Den foreliggende opfindelse angår et mellemprodukt til fremstilling af en række hidtil ukendte forbindelser med inhibe-rende virkning overfor biosyntesen af cholesterol, hvilke forbindelser er derivater af visse forbindelser med betegne 1-5 sen ML-236A og MB-530A.
Hyperlipidæmi, især hypercholesterolæmi, vides at være en af hovedårsagerne til cardiopati, såsom hjerteinfarkt eller arte-riesclerose. Som resultat heraf har en betydelig forskning 10 været udført med henblik på at finde frem til forbi ndel ser, der er i stand til at formindske lipidmængden, og især cholesterol mængden i blodet. En gruppe forbindelser af denne type er omhandlet i USA-patentskrift nr. 3.983.140, hvilke forbindelser isoleres fra mikroorganismer af slægten Penicillium.

I de tyske offentliggørelsesskrifter nr. 3.006.215 og nr. 3.006.216 er omhandlet en anden sådan forbindelse, som har fået betegnelsen Monacolin K eller MB-530B, og denne forbin-20 delse kan fremstilles ved dyrkning af mikroorganismer af slægten Monascus, især stammer af Monascus ruber.
Det har nu vist sig, at en række forbindelser, der er beslægtet med ML-236 og MB-530B, har en bedre virkningsudvikling som 25 resultat af deres forbedrede absorption ved oral administration og lettere står til rådighed som et for-lægemiddel (dvs. et lægemiddel, der omdannes til en aktiv eller mere aktiv form efter administration som følge af kemiske eller biokemiske reaktioner i legemet).

MB-530A kan fremstilles ved dyrkning af en MB-530A-produceren-de mikroorganisme af slægten Monascus, fortrinsvis en stamme 35 af Monascus ruber og mest foretrukket Monascus ruber SANK 15177. Denne stamme blev deponeret den 27. april 1979 under accessionsnummeret FERM 4956 hos The Fermentation Research 6 DK 167805 51
Institute, Agency of Industrial Science and Technology, Ministry of International Trade and Industry, Japan, og den 25. januar 1980 under accessionsnummeret NRRL 12081 hos The Agricultural Research Service Culture Collection, Northern 5 Regional Research Laboratory, Peoria, Illinois, U.S.A. Denne stammes morfologi og fysiologi er beskrevet detaljeret i tysk offentliggørelsesskrift nr. 3.006.215.
Den ønskede forbindelse MB-530A kan fremstilles ved dyrkning af den udvalgte mikroorganisme i et dyrkningssubstrat under 10 aerobe betingelser under anvendelse af den samme teknik, som er velkendt til dyrkning af svampe og andre mikroorganismer.
For eksempel kan den valgte stamme af Monascus først dyrkes på et egnet medium, og den producerende mikroorganisme kan derefter opsamles og podes på og dyrkes på et andet dyrkningsme-15 dium til fremstilling af den ønskede MB-530A-forbindelse. Det til formeringen af mikroorganismen benyttede dyrkningsmedium og det til produktionen af MB-530A benyttede dyrkningsmedium kan være ens eller forskellige.
Ethvert velkendt dyrkningsmedium til dyrkning af svampe kan 20 benyttes, forudsat at det, som det er velkendt, indeholder de nødvendige næringsstoffer, især en assimilerbar carbonkilde og en assimilerbar nitrogenkilde.. Eksempler på sådanne kilder for assimilerbart carbon indbefatter glucose, maltose, dextrin, stivelse, lactose, sucrose og glycerol. Af disse kilder er 25 glucose, glycerol og stivelse særlig foretrukne. Eksempler på egnede kilder for assimilerbart nitrogen er pepton, kødekstrakt, gær, gærekstrakt, soyabønnemel, jordnøddemel, majsstøbevæske, risklid og uorganiske nitrogenkilder. Af disse nitrogenkilder er majsstøbevæske og pepton særlig foretrukne. Et 30 uorganisk salt og/eller et metalsalt kan om nødvendigt sættes til dyrkningsmediet. Endvidere kan om nødvendigt en mindre mængde af et tungt metal også tilsættes.

