Description
- Die vorliegende Erfindung betrifft eine Gruppe von neuartigen Enantiomeren von 4,5-Dihydro-1H-pyrazolderivaten mit S-Konfiguration an der 4-Position ihres 4,5-Dihydropyrazolrings, Verfahren zur Herstellung dieser Verbindungen und pharmazeutische Zusammensetzungen, welche eine oder mehrere dieser Verbindungen als Wirkstoff enthalten.
- Die oben erwähnten (4S)-4,5-Dihydro-1H-pyrazole sind wirkkräftige Cannabis-1 (CB1) Rezeptor Antagonisten mit Nützlichkeit zur Behandlung von psychiatrischen und neurologischen Störungen.
- Cannabinoide sind im Indischen Hanf Cannabis Sativa L. vorhanden und sind seit Jahrhunderten als medizinische Wirkstoffe verwendet worden (Mechoulam, R.; Feigenbaum, J. J. Prog. Med. Chem. 1987, 24, 159). Jedoch hat erst in den vergangenen zehn Jahren die Forschung im Cannabinoid-Bereich entscheidende Informationen über Cannabinoid-Rezeptoren und ihre (endogenen) Agonisten und Antagonisten offenbart. Die Entdeckung und die nachfolgende Klonierung von zwei unterschiedlichen Subtypen von Cannabinoid-Rezeptoren (CB1 und CB2) stimulierte die Suche nach neuartigen Cannabinoid-Rezeptor Antagonisten (Munro, S.; Thomas, K. L.; Abu-Shaar, M. Nature 1993, 365, 61. Matsuda, L. A.; Bonner, T. I. Cannabinoid Receptors, Pertwee, R. G. Hrsg. 1995, 117, Academic Press, London). Zusätzlich interessierten sich pharmazeutische Firmen für die Entwicklung von cannabinoiden Arzneien zur Behandlung von Krankheiten in Verbindung mit Störungen des cannabinoiden Systems. Die weite Verteilung von CB1-Rezeptoren im Gehirn macht in Kombination mit der streng peripheren Lokalisierung der CB2 Rezeptoren den CB1-Rezeptor zu einem sehr interessanten molekularen Ziel für auf CNS gerichtete Arzneientdeckung in den Bereichen von sowohl psychiatrischen, als auch neurologischen Störungen (Consroe, P. Neurobiology of Disease 1998, 5, 534. Pop, E. Curr. Opin. In CPNS Investigational Drugs 1999, 1, 587. Greenberg, D. A. Drug News Perspect. 1999, 12, 458). Bisher sind drei Typen von unterschiedlichen CB1-Rezeptor Antagonisten bekannt. Sanofi offenbarte ihre Diarylpyrazol-Verwandten als selektive CB1-Rezeptor Antagonisten. Ein repräsentatives Beispiel ist SR-141716A, welches gegenwärtig Phase II der klinischen Entwicklung für psychotische Störungen durchläuft (Dutta, A. K.; Sard, H.; Ryan, W.; Razdan, R. K.; Compton, D. R.; Martin, B. R. Med. Chem. Res. 1994, 5, 54. Lan, R.; Liu, Q.; Fan, P.; Lin, S.; Fernando, S. R.; McCallion, D.; Pertwee, R.; Makriyannis, A. J. Med. Chem. 1999, 42, 769. Nakamura-Palacios, E. M.; Moerschbaecher, J. M.; Barker, L. A. CNS Drug Rev. 1999, 5, 43). Aminoalkylindole sind als CB1-Rezeptor Antagonisten offenbart worden. Ein repräsentatives Beispiel ist Lodopravadolin (AM-630), welches 1995 eingeführt wurde. AM-630 ist ein CB1-Rezeptor Antagonist, aber benimmt sich manchmal wie ein schwacher Teil-Agonist (Hosohata, K.; Quock, R. M.; Hosohata, Y.; Burkey, T. H.; Makriyannis, A.; Consroe, P.; Roeske, W. R.; Yamamura, H. I. Life Sc. 1997, 61, PL 115). Kürzlich beschrieben Forscher von Eli Lilly Aryl-aroyl substituierte Benzofurane als selektive CB1-Rezeptor Antagonisten (z. B. LY-320135) (Felder, C. C.; Joyce, K. E.; Briley, E. J.; Glass, M.; Mackie, K. P.; Fahey, K. J.; Cullinan, G. J.; Hunden, D. C.; Johnson, D. W.; Chaney, M. O.; Koppel, G. A.; Brownstein, M. J. Pharmacol. Exp. Ther. 1998, 284, 291). Kürzlich wurden 3-Alkyl-5,5'-diphenylimidazolidindione als Cannabinoid-Rezeptor Liganden beschrieben, von welchen angezeigt wurde, dass sie Cannabinoid-Antagonisten sind (Kanyonyo, M.; Govaerts, S. J.; Hermans, E.; Poupaert, J. H., Lambert, D. M.; Biorg. Med. Chem. Lett. 1999, 9, 2233). Interessanterweise ist von vielen CB1-Rezeptor Antagonisten berichtet worden, dass sie sich in vitro wie inverse Agonisten verhalten (Landsman, R. S.; Burkey, T. H.; Consroe, P.; Roeske, W. R.; Yamamura, H. I. Eur. J. Pharmacol. 1997, 334, R1). Kürzliche Übersichten sehen einen schönen Überblick über den gegenwärtigen Stand im Bereich der Cannabinoid-Forschung vor (Mechoulam, R.; Hanus. L.; Fride, E. Prog. Med. Chem. 1998, 35, 199. Lambert, D. M. Curr. Med. Chem. 1999, 6, 635. Mechoulam, R.; Fride, E.; Di Marzo, V. Eur. J. Pharmacol. 1998, 359, 1).
- Es ist nun überraschend herausgefunden worden, dass die neuartigen Enantiomere von 4,5-Dihydro-1H-pyrazolderivaten mit S-Konfiguration an der 4-Position ihres 4,5-Dihydropyrazolrings der Formel (I), Tautomere davon und Salze davon worin:
R und R1 gleich oder unterschiedlich sind und 3-Pyridyl oder 4-Pyridyl oder Phenyl darstellen, welches mit Halogen oder Methoxy substituiert sein kann,
R2 und R3 gleich oder unterschiedlich sind und Wasserstoff, Alkyl (1–3 C) oder Dimethylamino darstellen,
R4 für Phenyl steht, welches mit 1, 2 oder 3 Substituenten substituiert sein kann, ausgewählt aus der Gruppe Halogen, Trifluormethyl, Methoxy und Alkyl (1–3 C) viel wirkkräftigere und selektivere Antagonisten des Cannabis CB1-Rezeptors sind, als das entsprechende R-Enantiomer. - Aufgrund der wirkkräftigen CB1-antagonistischen Wirksamkeit sind die Verbindungen gemäß der Erfindung zur Verwendung bei der Behandlung von psychiatrischen Störungen wie Psychose, Angststörung, Depression, Aufmerksamkeitsdefiziten, Gedächtnisstörungen und Appetitstörungen, Fettsucht, neurologischen Störungen wie Demenz, Dystonie, Parkinson-Erkrankung, Alzheimer-Erkrankung, Epilepsie, Huntington-Erkrankung, Tourette-Syndrom, cerebraler Ischämie, als auch zur Behandlung von Schmerzstörungen und anderen CNS-Störungen, welche Cannabinoid-Neurotransmission einbeziehen, und bei der Behandlung von gastrointestinalen und kardiovaskulären Störungen brauchbar.
- Die Affinität der Verbindungen der Erfindung für Cannabinoid CB1-Rezeptoren wurde unter Verwendung von Membranpräparaten von Eierstockzellen von Chinesischen Hamstern (CHO) bestimmt, in welche der humane Cannabis CB1-Rezeptor in Verbindung mit [3H]CP-55,940 als Radioligard stabil transfiziert wird. Nach Inkubation eines frisch hergestellten Zellmembranpräparats mit dem [3H]-Ligand mit oder ohne Zugabe von Verbindungen der Erfindung wurde Trennung von gebundenem und freiem Ligand durch Filtration über Glasfaserfilter durchgeführt. Die Radioaktivität an dem Filter wurde durch Flüssigkeitsszintillationszählung gemessen.
