Claims (20)
- EERMENTERINGSBOUILLONFORMULERINGER1. Fremgangsmåde til fremstilling af en fermenteringsbouillonformulering, hvilken fremgangsmåde omfatter følgende trin: (i) fremstilling af en første blanding, der omfatter: (a) tilvejebringelse af én eller flere fermenteringsbouilloner, (b) tilsætning af en første organisk syrebestanddel, der omfatter mindst én organisk syre med 1-5-kulstoffatomer og/eller et salt deraf i en mængde fra 0,1 til 15 vægt-% af blandingen, og (c) tilsætning af en anden organisk syrebestanddel, der omfatter mindst én organisk syre med 6- eller flere kulstofatomer og/eller et salt deraf i en mængde fra 0,025 til 5 vægt-% af blandingen, (ii) inkubation af den første blanding i et tidsrum og under betingelser, der resulterer i mindst et 4 log-fald i levedygtige celler i den ene eller flere fermenteringsbouilloner, hvorved fermenteringsbouillonformuleringen fremstilles; hvor den anden organiske syrebestanddel er benzoesyre, et salt af benzoesyre, cyclohexancarboxylsyre, et salt af cyclohexancarboxylsyre, 4-methylvalerianesyre, et salt af 4-methylvalerianesyre, phenyleddikesyre, et salt af phenyleddikesyre eller en blanding af to eller flere af ovenstående.
- 2. Fremgangsmåde ifølge krav 1, eller sammensætning ifølge krav 15, hvor den første organiske syrebestanddel er i en mængde fra 0,2 til 1 %, fra 0,2 % til 0,5%, fra 0,1 % til 10 %, fra 0,25 % til 5 % eller fra 0,3 % til 3 vægt-% af den første blanding, og/eller den anden organiske syrebestanddel er i en mængde fra 0,04 % til 3 %, fra 0,2 % til 0,5 %, fra 0,1 % til 1 %, fra 0,25 % til 5 % eller fra 0,3 % til 3 vægt-% af den første blanding eller sammensætning,
- 3. Fremgangsmåde ifølge et hvilket som helst af kravene 1 eller 2, hvor tidsrummet er fra 8 timer til 36 timer, såsom fra 20 timer til 28 timer.
- 4. Fremgangsmåde ifølge et hvilket som helst af kravene 1 til 3, hvor betingelserne indbefatter en temperatur på 20 °C til 50 °C, såsom 25 °C til 40 °C, for eksempel 28 °C til 33 °C.
- 5. Fremgangsmåde ifølge et hvilket som helst af kravene 1 til 4, hvor betingelserne indbefatter et pH på 3,5 til 5, såsom et pH på 4 til 4,7, for eksempel et pH på 4,2 til 4,5.
- 6. Fremgangsmåde ifølge et hvilket som helst af kravene 1 til 5, hvor cellerne er svampeceller, for eksempel filamentøse svampeceller, såsom celler fra arterne Trichoderma, Aspergillus, Penicillium, Humicola, Chrysosporium eller Neurospora.
- 7. Fremgangsmåde ifølge et hvilket som helst af kravene 1 til 6, eller sammensætning ifølge et hvilket som helst af kravene 15 til 18, hvor den første organiske syrebestanddel omfatter eller består af eddikesyre, et salt af eddikesyre, myresyre, et salt af myresyre, propionsyre, et salt af propionsyre eller en blanding af to eller flere af ovenstående.
- 8. Fremgangsmåde ifølge et hvilket som helst af kravene 1 til 7, eller sammensætning ifølge et hvilket som helst af kravene 15 til 17, hvor den første organiske syrebestanddel omfatter et natrium-, kalium-, calcium- eller magnesiumsalt af den organiske syre med 1-5 kulstofatomer, og/eller hvor den anden organiske syrebestanddel omfatter et natrium-, kalium-, calcium- eller magnesiumsalt af den organiske syre med 6 eller flere kulstofatomer.
- 9. Fremgangsmåde ifølge et hvilket som helst af kravene 1 til 6, hvor den første organiske syrebestanddel omfatter eddikesyre ved en koncentration på 0,2 - 0,4 vægt-% og den anden organiske syrebestanddel omfatter natriumbenzoat ved en koncentration på 0,2 - 0,4 vægt-%, eventuelt hvor tidsrummet er 24 timer, betingelserne indbefatter en temperatur på 40 °C, og pH’et er mellem 4 og 4,6.
- 10. Fremgangsmåde ifølge et hvilket som helst af kravene 1 til 9, hvor mindst ét af det ene eller flere fermenteringsbouilloner indeholder ét eller flere proteiner, der er sekreteret af cellerne, hvor eventuelt det mindst ene af det ene eller flere proteiner udtrykkes rekombinant af cellerne, og/eller mindst ét af det ene eller flere proteiner er et enzym, såsom en exoglucanase, en endoglucanase, en hemicellulase eller en β-glucosidase; hvor eventuelt mindst én af den ene eller flere fermenteringsbouilloner indeholder en flerhed af enzymer, der er udtrykt rekombinant og sekreteret af cellerne, for eksempel hvor flerheden af enzymer hver er en exoglucanase, en endoglucanase, en hemicellulase eller en β-glucosidase.
