Bei der Verfahrensvarianten b) handelt es sich um ein oh
misch eigenartiges Verfahren, da überraschenderweise χιηΛ
unvorhersehbar der Imidazolrest nicht mit dem atom, das durch das austretende Bromatom eine freie
erhalten hat, sondern mit dem benachbarten Kohlenstoffatom der Methylgruppe reagiert.
In den Formeln II und III stehen als Alkyl- und Alkenylreste
R , R und R^ vorzugsweise geradkettige, oder verzweigte
Alkylreste mit 1 bis 6, insbesondere 1 bis 4 Kohlenstoffatomen oder geradkettige oder verzweigte Alkenylreste mit vorzugsweise

wäßrige Formaldehydlösung, in einem inerten organischen Lösiangsmittel, z. B» Äthanol, bei erhöhter
Temperatur, z. B. Siedetemperatur des Reaktionsgemisches
umsetzt und das gewünschte Produkt auf die übliche Weise isoliert und reinigt. Die Ketogruppe wird gegebenenfalls in üblicher
Weise in ein funktionelles Derivat umgewandelt.
Die bei der'Verfahrensvariante a) als Ausgangsstoffe verwendeten
Verbindungen der Formel II, in denen R für Halogen steht, sind noch nicht bekannt, können aber nach üblichen Methoden
hergestellt werden, z. B. indem man das Ätherketon der Formel
VI, wie oben erläutert, in Gegenwart von Alkali mit Formaldehyd oder Formaldehyd abgebenden Mitteln zur Reaktion
bringt und anschließend die erhaltene Verbindung der Formel II, in welcher R* für OH steht, mit einem Halogenierungsmittel*,
wie Thionylchlorid, in einem inerten, polaren.organischen
Lösungsmittel, z. B. Methylenchlorid, bei Raumtemperatur umsetzt und das gewünschte Bndprodukt in üblicher Weise isoliert
und gegebenenfalls reinigt. Die Ketogruppe wird gegebenenfalls
in üblicher Weise in ein funktionelles Derivat umgewandelt.

ca. 500C siedende allphatisehe und aromatische Kohlenwasserstoffe, wie Ligroin, Benzol oder Toluol.
Die Umsetzung (Verfahren a)j R steht für Hydroxyl) wird
vorzugsweise in Gegenwart eines wasserentziehenden Mittels, wie gebranntes Calciumcarbonat oder trockenes Natriumsulfat
oder mit Hilfe eines Wasserabscheiders vorgenommen.
Die Reaktionstemperaturen können beim Verfahren a), falls
R für Hydroxyl steht, in einem größeren Bereich variiert
werden. Im allgemeinen arbeitet man zwischen etwa 5O0C bis
etwa 1800C, vorzugsweise bei 8O0C bis 140°C.
Bei der Durchführung des erfindungsgemäßen Verfahrens a),
falls R* für Hydroxyl steht, setzt man auf 1 Mol der Verbindung der Formel II vorzugsweise1 bis 5» insbesondere
1 bis 1,5 Mol Imidazol und gegebenenfalls 0,5 bis 20,
insbesondere 1 bis 5 Mol des wasserentziehenden Mittels ein.
Die Isolierung der Verbindungen der Formel I wird nach allgemein bekannten, üblichen Methoden vorgenommen. Beispielsweise kann das Lösungsmittel im Vakuum abdestilliert und der
Rückstand mit Methylenohlorid aufgenommen werden. Die organische Phase wird anschließend abgetrennt. Nach dem Trocknen
über Natriumsulfat wird das lösungsmittel im Vakuum abdestilliert. Der erhaltene Rückstand wird durch Umkristallisation
gereinigt. Falls die Reinigung durch Umkristallisieren nicht ausreichend 1st, kann nach dem ersten Abdestillieren des Lösungsmittels das Salz hergestellt werden und dieses durch
Umkristallisieren gereinigt werden. Aus dem Salz kann in bekannter Weise mit Hilfe einer Base die Verbindung der Formel I
in Form der Base freigesetzt werden.

