Abstract
【解決手段】オステオプロテゲリン結合性蛋白、そのフラグメント、類似体もしくは誘導体を調製する為の、該ポリペプチドをコードする核酸、ベクターおよび宿主細胞、および結合アッセイ系を提供する。更に、骨粗鬆症、関節炎または転移による骨損失、高カルシウム血症、およびページェット病のような骨疾患の治療の為の組成物および方法も提供する。また、オステオプロテゲリン結合性蛋白の為の受容体、作用薬および拮抗薬を用いた骨疾患を治療する方法も提供する。

Description
腫瘍壊死因子ファミリーの新規メンバーが、親和性プローブとしての組換えOPG−Fc融合蛋白を用いてスクリーニングされたCOS細胞中に発現したマウスcDNAライブラリーから同定された。新しいポリペプチドは、長さが316アミノ酸であると予想される膜貫通OPG結合性蛋白であり、アミノ末端細胞質領域、膜貫通領域、およびカルボキシ末端細胞外領域を有する。本発明のOPG結合性蛋白は、膜に結合している、または可溶性状態である。
本発明は、OPGに特異的に結合すると共に破骨細胞の分化に関連する、OPG結合性蛋白と呼ばれるポリペプチドを提供する。マウス形態のポリペプチドをコードするcDNAが、マウス骨髄単球細胞系32−Dから調製されるライブラリーから同定され、COS細胞内にトランスフェクトされた。トランスフェクタントを、OPG[22−201]−Fc融合ポリペプチドに結合する能力についてスクリーニングした(実施例1)。核酸配列により、OPG結合性蛋白が、TNFファミリーの新規メンバーであり、AGP−1、すなわち1996年6月7日付けの共有の同時係属中の米国出願No.08/660,562に最も近く関係することが示された。(AGP−1と同等でTRAILと記載されるポリペプチドが、Wileyら著,Immunity第3巻,673〜682頁(1995年)に記載されている。)。OPG結合性蛋白は、アミノ末端における細胞質領域、膜貫通領域、およびカルボキシ末端細胞外領域を有するII型膜貫通蛋白であると予想される(図1)。アミノ末端細胞質領域は、図1に示すように(配列番号2)、残基1〜48の周辺に及び、膜貫通領域は残基49〜69の周辺に及び、細胞外領域は残基70〜316の周辺に及ぶ。膜結合性蛋白は、OPGに特異的に結合する(図2)。すなわち、OPG結合性蛋白およびOPGは、受容体−リガンド対の多くの特徴を共有するが、OPG結合性蛋白のための他の天然産受容体が存在する可能性もある。
本発明は、OPG結合性蛋白をコードする単離された核酸を提供する。ここで用いられる核酸と呼ばれる用語は、cDNA、ゲノムDNA、全体的または部分的に合成のDNA、およびRNAを含む。本発明の核酸は、下記の群から選択される:
a)図1(配列番号1)および図4(配列番号3)に示される核酸;
b)図1(配列番号1)および図4(配列番号3)に示される核酸のポリペプチドコード領域にハイブリッド形成し;高過酷条件下に核酸へのハイブリッド形成を維持する核酸;

本発明の核酸は、生物学的活性OPG結合性蛋白を発現するようにDNA配列と結合する。発現に必要な配列は当業者に知られており、RNA合成の開始のためのプロモーターおよびエンハンサー、転写終了部位、蛋白合成の開始のためのリボソーム結合部位、および分泌のためのリーダー配列を含む。OPG結合性蛋白の発現および分泌を導く配列は相同である、すなわち、配列は、OPG結合性蛋白発現および分泌に関連するゲノム内の配列に同一または類似である、またはそれらは非相同である。宿主細胞中のOPG結合性蛋白を発現するために種々のプラスミドベクターが利用できる(例えば、Methods
