N-Methylimidazol
Die Erfindung betrifft ein Verfahren zur Herstellung des Gadoliniumkomplexes von N-(1 -Hydroxymethyl-2,3-dihydroxypropyl)-1 ,4,7-thscarboxymethyl-1 ,4,7,10-tetra- azacyclododecans„Gadobutrol = Gadovist®" im Eintopfverfahren mittels DMF-acetal und N-Methylimidazol.
Gadovist ist ein Gadolinium-haltiges Kontrastmittel für die Kernspin-Tomographie und ist seit 2000 in Deutschland in der Indikation„Kontrastverstärkung bei der kranialen und spinalen Magnetresonanztomografie" zugelassen.
Das MRT-Kontrastmittei Gadovsst® 1.Ö zählt zu den neueren Entwicklungen im Bereich der Gadoiinium-haitigen MR- ontrastmitte! (EP 0448191 B1 ). Es wird bei Untersuchungen eingesetzt, die eine hohe Kontrastmittel Konzentration erfordern - z.B. zur Diagnose des Schlaganfalls sowie zur Untersuchung von Gefäßen, z.B. in Tumoren.
Die Kontrastgebende Wirkung beruht auf Gadobutrol, einem nichtionischen Komplex bestehend aus Gadolinium(lll) und dem makrozyklischen Liganden Dihydroxy- hydroxy-methylpropyl-tetraazacyclododecan-triessigsäure (Butrol).
Gadobutrol führt bei den klinisch empfohlenen Dosierungen zu einer Verkürzung der Relaxationszeiten von Protonen des Gewebewassers.

versch iedenen Wegen mögl ich. Trotz erzielter Fortsch ritte gegen ü ber den u rsprü ng l ichen Verfa h ren besteht weiterh i n e i n Bed arf fü r vor a l l em i m Betriebsmaßstab durchzuführende umweltfreundlichere und kostengünstigere Synthesemögl ich keiten .
Es wurde gefunden, dass überraschenderweise ausgehend von Cyclen (1 ,4,7,10-Tetraazacyclododecan) der Formel 1 (DE19608307), das es mittlerweise zu sehr günstigen Konditionen zu kaufen gibt, spezifikationskonformes Gadobutrol in hoher Ausbeute ohne Isolierung von Intermediaten hergestellt werden kann und somit den Verfahren,

die mit Zwischen-Isolierung auf Zwischenaufreinigung arbeiten deutlich überlegen sind, vor allem was den Durchsatz und die Produktionszeit betrifft. Das erfinderische Verfahren ist dem nächstliegenden Stand der Technik (Inorg. Chem. 1997, 36, 6086- 6093 und DE 19724186.7), sowie dem in EP 1343770 B1 beschriebenen Verfahren, bei dem der Butrol-Ligand als Lithium-Komplex isoliert ist, deutlich überlegen.
Ausgehend von Cyclen wird, wie in der Schrift EP 0596586 B1 beschrieben, mit 4,4- Dimethyl-3,5,8-trioxabicyclo-(5.1 .0)-octan umgesetzt, anschließend durch Zugabe von Wasser und Lithiumhydroxyd die Formylzwischestufe verseift, und mit Chloroder Bromessigsäure umgesetzt, wobei als Basenfänger Lithiumhydroxyd oder N- Methylimidazol dienen, im selben Topf mit Salzsäure oder Bromwasserstoffsäure sauer gestellt und mit Gadolinium komplexiert. Nach Umdestillieren auf Ethanol oder Isopropanol fällt der Gadolinium-Komplex aus, es wird abfiltriert und nach kurzer Zwischen-Reinigung des Reaktions-Rührwerks direkt vom Filter (noch feucht) mit Wasser gelöst und ins Rührwerk zurückgespült, um eine finale Kristallisation aus Ethanol durchzuführen . Das Verfahren verwendet keine Ionenaustauscher und vermeidet auch die Zwischen-Isolierung vom Butrol-Liganden in freier oder auch in als Lithium-Komplex vorliegender Form, wie in EP 1343770 B1 beschrieben ist.
