Claims (1)
- 【特許請求の範囲】 1.基本マトリックスおよび、システインを含む組み換えプロテインAを提示 するマトリックス結合基を含み、該基が式 −B−X−rProtein A-cys I 〔式中、Bは基本マトリックスに結合する橋およびXはrProtein A-cys由来の ヘテロ原子NまたはSを含む〕 に従い、Xがチオエーテル硫黄(−S−)および/または2級アミン(−NH−)で あることを特徴とする、分離媒体。 2.橋−B−がエーテルにより基本マトリックスに結合し、それが所望により 1個またはそれ以上の酸素/窒素原子で中断され、1個またはそれ以上のアミノ またはヒドロキシ基で置換されていてもよい、直鎖または環状飽和炭化水素鎖を 含み、1個および同じ炭素原子が最大1個の酸素原子または窒素原子と結合して いる、請求項1記載の分離媒体。 3.システインが末端ペプチドリンカーに含まれることを特徴とする、請求項 1−2に記載の分離媒体。 4.ペプチドリンカーがC−末端であることを特徴とする、請求項1−3のい ずれかに記載の分離媒体。 5.システインがrProtein A-cysのC−末端アミノ酸であることを特徴とす る、請求項1−4のいずれかに記載の分離媒体。 6.基本マトリックスがポリヒドロキシポリマー、好ましくは不溶性ポリサッ カライドであることを特徴とする、請求項1−5のいずれかに記載の分離媒体。 7.C−末端残基がシステインであることを特徴とする、組み換えプロテイン A−cys。