in Enzymology 185巻,Goeddel,D.V.編,Academic Press(1990年)を参照されたい)。哺乳動物宿主細胞中での発現のために、好ましい態様は、PCT出願No.90/14363に記載のプラスミドpDSRαである。バクテリア宿主細胞中での発現のために、好ましい態様は、ラックス(lux)プロモーターを有するプラスミドを含む(1995年12月22日付けの共有の同時係属中の米国出願No.08/577,778を参照されたい。)。さらに、ベタクーは、トランスジェニック動物中でのOPG結合性蛋白の組織特異的発現のために利用できる。レトロウイルスおよびアデノウイルス系遺伝子転移ベクターを、生体内治療のためにヒト細胞中でOPG結合性蛋白を発現させるために用いることができる(PCT出願NO.86/00922を参照)。
本発明は、外因性DNA配列の原核細胞または真核細胞発現の生成物としてのOPG結合性蛋白も提供する、すなわち、OPG結合性蛋白は組換えOPG結合性蛋白である。外因性DNA配列は、cDNA、ゲノムDNAおよび合成DNA配列を含む。OPG結合性蛋白は、バクテリア、酵母、植物、昆虫または哺乳動物細胞の発現の生成物、または細胞非含有転写系からの生成物であり得る。バクテリア細胞中で生成されるOPG結合性蛋白は、N−末端メチオニン残基を有する。本発明は、また、OPG結合性蛋白をコードする核酸を用いて形質転換またはトランスフェクトした原核または真核宿主細胞を増殖させ、核酸のポリペプチド発現生成物を単離することを含んでなるOPG結合性蛋白を製造する方法も提供する。
本発明のポリペプチドを特異的に結合する抗体も、本発明に含まれる。抗体を、全長OPG結合性蛋白、OPG結合性蛋白の可溶性体、またはそのフラグメントを用いる免疫化により生成することができる。本発明の抗体は、ポリクローナルまたはモノクローナルであってよく、または組換え抗体であってよく、例えば、軽および重鎖上のマウス一定領域がヒト配列により置換されたキメラ抗体、または相補的決定領域のみがマウス起源である、CDRが移植された抗体である。本発明の抗体は、例えば、ヒト抗体を生成することのできるトランスジェニック動物の免疫化により調製されるヒト抗体であってもよい(例えば、PCT出願No.WO93/12227を参照)。抗体は、生物学的サンプル中においてOPG結合性蛋白を検出するのに有用であり、それにより、蛋白を生成する細胞または組織が同定される。さらに、OPG結合性蛋白に結合すると共に他の結合化合物との相互作用を遮断する抗体が、破骨細胞分化および骨吸収の調整において治療的用途を有する。

OPG結合性蛋白は、OPGを検出し、OPGとの相互作用を特徴付けるための種々のアッセイにおいて用いることができる。通常、そのアッセイは、OPGをOGP結合性蛋白に結合させる条件下においてOPG結合性蛋白をOPGを含む生物学的サンプルと共にインキュベートすること、および結合の程度を測定することを含む。OPGは精製される、または体液もしくは培地のような混合物中に存在してよい。定性的または定量的であるアッセイを開発することができ、OPG結合性蛋白へのOPGの結合パラメーター(親和定数および動態)を決めること、および混合物中における生物学的活性OPGの水準を定量することには後者が有用である。OPG結合性蛋白のフラグメント、類似体および誘導体へのOPGの結合を評価するため、および新しいOPGおよびOPG結合性蛋白ファミリーメンバーを同定するためにアッセイを用いることもできる。
本発明は、OPG結合性蛋白と相互作用する受容体も提供する。より詳しくは、本発明は、破骨細胞分化および活性化受容体(ODAR)を提供する。ODARは、TNF受容体ファミリーメンバーであるCD40に最も高度の類似性を示す膜貫通ポリペプチドである。マウスODARの核酸配列およびコードされたポリペプチドを図10に示す。マウスODARのヒト類似体は、図10の核酸配列を有するゲノムライブラリーまたはヒトcDNAのハイブリッド形成スクリーニングにより容易に単離することができる。ヒトODARをクローニングする手順は、ヒトOPG結合性蛋白をクローニングするための実施例5に記載の手順に類似している。図10に示すポリペプチドのヒト類似体はAndersonらの文献(Nature 390巻,175〜179頁(1997年))に見られ、そこでRANKと呼ばれている。RANKは、TNF受容体ファミリーメンバーへの類似性を有するI型膜貫通蛋白として特徴付けられ、樹枝状細胞機能に含まれる。