- Die cannabinoide CB1-antagonistische Wirksamkeit von Verbindungen der Erfindung wurde durch funktionale Studien unter Verwendung von CHO-Zellen bestimmt, in welchen humane cannabinoide CB1-Rezeptoren stabil exprimiert werden. Adenylylcyclase wurde unter Verwendung von Forskolin stimuliert und durch Quantifizieren der Menge an akkumuliertem cyclischen AMP gemessen. Begleitende Aktivierung von CB1-Rezeptoren durch CB1-Rezeptor Agonisten (z. B. CP-55,940 oder (R)-WIN-55,212-2) kann die durch Forskolin herbeigeführte Akkumulierung von cAMP auf eine Konzentrationsabhängige Weise abmildern. Diese CB1-Rezeptor vermittelte Response kann durch CB1-Rezeptor Antagonisten wie den Verbindungen der Erfindung antagonisiert werden.
- Die Erfindung betrifft sowohl das E-Isomer, Z-Isomer, als auch E/Z-Gemische von Verbindungen mit der Formel (I).
- Die Verbindungen können mittels üblicher Verfahren unter Verwendung von Hilfssubstanzen und/oder flüssigen oder festen Trägermaterialien in Formen gebracht werden, welche zur Verabreichung geeignet sind.
- Die Verbindungen der Erfindung mit der Formel (III) (vide infra) können gemäß bekannter Verfahren erhalten werden, z. B.: a) EP-0021506; b) DE-25 29 689.
- Eine geeignete Synthese für die racemischen Verbindungen gemäß der vorliegenden Erfindung ist die Folgende:

- Schritt 1 von Weg A
- Umsetzung einer Verbindung mit der Formel (III) mit einer Verbindung mit der Formel (IV) worin R5 eine nied.-Alkylgruppe darstellt, wie zum Beispiel 2-Methyl-2-thiopseudoharnstoff, oder mit einem geeigneten Salz davon in Gegenwart einer Base. Diese Umsetzung ergibt ein 4,5-Dihydro-1H-pyrazol-1-carboxamidinderivat mit der Formel (V) worin die Symbole die Bedeutungen wie oben erwähnt haben. Verbindungen mit der Formel (V), worin R, R1, R2 und R3 die hierin oben für Verbindung (I) beschriebene Bedeutung haben, sind neuartig.
- Alternativ wird eine Verbindung mit der Formel (III) mit einem sogenannten Guanylierungsmittel umgesetzt. Beispiele für solche Guanylierungsmittel sind 1H-Pyrazol-1-carboxamidin und seine Salze (zum Beispiel das Hydrochloridsalz) und 3,5-Dimethyl-1H-pyrazol-1-carboxamidin und seine Salze (zum Beispiel das Nitratsalz) und Ähnliches. Diese Umsetzung ergibt ein Carboxamidinderivat mit der Formel (V).
- Alternativ wird eine Verbindung mit der Formel (III) mit einem sogenannten geschützten Guanylierungsmittel umgesetzt. Beispiele für solche geschützten Guanylierungsmittel sind N-(Benzyloxycarbonyl)-1H-pyrazol-1-carboxamidin, N-(tert-Butoxycarbonyl)-1H-pyrazol-1-carboxamidin und N,N'-Bis(tert-butoxycarbonyl)-1H-pyrazol-1-carboxamidin und Ähnliches. Diese Umsetzung ergibt nach Entschützung eine Verbindung der Formel (V).
- Schritt 2 von Weg A
- Die Verbindung mit der Formel (V) wird mit einer gegebenenfalls substituierten Verbindung der Formel R4-SO2X umgesetzt, worin R4 die oben erwähnte Bedeutung hat und X ein Halogenatom darstellt. Diese Umsetzung wird vorzugsweise in Gegenwart einer Base wie Triethylamin in einem aprotischen Lösungsmittel wie Acetonitril durchgeführt.

- Diese Umsetzung wird vorzugsweise in einem inerten organischen Lösungsmittel wie zum Beispiel Acetonitril durchgeführt.
- Diese Umsetzung ergibt ein Thiocarboxamidderivat mit der Formel (VII). Verbindungen der Formel (VII), worin R, R1 und R4 die wie hierin oben für Verbindung (I) beschriebene Bedeutung haben sind neuartig.
- Schritt 2 von Weg A1
- Umsetzung einer Verbindung mit der Formel (VII) mit einem Amin in Gegenwart eines Quecksilber(II)-Salzes, wie zum Beispiel HgCl2 ergibt eine Verbindung mit der Formel (I). Diese Umsetzung wird vorzugsweise in einem polaren organischen Lösungsmittel wie zum Beispiel Acetonitril durchgeführt.

