Die vorliegende Erfindung besieht sich auf ein Verfahren
zur Herstellung von Verbindungen der allgemeinen Formel

worin R Wasserstoff, nied.Alkyl, Trichloräthyl, Benzyl,
p-Nitrobenzyl, Acetoxymethyl, Pivaloyloxymethyl oder
Phthalidyl oder eine Gruppe der Formel -CH(CH3)OCOOC2H5
bedeutet und R1 Hydroxymethyl, Formyl, Mercaptomethyl,
Aminomethyl oder eine Gruppe der Formel -CH2OCOR2 #
-CH2OR3, -COOR2, -CH3SR4 oder -CH2NHCOR2 darstellt, wobei
R„ nied.Alkyl, Aryl oder eine heterocyclische Gruppe,
R_ nied.Alkyl, Benzyl oder Trityl und R. einen fünf- oder
sechsgliedrigen heterocyclischen Ring mit einem oder mehreren Heteroatomen bedeuten, und η Null oder 1 ist.
Verbindungen
der Formel(I) sind starke irreversible Inhibitoren von
verschiedenen ß-Lactamasen, was durch eine erhebliche Verminderung des MIC-Wertes von Ampicillin gegen ß-lactamaseproduzierende
Bakterien gezeigt wurde. Sie besitzen auch antibakterielle Wirksamkeit.
Gemäß der vorliegandcn Erfindung wird 4-Acetoxya*etidin-2-on
(II; diese und die folgenden Formeln sind im später folgenden Formelschema gezeigt) [K. Clauss, D. Grimm und
G. Prossel, Justus Liebigs Arm.Chem. 539 (1974)], von dem
bekannt ist, daß es i'i Stellung 4 mittels schwacher Basen
oder Säuren, wie Thiolen, Thioenolen oder Carbinolen, leicht

Tetrahedron Lett. 1978/ 4059] mit einem ß-Thioketoester
der Formel R 'CHn-C(CH)=CHCOOA't, wobei R ' eine Gruppe der
Formel^CH2OCOR2, CH2OR3, CCOR3 oder HH0SR4 darstellL und
R?, R_ und R4 die oL^n angegebene Bedeutung haben, in Anwesenheit
einer Base,- wie Natriumbicarbonat, Natriumcarbonat, Kaliumcarbonat, Natriumhydroxyd, Ka<
inmhydroxyd oder i'riäthylamiii,
in einem geeigneten Lösungsmittel, wie Wasser, Tetrahydrofuran, Dimethylformamid, Aceton, Hexamethylenphosphoramid
oder einer Mischung hievon, behandelt. Die auf diese Weise erhaltene Verbindung der Formel (III) wird mit einem
geeigneten Derivat von Glyoxylsäure der Formel CHOCOOR1,
worin R1 nied.Alkyl, Trichloräthyl, Benzyl, p-Nitrobenzyl,
Benzhydryl, Acetoxymethyl, Phthalidyl, Pivaloyloxymethyl, Trimethylsilyl oder Dimethyl-tert.butylsilyl oder eine
Gruppe der Formel CH(CH3)OCOOC2H5 bedeutet, entweder bei
einer Temperatur von 40 bis 100 C oder bei Umgebungstemperatur in Anwesenheit einer Base, wie Triäthylamin oder Pyridin,
kondensiert.
Die darauffolgende Chlorierung der erhaltenen diastereoisomeren Carbinole der Formel (IV), vorzugsweise unter Verwendung
von Thionylchlorid und einer Base, wie Pyridin, bei einer Temperatur von -78 bis 00C, ergibt die Chloride der
Formel (V), die mittels Triphenylphosphin und einer Base, wie Pyridin oder 2,6-Lutidin, bei einer Temperatur von etwa
25 bis 500C in die Phosphorylide der Formel (VI) übergeführt
werden.
Die vorsichtige Ozonolyse einer Verbindung der Formel
(VI) in Anwesenheit einer Säure, wie Trifluoressigsäure, als Schutz der Ylidgruppe ergibt nach Wiederherstellung des
Ylid-Teiles mit einer schwachen wässerigen Base das Phospho- .
ran der Formel (VII). ...
Andererseits ergibt die Ozonolyse der Chloride der Formel (V) den Chlorthioestor der Formel (VIII), der mittels
Triphenylphosphin und einer Base in das Ylid der Formel (VII) Übergeführt wird.
Schließlich wird das Ylid (VII) einfach durch Erhitzen

