Description
- Die Erfindung betrifft die Herstellung in hoher Ausbeute von β-Lactam-Antibiotika mit einem hohen Reinheitsgrad, dadurch gekennzeichnet, daß die Acylierungsreaktion des β-Lactam-Zwischenproduktes in Anwesenheit eines Chlorwasserstoffakzeptors Nikotin- oder Isonikotinbase ausgeführt wird. Insbesondere betrifft die vorliegende Erfindung die Herstellung in hoher Ausbeute von Derivaten der Cephalosporan- und Penicillansäuren der Formeln:

- worin x = -O-CO-CH&sub3;, mit gewöhnlichen Silylierungsmitteln, wie etwa Trimethylchlorsilan, Hexamethyldisilazan, Dimethyldichlorsilan und ähnlichen, gefolgt von Acylierung mit D-α-Aminosäurechloriden, in denen die Aminogruppe als Hydrochlorid blockiert ist, der allgemeinen Formel

- US-A-4 182 709 offenbart ein Verfahren zur Herstellung von halbsynthetischen Penicillin-Antibiotika, insbesondere Ampicillin, welches die Acylierung einer silylierten 6-Aminopenicillansäure in Gegenwart von primären oder sekundären Carboxamidbasen als Halogenidwasserstoffakzeptoren umfaßt.
- Beispielsweise kann gemäß der vorliegenden Erfindung die Silylierungsreaktion entweder mit Hexamethyldisilazan in Methylenchlorid oder mit Trimethylchlorsilan in Gegenwart einer Base, wie beispielsweise Trimethylamin oder Diethylamin, bei einer Temperatur von 0ºC bis 42ºC ausgeführt werden.
- Diese Chlorwasserstoffakzeptorbasen haben den Vorteil, daß sie eine sehr niedrige Toxizität zeigen im Vergleich zu denen, die gewöhnlich für solche Reaktionen verendet werden, wie etwa Pyridin und Dimethylanilin.
- Die Acylierungsreaktion der zuvor silylierten Verbindung (III) und (IV) wird nach an sich bekannten Verfahren unter Verwendung der Verbindungen der Formel (V) ausgeführt, jedoch in Gegenwart von solchen Basen wie (VI) oder (VII).
- Die Basen (VI) und (VII) haben weiterhin den Vorteil, in Wasser ebenso wie in organischen Lösungsmitteln löslich zu sein,und können daher leicht entfernt werden.
- Die Acylierungsreaktion kann bei einer Temperatur von -40ºC bis +40ºC ausgeführt werden.
- Indem man nach dem erfindungsgemäßen Verfahren vorgeht, kann die Gewinnung der herzustellenden Produkte leicht entweder durch Extraktion und Ausfällung des Zwitterions bei isoelektrischem pH oder durch Ausfällung des Hydrochlorids durch Zugabe von Alkoholen, wie etwa Methanol, Ethanol, Isopropanol, n-Propanol und Butanol, zu den Reaktionsgemischen bewirkt werden.
- Das Hydrochlorid kann dann in das Zwitterion umgewandelt werden, indem man eine wäßrige Lösung des obigen Produktes bis zum isoelektrischen Punkt mit einer geeigneten Base behandelt.
- Das Verfahren dieser Erfindung wird nun durch die folgenden Beispiele erläutert, die auf spezielle Cephalosporine und Penicilline beschränkt sind; der Fachmann könnte jedoch ohne jede Schwierigkeit dieses Verfahren zur Herstellung weiterer β-Lactam-Antibiotika einsetzen, ohne damit den Bereich der vorliegenden Erfindung zu verlassen.
- Zu einer Suspension von (6-APA) 6-Aminopenicillansäure (54 g; 0,25 Mol) in Methylenchlorid (450 ml) wird Diethylamin (36,5 g; 0,5 Mol) zugegeben. Zu der so erhaltenen Lösung wird bei einer Temperatur von 0 bis +5ºC Trimethylchlorsilan (54,5 g; 0,5 Mol) zugegeben.
