Description
Die vorliegende Erfindung betrifft Geldanamycinderivate
und Antitumormittel mit einem Gehalt derselben als
Wirkstoff.
Geldanamycin ist das Antiprotozoen-Antibiotikum, das von
dem Stamm Streptomyces hydroscopicus var. Geldanus var. nova
[Journal of Antibiotics, Band 23, Seite 442, (1970)] produziert
wird. Es hat die folgende Formel
[Journal of the American Chemical Society, Band 92, Seite
7591 (1970)].
Aufgabe der vorliegenden Erfindung ist es, neue Antitumormittel
zu schaffen.
Diese Aufgabe wird erfindungsgemäß gelöst durch
die Geldanamycinderivate gemäß den in Anspruch 1 definierten Formeln
(I), (II), und (III) und Mittel mit einem Gehalt
derselben.
Die Geldanamycinderivate der Formel (I) können dadurch
hergestellt werden, daß man Geldanamycin mit einem Amin
der Formel
umsetzt, wobei R⁷ oder R⁸
eine gesättigte Alkylgruppe mit wenigstens 2 Kohlenstoffatomen,
eine ungesättigte Alkylgruppe, eine substituierte
Niederalkylgruppe, die mit einem Halogenatom, einer Hydroxy-, Adamantyl- Amino-,
Morpholino-, Piperazino-, Pyridinyl, Pyrrolidino- oder Methoxygruppe substituiert ist,
eine Cycloalkylgruppe oder eine Aralkylgruppe
bedeutet und der andere der beiden Reste R⁷ und R⁸ ein
Wasserstoffatom bedeutet und wobei R⁷ und R⁸ unter
Bildung einer Tetramethylengruppe verbunden sein können.
Typische Gruppen, die als R⁷ oder R⁸ in Frage kommen,
umfassen Äthyl, Propyl, Butyl, Pentyl, Hexyl, Heptyl,
Octyl, Decyl, Dodecyl, Allyl oder substituierte Niederalkylgruppen
(substituiert mit Hydroxy, Amino, Adamantyl,
Pyrrolidino, Pyridinyl, Methoxy, Piperazino,
Morpholino, oder mit einem
Halogenatom); oder Cyclopropyl, Cyclopentyl, Cyclohexyl,
Cycloheptyl, Cyclooctyl, Benzyl, Phenäthyl.
Typische Amine umfassen Äthylamin, Propylamin, Butylamin,
Pentylamin, Hexylamin, Heptylamin, Octylamin, Decylamin,
Dodecylamin, Allylamin, β-Hydroxyäthylamin, β-Chloräthylamin,
Aminobutylamin, Adamantylmethylamin
Cyclopropylamin, Cyclopentylamin, Cyclohexylamin,
Cycloheptylamim, Benzylamin, Phenäthylamin,
Pyrrolidin, Aminoäthylamin,
β-Hydroxyäthylamin, β-Morpholinoäthylamin,
β-Piperazinoäthylamin, 2′-, oder 3′- oder 4′-Picolylamin,
β-Pyrrolidinoäthylamin, 2′-Pyridinyläthylamin,
β-Methoxyäthylamin.
Methylamin kann unter speziellen Bedingungen verwendet werden.
Die Umsetzung von Geldanamycin mit einem Amin kann in Gegenwart
eines organischen Lösungsmittels durchgeführt werden.
Geeignete organische Lösungsmittel umfassen Alkohole,
chlorierte Kohlenwasserstoffe, insbesondere Chloroform,
Dichloräthan und Methanol. Das Lösungsmittel kann eine
Mischung aus zwei oder mehreren Lösungsmitteln sein. Die
bei der Umsetzung eingesetzten Komponenten können in einem
angestrebten Verhältnis kombiniert werden, z. B. in
einem äquimolaren Verhältnis oder im Überschuß. Vorzugsweise
werden 1 bis 50 Mol-Äquiv. Amin/Mol Geldanamycin
eingesetzt. Die Umsetzung wird gewöhnlich 1 bis 48 Stunden
bei Zimmertemperatur durchgeführt.
