Abstract
Description
Claims (1)
- 【特許請求の範囲】 1.次の工程からなる哺乳類対象中の生物適合体の局在を検出する非侵襲的方 法: (a) その対象に、その生物適合体と発光成分の複合体を投与し、 (b) その複合体がその対象内で局在を達成しうる期間の後、その対象を光検出 装置の検出フィールド内に固定し、 (c) その対象を固定状態に維持し、 (d) 当該維持中に、その対象中に局在した発光成分からの光子放射を、光子放 射の画像を構築することができるまで、その光検出装置で測定し、 (e) その画像を構築する。 2.工程(b)から(e)までを選択した間隔で反復することをさらに含み、該反復 が対象中の当該生物適合体の局在を経時的に追跡するのに有効である請求項1の 方法。 3.該測定が増倍化電荷結合光検出素子を用いて行われる請求項1の方法。 4.該投与が腫瘍細胞ターゲッティング成分を含有する複合体の投与を含む、 哺乳類対象中の腫瘍細胞の局在を検出するための請求項1の方法。 5.該投与が炎症ターゲッティング成分を含有する複合体の投与を含む、哺乳 類対象中の炎症の局在を検出するための請求項1の方法。 6.該投与が感染ターゲッティング成分を含有する複合体の投与を含む、哺乳 類対象中の病原体による感染の局在を検出するための請求項1の方法。 7.該投与が血栓性プラークターゲッティング成分を含有する複合体の投与を 含む、哺乳類対象内の血栓性プラークの局在を検出するための請求項1の方法。 8.該投与が発光成分を含有する粒子である複合体の投与を含む請求項1の方法 。 9.動物モデル中の病原体による感染症の局在を検出するための請求項1の方法 であって、生物適合体がその病原体である方法。 10.病原体がサルモネラである請求項9の方法。 11.発光成分が発光性タンパク質である請求項1の方法。 12.タンパク質がルシフェラーゼ、黄色蛍光タンパク質およびフェレドキシン IVからなる群より選択される請求項11の方法。 13.該投与が、抗体断片と発光性タンパク質の融合タンパク質である複合体の 投与を含む請求項11の方法。 14.生物適合体が形質転換細胞であり、発光成分がその細胞から発現する異種 遺伝子の産物である請求項11の方法。 15.異種遺伝子の発現が活性化可能なプロモーターの制御下にある請求項14の 方法。 16.哺乳類対象中の生物適合体のレベルを経時的に検出する非侵襲的方法であ って、 (a) その対象に、その生物適合体と発光成分の複合体を投与し、 (b) その対象を光検出装置の検出フィールド内に置き、 (c) その対象をその装置の検出フィールド中に維持し、 (d) 当該維持中に、その対象中の発光成分からの光子放射を、その光検出装置 で測定し、 (e) (b)から(d)までの工程を、選択した間隔で反復する ことからなり、該反復が、その対象中の当該生物適合体のレベルの経時変化を検 出するのに有効である方法。 17.次の工程からなる哺乳類対象における導入遺伝子の組み込みを検出する非 侵襲的方法: (a) その対象に、哺乳類細胞中に有効に組み込まれ、かつ、導入遺伝子、発光 性タンパク質をコードする遺伝子および活性化可能なプロモーターを含有するベ クター構築物(ただし、該発光性タンパク質をコードする遺伝子は該プロモータ ーの制御下にある)を投与し、 (b) その構築物が組み込みを達成しうる期間の後、該プロモーターを活性化し 、 (c) その対象を光検出装置の検出フィールド内に置き、 (d) その対象をその光検出装置の検出フィールド内に維持し、 (e) 当該維持中に、その対象中の、発現した発光性タンパク質からの光子放射 のレベルを、その光検出装置で測定し、 (f) その光子放射のレベルがバックグラウンドより有意に高ければ、その導入 遺伝子の組み込みを確認する。 18.次の工程からなる動物中のプロモーター誘導事象の局在を検出する非侵襲 的方法: (a) その事象に反応する誘導性プロモーターとその制御下にある発光性タンパ ク質をコードする異種遺伝子とを持つトランスジェニック動物中で、その事象を 誘発し、 (b) その動物を光検出装置の検出フィールド内に置き、 (c) その動物を固定状態に維持し、 (d) 該維持中に、その動物内に局在する発現した発光性タンパク質からの光子 放射を、光子放射の画像を構築することができるまで、その光検出装置で測定し 、 (e) その画像を構築する。 19.発光性タンパク質を発現させる遺伝子で形質転換されたサルモネラ。