auf eine Temperatur von 30 bis 140 C in einem inerten Lösungsmittel,
wie Benzol, Toluol oder Xylol, cyclisiert.
Verbindungen der Formel (I), worin iv Wasserstoff, R,
Hydroxymethyi, Formyl, Mercaptomethyi oder Aminomethyl und
η 1 bedeuten, werden einfach durch Entfernen der verschiedenen
Schutzrjruppen R„, R^ und R., durch Reduktion der
Gruppe -COOR2, durch Hydrolyse der Gruppe COOR' und durch
Oxydation des S-Ato/ns erhalten.
Beispielsweise führt die vorsichtige selektive Reduktion der α,β-ungesättigten Estergruppe von Verbindungen (Ia,
R.J = COOC2H5) zu der Hydroxymethylgruppe (Ia, R. - CII9OH)
wie im Clavulansäur (P.H. Bentley, P.D. Berry, G. Brooks, M.L. Gilpin, E. Hunt und I.I. Zomaya, J.C.S.Chem.
Comm. 1977, 748).
Eine zusätzliche Bestrahlung von (Ia, R. = CH3OH) in
trockenem Benzol ermöglicht den E/Z-Austausch.
Die exocyclische (Ia) oder endocyclische (Ib) Stellung
der Doppelbindung hängt von der Elektronenanziehungsfähigkeit der Gruppen COOR und R1 ab, die mit den entsprechenden Kräften
des Ringsystems zusammenwirkt.

Die folgenden Beispiele sollen dio vorliegende Erfindung
näher erläutern, ohne daß diese hierauf beschränkt sein soll. In den in den Beispielen aufscheinenden Formeln bedeutet
Me Methyl, Ät Äthyl, Ph Phenyl und Bz Benzyl.
Beispiel 1: Diäthyl-3-mercapto-2-pentcin-1,5-dioat

Zu 170 ml abs.Äthanol, das mit wasserfreiem Chlorwasserstoff
bei 0°C (2 bis 2 1/2 h) gründlich gesättigt worden war, wurden 15 ml Diäthyl-ß-ketoglutarat zugesetzt.
Die Lösung wurde auf -30 C gekühlt und mit Schwefelwasserstoff gesättigt. Der Reaktionsbehälter wurde dicht verschlossen
und 48 h bei Raumtemperatur gehalten. Die Reaktionsmischung wurde in eine Natriumchloridlösung gegossen und gründlich
mit einer 1:1 Mischung von Diäthyläther und Hexan extrahiert.
Die organische Schicht wurde mit einer Natriumchloridlösung bis zur Neutralität gewaschen, auf wasserfreiem Natriumsulfat
getrocknet und im Vakuum eingedampft, wobei ein Rückstand erhalten wurde, der durch kurze Säulenchromatographie
auf Silikagel gereinigt wurde, wobei mit 5 % Äthylacetat:Hexan eluiert wurde.

Zu einer Mischung von 936 mg (7,23 mMol) 4-Acetcxyazetidin-2-on
und 1,58 g (7,25 mMol) Diäthyl-3-mercapto-2-penten-1,5-dioat
in 60 ml Aceton und 30 ml Wasser wurden 670 im, (8 mMol) Natriumbicarbonat untex Pühren bei Raumtemperatur
zugesetzt. Nach 1 h weiteren Rührens wurde die Lösung mit Natriumchlorid gesättigt, mit verdünnter Salzsäure angesäuert
und dreimal mit Äthylacetat »extrahiert. Pie voreinigten Extrakte wurden mit Natriurachloridiösung bis zur Neutralität
gewaschen, über wasserfreiem Natriumsulfat getrocknet und im Vakuum eingedampft, wobei ein öliger Rückstand erhalten
wurde. SäulencLrematographia über Silikagel ergab 1f94 g
(6,75 mMol, 93 %) Produkt als farbloses öl. NMR (CDCl3, 6): 1,27 (611, t, J= 7 Hz); 3,00 (1H, dq, J1 =