Mit einem Überschuß des Diniederalkylamins bei der Umsetzung
wird R¹ teilweise
in die korrespondierenden Dialkylaminogruppe überführt,
obwohl R¹ teilweise als Methoxygruppe verbleibt.
Geldanamycinderivate der Formel (II) können durch Methylierung
von Geldanamycin erhalten werden. Geeignete Methylierungsmittel
umfassen Methylhalogeniede, wie Methylchlorid,
Methylbromid und Methyljodid. Die Methylierung wird dadurch
durchgeführt, daß man Geldanamycin in einem organischen
Lösungsmittel, wie Chloroform, Methanol oder einer
Mischung derselben, auflöst und einen Überschuß, insbesondere
die 4- bis 6fache Menge, an Silberoxid als Suspension
zu der Lösung gibt und anschließend die 8- bis 10fache
Menge Methylhalogenid zusetzt und das Ganze vermischt. Im
allgemeinen läuft die Umsetzung bei Zimmertemperatur innerhalb
von 3 bis 24 Stunden vollständig ab.
Bei der Umsetzung können verschiedene Verbindungen der Formel
(II) gleichzeitig erhalten werden.
Geldanamycinderivate der Formel (III) können durch eine
Epoxidierung erhalten werden.
Die Verbindung (III) kann durch Epoxidierung mit einem
Oxidationsmittel in einem organischen Lösungsmittel, wie
Chloroform, Benzol oder einer Mischung derselben, erhalten
werden.
Geeignete Oxidationsmittel umfassen organische oder anorganische
Peroxide, wie Peressigsäure, Perbenzoesäure,
Chlorperbenzoesäure und Perphthalsäure; oder Alkylhydroperoxide,
wie t-Butylhydroperoxid oder Wasserstoffperoxid.
Es kann ein Überschuß des Oxidationsmittels,
vorzugsweise 1,1 bis 1,5 Mol-Äquiv./Geldanamycin eingesetzt
werden. Falls ein Hydroperoxid verwendet wird, wird
der Reaktionsmischung vorzugsweise eine katalytische Menge
Vanadium(IV)-oxyacetylacetonat einverleibt. Falls Wasserstoffperoxid
verwendet wird, werden vorzugsweise
2 Mol Wasserstoffperoxid mit 1 Mol Geldanamycin in Gegenwart
von 4 Mol p-Chlorphenylisocyanat umgesetzt. Die Umsetzung
wird 2 bis 80 Stunden bei Zimmertemperatur bis
zum Siedepunkt der Mischung durchgeführt.
Bei der Epoxidierung von Geldanamycin mittels des Peroxids
wird nur die Doppelbindung in der 8- und 9-Stellung
epoxidiert. Die Isolierung und Reinigung der angestrebten
Verbindung kann mittels herkömmlicher Verfahren durchgeführt
werden.
Bei Verwendung des Amins (IV) wird das überschüssige Amin
durch Waschen mit einer verdünnten Mineralsäure entfernt.
Die Lösung wird konzentriert und bei 35 bis 45°C unter
vermindertem Druck getrocknet. Der Rückstand wird durch
Chromatographie oder Umkristallisation gereinigt.
Geldanamycinderivate der Formel (I) sind kristalline Verbindungen
(rot, rötlich-violett, braun, blau). Die Verbindungen
sind in Wasser unlöslich, lösen sich jedoch in
organischen Lösungsmitteln, wie Methanol, Äthanol, Aceton,
Äthylacetat, halogenierten Kohlenwasserstoffen, Tetrahydrofuran,
Dioxan, Dimethylsulfoxid, Dimethylformamid
und Pyridin.
Geldanamycinderivate (I) werden durch ein Reduktionsmittel,
wie schweflige Säure, Dithionit, Zink-Essigsäure oder
Ascorbinsäure, reduziert. Sie nehmen dabei eine blaßgelbe
oder weiße Färbung an. Die reduzierten Produkte werden jedoch
leicht durch Kontaktieren mit Luft oder einem Oxidationsmittel
oxidiert, wobei sie wieder farbig werden und
wobei wieder die Geldanamycinderivate (I) gebildet werden.