der Formel II mit Imidazol) alle für das erfindungsgemäße Verfahren inerten, polaren organischen Lösungsmittel infrage.
Hierzu gehören vorzugsweise Äther, insbesondere cyclische Äther, wie Dioxan oder Tetrahydrofuran, litrile, insbesondere
niedere Alkylnitrile, wie Acetonitril, Amide, insbesondere
niedere Dialkyl amide, wie Dimethylformamid, und Sulfoxide,
insbesondere niedere Dialkylsulfoxide, wie Dimethylsulfoxid.
Die Umsetzung (Verfahren a); R steht für Chlor oder Brom)
wird vorzugsweise in Gegenwart eines Säurebinders vorgenommen. Zweckmäßigerweise verwendet man einen entsprechenden Überschuß
an Imidazol. Man kann aber auch alle übrigen üblicherweise
verwendbaren organischen Säurebinder einsetzen, wie niedere tertiäre Alkylamine, z. B* Triäthylamin, oder sekundäre organische
Basen, z. B. Pyridin.
Die Reaktionstemperaturen können beim Verfahren a), falls R^"
für Chlor oder Brom steht, in einem größeren Bereich variiert werden. Im allgemeinen arbeitet man zwischen etwa 5G0C bis.
etwa 1500C, vorzugsweise bei 8O0C bis etwa 150°C.
Bei der Durchführung des erfindungsgemäßen. Verfahrens a),
falls R für Chlor oder Brom steht, setzt man auf 1 Mol der Verbindung der Formel II vorzugsweise etwa 1 bis 5, vorzugsweise
1 bis'1,5 Mol Imidazol und zusätzlich etwa 1 bis 10,
vorzugsweise 1 bis 2 Mol des säurebindenden Mittels (z. B. Imidazol) ein.

Die neuen Wirkstoffe der Formel I sowie deren.Salze besitzen
eine sehr gute und breite antimykotische Wirkungj insbesondere
gegen Dermatophyten, Epidermatophyten und Sproßpilze sowie
biphasische Pilze und Hefepilze, z. B. Triohophyton und
Candida, Sie können deshalb init gutem Erfolg gegen Pilzinfektionen
bei Mensch und Tier eingesetzt werden.
Als Indikationsgebiete in der Humanmedizin können beispielsweise genannt werden:
Dermatomykosen und Systemmykosen durch Trichophyton mentagrophytes
und andere Trichophytonarten, Mikrosporon Arten, Epidermatophyton floccosum, Sproßpilze und biphasische Pilze
sowie Schimmelpilze.
Als Indikationsgebiete in der Tiermedizin können beispielsweise
aufgeführt werden:
Alle Dermatomykosen und Systemmykosen, insbesondere solche, die durch die oben genannten Erreger hervorgerufen werden.
Zur vorliegenden Erfindung gehören pharmazeutische Zubereitungen,
die neben nichttoxischen, inerten, pharmazeutisch geeigneten Trägerstoffen eine oder mehrere Verbindungen der
Formel I und/oder deren Salze enthalten oder die aus einer oder mehreren Verbindungen der Formel I und/oder deren Salzen
bestehen, sowie Verfahren zur Herstellung dieser Zubereitungen.
Zur vorliegenden Erfindung gehören auch pharmazeutische Zubereitungen
in Dosierungseinheiten. Dies bedeutet, daß die Zubereitungen in Form einzelner Teile, z. B. Tabletten,
Dragees-, Kapseln, Pillen, Suppositorien und Ampullen vorliegen, deren Wirkstoffgehalt einem Bruchteil oder einem

