Description
- Diese Erfindung betrifft Imidazolinon-Verbindungen und besonders Verfahren und Zwischenprodukte, die brauchbar sind zum Herstellen von -Carboxyarylimidazolinon-Verbindungen, die als Herbizide brauchbar sind.
- Neue herbizide Imidazolinyl-Benzoesäuren, -Nikotinsäuren und -Chinolin-3-carbonsäuren, Ester und Salze und deren Herstellung und Verwendung sind in den US-PSn 4,188,487; 4,297,128 und 4,638,068 offenbart.
- Die vorliegende Erfindung schafft ein Verfahren zum Herstellen von -Carboxyarylimidazolinon-Verbindungen durch Oxidieren der geeigneten 2-{[(1-Carbamoyl-1,2-dimethylpropyl)amino]methyl}benzoesäure, -nikotinsäure oder -chinolin-3-carbonsäure mit einem Bromierungsmittel.
- Die Erfindung schafft auch ein Verfahren zum Herstellen der 2-{[(1-Carbamoyl-1,2-dimethylpropyl)amino]methyl}-Zwischenprodukte durch Alkylieren von 2-Methylvalinamiden mit dem geeigneten -Halogenmethylarylcarboxylat. Im Falle von Pyridin-Halogenmethyl-Verbindungen wird ein 2-Chlor-4-halogenacetoacetatester mit einem α,β-ungesättigten Aldehyd oder -Keton umgesetzt, um einen 2-(Halogenmethyl)nikotinsäureester zu bilden.
- Die Erfindung schafft weiter gewisse 2-{[(1-Carbamoyl-1,2-dimethylpropyl)amino]methyl}-benzoesäure-, -nikotinsäure- oder -chinolin-3-carbonsäure-Verbindungen und gewisse 2-(Halogenmethyl)benzoesäureester, -nikotinsäureester, eine Chinolin-3-carbonsäureester-Verbindung, die als Zwischenprodukte in den obigen Verfahren brauchbar sind.
- Die Erfindung schafft auch ein Verfahren zum Herstellen von -Carboxypyridylimidazolin-Verbindungen durch eine Reihe von Stufen, ausgehend von den 2-Chlor-4- halogenacetoacetaten, durch Umsetzungen, wie sie oben beschrieben sind, bis zur Oxidation von 2-{[(1-Carbamoyl- 1,2-dimethylpropyl)amino]methyl}-nikotinsäureester mit einem Bromierungsmittel.
- Das Oxidationsverfahren dieser Erfindung, das auf dem Einsatz einer Bromquelle beruht, ergibt unerwartete Ergebnisse. Andere Oxidationsmittel, wie Schwefel, Chlor, Iod und Manganoxid, sind hinsichtlich des erwünschten Ergebnisses unwirksam.

