Description
- Eine Aufgabe dieser Erfindung ist es, ein effizientes und effektives Verfahren zur Herstellung von substituierten und unsubstituierten 2,3-Pyridindicarboxylaten unter Verwendung von Dialkylchlorsuccinat, einem dehydrohalogenierenden Agens, einer Ammoniakquelle und einem entsprechend substituierten α,β-ungesättigten Aldehyd oder Keton bereitzustellen.
- Die hier beschriebene Erfindung betrifft ein neues Verfahren zur Herstellung von substituierten und unsubstituierten 2,3-Pyridindicarboxylaten der Formel I
- worin R C&sub1;-C&sub6;-Alkyl ist; X und Z unabhängig voneinander Wasserstoff, Halogen, C&sub1;-C&sub4;-Alkyl, C&sub1;-C&sub4;-Alkoxy oder C&sub2;-C&sub5;-Alkenyl sind und Y Wasserstoff, Halogen, gegebenenfalls mit 1 bis 3 Halogenen, Hydroxygruppen, C&sub1;- C&sub4;-Alkoxygruppen oder C&sub1;-C&sub4;-Alkylthiogruppen, C&sub1;-C&sub4;-Alkoxycarbonyl, Aminocarbonyl, Phenylthio, substituiertem Phenylthio, Phenoxy, substituiertem Phenoxy, Phenyl oder substituiertem Phenyl substituiertes C&sub1;-C&sub6;- Alkyl ist. Verbindungen der Formel I sind als Zwischenprodukte bei der Herstellung von herbiziden 2-(2-Imidazolin-2-yl)nicotinaten geeignet. Unter den Herstellungsverfahren dieser herbiziden Nicotinate wird im Folgenden das im U.S.-Patent 4 758 667 beschriebene erläutert solvent
- worin R&sub1; Wasserstoff oder C&sub1;-C&sub4;-Alkyl ist; R&sub2; Wasserstoff, Halogen, C&sub1;- C&sub4;-Alkyl, C&sub3;-C&sub6;-Cycloalkyl ist oder R&sub1; und R&sub2; zusammengenommen ein gegebenenfalls mit Methyl substituiertes C&sub3;-C&sub6;-Cycloalkyl und X, Y und Z wie oben in Formel I beschrieben sind.
- Es wurde nun gefunden, daß Pyridin-2,3-dicarboxylate der Formel I effizient und effektiv aus Dialkyldichlorsuccinaten der Formel II durch Umsetzung dieser Succinate mit mindesten einem Mol-Äquivalent eines dehydrohalogenierenden Agens wie Ammoniumacetat oder Trialkylamin in Gegenwart eines Lösungsmittels wie einem niederen Alkylalkohol zur Bildung eines Zwischenprodukts, der Umsetzung dieses Zwischenprodukts mit einer Ammoniakquelle, gegebenenfalls bei erhöhten Temperaturen, unter Bildung eines zweiten Zwischenprodukts und Umsetzung dieses zweiten Zwischenprodukts mit mindestens einem Mol-Äquivalent eines α,β-ungesättigten Aldehyds oder Ketons der Formel III, worin X, Y und Z wie oben in der Formel I beschrieben sind, in Gegenwart einer Säure wie Ameisensäure oder Essigsäure oder dergleichen hergestellt werden können. Das Verfahren wird im Folgenden gezeigt.
- In dem erfindungsgemäßen Verfahren verwendbare dehydrohalogenierende Agentien sind solche, die auch als Ammoniak-Quelle dienen können wie Ammoniumacetat, Ammoniumcarbonate, Ammoniumcarbamate, Ammoniumsulfαmate und dergleichen. Ebenfalls geeignet sind Reagentien wie Trialkylamine, Pyridine, Picoline, quartäre Aminsalze wie Tetraalkylammoniumhalogenide und dergleichen. Selbstverständlich kann zusätzlich zu den obengenannten Ammoniakquellen im erfindungsgemäßen Verfahren auch Ammoniak selbst verwendet werden.
