Claims (1)
- 【特許請求の範囲】 1. 免疫グロブリンが非ヒト起源の抗原結合領域およびヒト起源の免疫グロブ リンの少なくとも一部を含有してなる、α4β7インテグリンに対する結合特異 性を有するヒト化免疫グロブリン。 2. ヒト起源の免疫グロブリンの一部がヒト定常領域に由来するものである請 求項1記載のヒト化免疫グロブリン。 3. ヒト定常領域がガンマ型のものである請求項2記載のヒト化免疫グロブリ ン。 4. 抗原結合領域が齧歯類起源のものである請求項2記載のヒト化免疫グロブ リン。 5. 抗原結合領域がAct−1モノクローナル抗体に由来するものである請求 項2記載のヒト化免疫グロブリン。 6. 抗原結合領域が齧歯類起源の相補性決定領域を含有し、ヒト起源の免疫グ ロブリンの一部がヒト枠組み領域に由来するものである請求項1記載のヒト化免 疫グロブリン。 7. 相補性決定領域がAct−1モノクローナル抗体に由来するものである請 求項6記載のヒト化免疫グロブリン。 8. α4β7を結合する非ヒト起源の抗体に由来する相補性決定領域およびヒ ト起源の軽鎖に由来する枠組み領域を含む軽鎖;ならびに α4β7を結合する非ヒト起源の抗体に由来する相補性決定領域およひヒト起 源の重鎖に由来する枠組み領域を含む重鎖 を含有してなる、α4β7インテグリンに対する結合特異性を有するヒト化免疫 グロブリン。 9. 前記免疫グロブリンがα4β7への結合に関してネズミAct−1と競合 することができるものである請求項8記載のヒト化免疫グロブリン。 10. 軽鎖がAct−1抗体の軽鎖に由来する3つの相補性決定領域を含有し 、重鎖がAct−1抗体の重鎖に由来する3つの相補性決定領域を含有してなる 請求項8記載のヒト化免疫グロブリン。 11. ヒト起源の軽鎖がGM607’CL抗体の軽鎖である請求項8記載のヒ ト化免疫グロブリン。 12. ヒト起源の重鎖がヒト21/28’CL抗体である請求項8記載のヒト 化免疫グロブリン。 13. ネズミAct−1抗体の軽鎖のCDR1、CDR2およびCDR3なら びにヒト軽鎖枠組み領域を含有してなるヒト化免疫グロブリン軽鎖。 14. ヒト枠組み領域がGM607’CL抗体の軽鎖に由来するものである請 求項13記載のヒト化免疫グロブリン軽鎖。 15. 配列番号:21の可変領域を含有してなる請求項14記載のヒト化免疫 グロブリン軽鎖。 16. 請求項15記載のヒト化免疫グロブリン軽鎖をコードする単離された核 酸。 17. 配列番号:20の可変領域コーディング配列を含有してなる請求項16 記載の単離された核酸。 18. Act−1抗体の重鎖のCDR1、CDR2およびCDR3ならびにヒ ト重鎖枠組み領域を含有してなるヒト化免疫グロブリン重鎖。 19. ヒト枠組み領域がヒト21/28’CL抗体の重鎖に由来するものであ る請求項18記載のヒト化免疫グロブリン重鎖。 20. 配列番号:19の可変領域を含有してなる請求項19記載のヒト化免疫 グロブリン重鎖。 21. 請求項20記載のヒト化免疫グロブリン重鎖をコードする単離された核 酸。 22. 配列番号:18の可変領域コーディング配列を含有してなる請求項21 記載の単離された核酸。 23. 軽鎖が図7(配列番号:12)に示された軽鎖可変領域アミノ酸配列の 少なくとも機能しうる部分を含有するアミノ酸配列を有する、ヒト化免疫グロブ リン軽鎖。 24. 軽鎖が図7(配列番号:12)に示されたシグナルペプチド配列および 図7(配列番号:12)に示された軽鎖可変領域アミノ酸配列の少なくとも機能 しうる部分を含有するアミノ酸配列を有する、請求項23記載のヒト化免疫グロ ブリン軽鎖。 25. 請求項23記載のヒト化免疫グロブリン軽鎖をコードする配列を含有し てなる単離された核酸。 26. 配列番号:11の可変領域コーディング配列を含有してなる請求項25 記載の単離された核酸。 27. 重鎖が図9(配列番号:15)に示された重鎖可変領域アミノ酸配列の 少なくとも機能しうる部分を含有するアミノ酸配列を有する、ヒト化免疫グロブ リン重鎖。 28. 