D ie Vorte i le d ieses Verfahrens sind hoher Durchsatz ohne Isol ierung und Zwischenreinigung von Intermediaten unter Verwendung von milden Basen wie Lithiumhydroxyd oder N-Methylimidazol. Lithiumsalze im Verfahren können mit Vorteil zurück gewonnen werden und anschließend in den Produktions-Kreislauf wieder eingespeist werden. Die Abfall-Bilanz ist gegenüber der Stand der Technik Verfahren noch günstiger, da alles in einem „Topf" läuft, dadurch entfallen Mutterlaugenaufarbeitungen, Reinigung von Filterapparaten etc. Durch eine genaue Gehaltsbestimmung des Ligandengehaltes vor Gadolinium-Komplexierung gelingt es, gadoliniumhaltige Abwässer zu vermeiden, da die Gadoliniummenge so dosiert werden kann, dass alles Metall vom Butrol-Liganden komplexiert wird. Das Verfahren erlaubt es mit einem Rührwerk und einem Filterapparat auszukommen . Eine Zwischenreinigung erfolgt lediglich mit Wasser, es braucht nicht getrocknet werden, sondern es kann direkt der nächste Ansatz gefahren werden. Dieses gewährleistet eine optimale Geräte-Ausnutzung und ermöglicht eine semikontinuierliche Fahrweise. Durch dieses neue erfinderische Verfahren ist es gelungen den Herstellungspreis für Gadobutrol nochmals signifikant zu reduzieren.
Das neue erfinderische Verfahren wird wie folgt angewendet:
Gadobutrol wird hergestellt, indem man Cyclen wie in EP 0596586 beschrieben mit 4,4-Dimethyl-3,5,8-trioxabicyclo-(5.1 .0)-octan bei Temperaturen zwischen 80 bis 200°C, bevorzugt bei 100-140°C , 8-40 h, bevorzugt 12-30 h umsetzt, anschließend mit Wasser aufnimmt und durch Zugabe von 1 bis 5 eq. Lithiumhydroxyd die Formyl- Zwischenstufe bei 50-100°C, bevorzugt bei 100°C, 2-24 h, bevorzugt 8-16 h verseift, anschließend mit Chlor-oder Bromessigsäure versetzt, bevorzugt Chloressigsaure und Lithiumhydroxyd bei Temperaturen zwischen 40-150°C, bevorzugt bei 40-90°C, einem pH-Wert zwischen 8-14, bevorzugt bei pH-Wert 9-13, innerhalb von 0,5 bis 24 h, bevorzugt 1 bis 6 Stunden umsetzt. Anschließend m it Salzsäu re oder Bromwasserstoffsäure auf pH 1 -4,5 , bevorzugt 2,0 - 4,0 stel lt, 0,5 bis 24 h , bevorzugt 0,5 - 5 h bei 20-100°C, bevorzugt bei 30-70°C rührt, anschließend eine Gehaltsbestimmung auf Butrol-Ligand durchführt und dann die stöchiometrische Menge eines Gadol in iu msalzes, wie Gadol in iu moxid , Gadol in iu mcarbonat, Gadoliniumchlorid, bevorzugt aber Gadoliniumoxid zusetzt und anschließend 1 bis 12 h, bevorzugt 1 -5 h bei 50-100°C, bevorzugt bei 70-100°C rührt. Nach beendeter Komplexierung wird durch Zugabe von Lithiumhydroxyd (fest oder als aqu. Lösung) auf pH 4-8, bevorzugt aber auf 6-7,5 gestellt. Anschl ießend wird im Vakuum
weitgehend eingeengt und in der H itze bei 70-80°C Ethanol oder Isopropanol, bevorzugt Ethanol zugesetzt und gegebenenfalls Wasser azeotrop abdestilliert. Man destilliert gegebenenfalls solange bis man einen Wasserwert von 1 -20 %, bevorzugt 5-10% erreicht hat Unter diesen Bedingungen fällt das Produkt Gadobutrol bereits in der Hitze aus. Man kühlt auf 0 bis 30°C, bevorzugt 5-20°C ab und filtriert vom Produkt ab. Das noch filterfeuchte Produkt wird mit wenig Wasser bei 20-60°C, bevorzugt 20-40°C vom Filter gelöst und final aus Ethanol umkristallisiert. Hierzu wird das Wasser gegebenenfalls azeotrop weitgehend entfernt, wobei das Produkt in der Siedehitze ausfällt. Es wird auf 0 bis 20°C abgekühlt, das Produkt abfiltriert und mit wenig kaltem Ethanol (bevorzugt 0 bis 20°C) nachgewaschen und anschließend getrocknet.
