5 De uitvinding heeft betrekking op een werkwijze ter bereiding van een geneesmiddel met een anti tumor-werking. De uitvinding heeft verder betrekking op het in het geneesmiddel te gebruiken geldanamycine-derivaat als actieve component en op de aldus gevormde geneesmiddelen. Ook heeft de uitvinding betrekking op een werkwijze ter bereiding van de geneeskrachtige 10 verbindingen.
Geldanamycine is een antiprotozoaan antibiotisch middel dat bereid wordt door streptomyces hygroscopicus var. geldanus var. novasoort ( Journal of Antibiotics Vol. 23, bladzijde 442(1970)) en deze stof komt overeen met formule 5, aangegeven op het formuleblad en is vermeld in Journal of American 15 Society Vol. 92, Bladzijde 7591 (1970 ).
Een van de doelstellingen volgens de uitvinding is het verkrijgen van nieuwe antitumor-geneesmiddelen.
Dit wordt bewerkstelligd door een werkwijze volgens de uitvinding ter bereiding van een geneesmiddel met een antitumor-werking, welke werk-20 wijze hierdoor wordt gekenmerkt dat men nieuwe geldanamycine-derivaten met formule 1, waarbij R1 een verzadigde alkylaminogroep voorstelt met tem minste 2 koolstof atomen, een onverzadigde alkylaminogroep, een gesubstitueerde lagere alkylaminogroep, cycloalkylamino, pyrrolidino of een aralkylaminogroep of 25 een methoxygroep, p R is een waterstofatoom, een halogeenatoom of een lagere alkylaminogroep, 1 2 wanneer R methoxygroep is dan is R een halogeenatoom of een lagere alkylaminogroep of derivaten met formule 2, aangegeven op het formule-30 blad, waarbij 3 R een groep is met formule 6 of 7, 4 ς 6 R , R en R respectievelijk voorstellen een waterstofatoom of 4 5 6 een methylgroep en ten minste een van ‘de groepen R ,R en R een methyl- groep is of een derivaat met formule 3 aangegeven op het formuleblad
Wanneer in de verbinding met formule 1, aangegeven op het formule-1 2 blad, R methoxygroep is dan is R bij voorkeur een halogeenatoom of een mono- of di- lagere alkylaminogroep.

De geldanamycine-derivaten met formule 1, waarbij R en R de bete- 800085? « * ' «* -2- 21093/Vk/mv kenis hebben zoals boven is vermeld worden hierbij gebruikt als actieve component in het geneesmiddel, en een voorbeeld van een dergelijk gelda- namycine-derivaat is 8,9-epoxygeldanamycine.
De geldananmycine derivaten met formule 1 kunnen worden bereid 5 door geldanamycine in reactie te brengen met een amine met formule 4, aan- 7 8 gegeven op formuleblad, bij R en R respectievelijk voorstellen een water- .
stofatoom of een verzadigde alkylgroep met tem minste 2 koolstofatomen, een onverzadigde alkylgroep, een gesubstitueerde lagere alkylgroep, een cyclo- 7 8 alkylgroep of een aralkylgroep en zowel R' als R kunnen niet een waterstof-7 8 10 atoom zijn en R en R . kunnen samen gebonden een alkyleengroep vormen.

Voorbeelden van de groepen R of R zijn ethyl, propyl, butyl, pentyl, hexyl, octyl, decyl, dodecyl, allyl of gesubstitueerde lagere alkyl-groepen (gesubstitueerd met hydroxyde, amino, methylaraino, pyrrolidino, pyridinyl, alkoxy, piperadino, morfolino, cycloalkyl of hydroxyalkoxy- 15 groep of een halogeenatoom); of cyclopropyl, cyclopentyl,cyclohexyl 7 8 cycloheptyl, cyclooctyl, benzyl, fenethylgroep. Wanneer R en R samen-gebonden zijn kunnen ze een etheen, tetramethyleen, pentamethyleen.of hexa-•methyleengroep vormen.