- Diese Umsetzung wird vorzugsweise in einem inerten organischen Lösungsmittel wie zum Beispiel 1,4-Dioxan durchgeführt.
- Diese Umsetzung ergibt ein 4,5-Dihydropyrazol-1-carboxamidderivat mit der Formel (IX). Verbindungen mit der Formel (IX), worin R, R1 und R4 die wie hierin oben für Verbindung (I) beschriebenen Bedeutungen haben, sind neuartig.
- Schritt 2 von Weg A2
- Umsetzung einer Verbindung mit der Formel (IX) mit einem Halogenierungsmittel wie zum Beispiel PCl5 ergibt ein 4,5-Dihydropyrazol-1-carboximidoyl-halogenidderivat mit der Formel (X) worin R7 ein Halogenatom darstellt, wie zum Beispiel Chlor. Diese Umsetzung wird vorzugsweise in einem inerten organischen Lösungsmittel wie zum Beispiel Chlorbenzol durchgeführt.
- Verbindungen mit der Formel (X), worin R, R1 und R4 die wie hierin oben für Verbindung (I) beschriebene Bedeutung haben und worin R7 ein Halogenatom darstellt, sind neuartig.
- Schritt 3 von Weg A2
- Umsetzung einer Verbindung mit der Formel (X) mit einem Amin ergibt eine Verbindung mit der Formel (I).
- Diese Umsetzung wird vorzugsweise in einem inerten organischen Lösungsmittel wie zum Beispiel Dichlormethan durchgeführt.

- Diese Umsetzung wird vorzugsweise in einem polaren organischen Lösungsmittel wie zum Beispiel Acetonitril durchgeführt.

- Schritt 2 von Weg A3
- Umsetzung einer Verbindung mit der Formel (XII) mit einem Amin ergibt eine Verbindung mit der Formel (I).
- Diese Umsetzung wird vorzugsweise in einem polaren organischen Lösungsmittel wie zum Beispiel Methanol durchgeführt.

- Teil A: Ein gerührtes Gemisch aus 3-(4-Chlorphenyl)-4,5-dihydro-4-phenyl-1H-pyrazol (5,13 Gramm, 20,0 mmol), 2-Methyl-2-thiopseudoharnstoff-hydroiodid (5,00 Gramm, 23,0 mmol) und Pyridin (10 ml) wird bei 110°C für 1 Stunde erhitzt. Nach einer Nacht Stehenlassen bei Raumtemperatur wird Diethylether zugegeben und der Niederschlag wird durch Filtration gesammelt. Dieser Niederschlag wird drei Mal mit Diethyletherportionen gewaschen, um einen Feststoff (9 Gramm) zu ergeben. Fließpunkt: ~230°C. Dieser Feststoff wird in Methanol (20 ml) gelöst. Zu der sich ergebenden Lösung werden nacheinander eine 2 N Natriumhydroxidlösung (12 ml) und Wasser (200 ml) zugegeben. Der gebildete Niederschlag wird durch Filtration gesammelt, zwei Mal mit Diethylether und darauf folgend mit Diisopropylether gewaschen. Der sich ergeben de Feststoff wird in vacuo getrocknet, um 3-(4-Chlorphenyl)-4,5-dihydro-4-phenyl-1H-pyrazol-1-carboxamidin (5,1 Gramm, 88% Ausbeute) zu ergeben. Fließpunkt: 187–189°C.
- Teil B: Zu einem gerührten Gemisch aus 3-(4-Chlorphenyl)-4,5-dihydro-4-phenyl-1H-pyrazol-1-carboxamidin (0,50 Gramm, 1,68 mmol) und 4-Fluorphenylsulfonylchlorid (0,34 Gramm, 1,75 mmol) in Acetonitril (10 ml) wird N,N-Dimethyl-4-aminopyridin (0,020 Gramm, 0,175 mmol) und Triethylamin (1 ml) zugegeben. Die sich ergebende Lösung wird bei Raumtemperatur für 30 Minuten gerührt. Nach Zugabe einer 2 N Natriumhydroxidlösung und Extraktion mit Ethylacetat (400 ml) wird die Ethylacetatschicht in vacuo konzentriert. Der sich ergebende rohe Rückstand wird mittels Blitzchromatographie (Petroleumether/Diethylether = 1/1 (Vol./Vol.), gefolgt von Ethylacetat) weiter gereinigt. Nachfolgende Konzentration in vacuo ergibt festes 3-(4-Chlorphenyl)-4,5-dihydro-N-((4-fluorphenyl)sulfonyl)-4-phenyl-1H-pyrazol-1-carboxamidin (0,55 Gramm, 72% Ausbeute). Fließpunkt: 214–215°C.