- Nach 90 Minuten bei 25 bis 30ºC ist die Silylierungsreaktion vollständig abgelaufen.
- Die Reaktionsmischung wird auf -30ºC gekühlt und Nikotinamid (36,6 g; 0,3 Mol) und D-α-Phenylglycinchlorid-hydrochlorid (51,5 g; 0,25 Mol) werden zugegeben.
- Die Suspension wird 2 Stunden bei -5ºC gehalten und dann 30 Minuten bei +10ºC.
- Sie wird mit Wasser behandelt (300 ml) und die Phasen werden getrennt.
- Die wäßrige Phase wird mit konzentriertem NH&sub4;OH bis zum isoelektrischen Punkt (pH = 4,5) behandelt.
- Nach 1 Stunde bei 0º/+5ºC wird das Antibiotikum abfiltriert und dann mit Wasser bis zum Verschwinden von Cl&supmin; gewaschen.
- Es wird unter Vakuum getrocknet, wodurch 85 g Ampicillin·3H&sub2;O erhalten werden.
- Ausbeute: 84,3% H&sub2;O = 13,4% (Karl Fischer) [α]²&sup0;D = +297º

- Unter den Bedingungen von Beispiel 1 wird unter Einsatz von äquimolaren Mengen von 7-ADCA anstelle von 6-APA Cephalexin·H&sub2;O (69 g) erhalten.
- Ausbeute: 77,5%
- H&sub2;O = 6,6% (Karl Fischer)

- Mikrobiologischer Titer = 930 ug/mg
- Spektrophotometrischer Titer = 920 ug/mg
- Acidimetrischer Titer = 98,7%
- Amintiter = 97,9%
- Unter den Bedingungen von Beispiel 3 wird unter Einsatz von p·OH-Phenylglycinchlorid-hydrochlorid, Cephadroxil·H&sub2;O (73,8 g) erhalten.
- Ausbeute: 77,5%
- H&sub2;O = 5,6 (Karl Fischer)
- Mikrobiologischer Titer = 930 ug/mg
- Spektrophotometrischer Titer = 933 ug/mg
- Acidimetrischer Titer = 99,1%
- Amintiter = 97,7%
- Zu einer Suspension von 6-APA (54 g; 0,25 Mol) und Nikotinsäure (30,75 g; 0,25 Mol) wird Et&sub3;N (75,9 g; 0,75 Mol) zugegeben.
- Zur so erhaltenen Lösung wird bei einer Temperatur von 0 bis 5ºC Trimethylchlorsilan (81,75 g; 0,75 Mol) zugegeben. Nach 90 Minuten bei 25 bis 30ºC wird die Reaktionsmischung auf -30ºC abgekühlt und danach wird D-α-Phenylglycinchlorid-hydrochlorid (51,5 g; 0,25 Mol) zugegeben.
- Die Suspension wird 2 Stunden bei -5ºC gehalten und dann eine weitere Stunde bei +10ºC gehalten.
- Sie wird mit Wasser behandelt (300 ml), und die Phasen werden getrennt. Die wäßrige Phase wird mit
- konzentriertem NH&sub4;OH zur Erreichung des isoelektrischen Punktes (pH = 4,5) behandelt.
- Das ausgefällte Produkt wird abfiltriert und dann mit Wasser gewaschen.
- Es wird unter Vakuum getrocknet, wodurch 78 g Ampicillin·3H&sub2;O erhalten werden.
- Ausbeute: 77,4%
- H&sub2;O = 14,78% (Karl Fischer)
- [α]²&sup0;D = +291º
- Mikrobiologischer Titer = 840 ug/mg
- Mercurimetrischer Titer = 830 ug/mg
- Acidimetrischer Titer = 96,8%
- Amintiter = 96,0%
- Unter den selben Bedingungen wie in Beispiel 5 wird unter Verwendung von (7-ADCA) 7-Aminodeacetoxycephalosporansäure anstelle von 6-APA Cephalexin·H&sub2;O (67 g) erhalten.