Voorbeelden van de amines zijn ethylaraine, propylamine, butyl-20 amine, pentylamine, hexylamine, heptylamine, octylmine, decylamine,, dode-cylamine, allylamine, fy -hydroxyethylamine, ^-chloorethylamine, β-glycoxy-ethylamine, aminobutylamine, adamanthylmethylamine, cyclopropylamine, cyclopen tylamine,cyclohexylamine, cycloheptylamine, benzylamine, fenethylamine, ethyleenimine, pyrrolidine, piperidine, dimethylamine, aminoethylamine, 25 -hydroxyethylamine, diglycolamine, £-morfolinoethylamine, £>-piperadino- ethylamine, 2’-, of 3’-, of 4'-picolylamine,.^-pyrrolidinoethylamine, 2’-py piridinylmethylamine,^methoxyethylamine, $ -N-methylaminoethylamine. Methyl-amine kan onder speciale omstandigheden worden toegepast.
De reactie van geldanamycine met een amine kan worden uitgevoerd 30 in aanwezigheid van eem organisch oplosmiddel.
Geschikte organische oplosmiddelen zijn alkoholen, gechloreerde koolwaterstoffen meti narae chloroform,dichloorethaan en methanol. Het oplosmiddel kan ook een mengsel zijn van. twee of meer oplosmiddelen.
De verbindingen die toegepast worden in de reactie kunnen gecombineerd 35 worden op een gewenste verhouding zoals een» equimolaire verhouding of een overmaat hiervan. Het verdient de voorkeur om 1-50 mol equivalent amine te gebruiken per mol geldanamycine. De reactie wordt gewoonlijk uitgevoerd bij kamertemperatuur gedurende 1-48 uren.

Wanneer een overmaat amine gebruikt wordt onder stringente ora- 1 2 standigheden worden zowel R als R gedeeltelijk omgezet tot de corresponderende aminogroepen. Wanneer een overmaat di-lager lakylamine gebruikt 1 2 wordt bij de reactie worden zowel R als R gedeeltelijk omgezet tot de cor- *1 5 responderende dialkylaminogroepen, hoewel R. gedeeltelijk behouden blijft als me thoxy groep.
De geldanamycine-derivaten met formule 1, waarbij R een methoxygroep 2 is en R een halogeenatoom zoals Br, J, Cl of F,· met name Br, J of Cl, kunnen worden verkregen door halogenering vanf geldanamycine. De halogenering 10 wordt uitgevoerd onder toepassing van een halogeneringsmiddel zoals pyri-dinebromide ,perbromide, jodium-pyridine en lithiumchloride en dergelijke, in een oplosmiddel zoals ethanol-chloroform mengsel, pyridene, ethylace-taat of methylethylketon bij een temperatuur van 0-100°C gedurende 1-10 uren.
15 Het geldanamycine-derivaat met de fonnule 2 kan worden verkregen door methylering van geldanamycine.
Geschikte methyleringsmiddelen omvatten methylhaliden zoals methyl-chloride, methylbromide en methyljodide. De methylering wordt uitgevoerd door geldanamycine op te lossen in een organisch oplosmiddel zoals chlo-20 roform, methanol of een mengsel hiervan en het toevoegen van een overmaat met name 4-6 keren zilveroxyde aan de oplossing in de vorm van een suspensie en het vervolgens toevoeren van 8-10 keren een overmaat aan methyl-halide en deze stoffen te mengen. In het algemeen wordt de reactie bewerkstelligd bij kamertemperatuur gedurende 3-24 uren.
25 Bij de reactie kunnen verschillende verbindingen gelijktijdig worden verkregen met formule 1. Het geldanamycine-derivaat met formule 3 kan worden verkregen door een epoxydering De verbinding met fonnule 2 kan worden verkregen door een epoxydevorming met een oxydatiemiddel in een organisch oplosmiddel zoals chloroform, benzeen of eèn mengsel hiervan.
30 Geschikte oxydatiemiddelen omvatten organische of anorganische peroxyden zoals perazijnzuur, perbenzoëzuur, chloorperbenzoëzuur, perfthaal-zuur en dergelijke of alkylhalide-peroxyden zoals t-butylhydroperoxyde of waters tofperoxyde.
