Description
- Diese Erfindung bezieht sich auf Verbindungen mit pharmazeutischer Wirkung, d.h. antiviraler Wirkung, ihre pharmazeutisch annehmbaren Salze und Solvate und pharmazeutische Zusammensetzungen, die die aktiven Verbindungen enthalten.
- Arildone, eine Verbindung, die durch die Formel
- dargestellt wird, ist in vitro gegen Herpesvirus und Poliovirus wirksam, ist gegen Rhinoviren jedoch nur geringfügig wirksam. Diana et al., J. Med. Chem. 28, 748 (1985), stellten in einem Versuch, Verbindungen mit Breitbandwirkung gegen Picornaviren herzustellen, einige alkylsubstituierte Isoxazolanaloga von Arildon her. Einige der Verbindungen waren sowohl gegen Rhinovirus Typ 2 als auch gegen Poliovirus Typ 2 wirksam.
- Andere antivirale Mittel sind in der Fachkunde bekannt. US-A- 3190888 beschreibt Aryloxyalkylpyrazolverbindungen mit antiviraler Wirkung. EP-A-0112292 beschreibt Verbindungen mit wachstumsregulierender und mikrobizider Wirkung, die terminale Azolylgruppen enthalten.
- Weitere pharmazeutische Zusammensetzungen sind in AU-A-56398/86 beschrieben und sind als Lipogenase-Inhibitoren, entzündungshemmende Mittel und Antiallergene wirksam.
- Es ist ein Ziel der vorliegenden Erfindung, antivirale Verbindungen bereitzustellen.
- Die Erfindung bezieht sich im großen und ganzen auf Verbindungen der Formel I, und Verbindungen der Formel I sind auch der Gegenstand des Europäischen Patents 0 274 867, das auf eine Anmeldung erteilt wurde, aus der der hier beschriebene und beanspruchte Gegenstand abgetrennt wurde. Verbindungen der Formel I haben die folgende Strukturformel

- X und Y jeweils unabhängig eine Bindung, -O-, -S-, -SO&sub2;-,

- W eine einwertige substituierte oder unsubstituierte Arylgruppe oder ein heterocyclischer einfacher oder kondensierter Ring mit 4 bis 10 Ringatomen ist, wobei wenigstens eines der Heteroatome ein Stickstoffatom ist und es sich bei den übrigen Ringatomen um wenigstens ein Kohlenstoffatom und gegebenenfalls Schwefel oder Sauerstoff handelt, wobei die Substituenten einer oder mehrere der Reste Hydroxy, Oxo, Amino, Carbamoyl, Carboxyl, Nitril, Nitro, Niederalkyl, Niederalkoxycarbonyl, Halogen, Sulfamyl, Niederalkoxycarbonylniederalkyl, Niederalkylthio, Niederalkoxy, Hydroxyniederalkyl, Aminoniederalkyl, Carboxyniederalkyl, Guanidino, Thioureido, Niederalkylsulfonylamino, Aminocarbonylniederalkyl, Allyloxycarbonylmethyl oder Carbamoyloxyniederalkyl sind, mit der Maßgabe, daß W nicht substituiertes oder unsubstituiertes Isoxazolyl sein kann,
- W' zweiwertiges W ist.
- Die Erfindung beinhaltet auch pharmazeutische Zusammensetzungen, die pharmazeutisch wirksame Mengen einer Verbindung der Formel II enthalten, sowie ein Verfahren zur Behandlung von Virusinfektionen unter Verwendung geeigneter pharmazeutischer Zusammensetzungen.
- Der hier verwendete Ausdruck "Niederalkyl" allein oder in kombinierter Form, z.B. "Niederalkoxy" oder "wiederalkandiyl", bedeutet geradkettige oder verzweigte Alkylgruppen mit 1 bis 10 Kohlenstoffatomen, z.B. Methyl, Ethyl, Propyl, Isopropyl, Butyl, t-Butyl, Pentyl, Neopentyl, Hexyl und dergleichen.
- Die Heterocyclen bei Z und W sind über ein Heteroatom, vorzugsweise Stickstoff, oder ein Kohlenstoffatom einwertig an X bzw. Y gebunden. Die Heterocyclen bei W' haben vorzugsweise zwei Stickstoffatome, von denen jedes an eine getrennte -Y-Q-X-Z- Struktureinheit gebunden ist.