オステオプロテゲリン(OPG)は、生体外および生体内の破骨細胞発生を抑制制御する。OPGは、TNFR関連蛋白であるので、TNF関連ファミリーメンバーと相互作用し易いと共にその効果を媒介する。1つの例を除いて、TNFスーパーファミリーの全ての既知のメンバーは、細胞表面に発現されるII型膜貫通蛋白である。OPG結合性蛋白の供給源を同定するために、種々の細胞系および一次造血細胞の表面に配されたOPG結合性蛋白についてスクリーニングするための免疫プローブとして、組換えOPG−Fc融合蛋白を用いた。

cDNAライブラリーを32D mRNAから調製し、哺乳動物発現ベクターpcDNA3.1(+)(Invitrogen,San Diego,CA)中に結合させた。組換えインターロイキン−3の存在下に維持された指数増殖期の32D細胞を収穫し、全細胞RNAを酸性チオシアン酸グアニジニウム−フェノール−クロロホルム抽出により精製した(ChomczynskiおよびSacchiのAnal.Biochem.第162巻,156〜159頁(1987年))。製造者の推薦手順を用いてDynabeads Oligo(dT)25(Dynal Corp製)への吸着またはそこからの溶離により、全RNA製剤からポリ(A+)mRNAフラクションを得た。指向的オリゴ−dTでプライミングしたcDNAライブラリーを、製造者の推薦手順を用いてSuperscript Plasmid System(Gibco BRL,Gaithersburg,Md)を用いて調製した。得られるcDNAを、Sal IおよびNot I制限エンドヌクレアーゼを用いて完全に消化し、次に、サイズ排除ゲルクロマトグラフィーにより分画した。最も分子量の大いフラクションを選択し、次に、プラスミドベクターpcDNA3.1(+)(Invitrogen,San Diego,CA)のポリリンカー領域に結合させた。このベクターは、複クローニング部位の上流のCMVプロモーターを含み、真核生物細胞中に高水準の発現を導いた。次に、ライブラリーを、電気穿孔によりコンピテント大腸菌中に入れ(ElectroMAX DH10B,Gibco,NY)、100ug/mlアンピシリンを含むLB寒天上に滴下した。次に、ライブラリーを、約1000クローン/プールを含む分離プール中に配置し、各プールの培地1.0mlを37℃で16〜20時間増殖させた。各培地からのプラスミドDNAを、製造者の推薦手順に従って、Qiagen Qiawell 96 Ultra Plasmid Kit(カタログ#16191)を用いて調製した。
cDNAクローン、即ち各々1000クローンを含む300のトランスフェクトしたプールをスクリーニングした。OPG−Fc融合蛋白により特異的に修飾される能力を獲得した細胞を含む単一のウエルを同定した。このプールを、同胞選択の連続的繰り返しにより分割し、単一プラスミドクローン32D−F3を得た(図1)。次に、32D−F3プラスミドDNAを、COS−7細胞中にトランスフェクトし、それを、FITC−結合ヤギ抗ヒトIgG2次抗体単独、ヒトOPG[22−201]−Fc融合蛋白+2次、またはATAR−Fc融合蛋白(HVEMとしても知られているATAR;Montgomeryら著,Cell 第87巻,427〜436(1996年))で免疫染色した(図2)。2次抗体単独はCOS−7/32D−F3細胞に結合せず、ATAR−Fc融合蛋白にも結合しなかった。OPG Fc融合蛋白のみが、COS−7/32D−F3細胞に結合し、このことは、OPG結合性蛋白をコードする32D−F3が発現細胞の表面に表示されることを示している。