Als overmaat aan oxydatiemiddel kan bij voorkeur 1,1-1,5 mol· equi-35 valenten per geldanamycine worden toegepast. Wanneer hydroperoxy-de wordt gebruiktfverdient het de voorkeur om een katalytische hoeveelheid vanadium(IV)oxyacetylacetonaat toe te voegen.Wanneer waterstofperoxydé wordt gebruikt verdient het de voorkeur om 2 mol waterstofperoxyde te doen .8 000357 ·&» "~” -4- 21093/Vk/mv reageren met 1 mol geldanamycine in aanwezigheid van 4 mol para-chloorfenyl isocyanaat. De reactie wordt uitgevoerd bij kamertemperatuur tot het kookpunt van het mengdel gedurende 2-80 uren.
Bij de epoxydatie van geldanamycine door het peroxyde wordt alleen 5 de dubbele binding bij de plaatsen 8 en 9 geëpoxydeerd.
De afscheiding en de zuivering van de gewenste verbinding kan op conventionele wijze worden uitgevoerd.
Wanneer het amine met formule 4, vermeld op het formuleblad, wordt gebruikt, wordt de overmaat aan amine verwijderd door wassen met een verdund 10 mineraal zuur. De oplossing wordt ingedampt en gedroogd bij 35-45°C onder verlaagde druk en de rest wordt chromatografisch gezuiverd of door het uitvoeren van een herkristallisatie.
De geldanamycine-derivaten met formule 1 zijn kristallijne verbindingen (rood, rood-violet, bruin, blauw),zijn niet oplosbaar in water maar 15 wel oplosbaar in een organisch oplosmiddel zoals methanol, ethanol, aceton, ethylacetaat, gehalogeneerde koolwaterstof, tetrahydrofuran, dioxaan, dime thylsulfoxyde, dimethylformamide en pyridine.
De geldanamycine-derivaten met formule 1 worden gereduceerd met een reductiemiddel zoals zwavelwaterstof, dithioniet, zink-azijnzuur of ascor-20 binezuur zodat een lichtgele of witte stof wordt verkregen, maar de gereduceerde producten worden gemakkelijk geoxydeerd door contact met lucht of met een oxydatiemiddel zodat ze gekleurd worden en de geldanamycine-derivaten met formule 1 vormen.
Wanneer de methylering wordt uitgevoerd ter verkrijging van de gel-25 danamycine-derivaten met formule 2, wordt de afscheiding en de zuivering hiervan uit het reactiemiddel uitgevoerd doorjhet afscheiden van onoplosbaar zilverjodide en overmaat zilveroxyde en het indampen van het filtraat tot'droog‘onder verlaagde druk en het zuiveren van het residu door een chro-matografische bewerking of door een herkristallisatie.
30 De geldanamycine-derivaten met formule 2, vermeld op het formule blad, zijn kristallijne verbindingen (geel of geelbruin) die niet oplosbaar zijn in water maar wel oplosbaar in een organisch oplosmiddel.
Wanneer de oxydatie wordt uitgevoerd ter verkrijging van geldanamycine derivaten met formule 3, vermeld op het formuleblad, wordt het afschei-35 den en zuiveren uit het reactiemengsel bewerkstelligd door het afscheiden van de overmaat van het oxydatiemiddel door wassen met een 5 J/o-waterige oplossing van natriumsulfiet en het drogen van de oplossing over watervrij natriumsulfaat en het indampen van het filtraat tot droog onder verlaagde 8000357 <· # -5- 21093/Vk/mv it druk en het zuiveren van het residu door een chromatografische bewerking of door herkristallisatie.
De geldanamycine-derivaten met formule 2 zijn kristallijne verbindingen (geelbruin of geel) die niet oplosbaar zijn in water maar wel oploste baar in genoemde organische oplosmiddelen.
De nieuwe geldanamycine-derivaten met formule 1, 2 of 3 volgens de uitvinding hebben een significante groei-remmende werking op kanker- . cellenW-2K-11, hetgeen algemeen bekende kankercellen zijn, en deze nieuwe derivaten zijn dan ook geschikt als antitumor-geneesraiddel.

Een of meer van de geldanamycine-derivaten met formule 1, 2 of 3 aangegeven op het formuleblad, kunnen worden toegediend als geneesmiddel tegen tumor.