Vielfachen einer Einzeldoais entsprechen. Die Dosierungseinheiten können z. B. 1, 2, 3 oder 4 Einzeldosen oder 1/2, 1/3
oder 1/4 einer Einzeldosis enthalten. Eine Einzeldosis enthält vorzugsweise die Menge V/irkstoff, die bei einer Applikation verabreicht wird und die gewöhnlich einer ganzen,
einer halben oder einem Drittel oder einem Viertel einer Tagesdoeia entspricht.
Unter nichttoxischen, inerten pharmazeutisch geeigneten Trägeretoff en sind feste, halbfeste oder flüssige Verdünnungsmittel, Füllstoffe und Formulierungehilfsmittel jeder Art
zu verstehen. Als bevorzugte pharmazeutische Zubereitungen seien Tabletten, Dragees, Kapseln, Pillen, Granulate, Suppositorien, Lösungen, Suspensionen und Emulsionen, Fasten,
Salben, OeIe, Cremes, Lotions, Fuder und Sprays genannt.
Tabletten, Dragees, Kapseln, Pillen und Granulate können den oder die Wirkstoffe neben den üblichen Trägerstoffen enthalten, wie (a) Füll- und Streckmittel, z. B. Stärken, Milchzucker, Rohrzucker, Glukose, Mannit und Kieselsäure,
(b) Bindemittel, z. B. Carboxymethylcellulose, Alginate, Gelatine, Polyvinylpyrrolidon, (o) Feuchthaltemittel, E. B.
Glycerin, (d) Sprengmittel, z. B. Agar-Agar, Calciumcarbonat und Natriumcarbonat, (e) Lösungsverzögerer, z. B. Paraffin,
und (f) Resorptionsbeschleuniger, z. B. quarternary Ammoniumverbindungen, (g) Netzmittel, z. B. Cetylalkohol, Glycerinmonostearat, (h) Adsorptionsmittel, z. B. Kaolin und Bentonit,
und (i) Gleitmittel, z. B. Talkum, Calcium- und Magnesiumstearat und feste Polyäthylenglykole oder Gemischt der unter
(a) - (i) aufgeführten Stoffe.
Die Tabletten, Dragees, Kapseln, Pillen und Granulate können
mit den üblichen gegebenenfalls Opakisierungsmittel enthaltenden überzügen und Hüllen versehen sein und auch so zusammenge-

setzt sein,, daß sie den oder die Wirkstoffe nur oder bevorzugt
in einem "bestimmten Teil des Intestinaltraktes gegebenenfalls
verzögert abgeben, wobei als Einbettungsmassen z. B. Polymersubstanzen
und Wachse verwendet werden können.
Der oder die Wirkstoffe können gegebenenfalls mit einem oder
mehreren der oben angegebenen !Prägerstoffen auch in mikroverkapselter Form vorliegen«,

Suppositorien können neben dem oder den Wirkstoffen die üblichen

Wirkstoffen die üblichen Trägerstoffe enthalten, z. B. tierische und pflanzliche Fette, Wachse, Paraffine, Stärken,
Tragant, Cellulosederivate, Polyäthylenglykole, Silicone,
Bentonite, Kieselsäure, Talkum und Zinkoxid oder Gemische
dieser Stoffe. N - .
Wirkstoffen die üblichen Trägerstoffe enthalten, z. B. tierische und pflanzliche Fette, Wachse, Paraffine, Stärken,
Tragant, Cellulosederivate, Polyäthylenglykole, Silicone,
Bentonite, Kieselsäure, Talkum und Zinkoxid oder Gemische
dieser Stoffe. N - .
Puder und Sprays können neben dem oder den Wirkstoffen die
üblichen Trägerstoffe enthalten, z. B. Milchzucker, Talkum,
Kieselsäure, Aluminiumhydroxid, Calciumsilikat und Polyamidpulver oder Gemische dieser Stoffe. Sprays können zusätzlich
die üblichen Treibmittel, z. B. Chlorfluorkohlenwasserstoffe, enthalten.

Kieselsäure, Aluminiumhydroxid, Calciumsilikat und Polyamidpulver oder Gemische dieser Stoffe. Sprays können zusätzlich

Lösungen und Emulsionen können neben dem oder den Wirkstoffen die üblichen Trägerstoffe, wie Lösungsmittel, Lösungsvermittler
und Emulgatoren, z. B, Wasser, Äthylalkohol, Isopropylalkohol,
Äthylcarbonat, Äthylacetat, Benzylalkohol, Benzylbenzoat,
Propylenglykol, 1,3-Butylenglykol, Dimethylformamid,

