Description
- Technisches Gebiet
- Die vorliegende Erfindung betrifft eine den Haarwuchs stimulierende Zusammensetzung, die Minoxidil als Wirkkomponente umfasst, und insbesondere betrifft die Erfindung eine den Haarwuchs stimulierende Zusammensetzung, die frei von Problemen wie Kristallausfällungen und dgl. bei niedrigen Temperaturen ist.
- Stand der Technik
- Der chemische Name für Minoxidil ist 6-(1-Piperidinyl)-2,4-pyrimidindiamin-3-oxid. Die Anwendung von Minoxidil als Haarwuchsmittel ist in
US 4,139,619 offenbart. Wegen des ausgezeichneten Haarwuchses, der durch die topische Anwendung von Minoxidil erhalten wird, sind Haarwuchsmittel, die Minoxidil enthalten (nachfolgend bezeichnet als "Minoxidil-Zubereitungen"), in breitem Umfang angenommen worden, und deren Verkaufsvolumen bricht Rekorde. - Zur Steigerung des Effekts dieser Minoxidil-Zubereitungen ist nach Zubereitungen mit höheren Minoxidil-Konzentrationen gesucht worden. Bisher sind Minoxidil-Zubereitungen mit einer Minoxidil-Konzentration von ca. 3 bis 5 Masse-% leicht hergestellt worden (JP-OS 150 211/1988).
- Das Patentdokument WO 99/53 923 offenbart den Haarwuchs stimulierende Zusammensetzungen, die 10 bis 50 Masse-% Wasser, Minoxidil und ferner 8 bis 30 Masse-% eines polyhydrischen Alkohols umfassen und einen pH-Wert von 5,5 bis 6,5 aufweisen (Beispiele 5, 7 und 10). Diese Zusammensetzungen enthalten 10 oder 12% Minoxidil.
- Offenbarung der Erfindung
- Enthält jedoch eine Minoxidil-Zubereitung mit hoher Konzentration (nachfolgend bezeichnet als "hoch konz. Minoxidil-Zubereitung") 10 bis 50 Masse-% Wasser zur Verbesserung des Gefühls bei Gebrauch, tritt Kristallausfällung in der Zubereitung auf, wenn sie über einen langen Zeitraum bei niedriger Temperatur aufbewahrt und gelagert wird. Daher treten, wegen des Zeitaufwands zur erneuten Auflösung der ausgefallenen Kristalle, Probleme auf, wenn die Haarwuchszusammensetzung in kalten Gegenden angewandt oder an kalten Plätzen gelagert wird.
- Aus diesem Grund bestand die Forderung nach einer stabilen hoch konzentrierten Minoxidil-Zubereitung, worin Kristallausfällungen nicht auftreten, sogar wenn die Zubereitung bei niedriger Temperatur gelagert wird. Demnach ist es die Aufgabe der vorliegenden Erfindung, eine derartige Zubereitung anzugeben und zur Verfügung zu stellen.
- Als Ergebnis umsichtiger Forschung bezüglich der Lösungsmittelzusammensetzung und dgl. einer hoch konzentrierten Minoxidil-Zubereitung zur Lösung der obigen Probleme, haben die hier auftretenden Erfinder herausgefunden, dass durch Auswahl eines spezifischen polyhydrischen Alkohols und durch dessen Anwendung bei einer spezifischen Konzentration als Lösungsmittel sowie durch Einstellung des pH-Wertes der Lösung in einem festgelegten Bereich in der Minoxidil-Lösung, worin das Minoxidil hoher Konzentration mit einer festgelegten Menge Wasser kombiniert ist, Kristalle nicht ausgefällt werden und das Minoxidil selbst stabil bleibt, wodurch die vorliegende Erfindung erfolgreich abgeschlossen wurde.
- Gegenstand der vorliegenden Erfindung ist es nämlich, eine den Haarwuchs stimulierende Zusammensetzung bereit zu stellen, die 10 bis 50 Masse-% Wasser und 3 bis 6 Masse-% oder mehr Minoxidil und ferner 8 bis 30 Masse-% eines oder mehrerer polyhydrischer Alkohole umfasst und einen pH-Wert von 5,5 bis 6,5 aufweist.
- Detaillierte Beschreibung der bevorzugten Ausführungsform
- Die den Haarwuchs stimulierende Zusammensetzung der vorliegenden Erfindung enthält 3 bis 6 Masse-% oder mehr (nachfolgend einfach bezeichnet als "%") der Wirkkomponente Minoxidil.