Ein so erhaltenes Produkt zeichnet sich durch hohe Qualität und Reinheit aus und entspricht den geforderten Ansprüchen der Spezifikation.
Beispiel
Zu 24,0 kg (139,34 mol) Cyclen (= 1 ,4,7,10 Tetraazacyclododecan) in 200 I Toluol gibt am unter Stickstoff 20 I Dimethylformamiddimethylacetal (DMF-acetal). Es wird langsam hochgeheitzt und das Azeotrop aus Methanol/Dimethylam in/Toluol abdestilliert. Anschließend wird unter Vakuum das Lösungsmittel vollständig abdestilliert. Das zurückbleibende Öl lässt man auf 50°C abkühlen und gibt dann 22,44 kg (147,86 mol) 4,4-Dimethyl-3,5,8-trioxabicyclo-(5.1 .0)-octan (ca. 95 %ig vom Gehalt) zu (ebenfalls unter Stickstoff). Dann wird 12 h bei 130°C Manteltemperatur gerührt. Man kühlt auf 40°C ab und gibt 200 I Wasser und 17,53 kg ( 418,0 mol) Lithiumhydroxid-monohydrat zu. Man kocht 8 h bei unter Rückfluss , anschließend wird ca. 140 I Wasser im Vakuum abdestilliert, auf Raumtemperatur abgekühlt und weiterverarbeitet.
46,66 kg (493,83 mol) Chloressigsäure werden in 50 kg Wasser gelöst und auf 5 °C abgekühlt. Zu dieser Lösung gibt man 20,73 kg (494,1 mol) Lithiumhydroxid- monohydrat zu . Anschl ießend wird d ie so hergestel lte Lösung zu der oben beschriebenen Lösung zugeben. Das Gemisch wird auf ca. 65°C Innentemperatur erwärmt und bei dieser Temperatur innerhalb von 2 h insgesamt 12,0 kg (286,1 mol) Lithiumhydroxid-monohydrat (ca. 5-6 Portionen) oder die äquivalente Menge an N-Methylimidazol zugesetzt. Danach wird 1 h bei 65 °C nachgerührt. Man stellt mit
konzentrierter Salzsäure auf pH 1 und rührt 30 Minuten bei 65 °C. Man kühlt auf 20°C ab, stellt mit Lithiumhydroxid-monohydrat auf pH 3,5 und führt anschließend eine Gehaltsbestimmung des Butrol-Liganden (= N-(1 -Hydroxymethyl-2,3- dihydroxypropyl)-1 ,4,7-thscarboxymethyl-1 ,4,7,10-tetra-azacyclododecan) per HPLC gegen externen Standard durch. Es ergibt sich umgerechnet ein Gehalt von 94,7 %. Anschließend gibt man 23,92 kg (65,97 mol) Gadoliniumoxid zu und rührt 1 h bei 90°C. Nach beendeter Komplexierung (aus der ursprünglichen Suspension wird eine klare Lösung) wird durch Zugabe von Lithiumhydroxid-monohydrat auf pH 7,0 gestellt. Man destilliert im Vakuum Wasser ab, solange bis eine noch rührbare viskose Lösung im Rührwerk übrig bleibt. Zu dieser Lösung gibt man in der Hitze (ca. 80°C) 1350 I Ethanol kocht 5 Stunden unter Rückfluss. Man kühlt auf 10°C ab und filtriert die ausgefallene Kristall-Suspension ab, wäscht 2 mal mit 100 I Ethanol nach. Der noch Ethanol-feuchte Filterkuchen wird auf dem Filter in 75 I Wasser gelöst und die Lösung über eine Filterkerze filtriert. Anschließend wird 750 I Ethanol zugegeben und 5 h unter Rückfluss erhitzt. Man kühlt auf 10°C ab und filtriert die ausgefallene Kristall-Suspension ab, wäscht 2 mal mit 75 kg Ethanol nach und trocknet im Vakuum bei 60°C
Ausbeute: 78,89 kg = 130,46 mol , dieses entspricht 84,6 % der Theorie, bezogen auf eingesetztes 1 ,4,7,10-tetraaza-cy c l o d o d e c a n ( a u f Wa s s e r u n d Restlösungsmittel korrigiert ) farbloses Kristallpulver.