- Heterocyclische Gruppen innerhalb des Umfangs dieser Erfindung für Z sind zum Beispiel Imidazolyl (wie Imidazol-1-yl, Imidazol- 2-yl, Imidazol-4-yl und Imidazol-5-yl), Dihydrothiazolyl (wie 4,5-Dihydrothiazol-2-yl), Tetrazolyl (wie Tetrazol-5-yl, Tetrazol-1-yl und Tetrazol-2-yl), Pyridinyl (wie Pyridin-2-yl), Triazolyl (wie 1,2,4-Triazol-1-yl), Tetrahydropyrimidinyl (wie 1,2,3,4-Tetrahydropyrimidin-1-yl), Dihydrooxazolyl (wie 4,5- Dihydrooxazol-2-yl), Pyrrolidinyl (wie Pyrrolidin-1-yl), Pyrazolyl (wie Pyrazol-1-yl und Pyrazol-2-yl), Morpholinyl und Azetidinyl. Alle möglichen Bindungspositionen der obigen heterocyclischen Gruppen sind innerhalb des Umfangs dieser Erfindung.
- Heterocyclische Gruppen innerhalb des Umfangs dieser Erfindung für W sind zum Beispiel alle oben für Z aufgeführten und außerdem Verbindungen mit kondensierten Ringen, zum Beispiel Benzamidazolyl (wie Benzamidazol-1-yl und Benzamidazol-2-yl), Naphthyridinyl (wie Naphthyridin-1-yl), Purinyl (wie Purin-9-yl und Purin-7-yl) und Chinolinyl.
- Heterocyclische Gruppen innerhalb des Umfangs dieser Erfindung für W' sind alle oben für Z und W aufgeführten, wobei sie jedoch zweiwertig sind. Wenn W' Tetrahydropyrimidinyl wäre, könnte es zum Beispiel 1,2,3,4-Tetrahydropyrimidin-1,3-diyl sein. W' als Benzamidazolylgruppe könnte zum Beispiel Benzamidazol-1,3-diyl sein. Mit anderen Worten, um eine mögliche W'-Gruppe aus den für Z und W aufgeführten Gruppen zu erhalten, wird die Endung "yl" des Z- und W-Restes durch "diyl" ersetzt.
- Der hier verwendete Ausdruck "Aryl" bezieht sich auf Phenyl und Naphthyl.
- Der hier verwendete Ausdruck "Halogen" bedeutet Chlor, Fluor, Brom oder Iod, wobei Chlor oder Fluor bevorzugt sind.
- "Cycloalkan" allein oder in kombinierter Form bedeutet einen 4-, 5-, 6- oder 7-gliedrigen gesättigten carbocyclischen Ring.
- "Niederalken" allein oder in kombinierter Form bedeutet eine verzweigte oder geradkettige Alkengruppe mit 2 bis 10 Kohlenstoffatomen.
- "Niederalkin" allein oder in kombinierter Form bedeutet eine verzweigte oder geradkettige Alkingruppe mit 2 bis 10 Kohlenstoffatomen.
- Der hier verwendete Ausdruck "pharmazeutisch annehmbare Salze" bedeutet Säureadditionssalze, die aus Mineralsäuren, wie Chlorwasserstoff-, Bromwasserstoff-, Phosphor- oder Schwefelsäure, gebildet sind oder aus organischen Carbon- oder Sulfonsäuren, wie Trifluoressig-, p-Toluolsulfon-, Malein-, Essig-, Zitronen-, Oxal-, Bemstein-, Benzoe-, Wein-, Fumar-, Mandel-, Ascorbin- und Äpfelsäure, gebildet sind, oder quartäre Salze, die aus organischen Halogeniden, wie Methyliodid, Ethyliodid, Benzylchlorid und dergleichen, hergestellt sind, obwohl alle pharmazeutisch annehmbaren quartären Salze in Betracht gezogen werden. Basenadditionssalze sind ebenfalls innerhalb des Umfangs dieser Erfindung.
- Die obigen Salze werden nach in der Fachkunde üblichen Methoden hergestellt, z.B. Reaktion der Verbindung mit einer geeigneten Säure, einem organischen Halogenid oder einer Base.