Falls die Methylierung durchgeführt wird, um die Geldanamycinderivate
der Formel (II) zu erhalten, können
Isolierung und Reinigung des angestrebten Produktes folgendermaßen
durchgeführt werden. Unlösliches Silberjodid
und überschüssiges Silberoxid werden abgetrennt, das Filtrat
wird unter vermindertem Druck zur Trockene eingeengt
und der Rückstand wird durch Chromatographie oder durch
Umkristallisieren gereinigt. Geldanamycinderivate (II)
sind kristalline Verbindungen (gelb oder gelblich-braun),
die sich nicht in Wasser, jedoch in einem der genannten
organischen Lösungsmittel lösen.
Falls eine Oxidation durchgeführt wird, um die Geldanamycinderivate
der Formel (III) zu erhalten, können Isolierung
und Reinigung des angestrebten Produktes aus der Reaktionsmischung
folgendermaßen durchgeführt werden. Der
Überschuß an Oxidationsmittel wird durch Waschen mit einer
5%igen wäßrigen Natriumsulfitlösung entfernt, die Lösung
wird über wasserfreiem Natriumsulfat entwässert und
das Filtrat wird unter vermindertem Druck zur Trockene
eingeengt. Der Rückstand wird mittels Chromatographie oder
durch Umkristallisieren gereinigt. Geldanamycinderivate
(III) sind kristalline Verbindungen (gelblich-braun oder
gelb), die sich nicht in Wasser, jedoch in den genannten
organischen Lösungsmitteln lösen.
Die erfindungsgemäßen Geldanamycinderivate (I), (II)
oder (III) zeigen einen signifikanten Wachstumsinhibitionseffekt
gegenüber der Krebszelle W-2K-11, die als
Modell für Krebszellen bekannt ist. Die erfindungsgemäßen Geldanamycinderivate
sind daher als Antitumormittel brauchbar.
Man kann eine oder mehrere Arten der erfindungsgemäßen
Geldanamycinderivate (I), (II) oder (III) als Antitumorarzneimittel
verabreichen.
Vorzugsweise wird der Wirkstoff mit einem geeigneten Hilfststoff
oder einem Zusatzstoff kombiniert, um pharmakologische
Mittel zu erhalten, die zur oralen Verabreichung
oder zur nicht-oralen Verabreichung geeignet sind. Die
Hilfsstoffe und Zusatzstoffe können folgende organische
oder anorganische Feststoffe oder Flüssigkeiten sein.
Geeignete Hilfsstoffe umfassen Wasser, Gelatine, Lactose,
Stärke, Calcium, Carboxymethylcellulose, mikrokristalline
Cellulose, Stearylalkohole, Magnesiumstearat, Talkum,
Pflanzenöl, Benzylalkohol, Propylenglykol, Kautschuk,
Polyalkylenglykol, Kerosin, Gelee und Cholesterin. Geeignete
Additive umfassen Konservierungsstoffe, Benetzungsmittel,
Emulgatoren, Lösungsbeschleuniger, den osmotischen
Druck einstellende Salze, Puffer, Bindemittel, Suspensionsmittel
und Dispersionsmittel.
Die pharmakologischen Mittel können als Puffer, Granulat,
Kapseln, Pastillen, Tabletten, mit Zucker überzogene
Tabletten, als Injektionsmittel, Suppositorien und Salben
vorliegen. Diese Darreichungsformen der Mittel können nach
herkömmlichen Verfahren hergestellt werden.
Das erfindungsgemäße Antitumor-Arzneimittel kann bei der
Humantherapie eingesetzt werden, jedoch in gleicher Form
auch als Tierarzneimittel verwendet werden. Bei der
Therapie unter Verwendung des erfindungsgemäßen Antitumor-
Arzneimittels liegt die Dosis an Antitumormittel bei
nicht-oraler Verabreichung (Injektion) gewöhnlich in einem
Bereich von 0,5 bis 80 mg/kg, vorzugsweise 1 bis
40 mg/kg, und bei oraler Verabreichung in einem Bereich
von 1 bis 100 mg/kg, vorzugsweise 2 bis 50 mg/kg.