先に単離した32D−F3は、約2.3kbのcDNA挿入部を含み(図1)、それを、製造者の推薦手順に従って、プライマー誘導Taq染料ターミネーター反応(Applied Biosystems)を用いてApplied Biosystems 373A自動DNA配列機において両方向に配列決定した。得られるヌクレオチド配列を、FASTAプログラム(GCG,ウイスコンシン大学)を用いてDNA配列データベースと比較し、「6方向(six−way)オープンリーディングフレーム」アプリケーション(Frames)(GCG,ウイスコンシン大学)を用いて長いオープンリーディングフレーム(LORF’s)の存在について分析した。メチオニンで始まっている316アミノ酸(aa)残基のLORFを適当な方向において検出し、このLORFに続いて約150bpの5’非翻訳領域が存在した。5’非翻訳領域は、予想される開始コドンの上流にフレーム内停止コドンを含んでいた。これは、32D−F3プラスミドの構造が、哺乳動物細胞中で316aa遺伝子生成物の発現を導くようにCMVプロモーター領域を利用するその性能に一致することを示している。
様様な複合ヒト組織ノーザンブロット(Clontech,Palo Alto,CA)を、32P−dCTP標識化32D−F3制限フラグメントを用いて検知することにより、ヒト転写の寸法を検出し、発現のパターンを決定した。ノーザンブロット体は、5×SSPE、50%ホルムアミド、5×デンハート溶液、0.5%SDS、および100μg/ml変性サケ精子DNA中で42℃で2〜4時間、プレハイブリダイゼーションを行った。次に、ブロット体を、5×SSPE、50%ホルムアミド、2×デンハート溶液、0.1%SDS、100μg/ml変性サケ精子DNA、および5ng/ml標識化プローブ中で42℃で18〜24時間、ハイブリダイゼーションを行った。次に、ブロット体を、2×SSC中にて室温で10分間、1×SSC中にて50℃で10分間、次に、0.5×SSCにて10〜15分間洗った。
OPG結合性蛋白のヒト相同体が、ヒト末梢リンパ節中に約2.5kbのmRNAとして発現され、緊縮ハイブリッド形成条件下にマウスcDNAプローブとのハイブリッド形成により検出される。ヒトOPG結合性蛋白をコードするDNAが、組換えバクテリオファージプラーク、または形質転換バクテリアコロニーハイブリッド形成法によりヒトリンパ節cDNAライブラリーをスクリーニングすることにより得られる(Sambrookら著,Molecular Cloning: A
Laboratory Manual,Cold Spring Harbor Press,New York(1989年))。このために、マウスOPG結合性蛋白クローン32D−F3を放射能で標識したプローブを用いて、そのファージまたはプラスミドcDNAライブラリーをスクリーニングする。プレート上のライブラリーから持ち上げられたニトロセルロースフィルターをプローブを用いてスクリーニングする。これらのフィルターはプレハイブリダイゼーションを行い、次に実施例4に特定された条件を用いてハイブリダイゼーションを行い、最終的にヒトOPG結合性蛋白cDNAの精製クローンを得た。任意のヒトOPG結合性蛋白クローンから得られた挿入物を、実施例3に記載のように配列決定および分析する。

プライマー対と以下に記載するテンプレートを用いるPCR増幅を利用して、種々の形状のマウスOPG結合性蛋白を合成する。各対の1つのプライマーは、遺伝子のカルボキシ末端に続いて、TAA停止コドンおよび独自のXhoIまたはSacII部位を導入する。各対の他のプライマーは、独自のNdeI部位、N−末端メチオニン、および遺伝子のアミノ末端部分への最適化コドンを誘導する。PCRおよび熱循環は、標準的組換えDNA法を用いて行う。PCR生成物を精製し、制限酵素で消化し、ベクターpAMG21(ATCCアクセス番号
98113)の独自のNdeIおよびXhoIまたはSacII部位に挿入し、原栄養大腸菌393または2596中に形質転換する。他の一般的に用いられる大腸菌発現ベクターおよび宿主細胞も発現に適している。形質転換後、クローンを選択し、プラスミドDNAを単離し、OPG結合性蛋白挿入体の配列を確認する。

