Den foreliggende opfindelse angår en analogifremgangsmåde til fremstilling af hidtil ukendte hydroxykarbonylfosfon-syremonoestere med den i krav l's indledning angivne almene formel I, hvor R betegner adamant-l-yl eller adamant- 2-yl, eller optiske antipoder og/eller fysiologisk acceptable salte deraf. Fremgangsmåden ifølge opfindelsen er ejendommelig ved det i krav l's kendetegnende del angivne.
Disse forbindelser kan indgå i farmaceutiske præparater og bruges til selektivt at bekæmpe vira såsom herpesvira, in-fluenzavira og RNA-tumorvira som bevirker forskellige sygdomme hos dyr og mennesker. Sådanne sygdomme omfatter både almindelige infektioner og neoplastiske tilstande, fx kræft.
Virkningen af vira på legemlige funktioner er slutresultatet af forandringer som forekommer på cellulært og sub-cellulært niveau. De patogene forandringer på det cellulære niveau er forskellige for forskellige kombinationer af vira og værtsceller. Medens vira bevirker en generel ødelæggelse (aflivning) af visse celler, kan andre transformere celler til en neoplastisk tilstand.
Betydningsfulde almindelige virusinfektioner er herpes dermatitis (herunder herpes labialis), herpes keratitis, herpes genitalis, herpes zoster, herpes encephalitis, infektiøs mononucleose og cytomegalovirus-infektioner, der alle bevir-kes af vira som hører til herpesvirusgruppen. Andre vigtige virussygdomme er influenza A og B som fremkaldes af henholdsvis influenza A og influenza B virus. Andre betydningsfulde almindelige virussygdomme er viral leverbetændelse, og navnlig er infektioner med hepatitis B virus vidt udbredte. Der behøves effektive og selektive antivirale midler til behandling af disse sygdomme.
Det er påvist at adskillige forskellige vira både af DNA-type og RNA-type fremkalder svulster hos dyr. Virkningen af cancerogene kemikalier kan hos dyr resultere i aktivering af latente tumorvira. Det er muligt at tumorvira er involveret ved svulster hos mennesker. De mest sandsynlige tilfælde hos mennesker der kendes for tiden, er leukæmier, sarcomer, brystcarcinomer, Burkitt-lymfomer, nasofaryngale carcinomer 2 148632 og halskræft hvor RNA-tumorvira og herpesvira er indikeret.
Dette medfører at det til behandling af kræft er vigtigt at søge at finde frem til selektive inhibitorer af tumorogene vira og deres funktioner.
Et overmåde vigtigt træk i samspillet mellem vira og celler er reproduktionen (formeringen) eller transkriptionen (overføringen) af de specifikke virale genetiske informationer der bæres af de virale nucleinsyrer. Disse virale nu-cleinsyrer er to slags, desoxyribonucleinsyre, DNA, og ribo-nucleinsyre, RNA. Cellens primære genetiske information bæres af celle-DNA. DNA-syntese og ENA-syntese involverer kom-plexe enzymer som henholdsvis kaldes DNA-polymeraser og RNA-polymeraser. Den genetiske information overføres til den nye nucleinsyre fra en model ("template") nucleinsyre. Der er fire generelle måder på hvilke disse nucleinsyrer kan reproduceres eller overføres.

Processerne 1 og 3 bruges af celler. DNA-vira såsom herpesvira bruger også proces nr. 1, men enzymet er anderledes end det fra cellen. RNA-vira såsom influenzavira bruger proces nr. 2 og RNA-tumorviruserne (retrovira) kan omdanne sit RNA til DNA i henhold til proces nr. 4.
De virale polymeraser og de virale nucleinsyresynteser er essentielle ikke blot for almindelige (produktive) virusinfektioner, men også for virale omdannelser af celler til en neoplastisk tilstand som fører til kræft (virus' tumorogene funktion). I sidstnævnte tilfælde kan DNA dannet af DNA-vira såsom herpesvirus eller omdannet fra RNA af RNA-tumor-vira og som bærer den genetiske information til celleomdan- 3 148632 nelse integreres i værtscellens DNA. Denne integration, eller senere begivenheder som følge af integrationen (fx samspil med cancerogene kemikalier) kan derefter føre til omdannelse af værtscellen.Implikationerne ved at inhibere revers transkriptase til celleomdannelse er også beskrevet i US-patent-skrift 3.979.511.