Het verdient de voorkeur om de actieve stof te mengen met een geschikte hulpstof of een additief ter vorming van een formacologische samenstel-^ ling die geschikt is voor orale toediening of niet-orale.toediening. De hulpstoffen en toevoegstoffen kunnen , bestaan uit de volgende organische of anorganische vaste stoffen of vloeistoffen.
Geschikte hulpstoffen zijn water, gelatine, lactose, zetmeel, calcium, carboxymethyl-cellulose, microkristallijne cellulose, stearylal-PO
cohol, magnesiumstearaat, talk, plantaardige olie, benzylalcohol, pro-pyleenglycol, rubber, polyalkyleenglycol, kerosine, gel- en cholesterol.
Geschikte additieven omvatten verduurzamingsmiddelen, bevochtigings-middelen, emulgeermiddelen, oplosversnellers, middelen die de osmotische druk regelen in de vorm van zouten, buffers, bindingsmiddelen, suspendeer-^ middelen en dispersiemiddelen.
De farmacologische samenstellingne kunnen verkregen worden in de vorm van een poeder, granule, capsule, pellet, tablet, met suiker-bedekt tablet, injecteervloeistof, zetpil en zalf. Deze samenstellingen kunnen op bekende wijze worden verkregen.
Het antitumor-geneesmiddel volgens de uitvinding kan gebruikt worden bij de mens maar ook als geneesmiddel bij dieren in dezelfde vorm.
Als geneesmiddel wordt het gedoseerd in een dagelijkse hoeveelheid van0,5-80 mg/kg, bij voorkeur 1-40 mg/kg bij een niet-orale toediening (injectie) en. een hoeveelheid van 1-100mg/kg, bij voorkeur 2-50 mg/kg bij een orale toediening.
De uitvinding zal nader worden toegelicht aan de hand van het volgende niet-beperkende voorbeelden.

Voorbeeld I
In 50 ml chloroform werd 280 rag geldanamycine opgelost en vervolgens 1 ral n-propylamine toegevoegd. Het mengsel werd"geroerd gedurende 1 uur bij kamertemperatuur en het mengsel werd overgebracht in 50 ml koud 5 water en vervolgens werd de pH ingesteld op 3 met 6N-HC1. De chloroform-laag werden afgescheiden en de waterlaag werd geëxtraheerd met 50 ral chloroform. De beide chloroformlagen werden samengevoegd en gedroogd over watervrij natriumsulfaat gedurende 2 uren. Chloroform werden afgedestilleerd onder verlaagde druk zodat een rood-violet vast product werd verkregen. Het ”*3 vaste product werd herkristalliseerd uit aceton-n-hexaan ter verkrijging van 286 mg 17-n-propylamino-geldanamycine in de vorm van een rood-violet naaldachtig kristal. De opbrengst komt overeen met 98%. Het smeltpunt bedroeg H3-1A5°C.

Voorbeeld II
In 100 ml chloroform werd 560 mg geldanamycine opgelost en vervolgens werd 2 ml »ethyleeniminetoegevoegd. Het mengsel werd bij kamerterape-25 ratuur geroerd gedurende 21 uren en vervolgens toegevoegd aan 100 ml koud water en de pH werd ingesteld op 3 met 6N-HC1. De chloroformlaag werden afgescheiden. De waterlaag werd geëxtraheerd met 100 ml chloroform. Beide chloroformlagen werden gecombineerd en gedroogd over watervrij natriumsulfaat en ingedampt tot droog onder verlaagde druk ter verkrijging van een 30 rood olie-achtig product. Het olie-achtige product werd gezuiverd over een -kolom met silicagel, zodat een chromatografische zuivering plaats had met 25% methanol-chloroform. De gewenste fractie werd verzameld met behulp van een fractie-verzamelinrichting en verdampt tot droog onder een ver laagde druk en het product werd herkristalliseerd uit aceton-n-hexaan ter 35 verkrijging van 530 mg 17-ethyleeniminogeldanamycine, ha tg. r-r wvervenkort met een opbrengst van 93%, in de vorm van rood-oranje kristallen.
Het smeltpunt onder ontleding bedroeg 261-262°C.