- Das bevorzugte Salz ist das Hydrochlorid.
- Der hier verwendete Ausdruck "Hydroxyschutzgruppe" bedeutet jede bekannte Hydroxyschutzgruppe, die durch herkömmliche Reaktionen, die die hergestellten Verbindungen nicht beeinträchtigen, entfernt wird. Typische geeignete Hydroxyschutzgruppen sind t-Butyldimethylsilyl (TBDMS) oder Tetrahydropyranyl.
- Es wurde gefunden, daß die Verbindungen dieser Erfindung gegen etherresistente RNA-Viren, d.h. Picornaviren, zu denen Enteroviren und Rhinoviren gehören, wirksam sind. Zu den Enteroviren gehören das Poliovirus, Coxsackie-Viren und Echoviren. Zu den Rhinoviren gehören die Viren, die mit der gewöhnlichen Erkältung und bestimmten anderen Krankheiten der Atemwege zusammenhängen. über hundert Serotypen wurden identifiziert. Obwohl die Verbindungen dieser Erfindung nicht gegen alle Rhinoviren wirksam sind, sind sie gegen eine große Zahl davon wirksam, einschließlich Rhinovirus 2. Die Verbindungen dieser Erfindung sind außerdem wirksam gegen die Enteroviren, wie Poliovirus 2, Coxsackie-Viren A und B3, ECHO und Hepatitis A.
- Außerdem sind die Verbindungen dieser Erfindung wirksam gegen bestimmte DNA-Viren, wie Herpesvirus und Cytomegalovirus. So zeigten sie Aktivität, als sie in in-vitro-Aktivitätsassays getestet wurden, d.h. Plaquereduktionsassays, die die Fähigkeit synthetischer Verbindungen messen, die Infektiosität von Viren, z.B. die Infektiosität von Picornavirus, zu neutralisieren. In Tests gegen das Coxsackie-Virus 3 variierten die IC&sub5;&sub0;-Werte der getesteten Verbindungen dieser Erfindung von etwa 0,7 µg/ml bis etwa 1,4 µg/ml. Der IC&sub5;&sub0;-Wert ist bei allen antiviralen Tests die Konzentration der Testverbindung in Mikrogramm pro Milliliter, die zu einer 50%igen Abnahme der Plaquebildungseinheiten im Vergleich zu einer unbehandelten Kontrolle führt.
- In einem modifizierten Standardtest, d.h. bei dem das Virus und die Testverbindung vor der Uberschichtung mit einem Agarmedium gemischt und inkubiert werden, hatten aktive Verbindungen dieser Erfindung IC&sub5;&sub0;-Werte von etwa 6,0 bis etwa 37,3 gegen Poliovirus 2, etwa 8,5 bis etwa 39,5 gegen Human-Rhinovirus 14 und etwa 2,4 bis 5,0 gegen Coxsackie-Virus B3.
- Der Standardtest beinhaltet das Überschichten von HeLa-Zellen mit Agarmedium, das abgemessene Konzentrationen der Testverbindung enthält, nach der Virusabsorption und dann das Inkubieren während 72 Stunden. Die resultierenden Plaques werden angefärbt, sichtbar gemacht und gemessen, um die Viruswachstumshemmung zu bestimmen, wie sie sich anhand der Plaquereduktion im Vergleich zu einer Kontrolle zeigt.
- Der modifizierte Standardtest wird als empfindlicher angesehen aufgrund seiner Fähigkeit, genauer zwischen den das Viruswachstum neutralisierenden Wirkungen von Verbindungen zu diskriminieren, deren IC&sub5;&sub0;-Werte nach dem Standardtest sehr nahe zusammen liegen.
- Von den antiviralen Verbindungen innerhalb des Umfangs von Formel II sind solche, die wasserlösliche Säureadditionssalze bilden, oral absorbierbar, zeigen gute Gewebespiegel und Serumstabilität.
- Die Verbindungen dieser Erfindung werden in herkömmlicher Weise für orale, parenterale, topische und transdermale Verwendung zubereitet, wobei oral bevorzugt wird.