Eftersom de virale polymeraser i de fleste tilfælde afviger fra de cellulære, er disse virale enzymer og virale nucleinsyresynteser gode mål for specifik antiviral kemoterapi, herunder kemoterapi over for kræft fremkaldt af vira. Det skal bemærkes at mange forbindelser der for tiden bruges i den kemoterapeutiske behandling af kræft er inhibitorer af nucleinsyresyntese. Det er derfor muligt at antivirale forbindelser som også er inhibitorer af nucleinsyresyntese, kan påvirke kræftceller direkte. Der er behov for et effektivt antiviralt middel fortrinsvis med selektiv inhiberende virkning på en specifik viral funktion af det virus som skal bekæmpes. Det er derfor et generelt formål for den foreliggende opfindelse at tilvejebringe en fremgangsmåde til fremstilling af forbindelser der kan give en ny metode til bekæmpelse af virusinfektioner ved hjælp af et antiviralt middel som udøver selektiv inhiberende virkning på virale funktioner, men som kun udøver forsvindende inhiberende virkning på værtscellernes funktioner.
De omhandlede forbindelser med formel I er sådanne forbindelser.
Forbindelserne med den almene formel I er estere af fosfonmyresyre Der er beskrevet forskellige estere af fosfon-myresyre i bl.a. US-patentskrifterne 3.943.201, 3.155.597 og 3.533.995 samt i Chem. Ber. 57 side 1023 (1924), Chem. Ber. 60B side 291 (1927) og i Chem. Pharm. Bull. 21 (5) side 1160 (1973).
Disse estere er imidlertid ikke blevet foreslået anvendt til noget farmakologisk formål.
Det er kendt fra LU patentskrift nr. 77.666 at fosfon-myresyrenfder er en forbindelse svarende til formel I, men hvor R er hydrogen, har antiviral virkning. Den fri syre og dens fysiologisk acceptable salte inhiberer således virale funktioner såsom polymeraser inklusive revers transkrip- 4 148632 tase og virusformering, og de har virkninger på virusinfektioner og svulster med relation til vira i dyremodeller.
Antivirale virkninger af trinatriumfosfonformiat er i øvrigt beskrevet af Helgstrand et al i Science 201, 819 (1072).
En fordel ved de ved fremgangsmåden ifølge opfindelsen fremstillede estere (og deres fysiologisk acceptable salte) i sammenligning med den fri syre og dens salte er at estrene har bedre vævspenetration, bedre absorption og forlænget aktivitet; det bemærkes at de kan hydrolyseres in vivo til fosfonmyresyre eller ioniserede former deraf.
Således er de omhandlede adamantylestre ved fosforsyregruppen generelt mere stabile i sure opløsninger end fosfon-myresyre og nedbrydes derfor vanskeligere i maven. De er endvidere mere lipofile end syren selv og dens salte og er derfor bedre egnede til behandling af virusinfektioner i organer hvor penetration gennem lipidbarrierer er af betydning.
Fra US patentskrift nr. 3.767.795 er det kendt at fosfoneddikesyre, dvs. en forbindelse med formlen 0 o a 11 II b

a h hvor R og R begge er hydrogen, har antiviral virkning. Imidlertid er det også kendt at fosfoneddikesyre er stærkt hudirriterende [Alenius et al, Antimicrobial Agents and Chemotherapy, vol. 14, nr. 3, 408-413, (1978)], og da en række virusinfektioner netop angriber huden og kræver topisk be-\ handling af denne, er fosfoneddikesyre uegnet til i hvert fald disse infektioner. Det samme vil også gælde estere af fosfoneddikesyre, idet de spaltes efter påføring eller indgift til den frie syre og dermed også virker hudirriterende.
Estere af fosfonmyresyre spaltes som nævnt tilsvarende, men fosfonmyresyre er ikke hudirriterende. Principielt er fos-fonmyresyre og derivater deraf bedre egnede som lægemidler, i hvert fald til hudbehandling end fosfoneddikesyre og dens derivater.
Fra US patentskrift nr. 4.052.439 er det kendt at mono- 5 148632 estere ved karboxylgruppen af fosfoneddikesyre, nemlig for-bindeiser med formel A hvor Ra er hydrogen og RD er C^g alkyl, aralkyl, cyklohexyl eller adamantyl, har antiviral virkning.
Fra FR patentskrift nr. 2.349.596 er det kendt at monoestere ved fosforsyregruppen af fosfoneddikesyre, nemlig forbindelser med den almene formel A hvor Ra er en C, 0 al-b kylgruppe og R hydrogen, har antiviral virkning.