発現されたマウスOPG結合性蛋白(158−316)を有する凍結バクテリア細胞を解凍し、20mM Tris−HCl pH7.0、10mM EDTA中に再び懸濁させた。次に、細胞懸濁液(20%w/v)を、マイクロフルイダイザイーを3回通過させることにより均質化した。溶解された細胞懸濁液を、JA14ローター内で10000rpmにて45分間遠心分離した。SDS−PAGE分析は、封入体および上清の両方において存在する分子量約18kdのバンドを示した。次に、可溶性フラクションを、10mM MES pH6.0で平衡させたPharmacia SP Sepharose 4FFカラムに適用した。OPG結合性蛋白を、MES pH6.0中に0〜0.4MのNaClを含む20カラム体積のグラジエントで溶離した。次に、OPG結合性蛋白を含むフラクションを、20mM MES pH6.0で平衡させたABX Bakerbondカラムに適用した。OPG結合性蛋白を、MES pH6.0中に0〜0.5MのNaClを含む15CVのグラジエントで溶離した。最終生成物は、SDS−PAGEによると95%を超える均質さであった。N−末端配列化により、次の配列が得られた:Met−Lys−Pro−Glu−Ala−Gln−Pro−Phe−Ala−His:これは、残基158で開始(メチオニンイニシエーター)するポリペプチドについて予想されるものと同様である。SDS−PAGE中の蛋白の相対分子量は還元時に変化しない。
組換えOPG蛋白が、先に、米国出願08/577,788に記載されているように、破骨細胞形成アッセイにおいて骨髄および脾臓前駆体からのビタミンD3−依存性破骨細胞形成を阻害することが示された。OPG結合性蛋白は、OPGに結合し、リガンドのTNFファミリーの新しいメンバーであるので、OPG生物学的活性の潜在的標的である。最少コアTNFα様領域を表す組換え可溶性OPG結合性蛋白(158〜316)を、破骨細胞前駆体からの破骨細胞の分化を調整する性能について試験した。骨髄細胞を成体マウス大腿骨から単離し、M−CSFで処理した。非接着性フラクションを、ビタミンD3およびデキサメタゾン両方の存在または不存在下にST2細胞と一緒に共培養した。先に示したように、破骨細胞は、基質細胞(ST2)、ビタミンD3およびデキサメタゾンを含む共培養からでないと発達しない。組換え可溶性OPG結合性蛋白を、0.16〜500ng/mlの種々の濃度で添加し、破骨細胞成熟を、TRAP溶液アッセイにより、および視覚的観察により測定した。OPG結合性蛋白は、破骨細胞分化および成熟を投与量に依存して強度に刺激し、1〜2ng/mlの範囲において最大の半分の効果を有しており、このことは生体外での破骨細胞発生の潜在的誘発剤として作用することを示している(図5)。OPG結合性蛋白の効果は、組換えOPGにより阻害される(図6)。
生体外研究に基づき、大腸菌内で生成した組換えマウスOPG結合性蛋白[158−316]は、骨髄性前駆体からの破骨細胞発達の潜在的誘発剤である。生体内におけるその効果を決めるために、4〜5週齢のオスBDF1マウス(Charles River Laboratories)に、OPG結合性蛋白[158−316]の皮下注射を、1日2回で3日間および4日目の朝に行った(0、1、2および3日目)。5つの群のマウス(n=4)にキャリアのみ、または一日当り1、5、25もしくは100μg/のOPG結合性蛋白[158−316]を与えた。さらに5つの群のマウス(n=4)に上記投与量のキャリアまたはOPG結合性蛋白[158−316]を与え、さらにヒトFc−OPG[22−194]を1日1回の皮下注射により1mg/kg/日(約20μg/日)の割合で与えた。全血イオン化カルシウムを、0日目の処理前、および1、2および3日目のOPG結合性蛋白[158−316]の最初日毎注射の3〜4時間後に決めた。3日目の最後の注射から4時間後、マウスを殺し、放射線撮影を行った。