- In einer Ausführungsform der Erfindung kann das verwendete Lösungsmittel ein polares Lösungsmittel wie ein Alkohol, ein Nitril wie Acetonitril, ein Carbonsäureamid wie N,N-Dimethylformamid, N-Methylpyrrolidon, ein Sulfoxid wie Dimethylsulfoxid oder ein Sulfon sein.
- Von den α,β-ungesättigten Aldehyden oder Ketonen der Formel III können Acrolein, Methacrolein, Ethacrolein, Crotonaldehyd, Methylvinylketon, α-n-Butylacrolein, 4-Methyl-2-hexanal, α-Methoxymethacrolein, α- Chlormethacrolein, α-Trifluormethacrolein, Zimtaldehyd, α-Ethoxyacrolein, Methyl-α-formylacrylat oder α-(2-cyanethyl)acrolein verwendet werden.
- In einer weiteren Ausführungsform der Erfindung kann die das obengenannte zweite Zwischenprodukt und den α,β-ungesättigten Aldehyd oder das Keton der Formel III enthaltende Reaktionsmischung mit einem Dehydrierungs-Katalysator behandelt werden, wie er im Stand der Technik bekannt ist, einschließlich Metallen oder Verbindungen von Platin, Pallαdium, Ruthenium, Iridium, Nickel, Eisen, Kupfer, Antimon, Kobalt, Rhodium und dergleichen. Der Dehydrierungskatalysator wird gewöhnlich in Form eines Dehydriermetalles oder dessen Verbindung verwendet, das auf einem geeigneten Träger wie Aluminiumoxid, Kohlenstoff, Ton, Zeolith, Chromoxid oder Zirkonoxid abgeschieden wird.
- Unter Anwendung des erfindungsgemäßen Verfahrens können Substituenten in den 4-, 5- und 6-Positionen enthaltende Pyridin-2,3-dicarboxylate bequem durch Mischen eines Dialkyldichlorsuccinats der Formel II mit mindestens einem Mol-Äquivalent eines dehydrohalogenierenden Mittels in einem geeigneten Lösungsmittel, gegebenenfalls Abfiltrieren des Unlöslichen, Behandeln des Filtrats oder des unfiltrierten Reaktionsgemischs mit wasserfreiem Ammoniak, gegebenenfalls Erhitzen dieser Reaktionsmischung bis die Bildung des Enamin-Zwischenprodukts vollständig ist, gegebenenfalls Abfiltrieren des Unlöslichen, Zugabe einer Säure wie einer Mineralsäure, Schwefelsäure, Phosphorsäure, einer organischen Säure wie Ameisensäure, Essigsäure oder Propionsäure und dergleichen zu dem Filtrat oder der unfiltrierten Reaktionsmischung und Behandlung mit mindestens einem Mol-Äquivalent eines α,β-ungesättigten Aldehyds oder Ketons der Formel III, gegebenenfalls unter Zugabe eines Dehydrierkatalysators, gegebenenfalls unter Aufheizen des Reaktionsgemischs bis die Bildung des Pyridindicarboxylats der Formel I vollständig ist, hergestellt werden.
- Nach Vervollständigung der Umsetzung kann das gewünschte Pyridin- 2,3-dicarboxylat mit herkömmlichen Verfahren wie Extraktion, Destillαtion, Umkristallisation, Chromatografie und dergleichen isoliert und gereinigt werden.
- Die Bildungsgeschwindigkeit des zweiten Enamin-Zwischenprodukts und des Produkts der Formel I ist temperaturabhängig, so daß die Reaktionszeit wirksam durch Erhitzen des Reaktionsgemischs auf Temperaturen von ungefähr 45ºC oder höher verkürzt werden kann.
- Um das Verständnis der Erfindung weiter zu erleichtern werden die folgenden Beispiele aufgeführt, was hauptsächlich zum Zwecke der Erläuterung bestimmter spezieller Einzelheiten geschieht, wobei die Erfindung dadurch jedoch nicht eingeschränkt wird. Die Ausdrücke ¹H-NMR und IR bedeuten Protonen-Kernresonanz und Infrarot. Wenn nicht anders definiert, bedeuten alle Teile Gewichtsteile.