Forbindelser kendt fra de to sidstnævnte patentskrifter er ifølge skrifterne afprøvet ved en ensartet teknik der tillader direkte sammenligning. Ved denne teknik har man målt den koncentration (i μg/ml) af testforbindelserne, der giver 50% inhibering af viral DNA-polymerase.
I en af de afprøvede forbindelser er Ra hydrogen og R 2-adamantyl; der er således tale om en forbindelse hvor estergruppen er knyttet til karboxylgruppen. Denne forbindelse har -den højeste koncentration til frembringelse af 50% inhibering af samtlige de afprøvede, nemlig 166 pg/ml, og dermed den ringeste antivirale virkning. Den bedste af de afprøvede forbindelser, nemlig den tilsvarende t-butylester, behøvede kun en koncentration på 4 pg/ml for at bevirke en inhibering af viral DNA-polymerase på 50% og var altså godt 40 gange så effektiv. En sammenligning mellem det amerikanske og det franske patentskrift tyder på en tendens til at monoestrene ved fosforsyregruppen er noget mindre effektive end monoestrene ved karboxylgruppen; propylesteren ved karboxylgruppen var således 1,7 gange så effektiv som propylesteren ved fosforsyregruppen (koncentration til at bevirke 50% inhibering henholdsvis 4,7 og 8,0 μg/ml).
Det er på den baggrund overraskende at adamantyles-tere ved fosforsyregruppen af fosfonmyresyre overhovedet har nævneværdig antiviral virkning; og dertil kommer den allerede nævnte fordel i forhold til fosfoneddikesyreestere at de ved spaltning til den fri syre (eller salte deraf) ikke virker hudirriterende.
Da forbindelserne med den almene formel I indeholder et asymmetrisk center, eksisterer de i form af optisk aktive former og kan opdeles i deres optiske antipoder ved kendte 6 148632 metoder.
X nærværende beskrivelse er de omhandlede forbindelser navngivet som derivater af forbindelsen hydroxy-karbonylfosfonsyre, der er den samme forbindelse der også kendes under betegnelsen fosfonmyresyre
Forbindelser med formel I og fysiologisk acceptable salte deraf er nyttige ved terapeutisk og/eller profylaktisk behandling af virussygdomme og kan være nyttige til terapeutisk og/eller profylaktisk behandling af kræft fremkaldt af vira.
De omhandlede forbindelser kan oparbejdes til farmaceutiske præparater der som virksom bestanddel indeholder en forbindelse med den almene formel I eller et fysiologisk acceptabelt salt deraf, vilkårligt i forbindelse med en farmaceutisk acceptabel bærer. Ved fremstilling af en medicin med antiviral virkning kan man således inkorporere en forbindelse med den almene formel I eller et fysiologisk acceptabelt salt deraf i en administrationsform som egner sig til terapeutisk formål og herved eventuelt fremstille et formet medikament.
Et særligt vigtigt område for anvendelse af præparater af de omhandlede forbindelser er til behandling af herpesvi-rusinfektioner. Blandt herpesviruserne kan nævnes Herpes simplex type 1 og 2, skoldkopper, Herpes zoster, virus som forårsager smitsom mononucleose (dvs. Epstein-Barr virus) og cytomegalovirus. Blandt sygdomme som fremkaldes af herpesvi-ra kan nævnes herpes dermatitis (herunder herpes labialis), herpes genitalis, herpes keratitis og herpes encephalitis.
Et andet betydningsfuldt område til anvendelse af de omhandlede præparater er ved behandling af infektioner fremkaldt af ortomyxovira, dvs. influenzavira af type A og type B. Et yderligere anvendelsesområde er behandling af infektioner fremkaldt af sådanne vira som hepatitis virus Δ og hepatitis virus B, papillomavira, adenovira og poxvira.
Andre mulige anvendelsesområder for præparater indeholdende de ved fremgangsmåden ifølge opfindelsen fremstillede forbindelser er ved behandling af infektioner som er fremkaldt af pivornavira, togavira inklusive arbovira, retrovira 7 148632 (fx leucovira), arenavira, coronavira, rhabdovira, paramyxo-vira, hepatitis-vira andre end hepatitis A og B, iridovira, papovarira, parvovira, reovira og bunyavira.