20 Voorbeeld XXXV
In 100 ml chloroform-methanol (3:2), werd 560 mg geldanamycine opgelost en vervolgens 30 ml 50%waterige oplossing van dimethylamine toegevoegd. Het mengsel werd geroerd bij kamertemperatuur gedurende 2 uren en vervolgens toegevoegd aan 100 ml koud water en de pH werd ingesteld op 25 3-4 met 6N-HC1. Het product werd 2 keer geëxtraheerd met chloroform en de geëxtraheerde chloroformlagen werden gewassen met water en gedroogd over watervrij natriumsulfaat en ingedampt tot droog onder verlaagde druk, ter verkrijging van een blauw-olie;-achtig product.
Het olie-achtige product werd kolomchromatografisch gezuiverd over 30 silicagel met 2% methanol-chloroform. De fracties te weten de l4e-32® fractie , werden verzameld en het oplosmiddel werd afgedestilleerd en het product herkristalliseerd uit ether ter verkrijging van 52 ml 17-demetho-xy_17,19-bisdimethylarainogeldanamycine in de vorm van bruine kristallen.
Het smeltpunt bedroeg 135-137°C.

In 60 ml ethanol en 60 ml chloroform werd 1,12 g geldanamycine op-.
gelosUHet mengsel werd geroerd onder afkoelen met ijswater en 960 mg py-30 ridinebromideperbromide werd toegevoegd. Het mengsel werd gedurende 1 uur geroerd. Nade reactie werd het mengsel verdund met een grote hoeveelheid chloroform en herhaaldelijk gewassen met chloroform en met een verzadigde waterige oplossing natriumchloride en gedroogd over watervrij natriumsul-3g faat. De chloroformlaag werd ingedampt tot drrog onder verlaagde druk ter verkrijging van een geel-oranje olie-achtig product. In 20 ml prydine werd 762 mg jodium opgelost en 1,12 geldanamycine.
15 Het mengsel werd geroerd bij kamertemperatuur gedurende 1 uur. Het reactie-mengsel werd verdund met ethylacetaat. De verdunde oplossing werd gewassen met 5% waterige oplossing natriumthiosulfaat, met een 1($ waterige oplossing azijnzuur en verzadigd met een waterige oplossing van natriumchloridé en vervolgens gedroogd over watervrij natriumsulfaat. De oplossing met ethylacetaat 20 -werd ingedampt tot droog onder verlaagde druk ter verkrijging van een

In 60 ml methylethylketon werd 726 mg 19-broomgeldanamycine opgelost en 3,5g lithiumchloride werd toegevoegd.Het mengsel werd gedurende 5 uren gekookt onder terugvloeikoelen onder roeren. Na de reactie werd 6000857 i.
*- -11- 21093/Vk/mv het reactiemengsel afgekoeld tot kamertemperatuur en verdund met een grote hoeveelheid chloroform en gewassen met een 5% waterige oplossing van na-triumthiosulfaat, met water, met een verzadigde waterige oplossing van na-triumchloride en gedroogd over watervrij natriumsulfaat. De chloroform op-5 lossing werd ingedampt tot droog ter verkrijging van een oranje-olieachtig product. Het olieachtige product werd kolomchromatografisch gezuiverd over silicagel met 3% methanol-chloroform en herkristalliseerd uit chloroform-n-hexaan ter verkrijging van 580mg 19-chloorgeldanamycine in de vorm van rode-oranje kristallen.
10 Smeltpunt: 157-159°C

20 Voorbeeld
In 200 ml chloroform en 200 ml methanol werd 5 g geldanamycine opgelost en vervolgens werd 20 g zilveroxyde toegevoegd en daarna 40 g methyl jodide onder roeren en het mengsel werd gedurende 5'uren geroerd. De onoplosbare stoffen werden afgescheiden door fitratie en het filtraat werd 25 ingedampt tot droog onder een verlaagde druk bij 40°C. Het verkregen roodo-ranje olieachtige product werd kolomchromatografisch over silicagel gezuiverd met 2,5% methanol-chloroform. De 42e-60e fractie werden verzameld en ingedampt tot droog eri herkristalliseerd uit ether-n-hexaan ter verkrijging van 600 mg 1-0- methylgeldanamycine in de vorm van geel-oranje 30 kristallen.
