Claims (11)
1. Affinitetskromatografimatrix, der omfatter en fast bærer med en affinitetskromatografiligand, der omfatter enten ét eller flere B-domæner af Staphylococcus aureus protein A (SpA) med aminosyresekvensen ifølge SEQ ID NO: 3 eller kodet for af nukleotidsekvensen ifølge SEQ ID NO: 9, eller ét eller flere Z-domæner af SpA med aminosyresekvensen ifølge SEQ ID NO: 6 eller kodet for af nukleotidsekvensen ifølge SEQ ID NO: 12, hvor mindst ét af det ene eller flere domæner er muteret for at deletere tre, fire eller fem konsekutive aminosyrer fra N-terminus, startende ved position 1 eller position 2 af SEQ ID NO: 3 eller SEQ ID NO: 6; hvor liganden udviser reduceret fragmentering i forhold til en vildtype-modpart, efter eksponering for 0,5 M NaOH i mindst 5 timer.
2. Affinitetskromatografimatrix ifølge et hvilket som helst foregående krav, hvor det ene eller flere B-domæner eller ene eller flere Z-domæner af liganden endvidere omfatter en aminosyremutation ved position 29 for at reducere Fab-binding, hvor positionen svarer til vildtype SpA B- eller SpA Z-domæneposition.
3. Affinitetskromatografimatrix ifølge krav 2, hvor amino syremutationen af B-domænet omfatter udskiftning af en glycin-aminosyrerest med en lysin-aminosyrerest.
4. Affinitetskromatografimatrix ifølge krav 2, hvor aminosyremutationen af Z-domænet omfatter udskiftning af en alanin-aminosyrerest med en lysin-aminosyrerest.
5. Affinitetskromatografimatrix ifølge et hvilket som helst foregående krav 1, der omfatter to eller flere B-domæner af SpA eller to eller flere Z-domæner af SpA eller en hvilken som helst kombination af B- og Z-domæner i en hvilken som helst rækkefølge.
6. Affinitetskromatografimatrix ifølge et hvilket som helst af de foregående krav, der omfatter mindst fem B-domæner af SpA eller mindst fem Z-domæner af SpA, hvor hvert af domænerne omfatter en mutation for at reducere Fab-binding såvel som deletion af 4 konsekutive aminosyrer fra N-terminus, startende ved position 1, hvor positionen svarer til vildtype SpA B-eller Z-domæneposition.
7. Affinitetskromatografimatrix ifølge et hvilket som helst af de foregående krav, hvor affinitetskromatografiliganden er bundet til den faste bærer via flerpunktsbinding.
8. Affinitetskromatografimatrix ifølge et hvilket som helst af de foregående krav, hvor matrixen bevarer mindst 95 % af dens oprindelige bindingsevne efter 5 timers inkubation i 0,5 M NaOH.
9. Affinitetskromatografimatrix ifølge afhængige krav 1-7, hvor matrixen bevarer mindst 95 % af dens oprindelige bindingsevne efter 25 timers inkubation i 0,1 M NaOH.
10. Affinitetskromatografimatrix ifølge et hvilket som helst af de foregående krav, hvor den faste bærer er udvalgt fra gruppen bestående af glas med kontrolleret porøsitet, siliciumdioxid, zirconiumoxid, titanoxid, agarose, polymethacrylat, polyacrylat, polyacrylamid, polyvinylether, polyvinylalkohol og polystyren og derivater deraf.
11. Fremgangsmåde til affinitetsoprensning af ét eller flere målmolekyler fra en prøve, hvilken fremgangsmåde omfatter følgende trin: (a) tilvejebringelse af en prøve, der omfatter ét eller flere målmolekyler; (b) etablering af kontakt mellem prøven og affinitetskromatografimatrixen ifølge et hvilket som helst af de foregående krav under forhold, således at det ene eller flere målmolekyler bindes til matrixen; og (c) genvinding af det ene eller flere bundne målmolekyler ved eluering.
