- 【特許請求の範囲】 1.コバラミン産生性原核細胞微生物の発酵によるコバラミンの生合成方法であ って、 O−ホスホ−L−トレオニンの生合成経路に関与する酵素をコードする少なく とも1つのDNAフラグメントによって形質転換された微生物を使用し、前記酵 素が発現しコバラミンが産生され得る条件下で前記微生物を培養する段階、及び /または、 前記微生物の培養培地にO−ホスホ−L−トレオニンを添加する段階、または、 5,6−ジメチルベンズイミダゾールの生合成経路に関与する酵素をコードす る少なくとも1つのDNAフラグメントによって形質転換された好気性微生物を 使用し、前記酵素が発現しコバラミンが産生され得る条件下で前記好気性微生物 を培養する段階、 から成ることを特徴とするコバラミンの生合成方法。 2.Rhodobacter capsulatusのbluE遺伝子及びbl uF 遺伝子を含みO−ホスホ−L−トレオニンの生合成経路に関与するポリペプ チドをコードするDNAフ ラグメントによって形質転換されるか、または、相同フラグメント、及び/また は、前記bluE遺伝子及びbluF遺伝子とハイブリダイズし且つO−ホスホ −L−トレオニンの生合成経路に関与する酵素をコードする機能を有しているフ ラグメントによって形質転換された微生物を使用することを特徴とする請求項1 に記載の方法。 3.Rhodobacter capsulatusのbluB遺伝子を含み5 ,6−ジメチルベンズイミダゾールの生合成経路に関与する酵素をコードするD NAフラグメントによって形質転換されるか、または、相同フラグメント、及び /または、前記bluB遺伝子とハイブリダイズし且つ前記遺伝子と同様に5, 6−ジメチルベンズイミダゾールの生合成経路に関与する酵素をコードする機能 を有しているフラグメントによって形質転換された微生物を使用することを特徴 とする請求項1に記載の方法。 4.前記DNAフラグメントが、プラスミドpER1の6.8kbのBamHI フラグメントから成ることを特徴とする請求項2または3に記載の方法。 5.前記DNAフラグメントが、プラスミドpER2の2.1 kbのEcoRI/ClaIフラグメントから成ることを特徴とする請求項2に 記載の方法。 6.前記DNAフラグメントが、プラスミドpAHW25またはプラスミドpE R2の1.6kbのEcoRI/EcoRVフラグメントから成ることを特徴と する請求項2に記載の方法。 7.請求項1から5のいずれか一項に記載のビタミンB12の製造方法。 8.前記微生物がPseudomonas denitrificansまたはAgrobacterium radiobacter の菌株であることを特徴 とする請求項1から7のいずれか一項に記載の方法。 9.遺伝子工学技術を用い、請求項1から6のいずれか一項に記載のO−ホスホ −L−トレオニンまたは5,6−ジメチルベンズイミダゾールの生合成経路に関 与する酵素をコードする少なくとも1つのDNAフラグメントによって微生物を 形質転換させることを特徴とするコバラミン産生性原核細胞微生物の組換え菌株 の製造方法。 10.請求項9に記載の方法によって得られた組換え微生物。 11.Pseudomonas denitrificans またはAgrobacterium radiobacterの菌株であること を特徴とする請求項10に記載の微生物。