マウスおよびヒトOPG結合性蛋白の全長クローンを、先に実施例2で記載したように哺乳動物細胞において発現することができる。また、哺乳動物細胞内で発現されたときに、分泌された状態の蛋白をコードするようにcDNAクローンを変性することができる。このために、開始コドンをコードし、膜貫通拡張領域を含む蛋白の最初の69のアミノ酸を略通過して伸びるcDNAの天然5’末端を、シグナルペプチドリーダー配列で置換することができる。例えば、既知の遺伝子の最初のコドンおよびシグナルペプチドをコードするDNA配列を切断して、アミノ酸残基68をコードする領域の後の任意の位置で始まるOPG結合性蛋白cDNA配列にすることができる。得られる組換えクローンは、動物細胞内で分泌状態のOPG結合性蛋白を生成することが予想され、グリコシル化のようなOPG結合性蛋白のC末端細胞外領域において通常起こる翻訳後変性を起こすべきである。この方式を用いて、マウスOPG結合性蛋白の5’末端およびその3’末端にヒトIgG1 Fc領域を有する分泌状態のOPG結合性蛋白を組み立てた。1995年12月22日付けの米国出願No.08/577,788に記載のプラスミドベクターpCEP4/muOPG[22−401]を、NotIで消化してOPGの3’末端とFc遺伝子との間を開裂させる。線状化したDNAを、次に、XmnIで部分的に消化してOPGの残基23と残基24との間のみを開裂してブラント末端を残した。制限酵素による消化物を、次に、CIPを用いて脱リン酸し、この消化物(OPGおよびFcの1〜23残基を含む)のベクター部分をゲル精製した。

1602〜61オリゴヌクレオチドが、遺伝子の5’末端を増幅し、人工的なStuI部位を含む。1602〜59プライマーが遺伝子の3’末端を増幅し、人工的なNotI部位を含む。得られるPCR生成物を、NotIおよびStuIを用いて消化し、次にゲルで精製した。精製されたPCR生成物をベクターと結合し、次に電気的コンピテント大腸菌DH10B細胞の形質転換に用いた。得られたクローンを配列させて、増幅した配列と制限部位接合部の一体性を確認した。次にこのプラスミドを用いてヒト293線維芽細胞をトランスフェクトし、OPG結合性蛋白−Fc融合蛋白を、先に1995年12月22日付けの米国出願No.08/577,788に記載されているように培地から収集した。

OPG結合性蛋白の特定領域への抗体を、OPG結合性蛋白からのペプチドを用いて免疫化することにより得ることができる。これらのペプチドを単独で用いて、または共役状態のペプチドを用いて免疫化することができる。
して[C.R.Gohら,前掲書]、リガンドTNFβの対応するループBB’およびEFが、TNFb:TNF−R55複合体の溶解した結晶構造中の受容体との接触体の主用部分を形成することがわかった。マウスOPG結合性蛋白のアミノ酸配列を、TNFαおよびTNFβのアミノ酸配列と比較した。BB’およびEFループに対応するマウスOPG結合性蛋白の領域を、この比較に基づき予想し、ペプチドを設計し、それを以下に記載する。
BB’ループペプチド
Na2CO3,0.035M NaHCO3)の1ml当り5μgの濃度の精製組換え蛋白または蛋白フラグメントで被覆する。蛋白フラグメントを、要すれば、ウシ血清アルブミン(BSA)に結合させることができる。50μlのagを、各ウエルに添加する。次に、プレートを、酢酸塩フィルム(ICN Biomedicals,Inc.,Costa Mesa,CA)で覆い、揺動台上で室温(rt)で2時間または4℃で一晩インキュベートする。BSA希釈剤/ブロッキング溶液濃縮液(Kirkegaad and Perry Laboratories,Inc.,Gaithersburg,MD)1部を脱イオン水(dH2O)1部と混合することにより調製した5%BSA溶液をウエル当り250μl用いて室温で30分間ブロッキングする。ブロッキング溶液を廃棄すると、血清2倍希釈液またはハイブリドーマ組織培地懸濁液の50μlを各ウエルに添加する。血清希釈剤は1%BSA(10%BSA希釈剤/ブロッキング溶液濃縮液が、ダルベッコリン酸塩緩衝生理生塩水D−PBS中に1:10で希釈されたもの;Gibco BRL、Grand Island、NY)であり、ハイブリドーマ上清を希釈しないで試験する。