- Eine gerührte Mischung aus Diethyldichlorsuccinat (12.2 g, 0.05 Mol) und Ammoniumacetat (15.4 g, 0.20 Mol) in absolutem Ethanol wird 6 Stunden auf 60ºC erhitzt, auf 20ºC abgekühlt, mit überschüssigem wäßrigem Ammoniak behandelt (5.0 g, 0.29 Mol), 4.5 Stunden auf 60ºC erhitzt, zur Entfernung überschüssigen Ammoniaks teilweise destilliert, auf 30ºC abge kühlt und filtriert. Der Filterkuchen wird mit Ethanol gewaschen, die Filtrate werden vereinigt und im Vakuum konzentriert. Die so konzentrierte Reaktionslösung wird mit Essigsäure behandelt, tropfenweise mit einer ethanolischen Lösung von 2-Ethacrolein bei 70ºC über einen Zeitraum von 1.5 Stunden behandelt, weiter bei 70ºC 4 Stunden erhitzt (bis die Umsetzung nach Gaschromatograf ie-Analyse vollständig ist) und im Vakuum konzentriert, um einen rohen öligen Rückstand zu erhalten. Der ölige Rückstand wird destilliert, um das Titelprodukt als braunes Öl mit dem Siedepunkt 151-152ºC bei 2 mm Hg zu erhalten.
- Eine Lösung von Diethyldichlorsuccinat (24.6 g, 0.10 Mol) in Ethanol wird mit Triethylamin (12.2 g, 0.12 Mol) 5 Minuten lang behandelt, 12 Stunden bei Umgebungstemperatur gerührt, mit wasserfreiem Ammoniak (7.3 g, 0.43 Mol) bei 20ºC behandelt, 3 Stunden auf 45-50ºC erhitzt und filtriert. Der Filterkuchen wird mit Ethanol gewaschen, die Filtrate werden vereinigt und im Vakuum konzentriert, um das Lösungsmittel und überschüssigen Ammoniak zu entfernen. Das erhaltene viskose Öl wird mit Ethanol und Essigsäure verdünnt, mit 2-Ethacrolein (19.5 g, 0.232 Mol) behandelt, 6 Stunden auf 70ºC erhitzt und bei 60ºC im Vakuum destilliert, um das Lösungsmittel zu entfernen. Die so erhaltene Flüssigkeit wird in Toluol aufgenommen, mit Wasser gewaschen und im Vakuum konzentriert, um einen rohen öligen Rückstand zu erhalten. Vakuumdestillation dieses Rückstands ergibt das Titelprodukt als braunes Öl mit dem Siedepunkt 151- 152ºC bei 2 mm Hg.
- Unter Verwendung des im Wesentlichen gleichen Verfahrens wie in Beispiel 2 unter Austausch des entsprechenden Dialkyldichlorsuccinats und des geeigneten α,β-ungesättigten Aldehyds werden die folgenden Pyridindicarboxylate der Formel I erhalten.
- Die obigen Verbindungen werden mittels ¹H-NMR- und IR-spektroskopischer Analyse identifiziert.