- werden hergestellt durch Umsetzen einer 2-{[(1-Carbamoyl- 1,2-dimethylpropyl)amino]methyl}-benzoesäure, -nikotinsäure oder -chinolin-3-carbonsäure der Formel II
- worin A, R&sub1;, R&sub2; und R&sub3; die oben für Formel I angegebene Bedeutung haben, mit mindestens zwei molaren Äquivalenten eines Bromierungsmittels, wie Brom, N-Bromsuccinimid, N-Bromacetamid und ähnlichem, in einem inerten organischen Lösungsmittel, wie Essigsäure, in Gegenwart eines Säureakzeptors, wie Natriumacetat, bei einer Temperatur im Bereich von etwa 20 bis 100ºC für eine genügende Zeitdauer, um im wesentlichen die Umsetzung auszuführen, wie sie im folgenden Fließdiagramm I dargestellt ist. - 2-{[(1-Carbamoyl-1,2-dimethylpropyl)amino]methyl}-Verbindungen der Formel II, die geeignet sind zum Einsatz im Verfahren dieser Erfindung, können hergestellt werden durch Umsetzen eines 2-(Halogenmethyl)-benzoesäureesters, -nikotinsäureesters oder -chinolin-3-carbonsäureesters der Formel III
- worin R&sub1; C&sub1;-C&sub1;&sub2;-Alkyl ist und A, R&sub2; und R&sub3; die oben für Formel I angegebene Bedeutung haben und X Cl oder Br ist, mit mindestens einem molarem Äquivalent eines razemischen 2-Amino-2,3-dimethylbutyramids oder eines einzelnen optischen Isomers davon, in einem inerten organischen Lösungsmittel, wie Dimethylsulfoxid, Aceton oder ähnlichem, in Gegenwart einer Base, wie Natriumbicarbonat, gegebenenfalls in Gegenwart einer katalytischen Menge von NaI, bei einer Temperatur im Bereich von 25ºC bis 100ºC, wie im folgenden Fließdiagramm II veranschaulicht Fließdiagramm II

- Verbindungen der Formel II, worin R&sub1; Wasserstoff ist, die geeignet sind zum Einsatz bei dem Verfahren dieser Erfindung, können leicht aus Verbindungen der Formel II, bei denen R&sub1; C&sub1;-C&sub1;&sub2;-Alkyl ist, durch Hydrolyse hergestellt werden.
- Die vorliegende Erfindung schließt neue Verbindungen der folgenden Formel II ein
- worin A CH oder N ist;
- R&sub1; H oder C&sub1;-C&sub1;&sub2;-Alkyl ist; R&sub2; H oder C&sub1;-C&sub6;-Alkyl ist; R&sub3; H, C&sub1;-C&sub6;-Alkyl ist oder, wenn R&sub2; und R&sub3; zusammengenommen werden, sie einen Ring bilden können, der repräsentiert ist durch
- -CH=CH-CH=CH-;
- sowie neue Verbindungen der Formel III
- worin R&sub1; C&sub1;-C&sub1;&sub2;-Alkyl ist und R&sub2; und R&sub3; die für die obige Formel II angegebene Bedeutung haben und X Cl oder Br ist, die brauchbare Zwischenprodukte für die Herstellung herbizider Imidazolinon-Verbindungen unter Einsatz der Verfahren dieser Erfindung sind. Eine bevorzugte Gruppe von neuen Verbindungen der Formel II und III sind solche, worin
- R&sub1; C&sub1;-C&sub3;-Alkyl ist;
- R&sub2; H oder C&sub1;-C&sub3;-Alkyl ist und
- R&sub3; H ist und
- X Cl für Verbindung III ist.
- Pyridin-halogenmethyl-Verbindungen der Formel III zum Einsatz beim Verfahren dieser Erfindung können hergestellt werden durch ein Verfahren, analog dem, das in der US-PS 4,723,011 von R. Doehner beschrieben ist, durch Umsetzen eines 2-Chlor-4-halogenacetoacetatesters der Formel IV
- worin R&sub1; C&sub1;-C&sub1;&sub2;-Alkyl ist und X Cl oder Br ist, mit einem α,β-ungesättigten Aldehyd oder -keton der Formel V