ハイブリドーマスクリーニングの場合、1つのウエルを結合体対照として維持し、第2のウエルを陽性ab対照とする。プレートを、再び揺動しながら室温で1時間インキュベートし、次に、dH2O中の20倍濃厚物(Kirkegaard and Perry Laboratories,Inc.,Gaithersburg,MD)の洗浄溶液の1倍製剤を用いて4回洗った。次に、1%BSA中に希釈したワサビ過酸化酵素結合2次ab(Boeringer Mannheim Biochemicals,Indianapolis,IN)を各ウエルにおいて30分間インキュベートする。プレートを前述のように洗い、吸取り紙で乾燥し、ABTS過酸化酵素単一成分基質(Kirkegaard and Perry Laboratories,Inc.,Gaithersburg,MD)を添加する。ミクロプレートEL310リーダー(Bio−tek Instruments,Inc.,Winooski,VT)を用いて各ウエルについて吸光度を405nmで読む。405における光学密度に対して血清希釈のlog10をプロットし、つぎにその血清により得られた最大光学密度の50%のポイントを外挿することにより、血清抗体の最大の半分の力価を計算する。光学濃度が前記バックグラウンドより5倍以上大きい場合、ハイブリドーマを陽性として選択する。このプロトコールへの調節を行い;例えば、特異性または非相互反応性について共役2次抗体を選択することができる。
Laboratories、Grand Island、NY)中で増殖させた、骨髄腫細胞の対数相培地、マウスまたはラット脾臓細胞溶融物のSp2/0−Ag14またはY3−Ag1.2.3(American Type Culture Collections;Rockville,MD)を同様にして洗う。脾臓細胞をミエローマ細胞と組み合わせて、再度ペレット化する。培地を細胞ペレットから吸引し、2mlのポリエチレングリコール1500(PEG1500;Boeringer Mannheim Biochemicals,Indianapolis,IN)を1分間かけて細胞中に穏やかに混入する。その後、等量の2×P/S/G DMEMをゆっくり添加する。細胞を37℃で2分間融合させ、次に、さらに6mlの2×P/S/G DMEMを添加する。細胞を再び37℃で3分保温する。最後に、35mlの2×P/S/G DMEMを細胞懸濁液に添加し、細胞を遠心分離によりペレット化する。培地をペレットから吸引し、細胞を完全培地中に穏やかに再懸濁する。細胞を、5mlのペレットから1回滴下することにより96ウエル平底組織培地プレート(Becton Dickinson Labware;Lincoln Park,NJ)上に分配する。プレートを、37℃で5%CO2の加湿条件において一晩インキュベートする。翌日、等体積の選択培地を各ウエルに添加する。選択培地は、完全培地中の0.1mMのヒポキサンチン、4×10−4mMのアミノプテリン、および1.6×10−2mMのチミジンからなる。融合プレートを7日間インキュベートし、次の3日間に培地を2回交換し;培地交換後にHAT選択培地を用いる。組織培地上澄みを、各ハイブリドーマ含有ウエルから、最後の培地交換から3〜4日後に取り出し、特異的抗体反応性についてEIAで試験する。このプロトコールは、HudsonおよびHay著,「Practical Immunology、第2版」,Blackwell Scientific Publicationsにおいて改良された。