Smeltpunt: 213-215°C

^ De fracties 61-82 verkregen door de tweede kolomchromatogra fische gel-zuivering bij ’de werkwijze volgens voorbeeld XLIII werden verzameld en herkristalliseerd uit chloroform-n-hexaan ter verkrijging van 334 mg 1-0-methyl-7-0C0NH-methyl-geldanamycine in de vorm van gele kristallen.

Voorbeeld XLV
In 800 ml 20% benzeen-chloroform werd 5,6 g geldanamycine opgelost en vervolgens werd 1,9 g methachloorbenzoëzuur toegevoegd. Het meng- 8000857 ψ -14- 21093/Vk/my i
sel werd geroerd bij kamertemperatuur gedurende 5 uren. Het reactiemeng-sel werd gewassen met 5% waterige oplossing natriumsulfiet en met water en met een verzadigde waterige oplossing van natriumchloride en gedroogd over watervrij natriumsulfaat en ingedampt tot droog onder verlaagde druk. Het 5 verkregen oranje-rode olieachtige residu werd kolömchromatografisch over silicagel gezuiverd met 3% methanol-chloroform. De gewenste fracties werden verzameld en ingedampt tot droog onder verlaagde druk en herkristalliseerd uit ether-n-hexaan ter verkrijging van 4,84 g 8,9-epoxygeldanamycine in de vorm van geel-oranje kristallen 10 Smeltpunt: 148-149°C

Voorbeeld XLVI
20- In 1000 ral benzeen werd 1,4 g geldanamycine opgelost en een kata lytische hoeveelheid vanadium(IV) oxyacetylacetonaat en het mengsel werd onder terugvloeikoeling gekookt op een oliebad en 360 mg 70% t-butyl-hydroperoxyde werd toegevoegd tijdens het koken onder terugvloeikoeling.
Na 6 uren werd het reactiemengsel afgekoeld tot kamertemperatuur en gewas-25 sen met 0,1 N-HC1, met 5% waterige oplossing natriumsulfiet, met water en met een verzadigde, waterige oplossing van natriumchloride en daarna gedroogd over watervrij natriumsulfaat en ingedampt tot droog onder verlaagde druk. Het verkregen residu werd kolomchromatografisch over silicagel gezuiverd met 2,5% methanol-chloroform. De gewenste fracties werden ver-30 zameld en ingedampt tot droog en herkristalliseerd uit ether ter verkrijging van 280 mg 8,9-epoxygeldanamycine in de vorm van geel-oranje kristallen.
Volgens de gegevens uit de dunne laag chromatografische silicagel bepaling, het smeltpunt en het IR spectrum bleek dat het product gelijk 35 was aan het product dat verkregen is in voorbeeld XLV*
Voorbeeld XLVII
Een preparaat werd bereid door 2500 g van elk product, 1375 g lactose, 775 giricrokristallijne > cellulose en 375g calciumcarboxymethylcel- 8Ό 0 0 8 5 7 -15- 21093/Vk/mv * lulose te zeven over een zeef van 16 mesh (1180 jum) zodat een gelijkmatig mengsel werd verkregen. Het mengsel werd toegevoerd aan een kneder en 3 liter 3% hydroxypropylcellulose in (isopropylalcohol: water =3:7) werd toegevoegd en het mengsel .gekneed. Het mengsel werd gegranuleerd met 5 behulp van èen extrusie-granuleermachine en aan de lucht gedroogd gedurende 6 uren bij een temperatuur van 50°C. De granules werden gezeefd in een fractie van 16-60 mesh ( 250-1180 yum) en magnesiumstearaat werd toege-voegd in een hoeveelheid van 0,3 gewichtsprocent en het mengsel werd getabletteerd ter verkrijging_van tabletten.
10 Daarna werden de volgende experimenten uitgevoerd waarbij kankercel len W-2K-11 werden gebruikt, verkregen door overbrengen van fibroblast C3H-2K verkregen uit de nieren van een.muis met behulp van een kankerogeen-virus. De kankercellen W-2K-11 werden gekweekt volgens de onderstaande methode.
