Description
Die Erfindung betrifft den Gegenstand des Patentanspruchs.
Ultraviolettstrahlen führen bekanntermaßen zu verschiedenen
Veränderungen der Haut. Unter dermatologischen Gesichtspunkten
wird eine grobe Einteilung der Ultraviolettstrahlen
auf der Basis der Wellenlängen vorgenommen, und
zwar in eine langwellige Ultraviolettstrahlung von 400
bis 320 nm, eine mittelwellige Ultraviolettstrahlung von
320 bis 290 nm und eine kurzwellige Ultraviolettstrahlung
unter 290 nm. Diese Strahlen werden als UV-A, UV-B
bzw. UV-C bezeichnet.
Eine typische Quelle für UV-Strahlen, denen ein Mensch
ausgesetzt sein kann, stellt das Sonnenlicht dar. Bei
den ultravioletten Strahlen aus dem Sonnenlicht, welche
die Erdoberfläche erreichen können, handelt es sich um
UV-A und UV-B. UV-C wird innerhalb der Ozonschicht absorbiert
und erreicht kaum die Erdoberfläche. Unter den
Ultraviolettstrahlen, welche die Erdoberfläche erreichen,
wirkt UV-B unter Bildung von Erythema und Blasen und
führt zu Hautveränderungen, wie einer beschleunigten
Bildung von Melanin oder einer Pigmentierung. Derartige
Erscheinungen treten auf, falls die Haut mit einer
Lichtmenge bestrahlt wird, welche ein bestimmtes Niveau
übersteigt. Im Gegensatz dazu hat man bisher angenommen,
daß UV-A keine wesentlichen Veränderungen der Haut bewirkt.
In jüngster Zeit wurde jedoch durch Elektronenmikroskopie
sowie durch histochemische Techniken festgestellt,
daß die Haut sogar durch UV-A-Bestrahlung verändert
wird. Es wurde speziell festgestellt, daß die Energie
von UV-A im Unterschied zu UV-B bis ins Innere der
Haut vordringt und zu geringfügigen chronischen Änderungen
bei den elastischen Fasern der Blutgefäßwände und
des Bindegewebes führt. Man nimmt an, daß diese Änderungen
zu einer beschleunigten Alterung der Haut führen. Es
ist ferner bekannt, daß UV-A auch im Sinne einer Melanisierung
der Haut wirkt (zeitlich beschränkte Melanisierung),
und zwar unmittelbar nach Bestrahlung der Haut,
und ferner im Sinne einer Steigerung der degenerativen
Wirkung von UV-B auf die Haut wirkt. Man nimmt daher an,
daß UV-A einer der Faktoren ist, der zur Entwicklung und
Ausbreitung von Muttermalen und Sommersprossen führt.
Es ist somit wichtig, die Haut nicht nur gegenüber UV-B,
sondern auch gegenüber UV-A zu schützen, und zwar unter
dem Gesichtspunkt, die Haut vor einer vorzeitigen Alterung
und der Entwicklung und Ausbreitung von Muttermalen
und Sommersprossen zu bewahren.
Bisher sind jedoch nur wenige Untersuchungen der Wirkung
von UV-A auf die Haut durchgeführt worden, und es
stehen lediglich wenige Substanzen zur Verfügung, welche
in der Lage sind, bei Applikation auf die Haut eine wirksame
Absorption von UV-A zu bewirken. Die bekannten Verbindungen
umfassen Dibenzoylmethan-Derivate und Zimtsäure-Derivate
(DE-OS 27 28 241 und 27 28 243 sowie
JS-OS 51-61641, 52-46056, 57-59840 und 57-197209).