- Auf analoge Weise sind die unten aufgelisteten Verbindungen der Formel (I) hergestellt worden:
4,5-Dihydro-N-((4-fluorphenyl)sulfonyl)-3-(4-methoxyphenyl)-4-(4-methoxy-phenyl)-1H-pyrazol-1-carboxamidin: Fließpunkt 155–156°C
4,5-Dihydro-3-(4-methoxyphenyl)-4-(4-methoxyphenyl)-N-((4-methoxy-phenyl)sulfonyl)-1H-pyrazol-1-carboxamidin: Fließpunkt 148–150°C
3-(4-Chlorphenyl)-4,5-dihydro-4-phenyl-N-((2,4,6-trimethylphenyl)sulfonyl)-1H-pyrazol-1-carboxamidin: Fließpunkt: 221–222°C. - Beispiel II
- N1,N1-Dimethyl-N2-((4-chlorphenyl)sulfonyl)-3-(4-chlorphenyl)-4,5-dihydro-4-phenyl-1H-pyrazol-1-carboxamidin
- Teil A: Ein gerührtes Gemisch aus 3-(4-Chlorphenyl)-4,5-dihydro-4-phenyl-1H-pyrazol (12,0 Gramm, 46,8 mmol), [(4-Chlorphenyl)sulfonyl]dithioimidocarbonsäure-dimethylester (CAS: 13068-12-7) (9,20 Gramm, 31,1 mmol) und Triethylamin (15 ml) in Acetonitril (200 ml) wird bei Rückfluss für 20 Stunden erhitzt. Eine zusätzliche Portion 3-(4-Chlorphenyl)-4,5-dihydro-4-phenyl-1H-pyrazol (12,0 Gramm, 46,8 mmol) wird zugegeben und das sich ergebende Gemisch wird bei Rückflusstemperatur für weitere 16 Stunden er hitzt. Nach Konzentration in vacuo wird Dichlormethan zugegeben und die sich ergebende Lösung wird zweimal mit Wasser gewaschen und über wasserfreiem Na2SO4 getrocknet. Nach Filtration und Eindampfen in vacuo wird der Rückstand durch Blitzchromatographie (Diethylether/Petroleumether = 1/1 (Vol./Vol.) weiter gereinigt, um 3-(4-Chlorphenyl)-N-((4-chlorphenyl)sulfonyl)-4,5-dihydro-4-phenyl-1H-pyrazol-1-carboximidothiosäure-methylester (12,5 Gramm, 80% Ausbeute, basierend auf [(4-Chlorphenyl)sulfonyl]dithioimidocarbonsäure-dimethylester) als einen amorphen Feststoff zu ergeben.
- Teil B: Zu einem gerührten Gemisch aus 3-(4-Chlorphenyl)-N-((4-chlorphenyl)sulfonyl)-4,5-dihydro-4-phenyl-1H-pyrazol-1-carboximidothiosäure-methylester (4,20 Gramm, 8,30 mmol) in Methanol (75 ml) werden Dimethylamin (10 ml) und Dichlormethan (75 ml) zugegeben und die sich ergebende Lösung wird bei Raumtemperatur für 6 Stunden gerührt. Eindampfen in vacuo und nachfolgende blitzchromatographische Reinigung (Diethylether/Petroleumether = 1/1 (Vol./Vol.), gefolgt von Diethylether) ergibt einen Feststoff, welcher durch Umkristallisation aus Diisopropylether weiter gereinigt wird, um N1-Dimethyl-N2-((4-chlor-phenyl)sulfonyl)-3-(4-chlorphenyl)-4,5-dihydro-4-phenyl-1H-pyrazol-1-carboxamidin (2,63 Gramm, 63% Ausbeute) zu ergeben. Fließpunkt: 182°C.
