Abstract
La presente invención está relacionada con la preparación de péptidos insulinotrópicos que se sintetizan utilizando una aproximación en fase sólida y en solución ("híbrida"). En general, la aproximación incluye la síntesis de tres fragmentos de péptido intermediarios diferentes utilizando la química de fase sólida. Entonces se utiliza la química de fase en solución para añadir material de aminoácidos adicional a uno de los fragmentos. Entonces se acoplan los fragmentos en la fase en solución sólida. La utlización de una pseudoprolina en uno de los fragmentos síntesis en fase sólida de ese fragmento y también facilita el subsiguiente acoplamiento en fase en solución de este fragmento con otros fragmentos. La presente invención es muy útil para la formación de péptidos insulinotrópicos como el GLP-1 7-36) y sus homólgos naturales y no naturales.