- 11. Fremgangsmåde ifølge et hvilket som helst af kravene 1 til 10, eller sammensætning ifølge krav 15 til 19, hvor proteiner udgør 5 til 15 vægtprocent af den første blanding.
- 12. Fremgangsmåde ifølge et hvilket som helst af kravene 1 til 11, der endvidere omfatter trinnet med tilsætning af et pH-justeringsmiddel, såsom phosphorsyre, svovlsyre eller natriumhydroxid, til den første blanding i løbet af inkubationsperioden.
- 13. Fremgangsmåde ifølge et hvilket som helst af kravene 1 til 12, hvor den første blanding endvidere omfatter ét eller flere antimikrobielle midler, såsom humleekstrakt, der indeholder iso-alfa-syrer, tetra-iso-alfa-syrer, og/eller beta-syrer, eventuelt i en mængde af 0,0005 til 0,05 vægt-% af den første blanding, såsom 0,001 til 0,025 vægt-% af den første blanding.
- 14. Fermenteringsbouillonformulering, der omfatter svampeceller, hvor fermenteringsbouillonformuleringen er opnået eller kan opnås ved hjælp af fremgangsmåden ifølge et hvilket som helst af kravene 1 til 13, der eventuelt omfatter ét eller flere ekstracellulære enzymer, der er udvalgt fra en exoglucanase, en endoglucanase, en hemicellulase og en β-glucosidase, hvilken bouillonformulering eventuelt har 2200 til 2800 CMC U/g endoglucanaseaktivitet og 525 til 775 pNPG U/g β-glucosidaseaktivitet, eller 2200 til 3200 CMC U/g endoglucanase, 300 til 800 pNPG U/g β-glucosidase aktivitet og 2000 til 5000 ABX U/g xylanaseaktivitet.
- 15. Sammensætning, der omfatter (a) én eller flere fermenteringsbouilloner, der omfatter svampeceller, for eksempel filamentøse svampeceller, såsom celler fra arterne Trichoderma, Aspergillus, Penicillium, Humicola, Chrysosporium eller Neurospora; (b) en første organisk syrebestanddel, der omfatter mindst én organisk syre med 1-5 kulstofatomer og/eller et salt deraf i en mængde af 0,2 til 1,5 vægt-% af sammensætningen, (c) en anden organisk syrebestanddel, der omfatter mindst én organisk syre med 6- eller flere kulstofatomer og/eller et salt deraf i en mængde af 0,04 til 0,6 vægt-% af sammensætningen, hvor den anden organiske syrebestanddel omfatter eller består af benzoesyre, et salt af benzoesyre, cyclohexancarboxylsyre, et salt af cyclohexancarboxylsyre, 4-methylvalerianesyre, et salt af 4-methylvalerianesyre, phenyleddikesyre, et salt af phenyleddikesyre eller en blanding af to eller flere af ovenstående; og hvor svampecellerne i overvejende grad eller fuldt ud er ikke-levedygtige celler, eventuelt hvor, hvis cellerne er levedygtige eller er til stede i sammensætningen, forholdet mellem ikke-levedygtige og levedygtige celler i sammensætningen er mindst 10:1, mindst 50:1, mindst 100:1, mindst 1000:1, mindst 10.000:1, mindst 100.000:1 eller mindst 1.000.000:1.
- 16. Sammensætning ifølge krav 15, der har et pH på 3,5 til 5, såsom et pH på 4 til 4,7, for eksempel et pH på 4,2 til 4,5.
- 17. Sammensætning ifølge et hvilket som helst af kravene 15 til 16, hvor den første organiske syrebestanddel omfatter eddikesyre ved en koncentration på 0,2 - 0,4 vægt-% og den anden organiske syrebestanddel omfatter natriumbenzoat ved en koncentration på 0,2 - 0,4 vægt-%.
- 18. Sammensætning ifølge et hvilket som helst af kravene 15 til 17, der omfatter ét eller flere proteiner, der er sekreteret af svampecellerne, hvor eventuelt mindst ét af det ene eller flere proteiner udtrykkes rekombinant af svampecellerne, og/eller hvor mindst ét af det ene eller flere proteiner er et enzym, såsom en exoglucanase, en endoglucanase, en hemicellulase eller en β-glucosidase; hvor eventuelt sammensætningen indeholder en flerhed af enzymer, der udtrykkes rekombinant og sekreteres af cellerne, såsom en flerhed af enzymer, der hvert er en exoglucanase, en endoglucanase, en hemicellulase eller en β-glucosidase.
- 19. Sammensætning ifølge et hvilket som helst af kravene 15 til 18, der endvidere omfatter ét eller flere antimikrobielle midler, såsom humleekstrakt, der indeholder iso-alfa-syrer, tetra-iso-alfa-syrer og/eller beta-syrer, eventuelt i en mængde af 0,0005 til 0,05 vægt-% af sammensætningen, såsom i en mængde af 0,001 til 0,025 vægt-% af sammensætningen.
- 20. Kit, der omfatter (a) emballering og (b) fermenteringsbouillonformuleringen ifølge krav 14 eller sammensætning ifølge et hvilket som helst af kravene 15 til 19.