Öle, insbesondere Baumwollsaatöl, Erdnußöl, Maiskeimöl,
Olivenöl, RicinuBöl und Sesamöl, Glycerin, Glyc.erinf örmal,
Tetrahydrofurfurylalkohol, Polyäthylenglykole und Fettsäureester des Sorbitans oder Gemische dieser Stoffe enthalten.
Zur parenteralen Applikation können die Lösungen und Emulsionen auch in steriler und blutlsotonischer Form vorliegen.
Suspensionen können neben dem oder den Wirkstoffen die üblichen Trägerstoffe, wie flüssige Verdünnungsmittel, z. B.
Wasser, Äthylalkohol, Propylenglykol, Suspendiermittel,
z. B. äthoxylierte Isostearylalkohole, Polyoxyäthylensorbit-
und -sorbitanester, mikrokristalline Cellulose, Aluminiummetahydroxid, Bentonit, Agar-Agar und Tragant oder Gemische
dieser Stoffe enthalten.
Die genannten Pormulierungsformeln können auch Färbemittel,
Konservierungestoffe sowie geruche- und geschmacksverbessernde Zusätze, z, B. Ffefferminzöl und Eukalyptusöl und
Süßmittel, z. B. Sacharin, enthalten.
Die therapeutisch wirksamen Verbindungen sollen in den oben aufgeführten pharmazeutischen Zubereitungen vorzugsweise in
einer Konzentration von etwa 0,1 bis 99,5, vorzugsweise von etwa 0,5 bis 95 Gewichtsprozent der Geaamtmischung vorhanden
sein.
Die oben aufgeführten pharmazeutischen Zubereitungen können außer Verbindungen der Formel I und/oder deren Salzen auch
andere pharmazeutische Wirkstoffe enthalten.
Die Herstellung der oben aufgeführten pharmazeutischen Zubereitungen erfolgt in üblicher Weise nach bekannten Methoden,
z. B. durch Mischen des oder der Wirkstoffe mit dem oder den Trägerstoffen.

Zur vorliegenden Erfindung gehfct aucia die Verwendung der
Verbindungen der Formel I und/oder deren Sals©n sowie von
pharmazeutischen Zuber®itasgeng die eine oder mehrere Verbindungen
der Formel I· und/oder deren Sals'© enthalten, in der Human- und Veterinärmedizin ζητ Verhütung, Besserung
und/oder Heilung der oben angeführten Erkrankungen.
Sie Wirkstoffe oder die pharmazeutischen Zubereitungen können
lokal, oral, parenteral, intraperitoneal und/oder rectal, vorzugsweise
oral oder lokal, appliai©rt
Im allgemeinen hat es eich sowohl in der Huuan- ale auoh in
der Veterinärmedizin als vorteilhaft erwiesen, den oder di©
Wirkstoffe in Mengen von etwa 30 bis etwa 200, vorzugsweise
50 bis 200 mgAg Körpergewicht je 24 Stunden, gegebenenfalls
in Form mehrerer Einzelgaben zur Erzielung der gewünschten
Ergebnisse zu verabreichen. Es kann jedoch erforderlich sein, von den genannten Dosierungen abzuweichen, und zwar in Abhängigkeit von der Art und der Schwäre der Erkrankung, der
Art der Zubereitung und der Applikation des Arzneimittel!},,
sowie dem Zeitraum bzw, Intervall, innerhalb welchem di© Verabreichung erfolgt. So kann es in einigen Fällen ausreichend
sein, mit weniger als der oben genannten Menge Wirkstoff auszukommen, während in anderen Fällen die oben
angeführte Wirkstoffmenge überschritten werden muß. Die Festlegung
der jeweils erforderlichen optimalen.Dosierung und Applikationsart der Wirkstoffe kann durch Jeden Faohmann auf