Mikroorganismen dyrkes fortrinsvis under aerobe betingelser under anvendelse af dyrkningsmetoder, der er velkendte inden for teknikken, f.eks. fast dyrkning, rystedyrkning eller dyrkning under luftning og omrøring. Mikroorganismen vil vokse 5 over et bredt temperaturinterval, f.eks. 7 til 40°C, men den foretrukne dyrkningstemperatur ligger inden for intervallet fra 20 til 30°C.
Under dyrkningen af mikroorganismen kan produktionen af 10 MB-530A måles ved prøveudtagning af dyrkningsmedium og måling af mediets fysiologiske akt ivi tet ved hjælp af velkendte forsøg. Dyrkningen kan derefter fortsættes, indtil en væsentlig akkumulering af MB-530A er blevet opnået i dyrkningsmediet, på hvilket tidspunkt MB-530A-forbindelsen derefter kan isoleres 15 og udvindes fra dyrkningsmediet og mikroorganismens cellevæv ved hjælp af enhver egnet kombination af isolationsteknik, der vælges under hensyntagen til dets fysiske og kemiske egenskaber. Ekstraktion af væsken fra dyrkningssubstratet med et hydrofilt opløsningsmiddel (såsom diethylether, ethylacetat, chloroform eller benzen), ekstraktion af organismen med et hydrofilt opløsningsmiddel (såsom acetone eller en alkohol), koncentre-25 ring, f.eks. ved afdampning af noget eller en del af opløsningsmidlet under formindsket tryk, opløsning i et mere polært opløsningsmiddel (såsom acetone eller en alkohol), fjernelse af urenheder med et mindre polært opløsningsmiddel (såsom petroleumsether eller hexan), gelfi 1 trer ing gennem en søjle af 30 et materiale, såsom "Sephadex" (et handelsnavn for et materiale, der fås fra Pharmacia Co. Limited, U.S.A.), absorptionskromatografi med aktivt carbon eller silicagel, og andre lignende metoder. Ved anvendelse af en passende kombination af disse metoder kan den ønskede MB-530A-forbindelse isoleres fra 35 dyrkningssubstratet som et rent stof.
De acylerede derivater fremstillet ud fra mellemproduktet MB-530A ifølge den foreliggende opfindelse kan fremstilles ved 8 DK 167805 B1 hjælp af enhver af de nedenfor anførte metoder.
Metode 1 5 MB-530A omsættes med et syrechlorid eller et syreanhydrid, der passer til det acylderivat, som det er hensigten at fremstille. Reaktionen udføres fortrinsvis i nærværelse af en base (der fungerer som et syrebindende middel), fortrinsvis en organisk amin, såsom pyridin, triethylamin, N,N-dimethylamino-10 pyridin, N-methylpyrrolidin eller N-methylmorfolin. Reaktionen udføres fortrinsvis i nærværelse af et opløsningsmiddel, og naturen af dette opløsningsmiddel er ikke afgørende, forudsat at det ikke på uheldig måde påvirker reaktionen. En carboxylsyre behandles med en chlorcarbonatester eller med et sulfonsyrechlorid i nærværelse af en base, f.eks. en af de ovenfor nævnte organiske aminer, til fremstilling af et blandet syreanhydrid, og dette omsættes på sin side med MB-530A. 30 Reaktionen foretages fortrinsvis i nærværelse af et opløsningsmiddel, hvis natur ikke er afgørende, forudsat at det ikke på uheldig måde påvirker reaktionen. Egnede opløsningsmidler indbefatter eksempelvis diethylether, benzen, chloroform og methylenchlorid. Reaktionen vil finde sted over et 35 bredt temperaturinterval, f.eks. fra -20°C til stuetemperatur, fortrinsvis fra -20°C til 0°C.

Metode 3 MB-530A omsættes med en carboxylsyre og et diazoalkyldicar-boxylat i nærværelse af f.eks. dicyklohexylcarbodiimid, tri-5 phenylphosphin eller dimethylphosphorsyrling-amid. Reaktionen foretages fortrinsvis i nærværelse af et opløsningsmiddel, hvis natur ikke er afgørende, forudsat at det ikke har en uheldig virkning på reakt ionen.Egnede opløsningsmidler indbefatter chloroform, methylenchlorid, benzen og diethylether.

Hver af de ovennævnte reaktioner vil normalt være afsluttet indenfor et tidsrum fra 30 minutter til 5 timer, selvom den nøjagtige tid, der kræves til reaktionen, vil afhænge af reagenserne og reaktionstemperaturen. Efter afslutningen af reak-15 tionen kan det ønskede produkt skilles fra reaktionsblandingen ved hjælp af konventionelle midler, f.eks. ved afdampning af opløsningsmidlet fra en opløsning indeholdende .det ønskede produkt (hvilken opløsning simpelthen kan være reaktionsblandingen eller kan være en opløsning opnået ved ekstraktion af 20 reaktionsblandingen med et organisk opløsningsmiddel), om ønsket efter vaskning og tørring af opløsningen, hvorefter produktet kan renses ved hjælp af konventionelle midler, f.eks. søjlekromatografi, tyndt 1agskromatografi eller rekrystal1 i sation eller ved hjælp af enhver kombination deraf.