Im folgenden wird die Erfindung anhand von Beispielen und
Vergleichsbeispielen näher erläutert.
280 mg Geldanamycin werden in 50 ml Chloroform aufgelöst.
Anschließend gibt man 1 ml n-Propylamin zu. Die Mischung
wird 1 h bei Zimmertemperatur gerührt dann in 50 ml kaltes
Wasser gegossen. Dann wird der pH-Wert mit 6N HCl auf 3
eingestellt. Die Chloroformschicht wird abgetrennt und
die Wasserschicht wird mit 50 ml Chloroform extrahiert.
Beide Chloroformschichten werden vereinigt und über wasserfreiem
Natriumsulfat 2 h entwässert. Das Chloroform
wird unter vermindertem Druck abdestilliert. Es bleibt ein
rötlich-violettes, festes Produkt zurück. Das feste Produkt
wird aus Aceton-n-Hexan umkristallisiert; man erhält
286 mg (Ausbeute 98%) 17-n-Propylamino-geldanamycin als
rötlich-violette, nadelförmige Kristalle, Fp. 143 bis
145°C.

560 mg Geldanamycin werden in 100 ml Chloroform aufgelöst
und anschließend werden 2 ml Äthylenimin zugesetzt. Die
Mischung wird 21 h bei Zimmertemperatur gerührt und dann
in 100 ml kaltes Wasser gegossen. Der pH-Wert wird mit 6 N
HCl auf 3 eingestellt. Die Chloroformschicht wird abgetrennt.
Die Wasserschicht wird mit 100 ml Chloroform extrahiert.
Beide Chloroformschichten werden vereinigt, über
wasserfreiem Natriumsulfat entwässert und unter vermindertem
Druck bis zur Trockene eingeengt. Man erhält ein
rotes, öliges Produkt. Das ölige Produkt wird durch Silikagel-
Säulenchromatographie mit 25% Methanol-Chloroform
gereinigt. Die angestrebte Fraktion wird mittels eines
Fraktionssammlers aufgefangen, unter vermindertem Druck
bis zur Trockne eingeengt und das Produkt wird aus
Aceton-n-Hexan umkristallisiert; man erhält 530 mg (Ausbeute
93%) 17-Äthylenimino-geldanamycin als rötlich-
orangefarbene Kristalle, Fp. 261 bis 262°C (Zers.)

Die Verfahren des Beispiels 1 oder des Bezugsbeispiels 2 werden wiederholt.
Dabei erhält man die in Tabelle 1 aufgeführten Verbindungen.
5 g Geldanamycin werden in 200 ml Chloroform und 200 ml
Methanol gelöst und dann mit 20 g Silberoxid versetzt.
Anschließend werden 40 g Methyljodid unter Rühren zugegeben
und die Mischung wird 5 h gerührt. Die unlöslichen
Materialien werden abfiltriert und das Filtrat wird bei
vermindertem Druck und bei 40°C bis zur Trockene eingeengt.
Das resultierende, rötlich-orangefarbene, ölige
Produkt wird mittels Silikagel-Säulenchromatographie mit
2,5% Methanol-Chloroform gereinigt. Die Fraktionen (42.
bis 60.) werden gesammelt und bis zur Trockene eingeengt.
Umkristallisation aus Äther-n-Hexan liefert 600 mg 1-0-Methyl-geldanamycin
in Form gelblich-oranger Kristalle,
Fp. 213 bis 215°C.