15 1) De bereiding van het cultuurmedium 9,4 g Eagle’s MEM cultuurmedium (bereid door Nissui Seiyaku K.K) werden opgelost in 900 ml gedestilleerd water en de oplossing werd gesterali-seerd onder hoge druk bij 120°C gedurende 15 1 inuten en afgekoeld. 100 ML kalfsserum en 3-5 ml 10% waterige oplossing an natriumbicarbonaat werd 20 toegevoegd aan de oplossing om de pH in te stellen op 7,1-7,2. Vervolgens werd 10 ml van een waterige oplossing van L-glutamine (2,02 g!/100 ml), afgefiltreerd over een.millipore filter,toegevoegd juist voordat de oplossing gebruikt werd als cultuurmedium.
2) De bereiding van de getransplanteerde celoplossing.
25 De kankercellen W-2K-11 werden bewaard in een vrieskast bij -0O°C
en voor gebruik bij kamertemperatuur gesmolten en gecentrifugeerd bij 670g gedurende 5 minuten. De afgescheiden cellen werden gedispergeerd in 50 ml cultuurmedium zoals boven beschreven onder 1). De suspensie werd toegevoerd aan'een Roux-kolf en het kweken had plaats bij een temperatuur van 37°C. Het vermenigvuldigen van de cellen werd gestart en na 3-4 dagen beëindigd. Het gekweekte mengsel werd gedecanteerd en 10 ml 0,2% tryp-sineoplossing werd toegevoegd en het mengsel werd op kamertemperatuur gehouden gedurende 2-3 minuten en het trypsinemengsel werd gedecanteerd en vervolgens werd 50 ml cultuurmedium (1) toegevoegd ter bereiding van een suspensie van cellen.
3) Incubatie van de cellen en het toevoegen van een monster 1,8 til van de celsuspentie (2) werd toegevoegd aan een Petri-schaal en geplaatst in een incubatie-inrichting met kooldioxyde (5% 95% 8000857 i -16- 21093/Vk/mv lucht) bijt 37°C. 24 Uren na het begin van de incubatie werd 0,2 ral van de ethanoloplossing van elk monster toegevoegd en de incubatie werd voortgezet. 48 Uren na de toevoeging van het monster werd.het aantal cellen die niet waren aangetast geteld onder een microscoop en de procentuele 5 remming van' de celverraenigvuldiging werd berekend. De hierbij verkregen resultaten zijn vermeld in tabel B.

Claims (13)
1, Werkwijze ter bereiding van een geneesmiddel met een werking tegen tumoren, met het kenmerk, dat men geldanamycine-derivaten met formule 1, aangegeven op het formuleblad, waarbij ^ R1 is een verzadigde alkylaminogroep, met ten. minste 2 koolstof- atomen, een onverzadigde alkylaminogroep, een gesubstitueerde lagere alkylaminogroep, cycloalkylamino, pyrrolidino of aralkylamino of een methoxy-groep, 0 R is een waterstofatoom halogeenatoom of een lagere alkylammo- 10 groep, 1 2 wanneer R is een methoxygroep dan.is R een halogeenatoom of een lagere alkylaminogroep, of verbindingen met formule 2,aangegeven op het formuleblad, waarbij R een groep is met formule 6 of 7, aangegeven op het formuleblad, 15 en R^, en R^ respectievelijk voorstellenc een waterstofatoom of een 4 5 6 methylgroep en ten minste een van de radicalen R , R en R een methyl groep is, of een verbinding met formule 3, aangegev n op het formuleblad, in een voor geneeskundige doeleinden geschikte toedieningsvorm 20 brengt. 2t Gevormd geneesmiddel met een antitumorwerking, verkregen door toepassing van de werkwijze volgens conclusie 1.
3. Werkwijze ter bereiding van geneeskrachtige verbindingen geschikt voor gebruik bij de werkwijze volgens conclusie 1, met het ken- 25 merk, dat men galdanamycine derivaten met formule 1, 2 of 3 bereidt op een voor analoge, verbindingen bekende wijze.
