Abstract
Un proceso para la producción de un producto alcohólico que comprende las fases secuenciales de: (a) proporcionar una pasta que comprende agua y almidón granular, (b) mantener dicha pasta en presencia de una alfa-amilasa ácida y una glucoamilasa a una temperatura de 0ºC a 20ºC por debajo de la temperatura de gelatinización inicial de dicho almidón granular durante un periodo de 5 minutos a 12 horas, (c) mantener dicha pasta en presencia de una alfa-amilasa ácida y una glucoamilasa y una levadura a una temperatura entre 10ºC y 35ºC durante un periodo de 20 a 250 horas para producir etanol y, (d) opcionalmente, recuperar el etanol.
Claims (37)
1. Un proceso para la producción de un producto
alcohólico que comprende las fases secuenciales de:
- (a)
- proporcionar una pasta que comprende agua y almidón granular,
- (b)
- mantener dicha pasta en presencia de una alfa-amilasa ácida y una glucoamilasa a una temperatura de 0ºC a 20ºC por debajo de la temperatura de gelatinización inicial de dicho almidón granular durante un periodo de 5 minutos a 12 horas,
- (c)
- mantener dicha pasta en presencia de una alfa-amilasa ácida y una glucoamilasa y una levadura a una temperatura entre 10ºC y 35ºC durante un periodo de 20 a 250 horas para producir etanol y,
- (d)
- opcionalmente, recuperar el etanol.
2. El proceso según la reivindicación 1, donde el
producto es etanol combustible, etanol potable y/o etanol
industrial.
3. El proceso según cualquiera de las
reivindicaciones precedentes en el que la temperatura en la fase
(c) es de entre 28ºC y 36ºC, como entre 29ºC y 35ºC, como entre
30ºC y 34ºC, como alrededor de 32ºC.
4. El proceso según la reivindicación 1, en el
que el producto es una cerveza.
5. El proceso según cualquiera de las
reivindicaciones 1 o 4 en el que la temperatura en la fase (c) es
de entre 11ºC y 17ºC, como entre 12ºC y 16ºC, como entre 13ºC y
15ºC, como alrededor de 14ºC.
6. El proceso según cualquiera de las
reivindicaciones precedentes en el que las actividades de la
alfa-amilasa ácida y de la glucoamilasa son añadidas
en la fase (b) y/o (c) en una proporción de entre 0.30 y 5.00
AFAU/AGU.
7. El proceso según cualquiera de las
reivindicaciones precedentes en el que la
alfa-amilasa ácida es una
alfa-amilasa ácida micótica.
8. El proceso según cualquiera de las
reivindicaciones precedentes en el que la
alfa-amilasa ácida micótica se obtiene de una
cepa de Aspergillus, preferiblemente de una cepa de
Aspergillus Niger o de una cepa de una cepa de
Aspergillus oryzae.
9. El proceso según cualquiera de las
reivindicaciones en el que la alfa-amilasa ácida es
una alfa-amilasa ácida que tiene una secuencia de
aminoácidos que tiene al menos el 70%, preferiblemente al menos el
75%, 80%, 85% o al menos el 90%, p. ej. al menos el 95%, al menos
el 97%, al menos el 98%, o al menos el 99% de homología con la SEC
ID NO: 1.
10. El proceso según cualquiera de las
reivindicaciones precedentes en el que la glucoamilasa se obtiene
de una cepa de Aspergillus, Talaromyces o
Clostridium.
11. El proceso según cualquiera de las
reivindicaciones precedentes en el que la glucoamilasa se obtiene
de una cepa de Aspergillus Niger.
12. El proceso según cualquiera de las
reivindicaciones precedentes en el que la
alfa-amilasa ácida es una
alfa-amilasa ácida bacteriana.
13. El proceso según cualquiera de las
reivindicaciones precedentes en el que la
alfa-amilasa ácida deriva de una cepa de
alfa-amilasa de B. licheniformis, B.
amyloliquefaciens, y B. stearothermophilus.
14. El proceso según cualquiera de las
reivindicaciones precedentes en el que la
alfa-amilasa ácida bacteriana deriva de una cepa de
alfa-amilasa de B. lichenfformis, B.
ayiloliquefaciens, y B. stearothermophilus.
15. El proceso según cualquiera de las
reivindicaciones precedentes en el que la
alfa-amilasa ácida bacteriana está derivada de una
cepa de Bacillus stearothermophilus, que tiene las
mutaciones 1181* + G182* + N193F en comparación con la secuencia de
aminoácidos de tipo salvaje expuesta en la SEC ID NO:2.
16. El proceso según cualquiera de las
reivindicaciones precedentes en el que la
alfa-amilasa ácida bacteriana es una
alfa-amilasa híbrida que comprende 445 residuos de
aminoácidos C-terminales de la
alfa-amilasa de Bacillus licheniformis
expuesta en la SEC ID NO:3 y los 37 residuos de aminoácidos
N-terminales de la alfa-amilasa
derivada de Bacillus amyloliquefaciens expuesta en la SEC ID
NO:4, que tiene la sustitución G48A + T491 + G107A + H 156Y + A181
T + N 190F + 1201F + A209V + Q264S.
17. El proceso según cualquiera de las
reivindicaciones precedentes en el que la
alfa-amilasa ácida bacteriana es una
alfa-amilasa que tiene la secuencia de aminoácidos
expuesta en la SEC ID NO:4, que tiene las mutaciones H154Y, A181T,
N190F, A209V, Q264S, y/o la supresión de dos residuos entre las
posiciones 176 y 179, preferiblemente la supresión de E178 y
G179.
