Meta-Halophenyl-bicyclo[2.2.2 Joctan-Derivate, Verfähren
zu ihrer Herstellung und diese enthaltende Arzneimittel
Die Erfindung betrifft eine neue Klasse von meta-Halophenyl-bicyclo[2.2.2joctan-Derivaten,
die besonders stark auf ..das. zentrale Nervensystem wirken. Die Erfindung betrifft
ebenfalls ein Verfahren zur Herstellung der neuen Derivate
wie auch pharmazeutische Zusammensetzungen, die diese Derivate enthalten.
• In der BE-PS 806 808 der gleichen Anmelderin wird eine Klasse von Phenyl-bicyclo[2.2.2]octanen beschrieben,
von denen berichtet wird, daß sie auf das zentrale Nervensystem wirken.
Es wurde nun überraschenderweise gefunden, daß eine neue kleine Klasse von Verbindungen, die ähnlich wie die
in der zuvor erwähnten belgischen Patentschrift beschriebenen Verbindungen sind, eine unerwartete und unerwartet
bessere Potenz bzw. Wirkung auf das zentrale Nervensystem besitzen. Verglichen mit den in der genannten Patentschrift
beschriebenen Verbindungen zeigen sie insbesondere eine wesentlich verbesserte Antidepressionsaktivität und antipsychotische Aktivität. Die erfindungsgemäßen Verbindungen
besitzen weiterhin sehr gute therapeutische Indices.
Gegenstand der Erfindung sind somit trans-Verbindungen
der Formel (I)

Der Ausdruck "Halogen" bedeutet Fluor, Brom, Chlor oder Jod.
Die - CH2N(Me)2- und m-Halophenyl-Gruppen stehen,
bezogen aufeinander, in trans-Konfiguration.
Die Verbindungen der Formel (i) können in zwei enantiomeren
Formen auftreten, und die Erfindung umfaßt die (+)- und (-)-Formen getrennt und die nichtracemischen Gemische
wie auch die racemischen Gemische der enantiomeren Formen.
Die Säureadditionssalze sind bevorzugt pharmazeutisch annehmbare, nichttoxische Additionssalze mit geeigneten Sau- \
ren, wie mit anorganischen Säuren, z.B. Chlorwasserstoffsäure, !
Bromwasserstoffsäure, Salpetersäure, Schwefelsäure oder Phos- j
phorsäure, oder mit organischen Säuren,wie mit organischen ! Carbonsäuren, z.B. Glykollsäure, Maleinsäure, Hydroxymaleinsäure,
Apfelsäure, Weinsäure, Citronensäure, Salicylsäure, o-Acetoxybenzoesäure, Nicotinsäure und Isonicotinsäure, oder
mit organischen Sulfonsäuren, z.B. Methansulfonsäure, Äthansulfonsäure,
2-Hydroxyäthansulfonsäure, p-Toluol-sulfonsäure
oder Naphthalin-2-sulfonsäure. Außer den pharmazeutisch
annehmbaren Säureadditionssalzen werden von dem Ausdruck "Säureadditionssalze" auch andere Salze, wie z.B. solche mit
Pikrinsäure oder Oxalsäure, umfaßt. Sie können als Zwischenprodukte
für äi.e Reinigung der Verbindungen oder für die Her-

stellung anderer, z.B. pharmazeutisch annehmbarer, Säureadditionssalze
dienen. Sie sind ebenfalls für die Identifi- ' zierung, Charakterisierung oder Reinigung der Basen nützlich.
Eine besonders bevorzugte erfindungsgemäße Verbindung ist N,N-Dimethyl-trans-2-(m-chlorphenyl)-bicyclo[2.2.2]oct-3-yl-methylamin,
beispielsweise in seiner Hydrochloridform.
Die überraschend erhöhte Potenz der neuen Verbindungen der Formel (I) und ihrer Säureadditionssalze kann anhand bekannter
Untersuchungsverfahren, wie dem Antagonismus der Reserpinhypcthermie bei der Maus und der Reserpinkatalepsie
bei der Ratte, gezeigt werden.
Gegenstand der Erfindung sind weiterhin pharmazeutische Zubereitungen, die als aktiven Bestandteile eine Verbindung
der Formel (I) oder ihr pharmazeutisch annehmbares Säureadditionssalz enthalten, zusammen mit einem pharmazeutisch
annehmbaren Träger dafür.
Für den pharmazeutisch annehmbaren geeigneten Träger gibt es keinerlei Beschränkungen«, Es können alle die in der
BE-PS 806 808 beschriebenen Träger und Verdünnungsmittel verwendet werden.
Die Erfindung betrifft weiterhin die Herstellung der Verbindungen der Formel (I) nach an sich bekannten Verfahren,
z.B. nach den in der BE-PS 806 808 beschriebenen Verfahren.
Bei einem bevorzugten Verfahren wird eine Verbindung
der Formel II