Es besteht somit ein großer Bedarf hinsichtlich der Entwicklung
von Schutzmitteln, welche in der Lage sind,
schädliche UV-A zu absorbieren und die menschliche Haut
in wirksamer Weise vor dieser Strahlung zu schützen. Derartige
Schutzmittel sollten den folgenden Anforderungen
genügen:
- (1) Es hat eine maximale Absorptionskapazität bei einer Wellenlänge im Bereich von 350 nm;
- (2) es hat einen befriedigend hohen molaren Absorptionskoeffizienten bei der obigen Wellenlänge;
- (3) es hat eine geringe Absorptionsfähigkeit für sichtbares Licht, da eine Anfärbung von kosmetischen Mitteln nicht erwünscht ist, d. h. die Absorptionsfähigkeit sollte oberhalb 400 nm nahezu Null sein (ε0);
- (4) es hat eine hohe Stabilität gegenüber Hitze und Licht;
- (5) es hat keine toxischen, reizenden oder andere schädliche Wirkungen gegenüber der Haut;
- (6) es hat eine gute Kompatibilität mit kosmetischen Substraten;
- (7) es wird bei Applikation auf die Haut nicht leicht perkutan absorbiert oder durch Schwitzen entfernt, d. h. seine Wirksamkeit ist dauerhaft.
Bei Untersuchungen haben die Erfinder festgestellt, daß
bestimmte Derivat-Typen des Chalkons, einer bekannten
Pflanzenkomponente, die obigen Anforderungen erfüllen
und somit wirksam zum Schutz der Haut gegenüber UV-A
eingesetzt werden können.
Es hat sich insbesondere gezeigt, daß bereits ein Gehalt von nur
2 Gewichts-% der neuen Verbindungen der unten stehenden Formel
(I) zu einer drastischen Erhöhung des Hautschutzfaktors gegen
UV-A-Strahlen führt. Die kürzeste UV-A-Bestrahlungszeit, die bei
einer nicht behandelten Haut zur Entwicklung von Erythema führt,
wird bei Anwendung eines erfindungsgemäßen Mittels auf mehr als
das 6fache verlängert. Ein Hautschutzfaktor von 6 oder mehr
bedeutet eine hervorragende UV-A-Schutzwirkung. Es ist hierbei zu
berücksichtigen, daß auf dem Markt der Sonnenschutzmittel bereits
Mittel mit einem Lichtschutzfaktor von 4 oder darunter wertvolle
Handelsprodukte darstellen. Derartigen Handelsprodukten sind die
erfindungsgemäßen Erzeugnisse mit einem Lichtschutzfaktor von
etwa 6 bis 8 weit überlegen.
Es ist andererseits kein Stand der Technik bekannt, der die
Lichtschutzwirkung der erfindungsgemäßen Mittel bzw. die erzielbaren
hohen Lichtschutzfaktoren nahelegt. Die Wirksamkeit der
erfindungsgemäßen Mittel und das Ausmaß der Wirksamkeit sind
somit im Hinblick auf den Stand der Technik überraschend.
Aufgabe der vorliegenden Erfindung ist somit die Schaffung eines
Ultraviolett-Absorbers mit hervorragender Schutzwirkung gegenüber
UV-A. Diese Aufgabe wird erfindungsgemäß gelöst durch Ultraviolett-Absorber,
die gekennzeichnet sind durch einen Gehalt von 0,1
bis 20 Gewichts-% eines Chalkon-Derivats der allgemeinen Formel
(I)
wobei R₁ für ein Wasserstoffatom oder eine Methylgruppe
steht und R₂ eine lineare oder verzweigte, aliphatische
Kohlenwasserstoffgruppe mit 2 bis 24 Kohlenstoffatomen
bedeutet.
In Fig. 1 sind als graphische Darstellung die SPF-Werte
der erfindungsgemäßen Produkte 1 bis 7 und der Vergleichsprodukte
1 und 2 dargestellt.
Die neuen Chalkon-Derivate der Erfindung können auf an
sich bekannte Weise hergestellt werden. Man kann insbesondere
gemäß der nachfolgenden Reaktionssequenz 4-Methoxybenzaldehyd
und 4-Hydroxyacetophenon einer Kondensationsreaktion
unterwerfen, um 4-Alkoxy-4′-hydroxychalkon
(II) zu erhalten, gefolgt von einer Acylierung, wobei
ein angestrebtes Chalkon-Derivat (I) erhalten wird.
wobei R₁ und R₂ jeweils die oben angegebene Bedeutung
haben.