Beispiel 3: Methy1-2-[(41R,S)-4'-(1"-äthoxycarbony
lmethyl-2"-äthoxycarbonylvinylthio)-2'-oxo-azetidin-1'-yl]-2-hydroxyacetate

Zu 926 mg (3,2 mMol) (4R,S)-4-(1'-Äthoxycarbonylmethyl·
2'-äthoxycarbonylvinylthio)-a2etidin-2-on und 0,450 ml
(3,2 mMol) Triäthylamin, gelöst in wasserfreiem Tetrahydrofuran, wurde ein molekularer Überschuß an Methylglyoxylat

tropfenweise zugesetzt.· Die Reakcioncmischung wurde in Anwesenheit
eines Molekularsiebe bei Raumtemperatur 3 h lang
gerührt. Dann wurde die Rciakvionsmir^hung mit A'thylacetat
verdünnt, rvi t gesättigter Natriumbicarbonatlösung und ui>.t
Natriumchloridlösnng gewaschen, über wasserfreiem Natriumsulfat
getrocknet und im Vakuum eingedampft.
Die Behandlung dec öliqen Rückstandes mit Diäthylather
ergab eines der beiden Diastereoisomeren in Form weißer
Kristalle (560 mg)'.

des anderen kristallinen Diastereoisomeren. NMR (CDCl3, δ): 1,27 (6H, t, J = 7 Hz); 3,14 (1H, dd, J1 =
15 Hz, J2 = 3 Hz); 3,59 (1H, dd, J1 = 15 Hz,
J3 = 5 Hz); 3,82 (3H, s); 4,12 (2H, q, J =
7 Hz); 4,13 (2H, q, J = 7 Hz); 4,48 (1H, d, J = 6 Hz); 5,27 (1H, dd, J9 = 3 Hz, J, =
5 Hz); 5,49 (1H, d, J -- 6 Hz] ; 5,88 (1H, s) Die Gesamtausbeute an den beiden Carbinolen betrug 88 %.
Beispiel 4: Methyl-2-[(4'R,S)-4'-(1"-äthoxycarbonylmethyl-2"-äthoxycarbonylvinylthio)-2'-oxoazetidin-1'-yl]-2-chloraceitate.

Eine Lösung von 491 mg (1,25 mMol) Methyl-2-[ (4'R,S}-4 '-(1"-äthoxycarbonylmethyl-2"-äthoxycarbonylvinylthio)-2'-oxoazetidin-1'-yl]-chloracetaten
in 80 ml Dichlormethan wurde auf -78 C abgekühlt und ozonisiert. Die erhaltene blaue Lösung wurde mit Natriummetabisulfit (2 h bei Raumtemperatur)
gerührt, dann mit Wasser gewaschen und über wasserfreiem Natriumsulfat getrocknet. Das Lösungsmittel wurde im
Vakuum abgedampft und der erhaltene Rückstand durch präparative Dünnschichtchromacographie gereinigt. Es wurden 165 ml
(0,51 mMol) wasserklares Oi isoliert. NMR (CDCl3, δ): 1,30 (311, s, J = 7 Hz); 3,00, 3,23 und 3,45

gelassen wurde. Nach 1 h weiteren Rühreis wurde die
Suspension abfiltriert und das Filtrat mit gesättigter Natriumbicarbonatiösung
und mit Wasser gewaschen und auf v/assrrfreiem Natriumsulfat getrcck.net. Das !lösungsmittel wurde abgedampft,
wobei in quantitativer Ausbeate die im Ti^l genannte
Verbindung erhalten wurde.

Andererseits wurde die im Titel genannte Verbindung aus Methyl-2-[(41R,S)-4'-äthoxycarbonylacetylthio-2'-oxoazetidin-1
'-yj]-2-chloracetaten gemäß einem ähnlichen Verfahren,
wie in Beispiel 6 beschrieben, erhalten.