- Auf analoge Weise sind die unten aufgelisteten Verbindungen der Formel (I) hergestellt worden:
N-Methyl-N'-((4-chlorphenyl)sulfonyl)-3-(4-chlorphenyl)-4,5-dihydro-4-(3-pyridyl)-1H-pyrazol-1-carboxamidin. Fließpunkt: 101–105°C
N-Methyl-N'-((4-chlorphenyl)sulfonyl)-3-(4-chlorphenyl)-4,5-dihydro-4-(4-pyridyl)-1H-pyrazol-1-carboxamidin. Fließpunkt: 112–115°C. - Beispiel III
- N-Methyl-N'-((4-chlorphenyl)sulfonyl)-3-(4-chlorphenyl)-4,5-dihydro-4-phenyl-1H-pyrazol-1-carboxamidin
- Teil A: Zu einer Lösung aus N-((4-Chlorphenyl)sulfonyl)carbaminsäure-methylester (CAS: 34543-04-9) (2,99 Gramm, 12,0 mmol) und Pyridin (4 ml) in 1,4-dioxan (20 ml) wird 3-(4-Chlorphenyl)-4,5-dihydro-4-phenyl-1H-pyrazol (3,39 Gramm, 13,2 mmol) zugegeben und das sich ergebende Gemisch wird für 4 Stunden bei 100°C gerührt. Nach Konzentration in vacuo wird der Rückstand in Di chlormethan gelöst, nacheinander mit Wasser, 1 N HCl und Wasser gewaschen, über wasserfreiem Na2SO4 getrocknet, filtriert und in vacuo zu einem Volumen von 20 ml konzentriert. Methyl-tert-butylether (60 ml) wird zugegeben und die sich ergebende Lösung wird zu einem Volumen von 20 ml konzentriert. Die gebildeten Kristalle werden durch Filtration gesammelt und aus Methyl-tert-butylether umkristallisiert, um 3-(4-Chlorphenyl)-N-((4-chlorphenyl)sulfonyl)-4,5-dihydro-4-phenyl-1H-pyrazol-1-carboxamid (4,75 Gramm, 76% Ausbeute) zu ergeben; Fließpunkt: 211–214°C.
- Teil B: Ein Gemisch aus 3-(4-Chlorphenyl)-N-((4-chlorphenyl)sulfonyl)-4,5-dihydro-4-phenyl-1H-pyrazol-1-carboxamid (3,67 Gramm, 7,75 mmol) und Phosphorpentachlorid (1,69 Gramm, 8,14 mmol) in Chlorbenzol (40 ml) wird bei Rückfluss für 1 Stunde erhitzt. Nach gründlicher Konzentration in vacuo wird das gebildete N-((4-Chlorphenyl)sulfonyl)-3-(4-chlorphenyl)-4,5-dihydro-4-phenyl-1H-pyrazol-1-carboximidoylchlorid in Dichlormethan suspendiert und mit kaltem Methylamin (1,5 ml) umgesetzt. Nach Rühren bei Raumtemperatur für 1 Stunde wird das Gemisch in vacuo konzentriert. Der Rückstand wird aus Diethylether kristallisiert, um N-Methyl-N'-((4-chlorphenyl)sulfonyl)-3-(4-chlorphenyl)-4,5-dihydro-4-phenyl-1H-pyrazol-1-carboxamidin (2,29 Gramm, 61% Ausbeute) zu ergeben; Fließpunkt: 96–98°C (zerf.).
- Auf analoge Weise sind die unten aufgelisteten Verbindungen der Formel (I) hergestellt worden:
N-Methyl-N'-((3-chlorphenyl)sulfonyl)-3-(4-chlorphenyl)-4,5-dihydro-4-phenyl-1H-pyrazol-1-carboxamidin. Fließpunkt: 156–160°C.
N-Propyl-N'-((4-fluorphenyl)sulfonyl)-3-(4-chlorphenyl)-4,5-dihydro-4-phenyl-1H-pyrazol-1-carboxamidin. Fließpunkt: 129–138°C.
N-(2-Propyl)-N'-((4-fluorphenyl)sulfonyl)-3-(4-chlorphenyl)-4,5-dihydro-4-phenyl-1H-pyrazol-1-carboxamidin. Fließpunkt: 110–112°C.
N-(2-Propyl)-N'-((4-chlorphenyl)sulfonyl)-3-(4-pyridyl)-4,5-dihydro-4-phenyl-1H-pyrazol-1-carboxamidin. Fließpunkt: amorph.