- Diese Erfindung beinhaltet innerhalb ihres Umfangs pharmazeutische Zusammensetzungen, die die Verbindungen dieser Erfindung im Gemisch mit einem pharmazeutisch annehmbaren Träger umfassen. Außerdem beinhaltet die vorliegende Erfindung auch die Verwendung der Verbindungen der Formel II zur Herstellung pharmazeutischer Zusammensetzungen, die sich zur Behandlung viraler Infektionen eignen. In den obigen Zusammensetzungen können die aktiven Verbindungen dieser Erfindung allein als einziger antiviraler Wirkstoff oder in Kombination mit anderen therapeutischen Mitteln verwendet werden.
- Für die bevorzugte orale Verabreichung werden die Verbindungen dieser Erfindung typischerweise in Form von Tabletten, Kapseln, Elixieren, Lösungen, Suspensionen und dergleichen, vorzugsweise Lösungen, zubereitet. Für die parenterale Verabreichung können sie zu Lösungen oder Suspensionen zubereitet werden. Topische Zubereitungen, wie Lotionen, Cremes, Salben, Sprays und mechanische Abgabevorrichtungen, z.B. transdermale, können ebenfalls mit den Verbindungen dieser Erfindung hergestellt werden.
- Typische pharmazeutisch annehmbare Träger zur Verwendung in den oben beschriebenen Zubereitungen sind zum Beispiel: Zucker, wie Lactose, Stärken, wie Maisstärke, Cellulose und Derivate, wie Natriumcarboxymethylcellulose, Ethylcellulose und Methylcellulose, sowie andere in der Fachkunde wohlbekannte Träger. Die Zusammensetzungen können auch Konservierungsmittel, Aerosoltreibmittel sowie Färbe-, Verdickungs-, Suspendier-, Dispergiermittel, Emulgatoren, Netzmittel, Stabilisatoren und Puffer enthalten.
- Die verabreichte Dosis der Verbindungen dieser Erfindung unterliegt dem Urteil des behandelnden Arztes und hängt von einer Vielzahl von Faktoren ab, z.B. dem Alter und Gewicht des behandelten Individuums, dem Verabreichungsmodus, der Wirksamkeit der verabreichten Verbindung, der Indikation, für die das Medikament verabreicht wird, und dem Ausmaß der behandelten Krankheit.
- Typischerweise entspricht die pro Tag verabreichte Dosis bei der Behandlung viraler Infektionen einer oralen Verabreichung von etwa 1 mg/kg bis etwa 75 mg/kg pro Tag in einzelnen oder geteilten Dosen, wobei etwa 1-25 mg/kg bevorzugt sind.
- Die Verbindungen dieser Erfindung werden nach den folgenden Verfahren hergestellt:
- (A) Zur Herstellung einer Verbindung der Formel I wird eine Verbindung der Formel
- Z-X-Q'-L¹,
- wobei Z und X wie zuvor definiert sind, Q' dasselbe wie das zuvor definierte Q ist oder, falls Q in Formel I wenigstens eine der Gruppen - H-, wobei jedes R unabhängig Wasserstoff oder Niederalkyl ist, enthalten soll, Q' auch dasselbe wie das zuvor definierte Q minus wenigstens eine der Gruppen - H- sein kann und L¹ eine Abgangsgruppe ist, mit einer Verbindung der Formel
- L²-Y'-W"
- umgesetzt, wobei L² eine Abgangsgruppe ist, W" wie W in Formel I definiert oder ein Tautomer davon ist und Y' dasselbe wie das in Formel I definierte Y ist oder, falls Q in Formel I wenigstens eine der Gruppen - H-, wobei jedes R unabhängig Wasserstoff oder Niederalkyl ist, enthalten soll, Y' auch dasselbe wie das in Formel I definierte Y plus wenigstens eine der Gruppen - H- sein kann, oder

- wobei Y' und W" wie zuvor definiert sind und L² eine Abgangsgruppe ist, mit einer Verbindung der Formel
- L&sup5;-Q"-L&sup6;
- umgesetzt, wobei L&sup5; und L&sup6; Abgangsgruppen sind und Q" zweiwertiges, wie oben definiertes Q' ist,
- wobei in den obigen Verfahren, falls notwendig oder gewunscht, jede reaktive Gruppe geschützt sein kann und sich, falls notwendig oder gewünscht, folgendes an die obigen Verfahren anschließt:
- (i) Entfernung gegebenenfalls vorhandener Schutzgruppen,
- (ii) Umwandlung einer so hergestellten Verbindung in eine andere Verbindung der Formel I oder Formel II,
- (iii) wenn mehr als eine Verbindung der Formel I oder II hergestellt wird, Trennung der so hergestellten Verbindungen oder
- (iv) Umwandlung einer der so hergestellten Verbindungen in ein Säureadditions-, Basenadditions- oder quartäres Ammoniumsalz oder ein pharmazeutisch annehmbares Solvat davon.