- Als Lösungsmittel zur Auflösung des Minoxidils wird eine Mischung aus polyhydrischem Alkohol und Wasser verwendet. Es ist erforderlich, dass die Endzusammensetzung 10 bis 50% Wasser und 8 bis 30% polyhydrischen Alkohol umfasst. Sind entweder der polyhydrische Alkohol oder das Wasser nicht vorhanden, ist es schwierig, die Ausfällung der Kristalle zu verhindern. Auch wenn sogar beide vorhanden sind, ist es schwierig, eine Zusammensetzung zu erzeugen, die ein gutes Gefühl bei Gebrauch ergibt und wirkungsvoll zur Unterdrückung der Ausfällung von Kristallen bei niedrigen Temperaturen ist, falls diese beiden Komponenten in einer Menge außerhalb der obigen Bereiche vorliegen. Durch die vorliegende Erfindung wird ein noch größerer Effekt erzielt, wenn die Wassermenge 18% oder mehr beträgt, weil andernfalls die Ausfällung der Kristalle dann leichter auftreten könnte. Der polyhydrische Alkohol wird in einer Menge gemäß dem obigen Bereich zugegeben, wobei 10 bis 20% bevorzugt und 15 bis 20% besonders bevorzugt sind.
- Es ist erwünscht, Ethanol dem Lösungsmittel zuzufügen. Die Endzusammensetzung enthält vorzugsweise Ethanol in einer Menge von 70% oder weniger.
- Als polyhydrischer Alkohol, der im obigen Lösungsmittel verwendet wird, können 1,3-Butylenglykol, Glycerin, Dipropylenglykol, Macrogol 400, Macrogol 600 und dgl. angegeben werden, wobei 1,3-Butylenglykol bevorzugt ist. Auch werden diese polyhydrischen Alkohole vorzugsweise in Kombinationen von zwei oder mehr Typen verwendet. Als besonders bevorzugte Kombinationen der polyhydrischen Alkohole können 1,3-Butylenglykol und Glycerin, die mit einem Gewichtsverhältnis von 10:1 bis 1:4 zusammengebracht werden, und Dipropylenglykol und Glycerin angegeben werden, die in einem Gewichtsverhältnis von 10:1 bis 1:4 kombiniert werden.
- Die den Haarwuchs stimulierende Zusammensetzung der vorliegenden Erfindung wird hergestellt, indem man 3% oder mehr der Wirkkomponente Minoxidil und weitere notwendige Wirkkomponenten und ergänzende Komponenten zum Lösungsmittel gibt, das den polyhydrischen Alkohol und Wasser enthält, und man dann diese Komponenten unter Rühren mit einem üblichen Verfahren vermischt. In diesem Fall ist es notwendig, den pH-Wert in einem Bereich von 5,5 bis 6,5 einzustellen. Wird Minoxidil lediglich zu einem Lösungsmittel vom wässrigen Typ gegeben, reagiert die entstandene Lösung neutral oder schwach alkalisch. Bei Herstellung der hoch konzentrierten Minoxidil-Zubereitung mit dieser Lösung tritt die Ausfällung von Kristallen leicht bei niedriger Temperatur auf. Wird die Lösung andererseits auf der sauren Seite bei einem pH-Wert von 5 oder weniger eingestellt, wird das Minoxidil selbst leicht zersetzbar, obgleich eine Ausfällung von Kristallen gar nicht auftritt.
- Der pH-Wert der den Haarwuchs stimulierenden Zusammensetzung der vorliegenden Erfindung wird mit einem üblichen Verfahren eingestellt. Als Mittel zur Einstellung des pH-Wertes werden vorzugsweise Zitronen-, Salz-, Milch-, Phosphorsäure und dgl. verwendet.
- Ferner können als medizinische Komponenten, die vorzugsweise zur den Haarwuchs stimulierenden Zusammensetzung der vorliegenden Erfindung gegeben und damit kombiniert werden, Komponenten, ausgewählt aus der Gruppe, bestehend aus Menthol, Vitamin E-Acetat, Pantothenylethylether, Hinokitiol, Glycyrrhetinsäure und aus Diphenhydramin-Hydrochlorid, angegeben und genannt werden (nachfolgend bezeichnet als "optionale Komponenten"). Unter diesen optionalen Komponenten besitzt Menthol eine besonders hohe Wirksamkeit. Die Zugabe von Menthol als Essentialkomponente ist sogar noch bevorzugter.