- worin R&sub2; H oder C&sub1;-C&sub6;-Alkyl ist und R&sub3; H ist, in Gegenwart von mindestens zwei molaren Äquivalenten eines Ammoniumsalzes in einem organischen Lösungsmittel, bei einer Temperatur im Bereich von Umgebungstemperatur bis zum Siedepunkt des Lösungsmittels, bis die Umsetzung im wesentlichen abgeschlossen ist, wie im folgenden Fließdiagramm III veranschaulicht. - Die vorliegende Erfindung schließt auch ein Verfahren zum Herstellen von -Carboxypyridylimidazolinon-Verbindungen der Formel I ein - worin
- R&sub1;H oder C&sub1;-C&sub1;&sub2;-Alkyl ist;
- R&sub2;H oder C&sub1;-C&sub6;-Alkyl ist und
- R&sub3;H ist,
- durch nacheinander Umsetzen eines 2-Chlor- 4-halogen-acetoacetatesters der Formel IV
- mit einem α,β-ungesättigten Aldehyd oder -Keton der Formel V
- in Gegenwart von mindestens zwei molaren Äquivalenten eines Ammoniumsalzes in einem organischen Lösungsmittel, bei einer Temperatur im Bereich von Umgebungstemperatur bis zum Siedepunkt des Lösungsmittels, bis die Umsetzung im wesentlichen abgeschlossen ist und Umsetzen des so gebildeten 2-(Halogenmethyl)-nikotinsäureesters der Formel III
- worin R&sub1;, R&sub2;, R&sub3; und X die oben für Formel IV und V angegebenen Bedeutungen haben- mit mindestens einem molarem Äquivalent von razemischem 2-Amino-2,3-dimethylbutyramid oder einem einzelnen optischen Isomer davon, in einem inerten organischen Lösungsmittel in Gegenwart einer Base, gegebenenfalls in Gegenwart einer katalytischen Menge von NaI, bei einer Temperatur im Bereich von etwa 25ºC bis 100ºC und weiteres Umsetzen des so gebildeten 2-{[(1-Carbamoyl-1,2-dimethylpropyl)amino]methyl}-nikotinsäureesters oder der entsprechenden Säure der Formel II
- worin R&sub1;, R&sub2; und R&sub3; die oben für Formel I genannten Bedeutungen haben, mit mindestens zwei molaren Äquivalenten eines Bromierungsmittels in einem inerten organischen Lösungsmittel in Gegenwart eines Säureakzeptors, bei einer Temperatur im Bereich von etwa 25ºC bis 100ºC, für eine genügende Zeitdauer, um die Umsetzung im wesentlichen abzuschließen.
- Die Herstellung anderer Zwischenprodukte der Formel III folgt Literaturverfahren, wie Oxidation eines 2-Methyl-3-pyridincarboxylats oder eines 2-Methyl-3-chinolincarboxylats, gefolgt von der Umlagerung des resultierenden N-Oxids mit POCl&sub3;, um Verbindungen der Formel III zu ergeben, wie sie im folgenden veranschaulicht sind.
- Die Erfindung wird weiter durch die folgenden Beispiele veranschaulicht.
- Eine Lösung von 61 g von Ethyl-2,4-dichloracetoacetat (0,306 mol) und 30 g von 2-Ethylacrolein (0,357 mol) in 500 ml absolutem Alkohol wurde mit 85,8 g Ammoniumsulfamat (0,75 mol) vermischt und die gerührte Mischung 90 Minuten lang am Rückfluß erhitzt. Die Reaktionsmischung wurde im Vakuum konzentriert und der Rest zwischen Ethylacetat und Wasser aufgeteilt. Die organische Phase wurde im Vakuum konzentriert und der Rest an Siliciumdioxidgel unter Einsatz von Hexan-Ethylacetat- Mischungen chromatographiert, wobei 50 g Rohprodukt erhalten wurden. Dieses Material wurde durch Extraktion in wässerige Chlorwasserstoffsäure, Waschen mit Hexanen, Basischmachen der wässerigen Phase mit kaltem Ammoniumhydroxid und Extrahieren mit Hexanen gereinigt, wobei 30 g des Titelproduktes als ein Öl (43%) erhalten wurden, dessen Elementaranalyse ergab:
- Errechnet für C&sub1;&sub1;H&sub1;&sub4;ClNO&sub2;: %C 58,03; H 6,20; N 6,15; Cl 15,57; Gefunden : %C 58,29; H 6,30; N 6,02; Cl 15,49.