Wassergehalt (Karl-Fischer): 4,12 %
Trockenverlust : 1 ,15 %
Elementaranalyse (auf Wasser korrigiert):

Claims
1 . Verfahren zur Herstellung des Gadoliniumkomplexes von N-(1 -Hydroxymethyl- 2,3-dihydroxypropyl)-1 ,4,7-thscarboxymethyl-l ,4,7,10-tetra-azacyclo- dodecans, indem man Cyclen m it 4,4-Dimethyl-3,5,8-trioxabicyclo-(5.1 .0)- octan und Dimethylformamid-dimethylacetal zur Reaktion bringt und durch Zugabe von Lithiumhydroxyd die Formyl-Zwischenstufe verseift, mit Chloroder Bromessigsäure versetzt und Lithiumhydroxyd oder N-Methylimidazol zur Reaktion bringt und mit Salzsäure oder Bromwasserstoffsäure auf einen sauren pH Wert einstellt, anschließend eine Gehaltsbestimmung auf Butrol- Ligand durchführt und die stöchiometrische Menge eines Gadoliniumsalzes zusetzt.
2. Verfahren nach Anspruch 1 , indem man Cyclen mit 4,4-Dimethyl-3,5,8- trioxabicyclo-(5.1 .0)-octan und Dimethylformamiddimethylacetal bei Temperaturen zwischen 80 bis 200°C zur Reaktion bringt und durch Zugabe von 1 bis 5 eq. Lithiumhydroxyd die Formyl-Zwischenstufe bei 50-100°C innerhalb von 2-24 Stunden verseift, mit Chlor-oder Bromessigsäure versetzt und Lithiumhydroxyd oder N-Methylimidazol bei Temperaturen zwischen 40- 150°C zur Reaktion bringt und anschließend mit Salzsäure oder Bromwasserstoffsäure auf einen pH Wert zwischen 1 - 4,5 einstellt, anschließend eine Gehaltsbestimmung auf Butrol-Ligand durchführt und die stöchiometrische Menge Gadoliniumoxid, Gadoliniumcarbonat oder Gadoliniumchlorid zusetzt und 1 bis 12 Stunden bei 50 -100°C rührt.
3. Verfahren nach Anspruch 1 oder 2, indem mit 4,4-Dimethyl-3,5,8-trioxabicyclo- (5.1 .0)-octan bei Temperaturen zwischen 80 bis 200°C, 8-40 h, bevorzugt 12 - 30 h umgesetzt wird, in Wasser löst und durch Zugabe von 1 bis 5 eq. Lithiumhydroxyd die Formyl-Zwischenstufe bei 50-100°C, bevorzugt bei 100°C, 2 - 24 h zur Reaktion bringt, anschließend mit Chlor- oder Bromessigsäure versetzt, und Lithiumhydroxyd oder N-Methylimidazol bei Temperaturen zwischen 40-150°C, bevorzugt bei 40-90°C, einem pH-Wert zwischen 8-14, bevorzugt bei pH-Wert 9-13, innerhalb von 0,5 bis 24 h, bevorzugt 1 bis 6 Stunden umsetzt, mit Salzsäure oder Bromwasserstoffsäure auf pH 1 -4,5 , bevorzugt 2,0 - 4,0 stellt, 0,5 bis 24 h, bevorzugt 0,5 - 5 h bei 20-100°C rührt, anschließend eine Gehaltsbestimmung auf Butrol-Ligand durchführt und die stochiometrische Menge eines Gadoliniumsalzes zusetzt und 1 bis 12 h bei 50-100°C, bevorzugt bei 70-100°C rührt, nach beendeter Komplexierung durch Zugabe von Lithiumhydroxyd auf pH 4-8, bevorzugt aber auf 6-7,5 stellt, anschließend im Vakuum einengt und in der Hitze bei 70-80°C Ethanol oder Isopropanol zugesetzt und gegebenenfalls Wasser azeotrop, bis zu einem Wasserwert von 1 -20 %, bevorzugt 5-10% abdestilliert, man kühlt auf 0 bis 30°C, bevorzugt 5-20°C ab , filtriert vom Produkt ab und kristallisiert aus Ethanol um.