- Es bestehen keine besonderen Einschränkungen bei der Menge dieser optionalen Komponenten, die zugegeben werden, und dies kann experimentell entschieden werden, wobei das Gefühl bei Gebrauch und die Stabilität des Minoxidils oder die Komponenten des Lösungsmittel-Typs zu berücksichtigen sind. Beispielsweise enthält die endgültige Zusammensetzung vorzugsweise 0,2 bis 0,4% Menthol, das eine bevorzugte optionale Komponente ist.
- Zusätzlich zu den obigen Komponenten kann die den Haarwuchs stimulierende Zusammensetzung der vorliegenden Erfindung verschiedene Wirk- und Ergänzungskomponenten, die im Normalfall in topischen Zubereitungen verwendet werden, in einer Menge enthalten, die den Effekt der vorliegenden Erfindung nicht beeinträchtigt. Beispiele dieser Komponenten schließen Füllstoffe, Vasodilatoren (Gefäßerweiterungsmittel) (Carproniumchlorid, Benzylnicotinat, Swertia-Gewürzextrakt, Panax-Ginsengextrakt, Capsicum-Tinktur und dgl.), Antihistamine (Isothipendyl-Hydrochlorid und dgl.), entzündungshemmende Mittel (Guaiazulen und dgl.), Keratolytika (Harnstoff, Salicylsäure und dgl.), antimikrobielle Mittel (Chlorhexidingluconat, Isopropylmethylphenol, quartäre Ammoniumsalze, Piroctonolamin und dgl.), Feuchtigkeitsmittel (Natriumhyaluronat, Chondroitinschwefelsäure und dgl.), tierische und pflanzliche Extrakte (Taxus cuspidata, Paeonia suffruticosa, Glycyrrhisa uralensis, Hypericum erectum, Aconitwurzel, Eriobotrya japonica, Artemisia capillaris, Symphytum officinale, Angelica keiskei, Crocus sativus, Gardeniae fructus, Rosmarinus officinalis, Salvia officinalis, Saussurea lappa, Aristolochia debilis, Lupuli strobilus, Samenleiste und dgl.), Vitamine (Retinolacetat, Pyridoxin-Hydrochlorid, Ascorbinsäure, Thiaminnitrat, Cyanocobalamin, Biotin und dgl.), Antioxidanzien (Dibutylhydroxytoluol, Natriumpyrosulfit, Tocopherol, Natriumedetat, Ascorbinsäure, Isopropylgallat und dgl.), Solubilisiermittel (Diisopropyladipat, Isopropylmyristat, pflanzliche Öle, tierische Öle, Alkylglycerylether, Kohlenwasserstoffe und dgl.), metabolische Aktivatoren (Panthenol und dgl.), Geliermittel (wasserlösliche hochmolekulare Verbindungen und dgl.), Klebemittel, Parfüme, Kühlmittel (Mentha-Öl, Kampfer und dgl.), Farbstoffe und dgl. ein.
- Allerdings ist es, da die Zugabe oberflächenaktiver Mittel die Absorption von Minoxidil durch die Haut beeinflusst und das Gefühl bei Gebrauch herabsetzt, bevorzugt, dass die den Haarwuchs stimulierende Zusammensetzung der vorliegenden Erfindung keine oberflächenaktiven Mittel enthält.
- Die so erhaltene den Haarwuchs stimulierende Zusammensetzung der vorliegenden Erfindung kann als geeignete topische Zubereitung wie als Lotion, Aerosol, Tonikum, Creme, Salbe, Gel und dgl. angewandt werden.
- Beispiele
- Die vorliegende Erfindung wird nun noch detaillierter durch Beispiele beschrieben, auf welche die vorliegende Erfindung allerdings nicht eingeschränkt sein soll.
- Beispiel 1
- 3,0 g Minoxidil, 10,0 g 1,3-Butylenglykol, 35 g Ethanol und 20 g gereinigtes Wasser wurden vermischt und bis zur Auflösung verrührt. Eine geeignete Menge Zitronensäure wurde zugegeben, um den pH-Wert der Lösung auf 5,5 einzustellen.
- Gereinigtes Wasser wurde erneut zugegeben, um 100 mL einer Lotionszubereitung (Produkt 1 der vorliegenden Erfindung) zu erhalten.