- Die Anwendung des obigen Verfahrens ergibt die in der folgenden Tabelle 1 aufgeführten Verbindungen der Formel III. Tabelle I Analyse (errechnet) niedrigschmelzender Feststoff Öl
- Eine Lösung von 9 g von Ethyl-2-methylnikotinat in 250 ml Methylenchlorid wurde bei Raumtemperatur gerührt, und 32 g 80%-iger m-Chlorperoxybenzoesäure wurden in einer Portion hinzugegeben. Die resultierende Lösung wurde drei Tage lang gerührt. Der ausgefallene Feststoff wurde abfiltriert und das Filtrat mit kaltem verdünntem wässerigem Natriumhydroxid gewaschen, getrocknet und im Vakuum konzentriert, wobei das rohe N-Oxid erhalten wurde. Dieses Material wurde in 75 ml von 1,2-Dichlorethan digeriert, und es wurden 15 ml-Phosphoroxychlorid hinzugegeben und die Lösung über Nacht am Rückfluß erhitzt. Die Lösung wurde im Vakuum konzentriert und der Rest in Methylenchlorid aufgenommen und mit wässerigem Natriumacetat neutralisiert. Die organische Phase wurde getrocknet, im Vakuum konzentriert und der Rest an Siliciumdioxidgel unter Einsatz von Hexan-Ethylacetat-Mischungen chromatographiert, wobei 1,2 g des Titelproduktes als ein Öl erhalten wurden, das ein NMR(CDCl&sub3;) δ(ppm) hatte: 1,4 (+, 3H), 4,5 (q, 2H), 5,1 (2H), 7,4 ( , 1H), 8,4 (dd, 1H), 8,8 (dd, 1H).
- Nach diesem Verfahren wurde auch Ethyl-2-chlormethylchinolin-3-carboxylat hergestellt;
- NMR(CDCl&sub3;) δ: 1,4 (+, 3H), 4,5 (q, 2H), 5,3 (5, 2H), 7,5-8,3 (m, 4H), 9,0 (5, 1H).
- Eine Mischung von 77 g von Ethyl-2-chlormethyl- 5-ethylnikotinat (0,338 mol), 44 g von α-Methylvalinamid (0,338 mol) und 33,5 g von Natriumbicarbonat (0,4 mol) in 60 ml Dimethylsulfoxid wurde gerührt und 16 Stunden bei 80ºC erhitzt. Die Reaktionsmischung wurde zwischen 1 : 1 Ethylacetat-Hexan und Wasser aufgeteilt. Die organische Schicht wurde gründlich mit Wasser gewaschen, getrocknet und im Vakuum zu einem Kautschuk konzentriert. Chromatographie dieses Kautschuks an Siliciumdioxidgel unter Einsatz von Hexan-Ethylacetat-Mischungen als Elutionsmittel ergab das Titelprodukt, F 68-72ºC.
- Die obige Umsetzung wurde mit Aceton als Lösungsmittel (mit Katalysator NaI) ausgeführt und ergab auch das Titelprodukt.
- Unter Anwendung des obigen Verfahrens mit analogen Halogemethyl-Verbindungen der Formel III ergab die in der folgenden Tabelle II aufgeführten Verbindungen. Tabelle II Feststoff mp = Schmelzpunkt
- Eine Mischung von 1 g von Ethyl-2-{[(1-carbamoyl-1,2-dimethylpropyl)amino]methyl}-5-ethylnikotinat (3,1 mmol) und 0,58 g wasserfreiem Natriumacetat (7 mmol) in 10 ml Essigsäure wurde auf 50ºC erwärmt, bei welcher Temperatur die Reaktionsmischung eine homogene Lösung war. Eine Lösung von 1 g Brom (6,2 mmol) in 2 ml Essigsäure wurde in zwei Portionen über zwei Minuten hinzugegeben und die Reaktionsmischung 16 Stunden lang bei 50ºC gerührt. Die Reaktionsmischung wurde im Vakuum konzentriert und der Rest zwischen Ethylacetat und Wasser verteilt. Die organische Schicht wurde mit wässerigem Natriumbisulfit gewaschen, über MgSO&sub4; getrocknet, mit einem gleichen Volumen von Hexanen verdünnt und durch ein Polster von Siliciumdioxidgel filterchromatographiert. Das Siliciumdioxidgel wurde mit zusätzlichem 1 : 1 Hexan-Ethylacetat weiter eluiert und die kombinierten Eluate im Vakuum konzentriert, um 0,6 g Produkt mit einem Schmelzpunkt (F) von 84,5ºC bis 86,5ºC zu ergeben. Das Titelprodukt wird auch erhalten unter Einsatz von N-Bromacetamid oder N-Bromsuccinimid anstelle von Brom.
