低1種の刺激リガンドをコードする核酸を含んでなり、前記既知の刺激分子の前記刺激リガンドとの結合が前記T細胞を刺激しかつ増殖させ、前記キットが、アプリケーター、および前記キットの使用のための説明資料をさらに含んでなる、上記キット。
- T細胞集団のサブセットを特異的に増殖させるためのキットであって、有効量のaAPCを含んでなり、かつ、前記aAPCはレンチウイルスベクター(LV)を使用して形質導入されたK562細胞を含んでなり、前記LVは前記T細胞集団上の共刺激分子を特異的に結合する最低1種の共刺激リガンドをコードする核酸を含んでなり、前記共刺激分子の前記共刺激リガンドとの結合が前記T細胞集団を刺激しかつ増殖させ、前記キットが、アプリケーター、および前記キットの使用のための説明資料をさらに含んでなる、上記キット。
- T細胞サブセットの活性化を特異的に誘導する、共刺激リガンド若しくは前記リガンドの組合せ物を同定するためのキットであって、複数のaAPCを含んでなり、かつ、各前記aAPCはレンチウイルスベクター(LV)を使用して形質導入されたK562細胞を含んでなり、前記LVは共刺激分子と特異的に結合する最低1種の既知の共刺激リガンドをコードする核酸を含んでなり、前記キットが、アプリケーター、および前記キットの使用のための説明資料をさらに含んでなる、上記キット。