Claims (7)
1. Ein Verfahren zur Herstellung einer durch die Formel I
gekennzeichneten Verbindung
worin R C&sub1;-C&sub6;-Alkyl ist; X und Z unabhängig voneinander Wasserstoff,
Halogen, C&sub1;-C&sub4;-Alkyl, C&sub1;-C&sub4;-Alkoxy oder C&sub2;-C&sub5;-Alkenyl sind und Y
Wasserstoff, Halogen, gegebenenfalls mit 1 bis 3 Halogenen, Hydroxygruppen, C&sub1;-
C&sub4;-Alkoxygruppen oder C&sub1;-C&sub4;-Alkylthiogruppen, C&sub1;-C&sub4;-Alkoxycarbonyl,
Aminocarbonyl, Phenylthio, substituiertem Phenylthio, Phenoxy,
substituiertem Phenoxy, Phenyl oder substituiertem Phenyl substituiertes C&sub1;-C&sub6;-
Alkyl ist, welches die Umsetzung eines Dialkyldichlorsuccinats der Formel
worin R C&sub1;-C&sub6;-Alkyl ist mit mindestens einem Mol-Äquivalent eines
dehydrohalogenierenden Agens, das auch als Ammonium-Quelle dient wie
Ammoniumacetat, Ammoniumcarbonat, Ammoniumcarbamat, Ammoniumsulfamat oder
einem dehydrohalogenierenden Agens wie Trialkylaminen, Pyridinen,
Picolinen und quartären Aminsalzen in Gegenwart eines polaren
Lösungsmittels um ein erstes Zwischenprodukt zu bilden, die Umsetzung dieses
ersten Zwischenprodukts mit Ammoniak oder einer oben erwähnten Ammoniak-
Quelle zur Bildung eines zweiten Zwischenprodukts, die Umsetzung dieses
zweiten Zwischenprodukts mit mindestens einem Mol-Äquivalent eines α,β-
ungesättigten Aldehyds oder Ketons der Formel III
worin X, Y und Z wie oben beschrieben sind und einer Säure in Gegenwart
eines Lösungsmittels sowie gegebenenfalls die Zugabe eines
Dehydrierkatalysators, einschließlich der Metalle oder Verbindungen von Platin,
Palladium, Ruthenium, Iridium, Nickel, Eisen, Kupfer, Antimon, Kobalt und
Rhodium umfaßt, um die Pyridincarboxylatverbindung der Formel I zu
bilden.
2. Das Verfahren gemäß Anspruch 1 zur Herstellung einer durch die Formel
I charakterisierten Pyridindicarboxylatverbindung, worin X und Z
Wasserstoff sind und Y Wasserstoff, Halogen oder gegebenenfalls mit einem bis
drei Halogenen, C&sub1;-C&sub4;-Alkoxygruppen oder C&sub1;-C&sub4;-Alkylthiogruppen
substituiertes C&sub1;-C&sub4;-Alkyl ist.
3. Das Verfahren gemäß Anspruch 1, worin das dehydrohalogenierende Agens
ein Ammoniumsalz ist, das Lösungsmittel ein polares Lösungsmittel ist,
die Säure Essigsäure ist und das zweite Zwischenprodukt bei einer
Temperatur von 45ºC oder höher gebildet wird.
4. Das Verfahren gemäß Anspruch 1, worin die
Pyridindicarboxylatverbindung der Formel I bei einer Temperatur von 45ºC oder höher gebildet
wird.
5. Das Verfahren gemäß Anspruch 1, worin der α,β-ungesättigte Aldehyd
oder das Keton der Formel III in einer Menge von mindestens einem Mol-
Äquivalent vorliegt.
6. Das Verfahren gemäß Anspruch 2, worin die hergestellte Verbindung der
Formel I aus der aus Dialkylpyridin-2,3-dicarboxylat, Dialkyl-5-methyl-
2,3-pyridindicarboxylat, Dialkyl-5-ethyl-2,3-pyridindicarboxylat,
Dialkyl-5-(methoxymethyl)-2,3-pyridindicarboxylat und Dialkyl-5-(chlormethyl)
-2,3-pyridindicarboxylat bestehenden Gruppe ausgewählt wird.
7. Das Verfahren gemäß Anspruch 6, worin die hergestellte Verbindung der
Formel I aus der aus Diethylpyridin-2,3-dicarboxylat, Diethyl-5-methyl-
2,3-pyridindicarboxylat, Diethyl-5-ethyl-2,3-pyridindicarboxylat,
Diethyl-5-(methoxymethyl)-2,3-pyridindicarboxylat und
Diethyl-5-(chlormethyl)-2,3-pyridindicarboxylat bestehenden Gruppe ausgewählt wird.