- Unter Anwendung des obigen Verfahrens auf verschiedene Verbindungen der Formel II erhielt man die in der folgenden Tabelle III aufgeführten Verbindungen der Formel I. Tabelle III

- Eine Lösung von 5,6 g von Ethyl-2-{[(1-carbamoyl-1,2-dimethylpropyl)amino]methyl}-5-ethylnikotinat (0,0174 mol) in 30 ml Methanol, enthaltend 2 g NaOH (0,05 mol) und 10 ml Wasser wurde zweieinhalb Stunden bei Raumtemperatur gerührt. Die Reaktionsmischung wurde im Vakuum konzentriert und wieder in 30 ml Wasser gelöst; die Einstellung des pH mit konzentrierter Chlorwasserstoffsäure auf 4 und das Konzentrieren im Vakuum ergab einen Kautschuk. Dieser Rest wurde in einer Mischung von Ethylacetat, Tetrahydrofuran und Methanol gelöst, über MgSO&sub4; getrocknet und im Vakuum konzentriert, wobei 6 g Schaum erhalten wurden. Das Kristallisieren aus Methanol-Ether ergab 3,1 g Produkt, F 180-181ºC.
- Eine Mischung von 0,8 g von 2-{[(1-Carbamoyl- 1,2-dimethylpropyl)amino]methyl}-5-ethylnikotinsäure (2,7 mmol) und 0,56 g Natriumacetat (6,8 mmol) in 10 ml Essigsäure wurde erwärmt, bis sie homogen war, und auf Raumtemperatur abgekühlt. Die Lösung wurde mit 0,88 g Brom (5,45 mmol) behandelt, und die Reaktionsmischung 16 Stunden lang bei 25ºC und dann drei Tage lang bei 75ºC gerührt. Die Reaktionsmischung wurde zwischen CH&sub2;Cl&sub2; und Wasser aufgeteilt und die organische Phase getrocknet und im Vakuum konzentriert. Der Rest wurde aus Ethylacetat- Hexanen umkristallisiert und ergab 0,4 g des Titelproduktes mit F 172-175ºC.
Claims (9)
1. Verfahren zum Herstellen von
-Carboxyarylimidazolinon-Verbindungen der Formel I
worin A CH oder N ist;
R&sub1; H oder C&sub1;-C&sub1;&sub2;-Alkyl ist;
R&sub2; H oder C&sub1;-C&sub6;-Alkyl ist;
R&sub3; H, C&sub1;-C&sub6;-Alkyl ist, oder wenn R&sub2; und R&sub3;
zusammengenommen werden, sie einen Ring bilden
können, der durch
-CH=CH-CH=
CHrepräsentiert ist;
umfassend das Umsetzen einer
2-{[(1-Carbamoyl-1,2-dimethylpropyl)amino]methyl}benzoesäure, -nicotinsäure oder
-chinolin-3-carbonsäure der Formel II
worin A, R&sub1;, R&sub2; und R&sub3; die für Formel I oben gegebene
Bedeutung haben, mit mindestens 2 molaren Äquivalenten
eines Bromierungsmittels in Gegenwart eines
Säureakzeptors.