- Ausbeute: 73,4%
- H&sub2;O = 7,5% (Karl Fischer)
- [α]²&sup0;D = +151º
- Mikrobiologischer Titer = 905 ug/mg
- Spektrophotometrischer Titer = 900 ug/mg
- Acidimetrischer Titer = 97,8%
- Amintiter = 96,9%
- Zu einer Suspension von 7-ADCA (53,5 g; 0,25 Mol) in Methylenchlorid (450 ml) wird Et&sub2;NH (36,5 g; 0,5 Mol) zugegeben.
- Zur so erhaltenen Lösung wird bei einer Temperatur von 0 bis +5ºC Trimethylchlorsilan (54,5 g; 0,5 Mol) zugegeben.
- Nach 90 Minuten bei -35ºC wird die Reaktionsmischung auf -30ºC abgekühlt, und Methylisonikotinat (34,5 g; 0,25 Mol) und D-α-Phenylglycinchlorid-hydrochlorid (51,5 g; 0,25 Mol) werden zugegeben.
- Man verfährt wie in Beispiel 5 und erhält Cephalexin·H&sub2;O (77 g).
- Ausbeute: 84,3%
- H&sub2;O = 7,5% (Karl Fischer)
- [α]²&sup0;D = +154º
- Mikrobiologischer Titer = 920 ug/mg
- Spektrophotometrischer Titer = 920 ug/mg
- Acidimetrischer Titer = 99%
- Amintiter = 98,2%
- Unter den selben Bedingungen wie in Beispiel 7 wird unter Anwendung von Methylnikotinat anstelle von Methylisonikotinat Cephalexin·H&sub2;O (78 g) erhalten.
- Ausbeute = 85%
- H&sub2;O = 7,4% (Karl Fischer)
- [α]²&sup0;D = +155º
- Mikrobiologischer Titer = 920 ug/mg
- Spektrophotometrischer Titer = 915 ug/mg
- Acidimetrischer Titer = 99,2%
- Amintiter = 98,5%.
Claims (4)
1. Verfahren zur Herstellung von beta-Lactam-Antibiotika
durch Acylierung eines beta-Lactam-Zwischenproduktes mit
einem Säurechlorid in Anwesenheit eines pharmakologisch
nicht-toxischen Chlorwasserstoffakzeptors Nikotin- oder
Isonikotinbase.
2. Verfahren gemäß Anspruch 1 zur Herstellung von Derivaten
der Cephalosporan- und Penicillansäuren der Formel
und
durch Silylierung der Verbindungen
und
worin X = -O-CO-CH&sub3;, gefolgt von Acylierung mit einem
Chlorid der Formel:
worin R = H, OH, dadurch gekennzeichnet,
daß die Acylierung in Anwesenheit eines
Chlorwasserstoffakzeptors Nikotin- oder Isonikotinbase
ausgeführt wird.
3. Verfahren gemäß Anspruch 1 oder 2, dadurch
gekennzeichnet, daß die
Acylierungsreaktion in Anwesenheit einer Verbindung der
Formeln:
ausgeführt wird, worin R&sub1; =
-COOR&sub4;; -COOH; -CN,
R&sub2; = R&sub3; = H, oder R&sub2; = H und R&sub3; = lineare oder verzweigte
C&sub1;&submin;&sub5;-Alkylkette; oder
R&sub2; = R&sub3; = lineare oder verzweigte C&sub1;&submin;&sub5;-Alkylkette; oder
R&sub2; ≠ R&sub3; = lineare oder verzweigte C&sub1;&submin;&sub5;-Alkylkette; und
R&sub4; = lineare oder verzweigte C&sub1;&submin;&sub5;-Alkylkette.
4. Verfahren gemäß Anspruch 2, dadurch
gekennzeichnet, daß die Derivate der
Cephalosporan- und Penicillansäuren der Formeln (I) und
(II) die folgenden sind: Amoxicillintrihydrat,
Ampicillintrihydrat, Cephalexinmonohydrat,
Cefadroxilmonohydrat, Cephaloglycin und p-
Hydroxycephaloglycin.