重鎖が図9(配列番号:15)に示されたシグナルペプチド配列および 図9(配列番号:15)に示された重鎖可変領域アミノ酸配列の少なくとも機能 しうる部分を含有するアミノ酸配列を有する、請求項27記載のヒト化免疫グロ ブリン重鎖。 29. 請求項27記載のヒト化免疫グロブリン重鎖をコードする単離された核 酸。 30. 配列番号:14の可変領域コーディング配列を含有してなる請求項29 記載の単離された核酸。 31. 遺伝子がα4β7インテグリンに対する結合特異性を有する非ヒト抗体 の軽鎖に由来するCDRおよびヒト起源の軽鎖に由来する枠組み領域をコードす るヌクレオチド配列を含む、ヒト化免疫グロブリン軽鎖をコードする融合遺伝子 を含有してなる発現ベクター。 32. 非ヒト抗体がネズミAct−1抗体である請求項31記載の発現ベクタ ー。 33. 請求項31記載の発現ベクターを含有してなる宿主細胞。 34. 遺伝子がα4β7インテグリンに対する結合特異性を有する非ヒト抗体 の重鎖に由来するCDRおよびヒト起源の重鎖に由来する枠組み領域をコードす るヌクレオチド配列を含む、ヒト化免疫グロブリン重鎖をコードする融合遺伝子 を含有してなる発現ベクター。 35. 非ヒト抗体がネズミAct−1抗体である請求項34記載の発現ベクタ ー。 36. 請求項34記載の発現ベクターを含有してなる宿主細胞。 37. ヒト化免疫グロブリン軽鎖をコードする第1の組換え核酸およびヒト化 免疫グロブリン重鎖をコードする第2の組換え核酸を含有してなる宿主細胞であ って、 前記第1の核酸はネズミAct−1抗体の軽鎖に由来するCDRおよひヒト起 源の軽鎖に由来する枠組み領域をコードするヌクレオチド配列を含み;ならびに 、 前記第2の核酸はネズミAct−1抗体の重鎖に由来するCDRおよびヒト起 源の重鎖に由来する枠組み領域をコードするヌクレオチド配列を含む、宿主細胞 。 38. 請求項37記載の宿主細胞をヒト化免疫グロブリンの発現に適した条件 下で維持し、それによりヒト化免疫グロブリン鎖を発現させ、ヒト化免疫グロブ リンを産生する工程を含む、ヒト化免疫グロブリンの調製方法。 39. ヒト化免疫グロブリンの単離工程をさらに含む請求項38記載の方法。 40. a)ネズミAct−1モノクローナル抗体に由来する抗原結合領域をコ ードする第1の核酸配列;および b) ヒト起源の免疫グロブリンの定常領域の少なくとも一部をコードする第2 の核酸配列 を含有してなるヒト化免疫グロブリン軽鎖または重鎖をコードする融合遺伝子。 41. α4β7を産生する第1細胞を請求項1記載のヒト化免疫グロブリンの 有効量と接触させる工程を含む、前記第1細胞とそのリガンドを産生する第2細 胞との相互作用を阻害する方法。 42. 請求項1記載のヒト化免疫グロブリンの有効量を患者に投与する工程を 含む、粘膜組織の白血球浸潤を阻害する方法。 43. 請求項1記載のヒト化免疫グロブリンの有効量を患者に投与する工程を 含む、MAdCAM−1分子を発現する組織の白血球浸潤に関連する疾患の治療 方法。 44. 該疾患がMAdCAM分子を発現する腸に関連した内皮への白血球の結 合の結果としての組織の白血球浸潤に関連する疾患である、請求項43記載の方 法。 45. 請求項1記載のヒト化免疫グロブリンの有効量を患者に投与する工程を 含む、炎症性腸疾患の患者における治療方法。 46. 治療または診断における使用のための請求項1記載のヒト化免疫グロブ リン。 47. 組織の白血球浸潤に関連する疾患(例えば、炎症疾患)の治療における 使用のための請求項1記載のヒト化免疫グロブリン。 48. 炎症性腸疾患の治療における使用のための請求項1記載のヒト化免疫グ ロブリン。 49. 組織の白血球浸潤に関連する疾患(例えば、炎症疾患)の治療用の医薬 の製造のための請求項1記載のヒト化免疫グロブリンの使用。 50. 炎症性腸疾患の治療用の医薬の製造のための請求項1記載のヒト化免疫 グロブリンの使用。 51. 請求項1記載のヒト化免疫グロブリンおよび適当な担体を含有してなる 医薬組成物。