生物学的活性組換えマウスOPG結合性蛋白[158−316]をフルオレセイン−イソチオシアネート(FITC)に結合させて蛍光プローブを形成した。組換えマウスOPG結合性蛋白[158−316]を、6−フルオレセイン−5−(および6)カルボキシアミドヘキサン酸スクシンイミジルエステル(Moleclar Probes、Eugen、OR)と、1:6のモル比にて4℃で12時間インキュベートすることにより蛍光標識化を行った。FITC標識化OPG結合性蛋白[158−316]をゲル濾過クロマトグラフィーによりさらに精製した。マウス骨髄細胞を単離し、実施例10に記載のようにCSF−1およびOPG結合性蛋白[158−316]の存在下に培地中でインキュベートした。マウス骨髄細胞をCSF−1(30ng/ml)およびOPG結合性蛋白[158−316](20ng/ml)中で一晩培養した。非接着性細胞を最初に除去し、氷上に貯蔵し、残りの接着性細胞を、細胞剥離緩衝液(Sigma Chemicals,St.Louis,MO)と一緒にインキュベートすることにより除去し、非接着性集合と共にプールし、次に、前述のようにFITC−OPG結合性蛋白で染色した。洗浄し、0.5%BSAを含むPBS中に再懸濁させた後、細胞をFITC−OPG結合性蛋白にさらし、洗い、次にFACSにより分類した。FITC−OPG結合性蛋白での染色に陽性である細胞の集合を集め、実施例2のようにmRNAを単離した。実施例2に記載の手順に従って、このmRNA製剤を用いてcDNAライブラリーを形成した。
ヒトIgGγ1のFc領域に融合している可溶性ODAR細胞外領域を、WO97/23614およびSimonetらの前掲書に既述しているようにFc融合蛋白の組み立ておよび発現のための手順を用いて処理した。哺乳動物細胞中に可溶性ODAR蛋白を発生させるために、マウスODAR(アミノ酸27〜211)のcDNAコード化細胞外領域を、以下のセットのオリゴヌクレオチドプライマーを用いてPCR増幅した:

ODARがOPG結合性蛋白により破骨細胞形成の刺激を抑制する性能を、CSF−1(30ng/ml)およびOPG結合性蛋白(5ng/ml)の存在下にマウス骨髄培地中で検定した。破骨細胞成熟を研究するためにマウス骨髄培地を使用する手順は、WO97/23614および実施例8に記載されている。実施例12に記載されているODAR−Fc融合蛋白を、65〜1500ng/mlの濃度になるように添加した。破骨細胞形成を、培地中で5日間後、抗酒石酸塩アルカリホスファターゼ(TRAP)細胞化学およびTRAP溶液アッセイにより検定した。
3〜4週齢の若い成長の早いオスBDF1マウスに、種々の投与量のODAR−Fc融合蛋白を、キャリアに溶かせ(PBS/0.1%BSA)て毎日1回4日間、皮下注射した。5日目にマウスにX線照射した。使用したODAR−Fc融合蛋白の投与量は0.5、1.5および5mg/kg/日であった。各処理において、その群、およびPBS/0.1%BSAを受けた対照群における全てのマウスを1枚のフィルム上でX線照射した。近位けい骨骨幹端領域を、対照対と処理けい骨との間で比較し、処理けい骨が、以下に示すスコアー8を与える対照よりも視覚評価で密度が高いと「+」と記録した。「陽性」と判断されるには5/8の観察スコアーを必要としている。(投与量はmg/kg/日で示す)(n=4)
マウスを殺した後、右けい骨を各動物から外し、近位けい骨骨幹端領域の骨密度を、末梢定量コンピューター補助断層撮影(pQCT)(Stratec,Germany)により測定した。けい骨の近位端部から1.5mmおよび2.0mmにおける2つの0.5mmの骨の断面を分析(XMICE5.2,Stratec,Germany)して、骨幹端の全骨鉱物密度を決めた。1500の軟質組織分離閾値を用いて、骨幹端骨の境界を定義した。
Claims (42)
- (a)図1(配列番号1)および図4(配列番号3)に示される核酸配列;
(b)図1(配列番号2)および図4(配列番号4)に示されるポリペプチドコード領域にハイブダイズし、高緊縮条件下に核酸へのハイブリダイズを維持する核酸;および
c)(a)または(b)の核酸に縮退である核酸
からなる群より選択されるオステオプロテゲリン縮合蛋白をコードする単離された核酸。 - cDNA、ゲノムDNA、合成DNAまたはRNAである請求項1に記載の核酸。
- 請求項1の核酸によりコードされるポリペプチド。
- 大腸菌での発現に好ましい1種またはそれ以上のコドンを含む請求項1に記載の核酸。