Das Ausgangsmaterial Hydroxychalkon (II), wobei R₁ für
eine Methylgruppe steht, ist bekannt [T. Szell et al.,
Can. J. Chem., 42, 2417 (1964); Can J. Chem., 43, 2134
(1965); D. Vorlander, Chem. Ber., 58, 118 (1925); G. Sipos
et al., Chem. Abstr., 59, 5059 (1963)]. Es kann unter
Verwendung von Säuren- oder Basenkatalysatoren in hoher
Ausbeute hergestellt werden.
Die Acylierung des Hydroxychalkons (II) kann durchgeführt
werden, indem man eine herkömmliche Acylierungsreaktion
anwendet. Man kann beispielsweise ein Verfahren
anwenden, bei dem ein korrespondierendes Carbonsäure
anhydrid [(R₂CO)₂O] mit der Verbindung (II) umgesetzt
wird, wobei man Pyridin oder ein Natriumsalz einer
korrespondierenden Carbonsäure einsetzt. Man kann auch
ein Verfahren anwenden, bei dem ein Säurechlorid (R₂COCl)
umgesetzt wird unter Verwendung von Basenkatalysatoren,
welche keine Nucleophilie aufweisen.
Bei der Verbindung der allgemeinen Formel (I) steht R₁
für ein Wasserstoffatom oder eine Methylgruppe. R₂ ist
eine lineare oder verzweigte aliphatische Kohlenwasser
stoffgruppe mit 2 bis 24 und vorzugsweise 4 bis 20 Koh
lenstoffatomen.
Die erfindungsgemäßen Ultraviolett-Absorber werden er
halten, indem man die Verbindung der allgemeinen Formel
(I) bekannten Substraten für Kosmetika auf herkömmliche
Weise zusetzt. Die Absorber können in Form von Cremes,
Lösungen, Ölen, Sprays, Stiften, Emulsionen, Fondcremes
und Salben bereitet werden.
Falls die erfindungsgemäßen Verbindungen mit Substraten
für Salben oder Cremes vermischt werden, erhält man
ölige oder nicht-ölige Anti-Sonnenbrandsalben oder Hautcremes.
In ähnlicher Weise erhält man bei Vermischen
der Verbindung mit Lösungsmitteln (oder gegebenenfalls
Emulgatoren) Anti-Sonnenbrandöl, -lotionen oder Haut
schutzmittel. Die für die obigen Zwecke geeigneten Substrate
oder Lösungsmittel umfassen Kohlenwasserstoffe,
wie feste oder flüssige Paraffine, Kristallöl, Ceresin,
Ozokerit und Montanwachs; Pflanzenöle, tierische Öle
und Fette sowie Wachs, wie Olivenöl, Erdwachs, Karnaubawachs,
Lanolin und Spermöl; aliphatische Säuren
und Ester derselben, wie Stearinsäure, Palmitinsäure,
Ölsäure, Glycerin-monostearat, Glycerin-distearat,
Glycerin-monooleat, Isopropylmyristat, Isopropylstearat und
Butylstearat; sowie Alkohole, wie Ethylalkohol,
Isopropylalkohol, Cetylalkohol, Stearylalkohol,
Palmitylalkohol und Hexyldodecylalkohol. Darüber
hinaus kann man mehrwertige Alkolole, welche als Feucht
haltemittel dienen, wie Glykol, Glycerin oder Sorbit,
ebenfalls verwenden.
Die Menge an Verbindung (I) in dem Ultraviolett-Absorber
kann je nach dem Typ der Präparation variieren und ist
nicht kritisch. Es reicht aus, wenn eine wirksame Menge
der Verbindung (I) vorliegt. Die Menge beträgt im allgemeinen
0,1 bis 20 Gew.-%, vorzugsweise 0,5 bis 10 Gew.-%,
bezogen auf das Mittel.
Die Verbindung (I) kann als alleiniger wirksamer Bestandteil
des erfindungsgemäßen Ultraviolett-Absorbers vorliegen.
Bei der Verwendung als Anti-Sonnenbrand-Kosmetikum
ist es jedoch bevorzugt, andere UV-B-Absorber in Kombination
zu verwenden. Bespiele der UV-B-Absorber umfassen
p-Methylbenzyliden-D,L-campher oder dessen Natriumsulfonat,
Natrium-2-phenylbenzimidazol-5-sulfonat, Natrium-
3,4-dimethylphenylglyoxylat, 4-Phenylbenzophenon, Isooctyl-
4-phenylbenzophenon-2′-carboxylat, p-Methoxycinnamat,
2-Phenyl-5-methylbenzoxazol und p-Dimethylaminobenzoat.