Produkte in 84 %iger Ausbeute erhalten.

Zu einer gerührten Lösung von 4,425 g (20,3 mMol) Diäthyl-3-mercapto-2-penten-1,5-dioat
in 70 ml wasserfreiem Tetrahydrofuran, auf -30 C gekühlt, wurden 920 mg (25,5 mMol)
Lithiumaluminiumhydrid in kleinen Portionen unter Stickstoff zugesetzt. Die erhaltene grünliche Aufschlämmung wurde
nach 1 h weiteren Rührens mit 1n Salzsäure zersetzt und dreimal mit Diäthyläther extrahiert. Die vereinigten Extrakte
wurden dreimal mit Natriumchloridlösung gewaschen, auf wasserfreiem Natriumsulfat getrocknet und im Vakuum eingedampft,
wobei ein Rückstand erhalten wurde, der durch Säulenchromatographie über Silikagel (Eluieren mit 20 - 25 %
Äthylacetat-Hexan) gereinigt wurde.
NMR (CDCl3, 6): 1,30 (3H, t, J = 7 Hz); 2,60 (2H + 111, br.t,

Eine Mischung von 1.-01 g (4,13 mMol) (4R,S)-4-(1 ' Hydroxyäthyl-2'-äthoxycarbonylvinylthio)-azetidin-2-on,
1 ml (10,6 mMol) Acetanhydrid und 0,7 ml (5 mMol) Triäthylamin, gelöst in 60 ml Dichlormethan, wurde über Nacht bei
Raumtemperatur stehen gelassen. Die Reaktionsmischung wurde dann dreimal mit einer Natriumchloridlösung gewaschen und
über wasserfreiem Natriumsulfat getrocknet. Bei Abdampfen des Lösungsmittels verblieb ein Rückstand, der nach Säuienchromatc
graphie über Silikagel (Elution mit 10 % fithylacetat:Benzol)
9bO mg (3,3 i niMol, 81 %) eines öligen Produktes ergab.

Eine Mischung von 2,392 g (8,35 mMol) (4R,S)-4-(1 ' Mthoxycarbonylmethyl-2'-äthoxycarbonylvinylthio)-azetidin-2-on
und 4 g Acetoxymethylglyoxylat (frisch hergestellt durch Ozonolyse des entsprechenden Fumarats) in 80 ml Benzol
wurde 4 h durch eine Dean-Stark-Vorrichtung am Rückfluß gehalten. Die gekühlte ReaKticnsi.ischung wurde auf eine Silikagelsäule
gegossen. Das Eluieren mit 10 % Äthylacetat-Benzol ergab 3,10 g (7,15 mMol, 88,2 %) der im Titel genannten
Verbindung als Diastereoisomorenmischung.

Zu 611 mg (1,4 mHol) der DiasLcrcoisomeren.iiischuug
von Acetoxymethyl -2-[(4'R,S)-4'-(1"-äthoxycarbonylmethyl-2"-äthoxycarbonyj
vinylthio) -2 ' -oxccizetidin- 1 ' -yl] -2-h^· Jroxyacetaten
und 0,30 ml Pyridin, gelöst in wasserfreiem Tetrahydrofuran, warden 2,& ipMoJ Thionylchlorid, gelöst in 10 ml
Tetrahydrofuran, tropfenweise bei -30 C unter Stickstoff zugesetzt. Nach 20 min Rühren wurde die Reaktionsmischung
in Wasser gegossen und dreimal mit Äthylacetat extrahiert. Die kombinierten Extrakte wurden viermal mit einer Natriumchloridlösung
gewaschen und über wasserfreiem Natriumsulfat getrocknet. Bei Abdampfen des Lösungsmittels im Vakuum wurden
die diastereoisomeren Chloride als gelbliches Öl erhalten (563 mg, 1,24 mMol, 89 % Ausbeute).