- Beispiel 2
- Mit den in Tabellen 1 und 2 angegebenen Komponenten wurden Lotionszubereitungen (Produkte der vorliegenden Erfindung) in der gleichen Weise wie in Beispiel 1 hergestellt. Mit den in Tabelle 3 angegebenen Komponenten wurden Lotionszubereitungen (Vergleichsprodukte) in der gleichen Weise wie in Beispiel 1 hergestellt. (Zusammensetzung) Tabelle 1

- Die Produkte der vorliegenden Erfindung und die Vergleichsprodukte wurden bei –10°C 1 Woche lang gelagert, und der Grad der Kristallausfällung wurde untersucht. Als Ergebnis wurde für die Produkte der vorliegenden Erfindung belegt, dass sie keine Anzeichen einer Kristallausfällung zeigen, obwohl die Konzentration von Minoxidil sogar hoch war. Für die Vergleichsprodukte wurde belegt, dass sie unterschiedliche Grade der Kristallausfällung zeigen und ergeben.
- Beispiel 3
- Ein Aerosol wurde durch Befüllen einer Aerosoldose mit 30 mL der in Beispiel 1 hergestellten Lotion und mit 70 mL Dimethylether zubereitet. In gleicher Weise wurde ein Aerosol mit der Lotion des Beispiels 2 zubereitet.
- Testbeispiel 1
- Lotionszubereitungen des Produkts 16 der vorliegenden Erfindung und des Vergleichsprodukts 3, welche in Tabelle 4 angegeben sind, wurden in gleicher Weise wie in Beispiel 1 hergestellt. Diese Produkte wurden unter den folgenden Bedingungen gelagert und bezüglich einer Kristallausfällung über die Dauer von 1 Woche beobachtet. Die Ergebnisse sind in Tabelle 5 angegeben.

Diese Produkte wurden unter den folgenden Bedingungen gelagert und bezüglich einer Kristallausfällung über die Dauer von 1 Woche beobachtet. Die Ergebnisse sind in Tabelle 8 angegeben.

Claims (12)
- Haarwachstum stimulierende Zusammensetzung, umfassend 10 bis 50 Massen-% Wasser und 3 bis 6 Massen-% Minoxidil, weiter umfassend 8 bis 30 Massen-% eines mehrwertigen Alkohols und mit einem pH von 5,5 bis 6,5.

- Haarwachstum stimulierende Zusammensetzung nach Anspruch 1, worin der mehrwertige Alkohol eine Mischung aus 1,3-Butylenglycol und Glycerin bei einem Gewichtsverhältnis von 10:1 bis 1:4 ist.
- Haarwachstum stimulierende Zusammensetzung nach Anspruch 1, worin der mehrwertige Alkohol eine Mischung aus Dipropylenglycol und Glycerin bei einem Gewichtsverhältnis von 10:1 bis 1:4 ist.
- Haarwachstum stimulierende Zusammensetzung nach einem der Ansprüche 1 bis 4 in der Form einer Lotion, weiterhin umfassend Ethanol.
- Haarwachstum stimulierende Zusammensetzung nach einem der Ansprüche 1 bis 4 in der Form eines Gels, weiterhin umfassend Ethanol.
- Haarwachstum stimulierende Zusammensetzung nach einem der Ansprüche 1 bis 6, worin die eingefügte Menge an Ethanol 70 Massen-% oder weniger ist.
- Haarwachstum stimulierende Zusammensetzung nach einem der Ansprüche 1 bis 7, worin die eingefügte Menge an Wasser 18 bis 50 Massen-% ist.
- Haarwachstum stimulierende Zusammensetzung nach einem der Ansprüche 1 bis 8, im wesentlichen nicht umfassend Tenside.
- Haarwachstum stimulierende Zusammensetzung nach einem der Ansprüche 1 bis 9, umfassend Zitronensäure, Salzsäure, Milchsäure oder Phosphorsäure als pH-Einstellmittel.
- Haarwachstum stimulierende Zusammensetzung nach einem der Ansprüche 1 bis 10, weiterhin umfassend eine oder mehrere Komponenten, ausgewählt aus der Gruppe bestehend aus Menthol, Vitamin E-Acetat, Pantothenylethylether, Hinokitiol, Glycyrrhetinsäure und Diphenhydraminhydrochlorid.
- Verfahren zum Verhindern des Ausfällens, umfassend das Einfügen von 8 bis 30 Massen-% eines mehrwertigen Alkohols in die Zusammensetzung, umfassend 10 bis 50 Massen-% Wasser und 3 bis 6 Massen-% Minoxidil, und Einstellen der Zusammensetzung auf einen pH von 5,5 bis 6,5.
