Claims (1)
- 【特許請求の範囲】 1.オリゴヌクレオチドを核酸標的にハイブリダイズさせる方法であって、 部分的に標的に相補的な核酸配列を有するが標的に対して少なくとも1つの 本来のミスマッチと、標的に対して少なくとも1つの人工的ミスマッチを含み 、人工的ミスマッチおよび本来のミスマッチが異なるヌクレオチド位置にある オリゴヌクレオチドを提供するステップと、 前記オリゴヌクレオチドと前記標的を選択したハイブリダイゼーション条件 下で結合させて生成物を形成させるステップであって、前記ハイブリダイゼー ション条件が、前記オリゴヌクレオチドおよび本来のミスマッチを含む第1の 二重鎖が、前記オリゴヌクレオチドを含むが本来のミスマッチを欠く第2の二 重鎖よりも安定でないステップを含む方法。 2.人工的ミスマッチがユニバーサルミスマッチヌクレオシドを含む、請求項1 に記載の方法。 3.ユニバーサルミスマッチヌクレオシドが1-(2'-デオキシ-β-D-リボフラノシ ル)-3-ニトロピロールである、請求項2に記載の方法。 4.人工的ミスマッチおよび本来のミスマッチが3あるいは4ヌクレオチド位置 隔てられている、請求項1に記載の方法。 5.オリゴヌクレオチドが2個の人工的ミスマッチを含む、請求項1に記載の方 法。 6.オリゴヌクレオチドが10塩基隔てられた2個の人工的ミスマッチを含む、 請求項5に記載の方法。 7.本来のミスマッチが、標的に対する、核酸の置換、挿入、欠失、再構成から なる群より選ばれる、請求項1に記載の方法。 8.標的に相補的なオリゴヌクレオチドの部分が約150ヌクレオチド未満である 、 請求項1に記載の方法。 9.オリゴヌクレオチドを含む二重鎖を検出するステップをさらに含む、請求項 1に記載の方法。 10.オリゴヌクレオチドを含む二重鎖を検出するステップが、続くPCR増幅断 片の生成のモニタリング、オリゴヌクレオチドのタグ付き型のモニタリング、 表面プラズモン共鳴のモニタリング、エバネセンス波蛍光の測定からなる群よ り選ばれる、請求項9に記載の方法。 11.第1の二重鎖を優先的に破壊するステップを含む請求項9に記載の方法。 12.破壊するステップが、第1の二重鎖の破壊に好適な条件下で結合ステップ の産物を洗浄するステップを含む、請求項11に記載の方法。 13.第2の二重鎖を形成させた後に核酸断片を選択的に増幅させるステップを さらに含む、請求項1に記載の方法。 14.第2の二重鎖を形成させた後に核酸断片を選択的に伸長させるステップを 含む、請求項1に記載の方法。 15.核酸を含むテストサンプル中の、第1の核酸標的と第1の標的に対して配列 変異を有する第2の核酸標的間を区別するための方法であって、 第1の標的に対して部分的に相補的であるが第1の標的に対して配列変異位置 以外の位置に少なくとも1個の人工的ミスマッチを含んでいるオリゴヌクレオ チドを供給するステップと、 前記オリゴヌクレオチドと核酸を、選択したハイブリダイゼーション条件下 で結合させて生成物を形成させるステップであって、生成物が(a)オリゴヌ クレオチドと第1の標的とを含む第1の二重鎖、(b)オリゴヌクレオチドと第 2の標的を含み第1の二重鎖よりも安定でない第2の二重鎖、(c)第1の二重鎖 と第2の二重鎖の両方を含む混合物、からなる群より選ばれるものであるステ ップと、 第1の二重鎖または第2の二重鎖を選択的に検出するステップとを含む方法。 16.第1の二重鎖または第2の二重鎖を検出するステップが、続くPCR増幅断片 の生成のモニタリング、オリゴヌクレオチドのタグ付き型のモニタリング、表 面プラズモン共鳴のモニタリング、エバネセンス波蛍光の測定からなる群より 選ばれる、請求項15に記載の方法。 17.第2の二重鎖を優先的に破壊するステップを含む、請求項15に記載の方 法。 18.破壊するステップが、結合ステップの生成物を第2の二重鎖の破壊に好適 な条件下で洗浄するステップを含む、請求項17に記載の方法。 19.人工的ミスマッチがユニバーサルミスマッチヌクレオシドを含む、請求項 15に記載の方法。 20.ユニバーサルミスマッチヌクレオシドが1-(2'-デオキシ-β-D-リボフラノ シル)-3-ニトロピロールである、請求項19に記載の方法。 21.人工的ミスマッチ及び配列変異が3または4ヌクレオチド位置離れている 、請求項15に記載の方法。 22.オリゴヌクレオチドが2個の人工的ミスマッチを含む、請求項15に記載 の方法。 23.オリゴヌクレオチドが10塩基離れた2個の人工的ミスマッチを含む、請求 項22に記載の方法。 24.配列変異が、標的に対する、核酸の置換、挿入、欠失および再構成からな る群より選ばれる、請求項15に記載の方法。 25.第1の標的に相補的なオリゴヌクレオチドの部分が約150ヌクレオチド未満 である、請求項15に記載の方法。
