Description
Claims (1)
- 【特許請求の範囲】 1. 非腫瘍細胞を含む細胞集団内から腫瘍細胞を除去する試験管内方法であっ て、 a) 前記腫瘍細胞を前記非腫瘍細胞から区別できるように、前記細胞の集団 をラベルする工程、 b) 全細胞集団を照明する工程、 c) 照明した細胞集団の映像を捕らえる工程、 d) 捕らえた映像及び前記ラベルを参照して細胞集団における腫瘍細胞の少 なくとも2次元座標を決定する工程、及び、 e) 前記腫瘍細胞が表面上の実質的に静止した位置にある間に、制御したエ ネルギー源からのパルスを前記腫瘍細胞の座標へ適用することによって前記腫 瘍細胞を殺す工程、からなる工程を含む方法。 2. 工程d)及びe)を前記非腫瘍細胞集団中のすべての腫瘍細胞に対して繰り返 す請求項1記載の方法。 3. 前記非腫瘍細胞集団が、造血細胞集団である請求項1又は2記載の方法。 4. 前記造血細胞集団が、造血幹細胞を増加させた造血細胞集団からなる請求 項1〜3のいずれか1項に記載の方法。 5. 前記造血幹細胞集団をCD34表面タンパク質の発現に基づき増加させる請求 項4記載の方法。 6. 前記造血幹細胞集団をThr-1、CD38、CD15、CD11b、Gly-A、CD3及びCD19か らなる群から選択される細胞マーカーの発現に基づき増加させる請求項4記載の 方法。 7. 前記増加させた細胞集団を深床式濾過法や、向流溶出法を使用することに よって作る請求項4記載の方法。 8. 工程d)及びe)をコンピューター制御のもとで行なう請求項1〜7のいずれか 1項に記載の方法。 9. 前記制御エネルギー源がレーザである請求項1〜7のいずれか1項に記載方 法。 10.前記レーザから放射した光の波長が375nm〜900nmの間である請求項9に記 載の方法。 11.前記ラベルが、前記腫瘍細胞に特異的である請求項1-10のいずれか1項に 記載の方法。 12.前記ラベルが、前記非腫瘍細胞に特異的である請求項1-11のいずれか1項 に記載の方法。 13.造血細胞集団において幹細胞の数を増加させるための試験管内方法であっ て、 a) 前記造血細胞集団を幹細胞に特異的なラベルでラベルする工程、 b) 全造血細胞集団を照明する工程、 c) 照明した造血細胞集団の映像を捕らえる工程、 d) 映像上の該細胞の位置を決定することによって造血細胞集団における いずれかの非ラベル細胞の少なくとも2次元座標を計算する工程、及び、 e) 前記幹細胞が表面上の実質的に静止した位置にある間に、前記集団の非 ラベル細胞の少なくとも1つの座標に高エネルギーレーザ光パルスを適用する こと、からなる方法。 14.前記適用工程が、前記集団の実質的にすべての非ラベル細胞へ前記高エネ ルギーレーザ光パルスを適用することからなる請求項13記載の方法。 15.前記幹細胞特異的ラベルがCD34である請求項13又は14記載の方法。 16.前記レーザから照射された光の波長が、375nmと900nmの間である請求項13 〜15のいずれか1項に記載の方法。 17.前記適用工程をコンピューターの制御下で行なう請求項13〜16のいずれか 1項に記載の方法。 18.患者に再導入するために単離した造血細胞を調製する試験管内方法であっ て、 a) 前記単離した造血細胞内の腫瘍細胞を腫瘍細胞に特異的なラベルでラベル すること、 b) 単離した造血細胞を照明すること、 c) 単離した造血細胞の映像を捕らえること、 d) 前記映像及び前記ラベルを参照することによって、単離した造血細胞内の 前記腫瘍細胞の少なくとも2次元座標を決定すること、 e) 前記腫瘍細胞が実質的に表面の静止した位置にある間に、決定した座標で のラベルした腫瘍細胞の少なくとも1つに高エネルギーレーザ光パルスを適用す ることからなる方法。 19.前記適用工程が、前記高エネルギーレーザ光パルスを前記集団中のすべて のラベルした細胞へ適用することなる請求項18記載の方法。 20.前記レーザから放射した光の波長が375nmと900nmとの間である請求項18 又は19記載の方法。 21.前記適用工程が、コンピューターの補助で自動的に行なう請求項18〜2 0のいずれか1項に記載の方法。 22.第2細胞集団から第1細胞集団を除去する試験管内方法であって、 a) 前記第1の細胞集団を前記第2細胞集団から識別できるように前記第1の 細胞集団をラベルすること、 b) 前記第1の細胞集団及び前記第2の細胞集団を表面の静止した位置に置く こと、 c) 前記ラベルを参照することによって前記第1の細胞集団の中の1つの細胞 の位置を確定すること、 d) 制御したエネルギー源から第1のパルスを前記1つの細胞へ適用すること 、 e) 前記1つの細胞が前記第1のパルスによって殺されたかどうか決定するこ と、及び、 f) 前記1つの細胞が殺されなかったなら、制御したエネルギー源からの第2 のパルスを適用すること、からなる方法。 23.前記第1の細胞集団をラベルする方法が、腫瘍細胞をラベルすることから なる請求項22記載の方法。 24.第2の細胞集団から第1の細胞集団を除去する方法が、第2の非腫瘍細胞 集団から第1の腫瘍細胞集団を除去することからなる請求項22又は23記載の 方法。 25.第1のパルスを適用する方法が、レーザパルスを適用することからなる請 求項22〜24のいずれか1項に記載の方法。 26.第2のパルスを適用する方法が、レーザパルスを適用することからなる請 求項22〜25のいずれか1項に記載の方法。 27.非腫瘍細胞を含む細胞集団内から腫瘍細胞を除去する方法であって、 a)非腫瘍細胞を前記腫瘍細胞から識別できるように非腫瘍細胞をラベルする工 程、 b)前記細胞集団中の非ラベル細胞を同定することによって、前記腫瘍細胞の1 つの位置を確定する工程、及び、 c)前記腫瘍細胞が表面上の実質的に静止した位置にある間に、制御したエネル ギー源からのパルスを、前記位置を確定した腫瘍細胞へ適用することによって前 記腫瘍細胞を殺す工程、からなる方法。 28.前記腫瘍細胞を殺す工程が、レーザパルスを適用することからなる請求項 27記載の方法。 29.前記腫瘍細胞を殺す工程が、前記腫瘍細胞が名目上平らな表面の静止した 位置にある間、制御したエネルギー源を前記位置を確定した腫瘍細胞へ適用する ことからなる請求項27又は28記載の方法。 30.前記腫瘍細胞を殺す工程が、前記細胞集団が集合している間に制御したエ ネルギー源を位置を確定した腫瘍細胞へ適用することからなる請求項27〜29 のいずれか1項に記載の方法。
