Abstract

Изобретение относится к новому способу биохимического синтеза 1,4-бутандиамина в микроорганизме, имеющем повышенный уровень активности ортининдекарбоксилазы (повышенная активность ODC) по сравнению с нативным уровнем орнитиндекарбоксилазы, где повышенную активность ODC получают посредством сверхэкспрессии гена, кодирующего орнитиндекарбоксилазу с увеличенной транскрипционной и/или трансляционной эффективностью и где производимый в микроорганизме 1,4-бутандиамина секретируется в среду ферментации и извлекается из среды ферментации. В предпочтительных воплощениях повышенная ферментативная активность получена путем сверхэкспрессии либо (i) гена speA, кодирующего аргининдекарбоксилазу, и гена speB, кодирующего агматиназу; (ii) гена speA, кодирующего аргининдекарбоксилазу, и гена aguA, кодирующего агматиниминогидролазу, и гена aguB, кодирующего N-карбамоилпутресцинамидогидролазу, и необязательно гена speB, кодирующего агматиназу. Изобретение относится также к векторам, плазмидам и к хозяевам, несущим при повышенном уровне активности одну или более упомянутых ферментативных активностей.