N1-Ethyl-N1-methyl-N2-((4-chlorphenyl)sulfonyl)-3-(4-chlorphenyl)-4,5-dihydro-4-phenyl-1H-pyrazol-1-carboxamidin. Fließpunkt: 184°C.
N1-Ethyl-N1-methyl-N2-((4-fluorphenyl)sulfonyl)-3-(4-chlorphenyl)-4,5-dihydro-4-phenyl-1H-pyrazol-1-carboxamidin. Fließ punkt: 173–176°C.
N1,N1-Dimethyl-N2-((4-trifluormethyl)phenyl)sulfonyl)-3-(4-chlorphenyl)-4,5-dihydro-4-phenyl-1H-pyrazol-1-carboxamidin. Fließpunkt: 195–196°C.
N1,N1-Dimethyl-N2-((3-methylphenyl)sulfonyl)-3-(4-chlorphenyl)-4,5-dihydro-4-phenyl-1H-pyrazol-1-carboxamidin. Fließpunkt: 195–198°C.
N1,N1-Dimethyl-N2-((3-methoxyphenyl)sulfonyl)-3-(4-chlorphenyl)-4,5-dihydro-4-phenyl-1H-pyrazol-1-carboxamidin. Fließpunkt: 204–206°C.
N-Ethyl-N'-((4-chlorphenyl)sulfonyl)-3-(4-chlorphenyl)-4,5-dihydro-4-phenyl-1H-pyrazol-1-carboxamidin. Fließpunkt: amorph.
N-Dimethylamino-N'-((4-chlorphenyl)sulfonyl)-3-(4-chlorphenyl)-4,5-dihydro-4-phenyl-1H-pyrazol-1-carboxamidin. Fließpunkt: 155–159°C.
N-Methyl-N'-((4-(trifluormethyl)phenyl)sulfonyl)-3-(4-chlorphenyl)-4,5-dihydro-4-phenyl-1H-pyrazol-1-carboxamidin. Fließpunkt: amorph.
N1,N1-Dimethyl-N2-((2-methylphenyl)sulfonyl)-3-(4-chlorphenyl-4,5-dihydro-4-phenyl-1H-pyrazol-1-carboxamidin. Fließpunkt: 148–151°C.
N-Methyl-N'-((2,4-difluorphenyl)sulfonyl)-3-(4-chlorphenyl)-4,5-dihydro-4-phenyl-1H-pyrazol-1-carboxamidin. Fließpunkt: 85°C. - Beispiel IV
- (–)-(4S)-N-Methyl-N'-((4-chlorphenyl)sulfonyl)-3-(4-chlorphenyl)-4,5-dihydro-4-phenyl-1H-pyrazol-1-carboxamidin
- (–)-(4S)-N-Methyl-N'-((4-chlorphenyl)sulfonyl)-3-(4-chlorphenyl)-4,5-dihydro-4-phenyl-1H-pyrazol-1-carboxamidin (7,16 Gramm, 0, 0147 mol)) ([α25 D] = –150°, c = 0, 01, MeOH) (Fließpunkt: 169–170°C) wurde durch chirale chromatographische Trennung von racemischem N-Methyl-N'-((4-chlorphenyl)sulfonyl)-3-(4-chlorphenyl)-4,5-dihydro-4-phenyl-1H-pyrazol-1-carboxamidin (18 Gramm, 0,037 mol) unter Verwendung eines Chiralpak AD, 20 μm chirale stationäre Phase erhalten. Die mobile Phase bestand aus einem Gemisch aus Hexan/Ethanol (80/20 (Vol./Vol.)) und 0,1% Ammoniumhydroxid (25% wässerige Lösung).
- Auf analoge Weise sind die unten aufgelisteten optisch reinen Verbindungen aus den entsprechenden Racematen hergestellt worden:
(–)-(4S)-N-Ethyl-N'-((4-chlorphenyl)sulfonyl)-3-(4-chlorphe nyl)-4,5-dihydro-4-phenyl-1H-pyrazol-1-carboxamidin: ([α25 D] = –126°, c = 0,01, CHCl3); Fließpunkt: 172–175°C. Stationäre Phase: Chiralcel OD. Mobile Phase: Ein Gemisch aus Heptan/2-Propanol (85/15 (Vol./Vol.)).