3,03, 3,32 und 3,60 (2H, m); 3,85 und 3,87 (2H, s); 4,17 und 4,19 (4H, q, J = 7 Hz);

gelöst in 20 ml Toluol, wurden 4 h unter Stickstoff am Rückfluß gehalten. Die gekühlte Lösung wurde
auf eine Silikagelsäule gegostieu. Bei Eluieren mit 5 %
Äthylacetat:Benzol wurden 250 mg (0,78 mMol, 71 %) eines
öligen Produktes erhalten.

Claims (1)
- Patentansprüche( \j Verfahren zur Herstellung von Verbindungen der r.llgemeinen Formel, (DCOORworin R Wasserstoff, nied.Alkyl, Trichloräthyl, Benzyl, p-Nitrobenzyl, Acetoxymethyl, Pivaloyloxymethyl, Phthalidyl oder eine Gruppe der Formel -CH(CH3)OCOOC2H5 bedeutet, R1 Hydroxymethyl, Formyl, Mercaptomethyl, Aminomethyl oder eine Gruppe der Formel -CH2OCOR2, -CH2OR3, -COOR2, -CH2SR4 oder -CH2NHCOr2 darstellt, wobei R2 nied.Alkyl, Aryl oder eine heterocyclische Gruppe, R3 nied.Alkyl, Benzyl oder Trityl und R. einen fünf- oder sechsgliedrigen heterocyclischen Ring mit einem oder mehreren Heteroatomen bedeuten, und η Null oder 1 ist, dadurch gekennzeichnet, daß man das 4-Acetoxyazetidin-2-on der FormelOAc(ID0-mit einem ß-Thioketoe^ter der allgemeinen FormelR1 1CH2-C(SH)=CHCOOC2H5 ,worin R1' eine Gruppe der Formel CH2QCOR2, CH3OR3, COOR3 oder CH^SR. ist, wobei R3, R3 und R. die obige Bedeutung haben, in Anwesenheit einer Base zu einer Verbindung der allgemeinen Formel030043/0764, (III)COOC2H5worin R1' die obige Bedeutung hat, kondensiert, welche dann mit einer Verbindung der allgemeinen FormalCHO.COOR1 ,worin R1 nied.Alkyl, Trichloräthyl, Ber.zyi, p-Nitrobenzyl, Benzhydryl, Acetoxymethyl, Pivaloyloxymethyl, Phthalidyl, Trimethylsilyl oder Dimethyl-tert.butylsilyl oder eine Gruppe der Formel -CH(CH-)OCOOC H bedeutet, zu einer Verbindung der allgemeinen Formel, (TV)OOC2H5COOR' worin R' und R1' die obige Bedeutung haben, kondensiert wird, worauf diese Verbindung in das entsprechende Chlorderivat der allgemeinen Formel(V)COOR1 übergeführt wird, das dann030043/0764a) mittels Triphenylphosphin in das Fhocphorylid der allgemeinen Formel, (VI)COOK'worin Ph Phenyl bedeutet und R1 und R ' die obige BedeuLung haben, umgewandelt wird, welches durch Ozonolyse in Anwesenheit einer Säure in das Phosphoran der allgemeinen Formel/ (VIl)worin R1, R1 1 und Ph die obige Bedeutung haben, übergeführt wird, das dann durch Erhitzen zur Verbindung der allgemeinen FormelCOOR', (I1)worin R1 und R^1 die obige Bedeutung haben, cyclisiert wird,b) zu einer Verbindung der allgemeinen Formel030043/0764, (VIII)COCR1worin R1 und R ' die obige Bedeutung haben ozonisiert welche dann mittels' Triphenylphosphin in das Ylid d-3r Formel (VII) umgewandelt wird, das durch Erhitzen zu einer Verbindung der allgemeinen Formel (I1) cyclisiert w^rd, worauf gegebenenfalls die verschiedenen Schutzgruppen R-f Rn und R. entfernt werden oder die Gruppe COOR„ reduziert wird oder die Gruppe COOR1 hydrolysiert wird oder das S-Atorn oxydiert wird, um eine Verbindung der allgemeinen Formel (I) zu erhalten, in der die exocyclische oder endocyclische Stellung der Doppelbindung von der Elektronenanziehungsfähigkeit der Gruppen COOR und R. abhängt.030043/0764