Außer den obigen Bestandteilen können die erfindungsgemäßen
Ultraviolett-Absorber außerdem verschiedene Additive
umfassen. Geeignete Additive sind beispielsweise
W/O- und O/W-Emulgatoren. diese Emulgatoren sind im Handel
erhältlich. Darüber hinaus seien erwähnt: Verdickungsmittel
oder Additive, wie Methylcellulose, Ethylcellulose,
Carboxymethylcellulose, Polyacrylsäure, Tragant,
Agar-Agar und Gelatine. Falls erforderlich, können
Parfüme, Konservierungsstoffe, Feuchthaltemittel, Emul
sionsstabilisatoren, medizinische Wirkstoffe und/oder
physiologisch zulässige Farbstoffe zugesetzt werden.
Im folgenden wird die Erfindung anhand von Untersuchungs
beispielen, Bezugsbeispielen und Beispielen näher erläutert.
Es werden Cremes mit einem Gehalt von 2% der erfindungs
gemäßen Verbindungen verwendet zur Bestimmung eines
Hautschutzeffekts gegenüber UV-A-Bestrahlung. Bei diesem
Test wird eine Creme mit der Formulierung gemäß Beispiel
9 (erfindungsgemäßes Produkt 4) eingesetzt. Ferner
werden Cremes eingesetzt, bei denen das in der Creme-
zusammensetzung von Beispiel 9 verwendete 4-Methoxy-4′-
palmitoyloxychalkon ersetzt wurde durch 4-Methoxy-4′-
methyl-verzweigt.-isostearoyloxychalkon (erfindungsgemäßes
Produkt 1), 4-Methoxy-4′-lineares-stearoyloxychalcon
(erfindungsgemäßes Produkt 2), 4-Methoxy-4′-
oleoyloxychalkon (erfindungsgemäßes Produkt 3), 4-Methoxy-
4′-octanoyloxychalkon (erfindungsgemäßes Produkt 5),
4-Methoxy-4′-hexanoyloxychalkon (erfindungsgemäßes Produkt
6) bzw. 4-Methoxy-4′-propanoyloxychalkon (erfindungsgemäßes
Produkt 7). Der Test wurde gemäß dem Verfahren
von Gschnait et al [Archives of Dermatological Research
263, 181-188 (1978)] durchgeführt. Dabei wurden, genauer
gesagt, Meerschweinchen auf dem Rücken rasiert, um die
Haut freizulegen. Die Empfindlichkeit gegenüber UV-A
wurde gesteigert, indem man den Meerschweinchen 8-Meth
oxypsoralen intracelial verabreichte. Anschließend wurden
die erfindungsgemäßen Cremes 1 bis 7 auf die freigelegte
Haut in einer Menge von 2 mg/cm² appliziert.
15 min nach der Applikation wird die UV-A-Bestrahlung
durchgeführt. 24 h nach der Bestrahlung wird die Haut
daraufhin untersucht, ob sich Erythema entwickelt haben
oder nicht. Die kürzeste UV-A-Bestrahlungszeit, welche
zum Auftreten von Erythema auf der Haut führt, wird be
stimmt. Diese kürzeste Zeit wird verglichen mit der
kürzesten UV-A-Bestrahlungszeit, die zur Entwicklung
von Erythema auf einer nichtbehandelten Haut führt. Gemäß
der folgenden Gleichung wird ein Sonnenschutz-Faktor
(im folgenden als SPF abgekürzt) berechnet. Auf der Basis
dieses Faktors wird der Hautschutzeffekt der jeweiligen
Verbindungen bestimmt. Zu Vergleichszwecken wird
die in Beispiel 9 verwendete Cremebasis allein (Ver
gleichsprodukt 1) sowie Vaseline (Vergleichsprodukt 2)
verwendet.