Die durch Silikagel-Säulenchromatographie bei dem Verfahren
von Beispiel 40 erhaltenen Fraktionen (3. bis 12.)
werden gesammelt und bis zur Trockene eingeengt. Das Produkt
wird durch Chromatographie mit 2% Methanol-Chloroform
gereinigt, um die 1. bis 40. Fraktion zu sammeln, und die
Fraktionen werden aus Aceton-n-Hexan kristallisiert; man
erhält 192 mg 7-OCONH-methyl-geldanamycin in Form gelber,
nadelförmiger Kristalle, Fp. 230 bis 232°C.

Die durch die zweite Gelsäulenchromatographie bei dem Verfahren
von Bezugs-Beispiel 41 erhaltenen Fraktionen (61. bis 82.)
werden gesammelt und aus Chloroform-n-Hexan umkristallisiert;
man erhält 334 mg 1-O-Methyl-7-OCONH-methyl-geldanamycin
in Form gelber Kristalle, Fp. 119 bis 121°C.

5,6 g Geldanamycin werden in 800 ml 20%igem Benzol-Chloroform
gelöst und dann mit 1,9 g m-Chlorperbenzoesäure versetzt.
Die Mischung wird 5 h bei Zimmertemperatur gerührt.
Das Reaktionsgemisch wird mit 5%iger wäßriger Natriumsulfitlösung,
mit Wasser und mit gesättigter, wäßriger Natriumchloridlösung
gewaschen und über wasserfreiem Natriumsulfat
entwässert und dann bei vermindertem Druck bis zur
Trockene eingeengt. Der resultierende, orange-rote, ölige
Rückstand wird durch Silikagel-Säulenchromatographie mit
3% Methanol-Chloroform gereinigt. Die angestrebten Fraktionen
werden gesamt, bei vermindertem Druck bis zur
Trockene eingeengt und aus Äther-n-Hexan umkristallisiert;
man erhält 4,84 g 8,9-Epoxy-geldanamycin in Form gelblich-
orangefarbener Kristalle, Fp. 148 bis 149°C.

1,4 g Geldanamycin werden in 1000 ml Benzol gelöst und
mit einer katalytischen Menge an Vanadium(IV)-oxyacetylacetonat
versetzt. Die Mischung wird auf einem Ölbad unter
Rückfluß erhitzt und während des Refluxierens werden 360 mg
eines 70%igen t-Butylhydroperoxids zugegeben. Nach 6stündigem
Refluxieren wird die Reaktionsmischung auf Zimmertemperatur
abgekühlt, mit 0,1 HCl, mit einer 5%igen wäßrigen
Natriumsulfitlösung, mit Wasser und mit einer gesättigten,
wäßrigen Natriumchloridlösung gewaschen, über wasserfreiem
Natriumsulfat entwässert und unter vermindertem Druck bis
zur Trockene eingeengt. Der resultierende Rückstand wird
durch Säulenchromatographie an Silikagel mit 2,5% Methanol-
Chloroform gereinigt. Die angestrebten Fraktionen werden
aufgefangen und bis zur Trockene eingeengt. Der Rückstand
wird aus Äther umkristallisiert; man erhält 280 mg 8,9-Epoxy-geldanamycin
in Form gelblich-orangefarbener Kristalle.
Durch Dünnschichtchromatographie an Silikagel und durch
Vergleich der Schmelzpunkte und der IR-Spektren wird sichergestellt,
daß das Produkt das gleiche wie das von
Beispiel 43 ist.
2500 g des jeweiligen Produktes, 1375 g Lactose, 775 g
mikrokristalline Cellulose und 375 g Calciumcarboxymethylcellulose
werden mit einem 16 Maschen/2,5 cm-Sieb (Taylor)
gesiebt und einheitlich vermischt. Die Mischung wird in
einen Kneter gegeben und es werden 3 l einer 3%igen Lösung
von Hydroxypropylcellulose (Isopropylalkohol: Wasser=3 : 7)
zugesetzt und die Mischung wird geknetet. Aus der Mischung
wird mittels einer Strangpresse-Granulierungsmaschine ein
Granulat hergestellt, und dieses wird 6 h bei 50°C luftgetrocknet.