- In Verfahren (A) ist L¹ vorzugsweise Brom oder am meisten bevorzugt Iod, und L² ist vorzugsweise ein Alkalimetall, wie Natrium, Kalium oder Cäsium Die Reaktion findet bei Temperaturen von etwa -20ºC bis 60ºC in einem inerten organischen Lösungsmittel, wie Dimethylsulfoxid (DMSO), Dimethylformamid (DMF) oder Tetrahydrofuran (THF), statt. In den meisten Fällen können die Endverbindungen durch herkömmliche Reaktionen in wasserlösliche Säureadditions- oder quartäre Salze umgewandelt werden, z.B. mit Salzsäure oder einem Quaternisierungsmittel, wie Methylsulfonsäure.
- Die Ausgangsverbindungen Z-X-Q'-L¹, wobei L¹ Halogen (Hal) ist, werden durch die folgende Reaktion hergestellt:
- ZXH + Hal-Q'-Hal -SäurefängerT Z-X-Q-Hal
- wobei Z, X und Q' wie oben definiert sind.
- Die Reaktion findet in Gegenwart eines Säurefängers, wie K&sub2;CO&sub3; oder organischer Basen, wie Collidin und auch Hunigs Base, statt. Die bevorzugte Hal-Gruppe ist Iod, obwohl auch Brom verwendet werden kann. Die Verbindung Hal-Q'-Hal, wobei Hal Iod ist, kann durch Umsetzen von Br-Q'-Br mit Natriumiodid hergestellt werden, außer wenn Q -CH&sub2;- oder -CH&sub2;-CH&sub2;- ist.
- Alternativ dazu kann ZXQ'-I durch Umsetzen von ZXH mit Br-Q'-Br, wobei man ZXQ'Br erhält, und dann Umsetzen von ZXQ'Br mit Natriumiodid, wobei man Z-X-Q'-I erhält, hergestellt werden. Oder bei Verbindungen, bei denen Q' -CH&sub2;- oder -C&sub2;H&sub4;- ist, kann Z-X-Q'-I hergestellt werden, indem man den entsprechenden einoder zweiwertigen Alkohol mit HI umsetzt.
- Wenn W eine stickstoffhaltige heterocyclische Struktureinheit ist, führen die oben beschriebenen Verfahren dazu, daß die Struktureinheit W an einem Stickstoffatom des heterocyclischen Ringes substituiert wird, es sei denn, das Stickstoffatom ist geschützt. Um eine Verbindung herzustellen, bei der der Heterocydus an einem Ringkohlenstoffatom substituiert ist, ist es notwendig, das Stickstoffatom mit einer Gruppe zu schützen, die sich nach der Durchführung der C-Substitution leicht entfernen läßt und während der C-Substitutionsreaktion nicht entfernt wird, z.B. die Tritylgruppe.
- So wird zum Beispiel bei der Herstellung eines 2-substituierten Imidazols das folgende Reaktionsschema befolgt: tritylchlorid Butyllithium
- wobei Q, Q', Z und X wie oben definiert sind. In diesem Fall wird die Methylgruppe an dem Imidazolring ein Teil von Q in der entstehenden Verbindung.
- Um bei Verfahren (B) eine Verbindung der Formel II herzustellen, sollte die Struktureinheit W' zwei Ringkohlenstoffatome haben, die anfangs mit Trimethylsilyloxygruppen substituiert sind. Wenn die Reaktion mit Z-X-Q'-I durchgeführt wird, ist das resultierende Produkt eine disubstituierte Struktureinheit W', wie es bei der folgenden Reaktion gezeigt ist, bei der die trimethylsilyloxysubstituierte Verbindung als ein Pyrimidin dargestellt ist.
- Die Reaktion wird bei Raumtemperatur durchgeführt.
- Wenn bei Verfahren (B) ein überschuß an der Verbindung Z-X-Q'-L¹ verwendet wird, wird das Produkt in erster Linie eines der Formel II sein. Wenn jedoch nur kleine Mengen der Verbindung Z-X-Q'-L¹ verwendet werden, entstehen Produkte der Formeln I und II. Die Reaktionsbedingungen sind dieselben wie bei Verfahren (A).
- Um eine Verbindung der Formel II herzustellen, bei der wenigstens eine der Einheiten Z, X oder Q nicht mit der anderen Einheit Z, X oder Q identisch ist, werden zwei unterschiedliche Verbindungen mit der allgemeinen Formel Z-X-Q'-L¹ verwendet. Das Ergebnis ist ein Gemisch von Verbindungen, die mit Standardtechniken isoliert werden können.
- Verfahren C, bei dem Verbindungen der Formel I entstehen, bei denen Z und W gleich sind und X und Y gleich sind, wird unter denselben Reaktionsbedingungen wie Verfahren (A) durchgeführt.
- Die Herstellung von Verbindungen, bei denen Q eine ungesättigte Kette ist, d.h. ein Alkindiyl oder Alkendiyl, wird durch die folgende Reaktion veranschaulicht:
- wobei R&sup8; Niederalkyl mit 2 bis 4 Kohlenstoffatomen ist und R' Niederalkinyl mit 3 Kohlenstoffatomen ist, Y eine Bindung ist, Pr eine Hydroxyschutzgruppe ist und hal Brom oder Iod ist.
- Das Alkendiyl wird hergestellt, indem man die Alkindiylverbindung durch katalytische Hydrierung partiell reduziert.
- Die Herstellung von Verbindungen, bei denen Q ein Alkyl ist, das ein Cycloalkan in der Kette enthält, wird durch die folgende Reaktion veranschaulicht: Mesylchlorid Triethylamin
- wobei hal Brom oder Iod ist und Z, X, Y und W wie oben für Formel I definiert sind.
- Eine Verbindung der Formel I oder II kann mit in der Fachkunde wohlbekannten Standardtechniken in eine andere Verbindung der Formel I bzw. II umgewandelt werden. Solche Umwandlungstechniken werden in den Beispielen veranschaulicht.
- Im allgemeinen beinhaltet das Herstellen der Verbindungen dieser Erfindung relativ einfache Verfahren, wie es durch die vielen Beispiele, die später in diesem Text erscheinen, veranschaulicht wird.
- Die folgende Tabelle I zeigt die Komponenten der in den Beispielen veranschaulichten Verbindungen der Formel II: Tabelle 1 Beispiel (Bind. = Bindung)
- Die Ausgangsverbindungen zur Verwendung bei der Herstellung der Verbindungen dieser Erfindung sind entweder im Handel erhältlich oder werden nach herkömmlichen, in der Fachkunde bekannten Methoden hergestellt.
- Die folgenden Beispiele erläutern die Erfindung. Massenspektren (MS) mit FAB (Fast Atom Bombardment) wurden auf einem doppeltfokussierenden Massenspektrometer Finnigan MAT 312 aufgenommen, das bei einer Beschleunigungsspannung von 3 kV arbeitete. Die MS-Proben wurden durch Beschuß mit Xenonatomen ionisiert, die von einer Sattelfeld-Ionenquelle von Ion Tech erzeugt wurden, die mit einem Röhrenstrom von 2 mA und einer Energie von 6 keV arbeitete. Die ¹H-NMR-Spektren (Protonen-NMR; kemmagnetische Resonanz) wurden bei 200 MHz auf einem Varian-XL-200-Spektrometer aufgenommen; alle Werte δ der chemischen Verschiebung sind in ppm gegenüber Tetramethylsilan zu tiefem Feld angegeben.
- (a) Etwa 500 mg Natriumiodid in 10 ml Aceton 5 bis 10 Minuten mit 350 mg 6-(2-Chlor-4-methoxyphenoxy)hexyl-1-bromid am Rückfluß halten, das Aceton entfernen, indem man Stickstoff durch das Reaktionsgemisch perlen läßt, Methylenchlorid hinzufügen, mit Wasser, dann Salzlösung waschen und über Natriumsulfat trocknen, wobei man 6-(2-Chlor-4-rnethoxyphenoxy)hexyl-1-iodid erhält.
- (b) 3 g 6- (2-Chlor-4-methoxyphenoxy)hexyl-1-iodid in 10 ml Acetonitril zu 1,2 g Rhodamin und 20 g Cäsiumcarbonat geben. Über Nacht rühren, dann das Acetonitril entfernen, Methylenchlorid hinzufügen, mit Wasser, dann salzlösung waschen und über Natriumsulfat trocknen. Auf einer Säule mit grobem Siliciumoxid mit Methylenchlorid und anschließend 5% Ethylacetat/Methylenchlorid und schließlich 10% Ethylacetat/Methylenchlorid eluieren, so daß die Titelverbindung isoliert wird.
- 350 mg 6-(2-Chlor-4-methoxyphenoxy)hexyl-1-bromid mit 10 ml Aceton und etwa 500 mg Natriumiodid 5 bis 10 Minuten am Rückfluß halten, das Aceton entfernen, indem man Stickstoff durch das Reaktionsgemisch perlen läßt, Methylenchlorid hinzufügen, mit Wasser, dann Salzlösung waschen und über Natriumsulfat trocknen, wobei man 6-(2-Chlor-4-methoxyphenoxy) hexyl-1-iodid erhält. 133 mg 5-Mercapto-1-methyltetrazol, 268 mg Cäsiumcarbonat und 3 ml Acetonitril zu dem Iodidprodukt geben, über Nacht rühren, Methylenchlorid hinzufügen und mit Wasser, Natriumcarbonat, Wasser, dann Salzlösung waschen und über Natriumsulfat trocknen.
- Das Lösungsmittel entfernen und die Titelverbindung in Form von Kristallen gewinnen.
- 300 mg des in Teil 1 oder 2 hergestellten Iodidprodukts, 1,83 g 2,4-Bis (trimethylsilyloxy)pyrimidin, wasserfreies DMF und 1,24 g Cäsiumfluorid in einem Reaktionskolben über Nacht bei Raumtemperatur rühren. Mit einem Wasserwaschschritt in Methylenchlorid aufarbeiten, auf einer Säule mit grobem Silicagel mit Methylenchlorid und anschließend 10% Ethylacetat/Methylenchlorid, dann 20% Ethylacetat/Methylenchlorid eluieren, so daß die Titelverbindung isoliert wird.
- 1 g 2-Hydroxybenzimidazol, 2 g des in Teil 1 oder 2 von Beispiel 1 hergestellten Iodids, 0,35 g Natriumhydroxid und 10 ml DMF in einem Reaktionskolben über Nacht bei Raumtemperatur rühren. Verteilung mit Wasser/Methylenchlorid. Auf einer Säule mit Siliciumoxid mit 100-% Methylenchlorid, dann 50/50 Methylenchlorid/Ethylacetat eluieren. Zwei Hauptfraktionen werden erhalten; NMR zeigt, daß die dem oberen Fleck entsprechende Fraktion N,N'-Bis[6-(2- Chlor-4-methoxyphenoxy)hexyl]-2-benzimidazolon ist, FAB-MS: m/z 615 (M&spplus;), freie Base; ¹H-NMR (200 MHz): ¹H (CDCl&sub3;), 1,35-1,65 (8H, m), 1,70-1,90 (8H, m), 3,75 (6H, s), 3,80-4,05 (8H, m), 6,7-7,2 (10H, m), und daß die dem unteren Fleck entsprechende Fraktion 1-[6-(2-Chlor-4-methoxyphenoxy)hexyl]-2-hydroxybenzimidazol ist, FAB-MS: m/z 375 (M&spplus;), freie Base; ¹H-NMR (200 MHz) δH (CDCl&sub3;), 1,4-1,70 (4H, m), 1,7-1,9 (4H, m), 3,75 (3H, s), 3,85-4,05 (4H, m), 6,7-7,2 (7H, m).

bei der Herstellung eines antiviralen Medikaments.