(–)-(4S)-N-Dimethylamino-N'-((4-chlorphenyl)sulfonyl)-3-(4-chlorphenyl)-4,5-dihydro-4-phenyl-1H-pyrazol-1-carboxamidin: ([α25 D] = –132°, c = 0,01, CHCl3); Fließpunkt: 218–224°C. Stationäre Phase: Chiralcel OD. Mobile Phase: ein Gemisch aus Heptan/2-Propanol (85/15 (Vol./Vol.)).
(–)-(4S)-N-Methyl-N'-((4-(trifluormethyl)phenyl)sulfonyl)-3-(4-chlorphenyl)-4,5-dihydro-4-phenyl-1H-pyrazol-1-carboxamidin: ([α25 D] = –131°; c = 0, 01, CHCl3); Fließpunkt: 157–160°C. Stationäre Phase: Chiralcel OD. Mobile Phase: ein Gemisch aus Heptan/2-Propanol (85/15 (Vol./Vol.)).
(–)-(4S)-N1,N1-Dimethyl-N2-((2-methylphenyl)sulfonyl)-3-(4-chlorphenyl)-4,5-dihydro-4-phenyl-1H-pyrazol-1-carboxamidin: ([α25 D] = –88°, c = 0,01, MeOH); Fließpunkt: amorph. Stationäre Phase: Chiralpak AD. Mobile Phase: Ethanol.
(–)-(4S)-N-Methyl-N'-((2,4-difluorphenyl)sulfonyl)-3-(4-chlorphenyl)-4,5-dihydro-4-phenyl-1H-pyrazol-1-carboxamidin: ([α25 D] = –129°, c = 0,01, MeOH); Fließpunkt: amorph. Chiralpak AD. Mobile Phase: Methanol.
Claims (8)
- Enantiomer mit S-Konfiguration an der 4-Position des 4,5-Dihydropyrazolrings einer Verbindung der Formel (I) worin: R und R1 gleich oder unterschiedlich sind und 3-Pyridyl oder 4-Pyridyl oder Phenyl darstellen, welches mit Halogen oder Methoxy substituiert sein kann, R2 und R3 gleich oder unterschiedlich sind und Wasserstoff, Alkyl (1–3 C) oder Dimethylamino darstellen, R4 für Phenyl steht, welches mit 1, 2 oder 3 Substituenten substituiert sein kann, ausgewählt aus der Gruppe Halogenatome, Trifluormethyl, Methoxy und Alkyl (1–3 C) und Tautomere und Salze davon.
- Verbindung mit der Formel (I) wie in Anspruch 1 beansprucht, worin R für die Gruppe 4-Chlorphenyl steht, R1 für Phenyl steht, R2 für Wasserstoff steht, R3 für Methyl steht und R4 4-Chlorphenyl darstellt, und Salze davon.
- Pharmazeutische Zusammensetzung, welche mindestens eine Verbindung wie in Anspruch 1 beansprucht als Wirkstoff enthält.
- Verbindung wie in Anspruch 1 beansprucht, oder ein Salz davon, zur Verwendung in Medizin.
- Verfahren zur Herstellung von Verbindungen mit der Formel I, dadurch gekennzeichnet, dass das racemische Gemisch einer Verbindung mit der Formel I in die linksdrehenden und die rechtsdrehenden Enantiomere getrennt wird.
- Verwendung einer Verbindung wie in Anspruch 1 beansprucht zur Herstellung einer pharmazeutischen Zusammensetzung zur Behandlung von psychiatrischen Störungen wie Psychose, Angststörung, Depression, Aufmerksamkeitsdefiziten, Gedächtnisstörungen und Appetitstörungen, Fettsucht, neurologischen Störungen wie Parkinson-Erkrankung, Demenz, Dystonie, Alzheimer-Erkrankung, Epilepsie, Huntington-Erkrankung, Tourette-Syndrom, Ischämie, Schmerz und anderen CNS-Störungen, welche Cannabinoid-Neurotransmission einbeziehen.
- Verwendung einer Verbindung wie in Anspruch 1 beansprucht für die Herstellung einer pharmazeutischen Zusammensetzung zur Behandlung von gastrointestinalen Störungen, welche Cannabinoid-Neurotransmission einbeziehen.
- Verwendung einer Verbindung wie in Anspruch 1 beansprucht für die Herstellung einer pharmazeutischen Zusammensetzung zur Behandlung von kardiovaskulären Störungen, welche Cannabinoid-Neurotransmission einbeziehen.
