【解決手段】2005年12月5日にDSMZに寄託されたバシラス・サブチリス変異株GN13/72(DSM17766)の培養を含む、ビタミンK2(MK−7)の製造方法。

Description
バシラス・サブチリスGN13/27(DSM17766)を、以下の組成を有する発酵培地「F10」が、幾何学的スケール上、20l入った30l容の発酵槽において好気的に増殖させる。
・微粉化大豆ミール 15.00g/l
・酵母エキス 1.00g/l
・グリセロール 10.00g/l
・K2HPO4 0.05g/l
・NaCl 5.00g/l
・pH7.3(±0.1)
・殺菌 121℃、30分
発酵槽に、培地「S3」が以下の組成を有する8%(1600ml)の16時間(±2時間)シードを接種する。
・大豆ペプトン 10.00g/l
・グリセロール 5.00g/l
・pH7.2(±0.1)
次に、シードに、精製水10mlで洗浄したスラント(斜面培養株)からのバシラス・サブチリス懸濁液5mlを接種した。
増殖条件:
・シード:
培地800mlが入った3l容の撹拌フラスコ
インキュベーション温度 37℃
撹拌 90rpm
時間 17時間(±2時間)
・発酵槽:
撹拌 120rpm
気流 0.6vvm
圧力 0.025MPa(0.25bar)
温度 37℃
シリコン消泡剤
時間 144時間(±4時間)
前記条件下で得られたバイオマスは、乾燥重量で8(±1)g/lであり、MK−7含有量が1100(±100)ppmであった。
(実施例2)
バシラス・サブチリスGN13/72(DSM17766)を実施例1と同様に培養するが、発酵培地「F12」は以下の組成を有する。
・大豆ペプトン 10.00g/l
・グリセロール 10.00g/l
・酵母エキス 1.00g/l
・K2HPO4 0.05g/l
・NaCl 5.00g/l
発酵期間は、140時間(±2時間)である。
(実施例3)
バシラス・サブチリスGN13/72(DSM17766)を実施例1と同様に培養するが、発酵培地「F13」は以下の組成を有する。
・大豆ペプトン 12.00g/l
・酵母エキス 0.50g/l
・デキストリン 60.00g/l
・K2HPO4 0.05g/l
・NaCl 5.00g/l
発酵期間は、140時間(±1時間)である。
(実施例4)
バシラス・サブチリスGN13/72(DSM17766)を実施例3と同様に培養するが、炭素源は、デキストリンの代りにマルトデキストリンからなる。
(実施例5)
バシラス・サブチリスGN13/72(DSM17766)を実施例4と同様に培養するが、消泡剤は大豆油とする。
(実施例6)
バシラス・サブチリスGN13/72(DSM17766)を、培地「F13」が225lの実用的な量であり、pHがNaOHで自動的に7.2に維持され、撹拌が100rpmであり、気流が0.5vvmであり、圧力が0.03MPa(0.3bar)であり、消泡剤の大豆油が自動制御される300l容の発酵槽において培養する。
(実施例7)
バシラス・サブチリスGN13/72(DSM17766)を、実施例6と同様に培養する。定常期に達する前に発酵を止めた。乾燥重量で7.0(±1.5)g/lが得られ、MK−7の生産力は21000ppmである。バイオマスを二部に分割し、一部を凍結乾燥して17000(±350)ppmの粉末を得、もう一部を噴霧乾燥して20000(±500)ppmを得た。
噴霧乾燥機の動作条件:
・吸気温度 200℃
・排気温度 80℃
(実施例8)
実施例7の手順に従うと、発酵の有効量は2m3であった。噴霧乾燥した最終製品を、真空下で250gの小袋(sachet)として包装した。

- バシラス・サブチリス(Bacillus subtilis)変異株GN13/72(2005年12月5日、DSMZに寄託;DSM17766)を培養する工程を含む、ビタミンK2(MK−7)の製造方法。
- 前記培養に使用される炭素源が、デキストリンである、請求項1に記載の方法。
- 前記培養に使用される炭素源が、マルトデキストリンである、請求項1に記載の方法。