worin X die zuvor gegebene Bedeutung besitzt und R Wasserstoff
oder Methyl bedeutet, mono- oder dimethyliert. Die
Art der Reagentien und der sonstigen Bedingungen für diese Umsetzung sind an sich bekannt.
Die folgenden Beispiele erläutern die Erfindung, ohne sie zu beschränken.
Trans-m-Chlorcinnamonitril
210,75 g (1,5 Mol) m-Chlorbenzaldehyd, 127,5 g
(1,5 Mol) Cyanoessigsäure und 150 ml Pyridin werden 2 Tage am Rückfluß erwärmt. Die Lösung wird eingedampft, wobei man
ein Öl erhält, das nach dem Destillieren ein Gemisch der Isomeren (72% trans/23% eis; 5% Aldehyd) ergibt; Ausbeute
129 g (5350, Kp. 70 bis 85°C bei 0,8 mm. Das Material wird in Isopropanol gelöst und kann fraktioniert bei O0C
kristallisieren. Der Feststoff wird filtriert und gefriergetrocknet. Die Ausbeute an reinem trans-Isomer beträgt
15 g (6,1%), Kp. 104 bis 1060C bei 1 mm, Fp. 26 bis 28°C.
Trans-6-(m-Chlorphenyl)-bicyclo[2.2.2]oct-2-en-5-carbonitril
9,2 g (0,056 Mol) trans-m-Chlcrcinnamonitril, hergestellt
wie oben beschrieben, und 8,03 ml (0,084 Mol) 1,3-Cyclohexadien
werden in ein verschlossenes Rohr mit Spuren von Hydrochinon und 1^-Dichlorbenzol gegeben
und 2 Wochen bei 150 bis 16O0C ex*wärmta Eindampfen der
Lösung ergibt ein goldfarbenes Öl (12 g), das mehrere Male

mit kaltem Benzin (40 bis 60°C) und dann wiederholt mit heißem Benzin (60 bis 80°C) gewaschen wird. Die letzteren
Waschlösungen werden gesammelt und zu einem viskosen Öl reduziert (10,2 g; 7596).
Trans-2-(m-Chlorphenyl)-bicyclo[2.2.2]octan-3-carbonitril
9,4 g trans-6-(m-Chlorphenyl)-bicyclo[2.2.2]oct-2-en-5-carbonitril,
hergestellt wie oben beschrieben (0,038 Mol), werden in Äthanol bei Atmosphärendruck in Anwesenheit von
1,0 g 5%igem Palladium-auf-Tierkohle hydriert. Nach Aufnahme
der theoretischen Wasserstoffmenge wird der Katalysator abfiltriert und die Lösung zu einem Öl reduziert (9,1 g;
97%)· Dieses wird durch Auflösen in Äthanol kristallisiert,
Fp. 112 bis 113°C.
Analyse:
Berechnet: C 73,2.2% H 6,56% N 5,7% Cl 14,43%
Gefunden : 73,43 6,41 5,82 14,38.
Trans-2-(m-Chlorphenyl)-bicyclo[2.2.2]oct-3-yl-methalaminhydrοchlorid
8,0 g (0,032 Mol) trans-2-(m-Chlorphenyl)-bicyclo-[2.2.2]octan-3-carbonitril,
hergestellt wie oben beschrieben, in 20 ml trockenem Tetrahydrofuran werden tropfenweise zu
einer gekühlten und gerührten Lösung aus 1,61 g (0,042 Mol) Lithium-aluminiumhydrid in 20 ml trockenem Tetrahydrofuran
gegeben. Nach der Zugabe kann sich die Lösung auf Zimmertemperatur erwärmen und wird über Nacht gerührt. 1,6 ml 5n Natriumhydroxid
und 7 ml Wasser werden vorsichtig zugegeben. Es fällt ein feiner, weißer Niederschlag aus, der abfiltriert
wird. Das Filtrat wird über Magnesiumsulfat getrocknet und nach der Filtration zu einem Öl eingedampft. Beim Auflösen
in äthanolischer Chlorwasserstoffsäure und bei langsamer Zugabe
von Äther fällt das Hydrochloridsalz (6,8 g; 74%) aus;
Fp. 220 bis 222°C.