4. Geldanamycine-derivaten , met het kenmerk, dat dit verbindingen 1 2 3 4 5 6 zijn met formule 1, 2 of 3, waarbij R , R , R , R , R en R de betekenis hebben zoals aangegeven in conclusie 1.
5. Geldanamycine-derivaten volgens conclusie 4, met het kenmerk, dat dit een geldanamycine-derivaat is met formule 1, waarbij R een verzadigde alkylaminogroep is met temminste 2 koolwaters tof atomen, een onverzadigde alkylaminogroep, een gesubstitueerde lagere alkylaminogroep, cycloalkylamino, pyrrolidino, arylalkylamino of een methoxygroep, 35 r2 een waterstofatoom, een halogeenatoom of een lagere alkylamino groep is, 1 2 en wanneer R een methoxygroep is dan is R een halogeenatoom of 8000857 J -20- 21093/Vk/mv een lagere alkylaminogroep. 6’. Geldanamycine derivaten, volgens conclusie 4, met het kenmerk, dat dit derivaten zijn met formule 1, waarbij R een methoxygroep is,en 2 R een halogeenatoom of een lagere alkylaminogroep.
7. Geldanamycine-derivaten, volgens conclusie 4, met het kenmerk, dat dit derivaten zijn met formule 2, aangegeven op het formuleblad, waarbij R·^ een groep voorstelt met formule 6 of 7, aangegeven op het formuleblad, en R^, R^ en R^ respectievelijk voorstellen een waterstofatoom of een 4 5 6 methylgroep en ten ’minste een van de groepen R.,R en R een methylgroep 10 is. ^
8. Geldanamycine derivaten volgens conclusie 4, met het kenmerk, dat dit derivaten zijn met formule 3, aangegeven op het formuleblad.
9. Werkwijze ter bereiding van geldanamycine*derivaten , met het kenmerk, dat derivaten met formule 1 worden bereid door geldanamycine in 15 reactie te brengen met een amine met formule 4, aangegeven op het formu-leblad, waarbij R en R respectievelijk een waterstofatoom voorstellen* of een verzadigde alkylgroep met tem minste 2 koolstofatomen,een onverzadigde alkylgroep, een gesubstitueerde lagere alkylgroep , een cycloal- 7 8 kylgroep of een aralkylgroep en ; R' en R niet gelijktijdig een wa- 7 8 20 terstofatoom kunnen zijn, en R en R samengebonden kunnen zijn ter vorming van een alkyléengroep.
10. Werkwijze volgens conclusie 9> met het kenmerk, dat het amine gekozen is uit ethylamine, propylamine, butylamine, pentylamine, hexylamine, heptylamine, octylamine, decylamine, dodecylamine, allylamine, |8-hydroxy- 25 ethylamine, ^chloorethylamine, ^-glycoxyethylamine, aminobutylamine, adamanthylmethylamine, cyclopropylamine, cyclopentylamine, cyclohexylamine, cycloheptylamine, benzylamine, fenethylamine,ethyleenimine pyrrolidine, piperidine, dimethylamine, amino-ehtylamine,^-hydroxye thylamine, digly-colamine, β-morfolinoethylamino, ^β-piperadinoethyl'amino, 2'-, of 3’-, of 30 4,-picolylamine, β-pyrrolidinoethylamine, 2f-pyridinylethylamine, ^-metho-xyethylamine of ^-N-methylaminoethylamine.
11. Werkwijze volgens conclusie 9> met het kenmerk, dat de reactie tussen het geldanamycine en een amine wordt uitgevoerd in aanwezigheid van een organisch oplosmiddel.
12. Merkwijze volgens conclusie 9, met het kenmerk , dat een overr maat aan lager alkylamine wordt gebruikt bij de reactie zodat zowel R als 2 R gedeeltelijk worden omgezet tot het corresponderende alkylamino ter-1 wijl R gedeeltelijk behouden blijft als methoxygroep. 8000857 -21- 21093/Vk/mv
13, Merkwijze ter bereiding van geldanamycine derivaten, met het kenmerk ,dat derivaten met formule 1, worden verkregen waarbij R een me- Λ thoxygroep is en een halogeenatoom, door het halogeneren van geldana-mycine.