18. El proceso según cualquiera de las
reivindicaciones precedentes, en el que la actividad de la
alfa-amilasa ácida está presente en una cantidad de
50-500 AFAU/Kg de DS (sólidos secos).
19. El proceso según cualquiera de las
reivindicaciones precedentes, en el que la actividad de
glucoamilasa está presente en una cantidad de 20-200
AGU/Kg de DS.
20. El proceso según cualquiera de las
reivindicaciones precedentes, en el que la proporción entre la
actividad de la alfa-amilasa ácida y actividad de la
glucoamilasa está entre 0.35 y 5.00 AFAU/AGU.
21. El proceso según cualquiera de las
reivindicaciones precedentes, en el que la fase (b) y/o la fase
(C) se realizada en presencia de una actividad enzimática
seleccionada de la lista consistente en xilanasa, celulasa y
fitasa.
22. El proceso de la reivindicación precedente,
en el que la pasta de almidón tiene 5-60% de
almidón granular de DS, preferiblemente 10-50% de
almidón granular de DS, más preferiblemente 20-40%
de DS, especialmente alrededor del 30% de almidón granular de
DS.
23. El proceso de cualquiera de las
reivindicaciones precedentes, en el que el pH durante la fase (b)
y/o (c) está en la gama de 3.0 a 7.0, preferiblemente de 3.5 a 6.0,
o más preferiblemente de 4.0-5.0, como de 4.3 a
4.6.
24. El proceso de cualquiera de las
reivindicaciones precedentes, en el que el almidón granular se
obtiene a partir de tubérculos, raíces, tallos, frutos, semillas o
del grano entero.
25. El proceso de cualquiera de las
reivindicaciones precedentes, en el que el almidón granular se
obtiene a partir de maíz, mazorcas, trigo, cebada, centeno, milo,
sagú, yuca, mandioca, tapioca, sorgo, arroz o patatas.
26. El proceso de cualquiera de las
reivindicaciones precedentes, en el que el almidón granular se
obtiene a partir de cereales.
27. El proceso de cualquiera de las
reivindicaciones precedentes, en el que el almidón granular se
obtiene a partir de la molienda en seco o molienda húmeda del grano
entero.
28. El proceso según cualquiera de las
reivindicaciones precedentes en el que el tiempo de mantenimiento
en la fase (b) es de 5 minutos a 12 horas, preferiblemente de 10
minutos a 6 horas, más preferiblemente de 15 minutos a 3 horas,
incluso más preferiblemente de 20 minutos a 1 ½ hora, como de 30
minutos a 1 ¼ hora, de 40 a 70 minutos, e incluso de 50 a 60
minutos.
29. El proceso según cualquiera de las
reivindicaciones precedentes, en el que el tiempo de mantenimiento
en la fase (c) es de 25 a 190 horas, preferiblemente de 30 a 180
horas, más preferiblemente de 40 a 170 horas, incluso más
preferiblemente de 50 a 160 horas, aún más preferiblemente de 60 a
150 horas, incluso aún más preferiblemente de 70 a 140 horas, y más
preferiblemente de 80 a 130 horas.
30. El proceso según cualquiera de las
reivindicaciones precedentes, en el que la temperatura en la fase
(b) es de 45ºC a 75ºC.
31. Una composición enzimática que comprende la
actividad de la alfa-amilasa ácida y la actividad
de la glucoamilasa en una proporción de entre 0.30 y 5.00 AFAU/AGU,
en la que una actividad enzimática adicional está presente; dicha
actividad enzimática está seleccionada de la lista consistente en
celulasa, xilanasa y fitasa.
32. Uso de una composición enzimática según la
reivindicación precedente para un proceso de un producto alcohólico
o un proceso de hidrólisis de almidón.
33. Uso de una composición enzimática que
comprende la actividad de la alfa-amilasa ácida y
la actividad de la glucoamilasa en una proporción de entre 0.30 y
5.00 AFAU/AGU, en un proceso para un producto alcohólico que
comprende la hidrólisis enzimática del almidón granular.
34. Un proceso de maceración que comprende la
aplicación de una alfa-amilasa ácida.
35. El proceso según la reivindicación
precedente, en el que la alfa-amilasa ácida está
derivada de un hongo dentro del Aspergillus, preferiblemente
de A.niger.
36. El proceso según cualquiera de las
reivindicaciones precedentes, en el que la
alfa-amilasa ácida tiene al menos el 50%, al menos
el 60%, al menos el 70%, al menos el 80%, al menos el 90% de
homología con la secuencia de aminoácidos mostrada en la SEC ID
NO:1
37. El proceso según cualquiera de las
reivindicaciones precedentes, que comprende:
- a.
- formar un macerado que comprenda entre el 5% y el 100% de malta de cebada (w/w de la malta molida);
- b.
- antes de, durante o después de a) añadir al menos una enzima seleccionada de la lista que comprende: una enzima generadora de una proteasa (E.C. 3.4.), celulasa (E.C. 3.2.1.4) y maltosa.
- c.
- alcanzar en 15 minutos a) una temperatura de incubación inicial de al menos 70ºC;
- d.
- después de c) incubar el macerado a una temperatura de al menos 70ºC durante un periodo de tiempo suficiente para conseguir una recuperación del extracto de al menos el 80%; y,
- e.
- separar el mosto de los granos de bagazo.