2. Verfahren zum Herstellen von 2-{[(1-Carbamoyl-1,2-
dimethylpropyl)amino]methyl}-Verbindungen der Formel II
worin A CM oder N ist;
R&sub1; H oder C&sub1;-C&sub1;&sub2;-Alkyl ist;
R&sub2; H oder C&sub1;-C&sub6;-Alkyl ist;
R&sub3; H, C&sub1;-C&sub6;-Alkyl ist, oder, wenn R&sub2; und R&sub3;
zusammengenommen werden, sie einen Ring bilden
können, der durch
-CH=CH-CH=
CHrepräsentiert ist;
umfassend das Umsetzen eines
2-(Halogenmethyl)benzoesäureesters, -nicotinsäureesters oder
-chinolin-3-carbonsäureesters der Formel III
worin R&sub1; C&sub1;-C&sub1;&sub2;-Alkyl ist und A, R&sub2; und R&sub3; die oben für
Formel II angegebene Bedeutung haben, und X Cl oder Br
ist, mit mindestens einem molaren Äquivalent von
racemischem 2-Amino-2,3-dimethylbutyramid oder einem
einzelnen optischen Isomer davon in Gegenwart einer Base.
3. Verfahren nach Anspruch 2, worin eine katalytische
Menge von NaI vorhanden ist.
4. Verfahren nach Anspruch 2, worin die Umsetzung in
einem inerten Lösungsmittel, Dimethylsulfoxid oder
Aceton, ausgeführt wird.
5. Verfahren zum Herstellen von
-Carboxypyridylimidazolinon-Verbindungen der Formel I
worin
R&sub1; H oder C&sub1;-C&sub1;&sub2;-Alkyl ist;
R&sub2; H oder C&sub1;-C&sub6;-Alkyl ist und
R&sub3; H ist;
durch eine Stufenfolge, umfassend
Umsetzen eines 2-Chlor-4-halogenacetoacetatesters der
Formel IV
worin R&sub1; C&sub1;-C&sub1;&sub2;-Alkyl und X Cl oder Br ist, mit einem
α,β-ungesättigten Aldehyd oder Keton der Formel V
worin R&sub2; H oder C&sub1;-C&sub6;-Alkyl und R&sub3; H ist, in Gegenwart
von mindestens zwei molaren Äquivalenten von
Ammoniumsalz, bis die Umsetzung im wesentlichen vollständig
abgeschlossen ist und
Umsetzen des so gebildeten
2-(Halogenmethyl)-nicotinsäureesters der Formel III
worin R&sub1;, R&sub2;, R&sub3; und X die oben für Formel IV und V
angegebene Bedeutung haben, mit mindestens einem molaren
Äquivalent von racemischem 2-Amino-2,3-dimethylbutyramid
oder einem einzelnen optischen Isomer davon, in Gegenwart
einer Base, gegebenenfalls in Gegenwart einer
katalytischen Menge von NaJ und weiter
Umsetzen des so gebildeten
2-{[(1-Carbamoyl-1,2-dimethylpropyl)amino]methyl}nicotinsäureesters oder der davon
abgeleiteten Säure der Formel II
worin R&sub1;, R&sub2; und R&sub3; die oben für Formel I angegebene
Bedeutung haben, mit mindestens zwei molaren Äquivalenten
eines Bromierungsmittels.

7. Verbindung nach Anspruch 6, worin
R&sub1; H oder C&sub1;-C&sub3;-Alkyl ist;
R&sub2; H oder C&sub1;-C&sub3;-Alkyl ist und
R&sub3; H ist.
8. 2-Halogenmethyl-nicotinsäureester der Formel III
worin
R&sub1; C&sub1;-C&sub1;&sub2;-Alkyl ist;
R&sub2; C&sub1;-C&sub6;-Alkyl oder H ist;
R&sub3; H, C&sub1;-C&sub6;-Alkyl ist, oder, wenn R&sub2; und R&sub3;
zusammengenommen werden, sie einen Ring bilden
können, der durch -CH=CH-CH=CH- repräsentiert
ist;
X ist Cl oder Br.
9. Verbindung nach Anspruch 8, worin
R&sub1; C&sub1;-C&sub3;-Alkyl ist;
R&sub2; H oder C&sub1;-C&sub3;-Alkyl ist;
R&sub3; H ist und
X Cl ist.