N, N-Dimethyl-trans-2- (m-chlorphenyl) -Mcyclo [2.2.2]oct-3-yl-methylamin-hydrochlorid
Zu 3,0 g (0,011 Mol) trans-2-(m-Chlorphenyl)-bicyclo[2.2.2]oct-3-yl-methylamin-hydrochlorid,
hergestellt wie oben beschrieben, gibt man 0,92 g (0,011 Mol) Natriumhydrogencarbonat
und 30 ml Dimethylformamid. Der Kolben wird auf O0C gekühlt und ein Gemisch aus 2,29 ml (0,055 Mol)
90%iger Ameisensäure und 3,78 ml (0,055 Mol) 37- bis 40%igem Formaldehyd wird langsam zugegeben. Nach der Zugabe wird
die Lösung langsam auf Rückflußtemperatur erwärmt. Nach 5 Stunden wird die Lösung abgekühlt, zu 80 ml ¥asser gegeben
und mit festem Kaliumhydroxid bis zur alkalischen Reaktion (pH = 8)versetzt. Sie wird dann mit 3 x 30 ml Äthylacetat
extrahiert. Die vereinigte organische Phase wird mit 2 χ 30 ml ¥asser gewaschen, über Magnesiumsulfat getrocknet,
filtriert und das Lösungsmittel wird im Vakuum entfernt; man erhält 2,10 g (61%) eines Öls. Das unter Verwendung von
äthanolischer Chlorwasserstoffsäure gebildete Hydrochloridsalz
und die Titelverbindung werden aus Äthanol und Äther umkristallisiert. Man erhält 2,1 g, Fp. 217 bis 219°C
Analyse:
Berechnet: C 64,96% H 8,01% N 4,45% Cl 22,56% Gefunden : 64,67 7,82 4,44 22,64.
In den folgenden Beispielen werden pharmazeutische Zubereitungen, die Verbindungen der Formell, (i) enthalten,
näher erläutert. Als aktiver Bestandteil wird N,N-Dimethyltrans-2-(m-chlorphenyl)-bicyclo[2.2.2]o
ct-3-yl-methylaminhydrochlorid verwendet. Diese Verbindung kann jedoch durch
andere aktive feste erfindungsgemäße Verbindungen ersetzt
werden.

Tabletten, die je 10 mg aktiven Bestandteil enthalten,
werden folgendermaßen hergestellt:
Aktiver Bestandteil Kartoffelstärke Lactose

Magnesiumstearat Talk
insgesamt 50,0 mg
Die Stärke, Lactose und der aktive Bestandteil werden durch ein Sieb gegeben und gut vermischt. Die Lösung aus
Polyvinylpyrrolidon wird mit dem entstehenden Gemisch vermischt und das Gemisch wird durch ein Sieb mit einer lichten
Maschenweite von 1,405 mm (Nr. 12 mesh B.S.) gegeben. Die so gebildeten Granulate v/erden bei etwa 55°C getrocknet und
durch ein Sieb mit einer lichten Maschenweite von 1,003 mm (Nr. 16 mesh B.S.) gesiebt. Magnesiumstearat, Natriumstärkeglykolat
und Talk, die man zuvor durch ein Sieb mit einer lichten Maschenweite von 0,25 mm (Nr. 60 mesh B.S0) gesiebt
hatte, werden dann zu dem Granulat gegeben. Nach dem Vermischen wird das Gemisch auf einer Tablettenvorrichtung verpreßt.
Man erhält Tabletten, die je 50 mg wiegen.
Kapseln, die je 15 mg Medikament enthalten, werden folgendermaßen hergestellt:

Die Lactose, Stärke, Magnesiumstearat und der aktive
Bestandteile werden durch ein Sieb mit einer lichten Maschenweite von 0,35 mm(Nr. 44 mesh B0S.) durchgesiebt und in Hartgelatinekapseln
in 100 mg-Mengen gefüllt.
Suppositorien, die je 30 mg aktiven Bestandteil enthalten,
werden folgendermaßen hergestellt:
Aktiver Bestandteil 30 mg
gesättigte Fettsäureglyceride bis zu 2 000 mg
Die aktiven Bestandteile werden durch ein Sieb mit einer lichten Maschenweite von 0,25 mm (Nr. 60 mesh B.S.)
gegeben und in gesättigten Fettsäureglyceriden, die zuvor
unter so wenig Wärme wie möglich geschmolzen wurden, suspendiert. Das Gemisch wird in eine Suppositorenform mit nominal
2 g Kapazitit gegossen und kann dann abkühlen.

Verfahren zur Herstellung einer trans-Verbindung der Formel (i)CH9N(Me).(Dworin X Halogen bedeutet,oder ihres Säureadditionssalzes, dadurch g e k e η η zeichnet , daß man eine trans-Verbindung der Formel (II)609 8 86/1208(II)X Halogen undR Wasserstoff oder Methyl bedeuten, mono- oder dimethyliert.
- 5· Verfahren nach Anspruch 4, dadurch gekennzeichnet, daß X Chlor bedeutet.
- 6· Trans-Verbindung der Formel (II)(II)worin X Halogen und R Wasserstoff oder Methyl bedeuten.
- 7. Verbindung nach Anspruch 6, dadurch gekennzeichnet, daß X Chlor bedeutet.
- 8. Pharmazeutische Zubereitung, enthaltend als aktiven Bestandteil eine Verbindung der Formel (I) nach mindestens einem der Ansprüche 1 bis 3 oder ihr pharmazeutisch annehmbares Additionssalz sowie gegebenenfalls pharmazeutisch annehmbare Träger und/oder Verdünnungsmittel dafür.
- 9. Pharmazeutische Zubereitung nach Anspruch 8, dadurch gekennzeichnet , daß sie in Tabletten-, Kapseloder Suppositorienform vorliegt.609 8 8 6/1208