Das Granulat wird auf eine Teilchengröße im Bereich
von 16 bis 60 Maschen/2,5 cm (Taylor) gebracht. Dem
Granulat wird in einem Verhältnis von 0,3 Gew.-% Magnesiumstearat
zugemischt und die Mischung wird zu Tabletten verarbeitet.
Aus Mäusenierenzellen durch ein cancerogenes Virus abgeleitetes
Fibroplast C3H-2K-Klon wird in Krebszellen W-2K-11
überführt. Die Krebszellen W-2K-11 werden mittels folgendem
Verfahren kultiviert.
9,4 g Eagle′s MEM-Kulturmedium (hergestellt von Nissui
Seiyaku K.K.) werden in 900 ml destilliertem Wasser aufgelöst.
Die Lösung wird 15 min bei hohem Druck bei 120°C
sterilisiert und abgekühlt. 100 ml Kalbserum und 3 bis
5 ml einer 10%igen wäßrigen Natriumcarbonatlösung werden
zur Einstellung des pH-Wertes auf 7,1 bis 7,2 zu der
Lösung gegeben. 10 ml einer wäßrigen L-Glutaminlösung
(2,92 g/100 ml), die durch ein milliporöses Filter filtriert
wurden, werden unmittelbar vor Verwendung der Lösung
als Kulturmedium zugesetzt.
Die in einer Gefriertruhe bei -80°C gelagerten Krebszellen
W-2K-11 werden bei Zimmertemperatur geschmolzen
und 5 min bei 670 g mit einem Zentrifugenseparator behandelt.
Die niedergeschlagenen Zellen werden in 50 ml des
Kulturmediums (1) dispergiert. Die Suspension wird in eine
Roux-Flasche gegeben und bei 37°C kultiviert. Die Vermehrung
der Zellen wird initiiert und nach 3 bis 4 Tagen
beendet. Die kultivierte Mischung wird dekantiert, und
es werden 10 ml einer 0,2%igen Trypsinlösung zugegeben.
Die Mischung wird 2 bis 3 min bei Zimmertemperatur gehalten,
die Trypsinmischung wird dekantiert und anschließend
werden 50 ml des Kulturmediums (1) zugegeben, um eine Zellsuspension
herzustellen.
1,8 ml der Zellsuspension (2) werden in jede Petrischale
gegeben und in einem Kohlendioxidgas-Inkubator (5% CO₂;
95% Luft) bei 37°C angeimpft. 24 h nach dem Animpfen werden
jeweils 0,2 ml einer Äthanollösung jeder Probe zugegegen
und die Kultivierung dann fortgesetzt. 48 h nach
Zugabe der Probe wird die Zahl der überlebenden Zellen
unter einem Mikroskop gezählt und jeweils die prozentuale
Hemmung der Zellvermehrung berechnet. Die Ergebnisse sind
in Tabelle 2 aufgeführt.
Bei jeder Probe wird der LD₅₀-Wert bei Mäusen bestimmt.
Jede Probe wird intraperitoneal injiziert. Die Ergebnisse
sind in Tabelle 3 aufgeführt.

Claims (2)
1. Geldanamycinderivate der folgenden Formel
wobei R¹ eine gesättigte Alkylaminogruppe mit wenigstens
2 Kohlenstoffatomen, eine ungesättigte Alkylaminogruppe,
eine substituierte Niederalkylaminogruppe, die mit einem
Halogenatom, einer Hydroxy-, Adamantyl-, Amino-, Morpholino-,
Piperazino-, Pyridinyl-, Pyrrolidino- oder
Methoxygruppe substituiert ist, eine Cycloalkylaminogruppe,
eine Pyrrolidinogruppe oder eine Aralkylaminogruppe bedeutet;
oder der Formel wobei R³ ein Wasserstoffatom oder eine Methylgruppe bedeutet;
oder der Formel
oder der Formel wobei R³ ein Wasserstoffatom oder eine Methylgruppe bedeutet;
oder der Formel
2. Antitumormittel, gekennzeichnet durch Geldanamycinderivate
der Formel (I), (II) oder (III) als Wirkstoff.