Da udgangsmaterialet MB-530A for acyleringsreaktionen indeholder to hydroxygrupper, kan de ovenfor beskrevne reaktioner føre til 3-monoesteren, 3,8'-diesteren eller 8'-monoesteren eller en blanding deraf. Arten af det opnåede produkt kan 30 påvirkes ved variering af mængderne af reagenser, reaktionstemperaturen og andre reaktionsbetingelser.· Fagmanden kan imidlertid let bestemme, hvad der er de optimale betingelser til fremstilling af den ønskede 8'-monoester. 1 8' --monoesteren vil overvejende blive produceret ved udførelse af reaktionen ved som acy1 er ingsmiddel at anvende et syrean- hydrid eller et syrehalogenid i nærværelse af en organisk base DK 167805 Bl 10 ved en temperatur fra -20°C til 0°C. 5 Forbindelserne fremstillet ud fra mellemproduktet ifølge opfindelsen har vist sig at have en specifik inhiberende virkning over for 3-hydroxy-3-methylglutarylcoerizym A reduktase (HMG-CoA reduktase), som er det hastigheds-kontrollerende enzym ved biosyntesen af cholesterol. De inhiberende virknin-10 ger af nogle af forbindelserne fremstillet ud fra mellemproduktet ifølge opfindelsen over for biosyntesen af cholesterol er vist i den efterfølgende tabel udtrykt som deres 150-vær-dier (det vil sige den koncentration i pg/ml, som resulterer i en 50% inhibering af cholesterolbiosyntesen), målt ved hjælp 15 af fremgangsmåden ifølge Knaus et al., J. Biol. Chem. 234, 2835 (1959).

Fremstilling af mellemproduktet MB-530A ifølge opfindelsen

300 1 af et dyrkningsmedium med en pH-værdi på 5,5 før sterilisering og indeholdende 5% vægt/volumen glucose, 0,5% vægt/ volumen majsstøbevæske og 2% vægt/volumen pepton (Kyokuto fa-brikat, fås fra Kyokuto Seiyaku KK, Japan) og 0,5% ammonium-
O O
chlorid blev fyldt på en 600 1 gæringsbeholder og podet med en kultur af Monascus ruber SANK 15177 (FERM 4956, NRRL 12081).

Dyrkning af mikroorganismen blev fortsat i 120 timer ved 27°C med en beluftningshastighed på 300 1/min og omroring med 190 omdrejninger per minut.
5 Ved afslutningen af dette tidsrum blev dyrkningssubstratet filtreret i en filterpose til opnåelse af et filtrat og en filterkage omfattende våde celler af mikroorganismen.
Filtratet blev indstillet til en pH-værdi på 3,0 ved tilsæt-10 ning af 6N saltsyre og derpå ekstraheret med 400 1 ethylace-tat. Ekstrakten (ca. 400 1) blev koncentreret ved inddampning under formindsket tryk og derefter afvandet over vandfrit natriumsulfat, hvorefter den blev inddampet til tørhed til opnåelse af ca. 60 g af et olieagtigt produkt. Dette olieagtige 15 produkt blev vasket med ethylcyklohexan og med hexan, og resten (20 g) blev separeret ved hjælp af kromatografi under anvendelse af et væskekromatografi apparat til udtagning af prøver med stort volumen (System 500 væskekromatografi, fremstillet af Waters Co., USA), idet der blev elueret med 60% volu-20 men/volumen vandig methanol. Fraktioner med en kromatografisk retentionstid på 6 minutter blev opsamlet og koncentreret ved inddampning under formindsket tryk til opnåelse af 100 mg af den ønskede MB-530A-forbindel se som et olieagtigt produkt. Denne olieagtige MB-530A-forbindelse blev rekrystalliseret fra 25 en blanding af acetone og diethylether til opnåelse af 57 mg af det ønskede produkt i form af farveløse nåle med følgende egenskaber: 1 2 3 4 5 6
Smeltepunkt: 92 - 93°C.

10 10. Inhiberende virkning overfor biosyntesen af cholesterol:
En 50% inhibering af chol esterolsyntesen i rottelever iagttages ved en koncentration på 0,04 pg/ml.
Eksempel 2 15 MB-530A 81-butvrat MB-530A (309 mg) og pyridin (0,36 ml) blev opløst i roethylen-chlorid (10 ml). Den resulterende opløsning blev kølet til 0°C 20 og butyrylchlorid (0,16 ml) blev dråbevis sat dertil. Efter afslutning af reaktionen blev vand sat til reaktionsblandingen, og det fraskilte methylenchloridlag blev vasket med vand og derefter tørret over vandfrit natriumsulfat. Den ved afdampning af methylenchloridet opnåede rest blev underkastet 25 separation ved hjælp af søjlekromatografi under anvendelse af silicagel (10 g) efterfulgt af rekrystallisation fra diethyl-ether til opnåelse af 154 mg af det ønskede produkt.