Die Ergebnisse dieses Tests zeigen, daß die Cremebasis
allein und Vaseline die Haut vor ultraviolettem Licht
nicht schützen können. Andererseits zeigen die Cremes
mit einem Gehalt an 2% der erfindungsgemäßen Verbindungen
SPF-Werte von etwa 6 bis 8 und schützen somit die
Haut wirksam gegen UV-A-Licht.
In 5 l einerEthanollösung, umfassend 500 g (3,67 Mol)
Anisaldehyd und 500 g (3,67 Mol) 4-Hydroxyacetophenon,
tropft man eine wäßrige Lösung (600 ml) von 250 g Natrium
hydroxid ein, und zwar während etwa 80 min in einem
Stickstoffstrom. Anschließend wird das Gemisch auf 50°C
erhitzt und 24 h gerührt. Das Reaktionsgemisch wird auf
Zimmertemperatur abgekühlt. Es werden 2 l 12%ige Chlor
wasserstoffsäure zugesetzt und dann 4 l Wasser. Danach
scheiden sich Kristalle ab. Die Kristalle werden durch
Filtration gesammelt und rasch mit Ethanol gewaschen.
Anschließend werden sie unter vermindertem Druck getrocknet,
wobei man 750 g der angestrebten Verbindung erhält
(2,95 Mol, Ausbeute 80%). Die Verbindung wird aus Ethanol
umkristallisiert. Man erhält hellgelbe, prismatische,
gereinigte Kristalle. Der Schmelzpunkt und verschiedene
spektrale Daten der Verbindung stimmen mit denen
einer Referenzverbindung überein; Fp. 189,5°C (188
bis 190°C bei der Referenzverbindung).
10,17 g (0,04 Mol) 4-Methoxy-4′-hydroxychalkon, erhalten
in Bezugsbeispiel 1, und 6,33 g (0,080 Mol) Pyridin werden
in 100 ml Methylenchlorid aufgelöst, das zuvor mit
Calciumchlorid getrocknet wurde. Dazu tropft man eine ge
trocknete Methylenchlorid(20 ml)-Lösung von Methyl-verzweigt.-
isostearoylchlorid innerhalb von 10 min unter
Eiskühlung und unter Rühren. Das Gemisch wird über Nacht
bei Zimmertemperatur stehengelassen und 20 min refluxiert,
um die Reaktion zu vervollständigen. Das Reaktionsgemisch
wird in eiskaltes Wasser gegossen, dem konz. Chlorwasser
stoffsäure zugesetzt wurde, um das Gemisch sauer zu machen
(pH=1). Anschließend wird mit Chloroform extrahiert.
Die Chloroformphase wird mit einer gesättigten,
wäßrigen Natriumbicarbonatlösung gewaschen, nachfolgend
mit wasserfreiem Natriumsulfat getrocknet und das Lö
sungsmittel durch Destillation unter vermindertem Druck
entfernt. Man erhält die angestrebte Verbindung im wesentlichen
quantitativ. Der Schmelzpunkt und die spektralen
Daten der Verbindung sind in den Tabellen 1 und 2
angegeben.
Das Verfahren des Beispiels 1 wird wiederholt. Anstelle
des Methyl-verzweigt.-isostearoylchlorids verwemdet man
jedoch Propionylchlorid, Hexanoylchlorid, Octanoylchlorid,
Palmitoylchlorid, Stearoylchlorid bzw. Oleoylchlorid,
wobei man die jeweils angestrebten Verbindungen
erhält. Die Schmelzpunkte und verschiedene
spektrale Daten dieser Verbindungen sind in den Tabellen
1 und 2 angegeben.
Eine Masse der folgenden Rezeptur wird auf herkömmliche
Weise verarbeitet, um eine Creme vom O/W-Typ zu erhalten.

- Ultraviolett-Absorber, gekennzeichnet durch einen Gehalt von 0,1 bis 20 Gewichts-% eines Chalkon-Derivats der allgemeinen Formel (I) wobei R₁ für ein Wasserstoffatom oder eine Methylgruppe steht und R₂ eine lineare oder verzweigte, aliphatische Kohlenwasserstoffgruppe mit 2 bis 24 Kohlenstoffatomen bedeutet.