Ifølge opfindelsen foretrækkes det i særlig grad at fremstille en forbindelse med formel I, hvor R er 1-ada-mantyl, eller optiske antipoder og/eller et fysiologisk acceptabelt salt deraf.
Fysiologisk acceptable salte af de virksomme stoffer med formel I fremstilles ved i og for sig kendte metoder som belyses i det følgende.
Eksempler på metalsalte der kan fremstilles af salte indeholdende Li, Na, K, Ca, Mg, Zn, Mn og Ba. Et mindre opløseligt metalsalt kan udfældes fra en opløsning af et mere opløseligt salt ved tilsætning af en passende metalforbindelse. Således kan fx salte af de virksomme stoffer med Ca, Ba, Zn,
Mg og Mn fremstilles ud fra natriumsalte deraf. Metalionen i et metalsalt af de virksomme stoffer kan udskiftes med hy-drogenioner, andre metalioner, ammoniumionen eller ammoniumioner substitueret med et eller flere organiske radikaler ved hjælp af en kationbytter.

Yderligere eksempler på andre nyttige salte der kan fremstilles ved ionbytterteknikken er kvaternære ammoniumsalte af de virksomme stoffer, dvs. salte hvor hydrogenatomerne i de virksomme stoffer (strukturformel I) er blevet substitueret med kvaternære ammoniumioner såsom (CHgJ^N, (Ο^ί^Ν, (c4H9)4n, (C5Hi;L}4Nr (C6H13) 4N og C2H5N(CH2CH2OH) 3. Lipofile salte af denne type kan også fremstilles ved at man danner et salt af det virksomme stof med et kvaternært ammoniumsalt i vand og ekstraherer det resulterende kvaternære ammoniumsalt af det virksomme stof med et organisk opløsningsmiddel såsom diklormetan, kloroform, ætylacetat og metylisobutylketon.
De omhandlede forbindelser kan.oparbejdes til brug i human- og veterinærmedicinen til terapeutisk og profylaktisk brug. Forbindelserne kan bruges i form af et fysiologisk acceptabelt salt. Egnede salte er fx aminsalte såsom dimetyl-amin- og triætylaminsalte, ammoniumsalt, tetrabutylammonium-salt, cyklohexylaminsalt, dicyklohexylaminsalt samt metalsal-- te såsom mono- og dinatriumsalt, mono- og dikaliumsalt, magniumsalt, kalciumsalt og zinksalt.
I klinisk praksis vil forbindelsen normalt blive administreret topisk, oralt, intranasalt, ved injektion eller ved inhalation i form af et farmaceutisk præparat omfattende den virksomme bestanddel i form af den oprindelige forbindelse eller eventuelt i form af et farmaceutisk acceptabelt salt deraf, og sammen med en farmaceutisk acceptabel bærer, som kan være et fast, halvfast eller flydende fortyndingsmiddel eller en" kapsel som kan siuges.
Den dosis hvormed de virksomme bestanddele administreres kan variere inden for et bredt område og vil afhænge af forskellige faktorer som fx infektionens styrke, patientens alder etc., Og den må reguleres individuelt. Et muligt område for den mængde af det virksomme stof som kan indgives pr. dag kan nævnes at være fra ca. 0,1 mg til omkring 2000 mg, eller fra 1 mg til ca. 2000 mg, eller fortrinsvis fra 1 mg til ca.
2000 mg til topisk administration, fra 50 mg til ca. 2000 mg eller fra 100 til 1000 mg for oral indgift og fra 10 mg til ca. 2000 mg eller fra 50 til 500 mg til injektion. I alvorli- 9 148632 ge tilfælde kan det være nødvendigt at forøge disse doser fem gange til ti gange. I mindre alvorlige tilfælde kan det være tilstrækkeligt at bruge op til 500 eller 1000 mg.
Som det fremgår af krav 1 fremstilles forbindelserne ved hydrolyse af hydroxykarbonylfosforsyretriestere II, hvor 2 3 R og R er hydrolyserbare grupper.

Eksempler på hydrolyserbare estergriipper R og 3 R kan ifølge opfindelsen være C^_g alkylgrupper, C3_g cyklo-alkylgrupper, C4_g cykloalkylalkylgrupper, benzylgrupper og fenylgrupper der eventuelt er substitueret med 1 eller 2 sub-stituenter udvalgt blandt halogen, alkyl og C2-7 alkoxy- karbonyl, eller med en uforgrenet, mættet C3_4 alkylenkæde bundet til fenylringens stilling 2,3 eller 3,4, jfr. krav 2.