Antimykotische Wirkung der erfindungsgemäßen Verbindungen
der Formel I im Tierexperiment
a) Lοkaianwendung bei der experimentellen Meerschweinchen-Trichophytie
(Erreger: Trichophyton mentagrophytes)
Versuohsbeachreibung:
Eine 1 #ige Lösung der Wirkstoffe in einem Dimethylsulfoxid/
Glycerin/Wasser-Gemiseh (1 : 3 : 6) oder in Polyäthylenglykol
400 wurde während 11 bis 14 Tagen einmal täglich lokal appliziert,
nachdem die Trichophytie experimentell erzeugt -worden war. ;
Die VersuchBergebnisse werden durch die Tabelle B wiederge- .
geben:

b) Wirkung (orale Applikation) "bei der Quinckeanum-Trichophytie
der weißen Maus
Mit Dosen von 2 χ 100 mg/kg Körpergewicht, täglich einmal
bis zum achten Tag der Infektion per os gegeben, konnte
das Angehen der Quinckeanum-Infektion bei Mäusen unterdrückt
werden.
Das Ergebnis ist aus Tabelle C zu entnehmen:

c) Candidose der Mäuse
Versuchsbeschreibung:
Mäuse vom Typ SPF-C1 wurden intravenös mit 1 - 2 χ
logarithmisch wachsenden Candida-Zellen, die in physiologischer Kochsalzlösung suspendiert waren, infiziert.
Die Tiere wurden zweimal täglich post infektionem mit 100 mg/kg Körpergewicht der Präparate oral behandelt.
ünbehandelte Tiere starben 3 bis 6 Tage post infektionem
an der Infektion. Die Überlebensrate am 6. Tage post infektionem betrug bei den Kontrolltieren 2 bis 3 pro
20 Tiere (-10 - 15 #).
Die Versuchsergebnisse sind in.Tabelle D zusammengefaßt:

Die erfindüngagemäßen Verfahren seien anhand der folgenden
Beispiele erläutert:

16,0 g (0,05 Mol) 2-(4-Chlorphenoxy)-2-brom-4|4-dimethylpentan-3-on werden in 120 ml Acetonitril mit 12 g (0,207 Μοί)
Imidazol 12 Stunden am Rückfluß zum Sieden erhitzt. Dann wird das Lösungsmittel im Vakuum bis fast zur Trockne abdestilliert, anschließend werden 50 ml Äther und 50 ml
gesättigte ätherische Chlorwasserstofflösung zugegeben. Das entstandene Ul wird abdekantiert, mit dreimal je 50 ml
Äther aufgekooht und die Ätherphase abdekantiert.
Das hinterbleibende Ul wird mit 120 ml Methylenchlorid aufgenommen, mit 50 ml Wasser versetzt, 20 g festes Natriumbioarbonat zugegeben, die organische Phase abgetrennt und die
wäßrige Phase mit zweimal je 50 ml Methylenchlorid extrahiert. Die vereinigten organischen Phasen werden mit je 50 ml Wasser
zweimal gewaschen, über Natriumsulfat getrocknet und im Vakuum
abdestilliert. Das erhaltene öl wird mit Ligroin/Petroläther angerieben, wobei es kristallisiert. Nach dem "Umkristallisieren aus Ligroin/Petroläther erhält man 2,6 g 2-(4-Chlorphenoxy) -1-imidazolyl-(1)-4,4-dimethyl-pentan-3-on (17 # der
Theorie) vom Schmelzpunkt 68 - 730C.

Das Ausgangsprodukt 2-(4-Chlorphenoxy)-2-brom-4,4-dimethylpentan-3-on
wird durch Bromierung von 2-(4-Chlorphenoxy)-4,4-dimethyl-pentan-3-on
mit elementarem Brom in Tetrachlorkohlenstoff bei 40° - 5O0C gewonnen? Schmelzpunkt ι 95°C.
Die übrigen Ausgangsstoffe sind auf die gleiche Weise erhältlich.

g (0, Mol) 2-(2,5-Dichlorphenoxy)-1-hydroxy-4,4-dimethyl-pentan-3-on
werden in 200 ml Toluol aufgenommen, dazu 10,2 g (0,14 Mol) Imidazol zugetropft und die ReaktionslOsung 3 Stunden am Wasserabscheider gekocht. Danach wird
das Lösungsmittel in Vakuum abdestilliert, da« erhalten· Ol
wird alt 100 ml Wasser versetzt und zweimal Bit je 100 el
Methylenchlorid extrahiert.
Sie organische Phase wird mit zweimal 50 ml Wasser gewaschen,
über Natriumsulfat getrocknet und das Lösungsmittel im Vakuum abdestilliert.