Andre eksempler på hydrolyserbare grupper er triorgano-silylgrupper, fx trimetylsilyl eller butyldifenylsilyl.

Trimetylsilylestergruppen hydrolyseres som vist i skemaet med vand, og den fri syregruppe omdannes derefter til et 6 salt med en svag kationbytter (M+) eller med en vandig base såsom MHC03, M2C03 eller MOH.

Fortrinsvis er fosfittet en bis-(trimetylsilyl)-fosforsyrling-triester.
Halogenmyresyresilylestrene kan fremstilles i henhold til skemaet C0C12 + HOSi(R7)3-* Cl-C02-Si(R7)3 + HC1 7 hvor R har den ovenfor angivne betydning.
Reaktionen udføres under vandfri betingelser og fortrinsvis bruges der en base som fx Ν,Ν-dimetylanilin til binding af det frigivne hydrogenklorid. Reaktionen udføres fortrinsvis i et inert opløsningsmiddel som fx toluen eller æter og ved fx -10 til 25°C i 1-25 timer.

Fremstilling af en triester af hydroxykarbonylfosfonsyre til anvendelse som udgangsmateriale:_
Metyl, 1-adamantyl-metoxykarbonylfosfonat fremstilledes ud fra 24,4 g (0,1 mol) dimetyl-l-ada-mantylfosfit og 18,9 g (0,2 mol) metylklorformiat (90°C, 2 timer).
NMR (CDC13> δ: 1,63 og 2,16 (brede singletter, ciqH15^' 3,83 (s, C02CH3), 3,88 (d, J 12Hz, 0CH3).
IR (rent) cm”1: 1730, 1290, 1230, 1060, 1020, 990.
En analyseprøve blev destilleret i vakuum. Kp._
π 25 UjO-L
125-127°C. n^ = 1,4922.
Eksempel 1-Adamantyl-dinatriumoxykarbonylfosfonat 11,5 g (0,04 mol) metyl-l-adamantylmetoxykarbonylfos-fonat og 12,5 g (0,08 mol) brcmtrimetylsilan omrørtes under nitrogenatmosfære ved stuetemperatur natten over. Overskud af bromtrimetylsilan afdampedes i vakuum (ca. 0,3 mm) ved 50°C. Remanensen omrørtes med 80 ml (0,08 mol) IN vandigt NaOH ved stuetemperatur i 2 timer, hvorefter opløsningsmidlet afdampedes i vakuum.
Remanensen (12,06 g) opløstes i 150 ml vand og filtreredes og der sattes 350 ml ætanol til opløsningen. Bundfaldet frafiltreredes (5,47 g) og genopløstes i 100 ml vand og der tilsattes. 60 ml ætanol. Et yderligere bundfald (A) frafiltreredes (2,83 g) og der sattes yderligere 350 ml ætanol til opløsningen hvorved der fremkom endnu 2,15 g bundfald (B).
Bundfald B genopløstes i 50 ml vand; der tilsattes 20 ml ætanol og bundfaldet kasseredes. Der tilsattes yderligere 400 ml ætanol, bundfaldet opsamledes ved centrifugering og gav efter tørring 1,49 g af den i overskriften angivne forbindelse. Ved to gange at gennemføre denne rensningssekvens på bundfald A kunne der opsamles yderligere 1,92 g af den i overskriften angivne forbindelse. Begge fraktioner kunne ved t.l.c. (polyætylenimin, 1M LiCl, molybdatsprøjtning) anslås til at indeholde <1% trinatriumoxykarbonylfosfonat.

Biologisk prøve
Inhibering af kutan herpes på marsvin
Virkningen på kutane infektioner af herpes simplex type 1 er blevet målt i en marsvinemodel beskrevet af Hubier et al., J. Invest. Dermatol. jS9 (1974) 92. Forbindelserne er blevet afprøvet som topiske påføringer af 30 μliter 2%s opløsning af forbindelsen i 45 rumfangsSs isopropanol, 10%s glycerol og 45%s vand to gange om dagen i 4 dage begyndende 24 timer efter infektionen. Udseendet af et inficeret behandlet område og et på tilsvarende måde inficeret, ubehandlet (kun isopropa-nol-glycerol-vand) område blev bedømt dagligt efter en skala fra 0 til 3. Den samlede virkning vurderedes på grundlag af bedømmelsen på dag nr. 5.