Man erhält ein öl, das in 50 ml Äther aufgenommen und mit
50 ml, mit trockenem Chlorwasserstoff gesättigtem Äther versetzt wird. Das Lösungsmittel wird im Vakuum abdestilliert,
das resultierende Öl mit einem Gemisch aus 500 ml Ligroin und 3OD ml Essigester aufgenommen und unter Rückfluß
zum Sieden erhitzt. Nach dem vorsichtigen Abgießen der entstandenen Lösung und Abkühlen derselben fallen 18,5 g (49 %
der Theorie) 2-(2,5-Dlchlorphenoxy)-4i4-dimethyl-1-(1-iinidazolyl)-pentan-3-on-hydrochlorid
als farblose Kristalle aus, die durch Abfiltrieren gewonnen werden. Schmelzpunkt! 1620C (Zersetzung) .
26,1 g (0,1 Mol) 1-(2,5-Dichlorphenoxy)-3,3-dimethyl-butan-2-on
werden in 200 ml Äthanol gelöst, dazu 20 g (0,24 Mol)
40 #ige Pormaldehydlöaung und danach etwa 5 ml 10 ^ige Natronlauge
bis zum P11 9 zugegebene Bis !©aktionsmischung wird
3 Stunden unter Rückfluß erhitzt und das Lösungsmittel im Vakuum abdestilliert. Der entstandene Niederschlag wird abfiltriert
und mit Petroläther gut naehgewaschen. Das Eiltrat
wird im Vakuum eingeengt. Zurück bleibt ein Öls rohes 2-(2,5-Dichlorphenoxy
) -1 -hydroxy-4,4-dimethyl-pentan-3-;on·
Die übrigen Ausgangsstoffe sind auf die gleiche Weise erhältlich.
In vielen Fällen können die Verbindungen der Formeln IV
und V unter den oben angegebenen Bedingungen über die Verbindungen
der Formel II in einem Eintopfverfahren. Mit Imidazol
zu Verbindungen der Formel I umgesetzt werden.

24 g (0,1 Mol) i-Chlor-2-phenoxy-4,4-dimethyl-pentan-3-on

vom Brechungsindex η ^= 1,5081 werden zu einer Lösung von 13,6 g (0,2 Mol) Imidazol in 150 ml wasserfreiem Acetonitril gegeben. Die entstandene Lösung wird 4 Stunden unter Rückfluß zum Sieden erhitzt; danach wird das Lösungsmittel im Vakuum abdestilliert. Der Rückstand wird mit Wasser gewaschen, mit Methylenchlorid aufgenommen und über Natriumsulfat getrocknet. Nach dem Abdestillieren des Lösungsmittels erhält man 25,3 g (93 # der Theorie) 1-Imidazolyl-(1)-2-phenoxy-4,4-dimethyl-pentan-3-on als öl vom Brechungsindex η^ « 1,5193.
vom Brechungsindex η ^= 1,5081 werden zu einer Lösung von 13,6 g (0,2 Mol) Imidazol in 150 ml wasserfreiem Acetonitril gegeben. 22,3 g (0,1 Mol) 1-Hydroxy-2-phenoxy-4,4-dimethyl-pentan-3-on (Herstellung gemäß Beispiel 2) werden in 120 ml Methylenchlorid gelöst und zu dieser Lösung 7,3 ml (0,1 Mol) Thionylchlorid bei Raumtemperatur getropft. Der Beginn der Reaktion
kann durch gelindes Erwärmen beschleunigt werden. Nach dreistündiger Reaktionszeit bei Raumtemperatur wird das Lösungsmittel im Vakuum abdestilliert und der ölige Rückstand im
Hochvakuum entgast.

Man erhält 21,8 g (9.1 ί> der Theorie) l-Chlor^-phenoxy^^-

dimethyl-pentan-3-on als öl vom Brechungsindex η ^ = 1,5081. .
Die übrigen Ausgangsstoffe können auf die gleiche Weise erhalten
werden.
Nach den Verfahren a) und b) werden in jeweils guten Ergebnissen
gemäß den Beispielen 1 bis 3 die folgenden Beispiele der Tabelle 1 erhalten:
