Prostaglandine sind eine Gruppe von Naturstoffen, die aus verschiedenen Tiergeweben isoliert wurden, !n
Säubern sind sie für eine Vielzahl von physiologischen Wirkungen verantwortlich. Die natürlichen Prostaglandine
besitzen ein Kohlenstoffgerüst von im allgemeinen 20 C-Atomen und unterscheiden sich vornehmlich
durch den Mehr- oder Mindergehalt an Hydroxylgruppen uz\/. Doppelbindungen im Cyclopentanring (bezüglich
der Struktur und Wirkung von Prostaglandinen siehe unter anderem M. F. Cuthbert »The Prostaglandins,
Pharmacological and Therapeutic Advances«, William Heinemann Medical Books LTD, London 1973).
Die Synthesen von nicht natürlich vorkommenden Analoga von Prostansäuren, in denen die Vielzahl der
pharmakologischcn Wirkungen der natürlichen Prostansäuren differenziert sind, gewinnt zunehmend an
Bedeutung.
Die vorliegende F.rfindung betrifft neue, nicht natürlich vorkommende Analoga von Prostansäuren der Formal

welche sowohl die optisch aktiven Verbindungen der natürlichen Konfiguration als auch die niccmischcn Verbindungen
umliil.it und in welcher X einen gesättigten, ver/.weigten Alkylenresl mit I 5 C-Atomen und K die
Melhy I-, Äthyl-, l'ropyl- oder I sob u ty !gruppe bedeuten, sowie ihre physiologisch verträglichen Salze mit organischen
und anorganischen Basen und ihre Alkylestcr mit 1-8 C-Atomen im Esterteil.
Die Erfindung betrifft weiterhin ein Verfahren zur Herstellung der neuen, nicht natürlich vorkommenden
Analoga von Prostansäuren der Formel I1 ihre physiologisch verträglichen Salze mit organischen und anorganischen
Basen und ihre Alkylester mit 1 -8 Kohlenstoffatomen im Esterteil sowie pharmazeutische Präparate, die
diese Wirkstoffe enthalten.
Das Verfahren ist dadurch gekennzeichnet, daß man

Das crfindungsgcmäUc Verfahren gehl aus von einem bereits bekannten primären Acyclischen Alkohol der
Formel II, der nach H.J. Corey et al. (J. Arn. Chcm. Soc. 93, 1491 bis 1493 (1971)) hergestellt Werden kann.
Hin solcher Alkohol wird in der ersten Stufe des erfindungsgemäßen Verfahrens mit einem Oxydationsmittel, jo
vorzugsweise mit dem Komplex aus Thioanisol und Chlor oder der Komplexverbindung aus CrOj und Pyridin in
einem aprotischcn Lösungsmittel bei Temperaturen zwischen -500C und Raumtemperatur, bevorzugt zwischen
-300C und -5°C in inerter Atmosphäre zum Aldehyd der Formel IH oxydiert. Als Lösungsmitte! kommen
dabei z. B. aromatische Kohlenwasserstoffe wie Benzol oder Toluol oder z. S. chlorierte aliphatische Kohlenwasserstoffe
wie Tetrachlorkohlenstoff in Frage.
in der nächsten Stufe wird der erhaltene Aldehyd der Formel III nach Horner, Wittig und Emmons mit einem
Phssphonsäureesier tier Formel IV zu einem ungesättigten Keton der Formel V umgesetzt, wobei eine bevorzugte
Ausführungsform der Reaktion darin besteht, daß man das Natriumsalz des Phosphonsäureesters mit
Nalriumhydrid in Glykoldimethyläther herstellt, anschließend einen Aldehyd der Formel III hinzugibt und bei
Raumtemperatur 2 bis 6 Stunden reagieren läßt.
Die Phosphonsäurcestcr der Formel IV können durch Umsetzen eines Esters der Formel R-OX-CQv-A'kyI
in Gegenwart von überschüssigem Butyllithium und Methylphosphonsäuredimethyiester (z.B. nach Corey,
J. Am. Chcm. Soc. 88, 5654 (1966)) hergestellt werden.
Aus dem Keton der Formel Verhält man durch Reduktion mit einem komplexen Metallhydrid, vorzugsweise
mit einem Alkali- oder Zinkboranat in ätherischer Lösung, bevorzugt bei Temperaturen zwischen 0°C und
Raumtemperatur, das Epimerengeraisch der Alkohole der Formel VI. Das Zinkboranat wird bevorzugt in situ
aus Zinkchlorid und Natriumborhydrid in absoluter ätherischer Lösung dargestellt.
Die Alkohole der Formel VI eignen sich besonders für eine Trennung in die S- und R-Epimeren, vorzugsweise
mittels Säulcnchromatographie an Kieselgel, jedoch kann die weitere Umsetzung auch mit dem Epimerengemisch
durchgeführt und die Epimerentrennung auf der Stufe des Endproduktes vorgenommen werden.
Die anschließende hydrolytische Abspaltung der p-Phenylbencoylgruppe des Alkohols der Formel VT wird in
alkoholischem Medium mit Hilfe von Alkali- oder Erdalkalicarbonaten durchgeführt. Eine vorteilhafte Ausführungsform
besteht darin, den Alkohol bzw. das entsprechende. Epinverengemisch in absolutem Methanol bei
Raumtemperatur mit wasserfreiem Kaliumcarbonat zu behandeln, wobei ein Diol der Formel VJI entsteht.
Die I Icrstellung des Di-TetrahycIropyFanyläthers der Formel VIIl geschieht in einer ätherischen oder benzolisehen
Lösung der Alkohole der Formel VII in Gegenwart üblicher saurer Katalysatoren, wie z. B. Toluolsuifonsäurc.
Die Verbindung der Formel VIII wird mit einem komptexen Aluminiumhydrid in einem aprotischen
Lösungsmittel zu einem Lactol der Formel IX reduziert. Bevorzugt wird mit Diisobutylaluminiumhydrid in
Toiuol bei -6O0C bis -700C gearbeitet. 6t>
Das erhaltene Lacton der Formel IX kann ohne weitere Reinigung nach Wktig zu einer Carbonsäure der Formel
X umgesetzt werden. Die bevorzugte Ausführungsform folgi dabei der Sn J.Org.Chem. 28, 1128 (1963)
gegebenen Vorschrift
Zur Darstellung eines Prostaglandins der Ε-Reihe wird eine Verbindung der Formel X bei Temperaturen von
-40° C bis 0° C oxydiert, bevorzugt mit Jones-Reagenz (LösuTiS von Chrom (Vl)-oxid in Schwefelsäure) in Aceton
oder einer Komplexverbindung von Chrom-{VI)-oxid mit Pyridin in Methylenchlorid als Lösungsmittel bei
-20° C. Die erhaltene Verbindung der Formel XI wird durch Extraktion abgetrennt und, falls erforderlich, durch
Säulenchromatographie gereinigt.
Die Abspaltung der Ätherschutzgruppen in einer Verbindung der Formel XI erfolgt durch schonende saure
Hydrolyse der Tetrahydropyfanyläthergruppen durch wäßrige organische Säuren, vorzugsweise in 2%iger wäßrig-alkoholischer
Oxalsäurelösung bei 20° bis 5O0C oder durch 1 bis 2stündigcs Erhitzen in 60- bis 70%igcr
Essigsäure auf 40°C, wobei eine Carbonsäure der Formel I entsteht.
S Fails keine Epimerentrennung auf der Stwfe der Alkohole der Formel Vl erfolgt ist, kann eine Trennung des
15-S- von dem 15-R-Epimeren bevorzugt auf der Stufe einer Verbindung der Formel I vorgenommen wsrdcn.
Dabei erfolgt die Trennung vorzugsweise an Kieselgel (Merck®, 70-230 mesh), wobei das S5-S-Epimere meist
nach dem 15-R-Epimeren eluiert wird.
Als Elutionsmittel eignen sich Tür die säulenchromatographische Trennung der Verbindungen der Formel I
Mischungen von Chloroform und Methanol im Verhältnis 15 :1 bis 3 :2.
Die erfindungsgemäßen Verbindungen der allgemeinen Forme! I stellen nicht natürlich vorkommende Analoga
von Prostansäuren dar, die aufgrund ihrer pharmakologischen Wirkungen als Arzneimittel eingesetzt werden
können.
Die natürlichen Prostaglandine PGE10, PGE2,,, PGF3n oder PGA2 haben den Nachteil, daß sie im lebenden
Körper so schnell desaktiviert werden, daß sie ihre pnarmakologische Wirkung während der für die Therapie
eriorderlichen Zeit nicht aufrecht erhalten können.
Im Gegensatz dazu zeichnen sich die erfindungsgemäßen Verbindungen durch eine längere Wirkdauer und
eine stärkere Wirkung aus.
Die erfindungsgemäßen Verbindungen können als Arzneimittel mit blutdrucksenkender sowie diurelischcr
2Q Wirkung, prophylaktischer und therapeutischer Wirkung bei Thrombosen, geburtseinleitendcr Wirkung, als
Aborf !va, Kontraceptiva, als Mittel zur Hemmung der Magensaftsekretion sowie als Mittel gegen Magcn-ulecra
und gegen Asthma verwendet werden.
Die erfindungsgemäßen Verbindungen zeichnen sich besonders durch ihre cytoprotektive Wirkung aus und
sind daher insbesondere zur Behandlung und Verhinderung von Ulcera im Magen und Zwölffingerdarm gccignet.
Die cytoprotektive Wirkung auf die Magenschleimhaut wurde nach Robert, A., et al., (Gastrocnterology 77
(1979), S. 433-443) an Ratten geprüft. Da die Toxizität der erfindungsgemäßen Verbindung gering und daher
hinsichtlich der zu verabreichenden Mengen als unerheblich anzusehen ist, wurde als unerwünschte Nebenwirkung
die Erzeugung von Diarrhöen gewählt. Die Werte dafür - ausgedrückt als ED, t m„ d. h. der Menge an Substanz,
die an Ratten das intestinal Flüssigkeitsvolumen um 200% vergrößern - wurde irn Enteropoolingassay
nach Robert, A., et al., (Prostaglandins 11 (1976), S. 809-828) bestimmt.
Als Vergleichssubstanz wurde das mit dergleichen Indikation bereits im Handel erhältliche 16,16-Dimclhyl-PGE:
gewählt. Es ergeben sich die folgenden Werte:

Aus der Tabelle geht hervor, daß die erfindungsgemäßen Verbindungen einen wesentlich höheren therapeutischen
Quotienten (im Hinblick auf die Erzeugung von Diarrhöen) als die Vergieichssubstanz besitzen und damit
therapeutisch überlegen wirksam sind.
Die Verbindung gemäß Beispiel 11 a) ist ferner am Katzenmagenstreifen mit einem EDä0-Wert von 4,0 [.ig/ml
wesentlich schwächer spasmogen wirksam als die Vergieichssubstanz 16,16-Dimethyl-PGEj (ED50 =
0,27 ;xg/ml).
Aus der aktiven Verträglichkeitsprüfung an Mäusen ergibt sich weiterhin, daß Gaben von 1500 mg/kg der Verbindung
gemäß Beispiel 11 a) noch nicht letal wirken, was für diese Verbindung eine äußerst niedrige Toxi/'ilät
bedeutet im Vergleich zu verwandten Verbindungen (z.B. PGE2 : LD^0 = 70 mg/kg); 16,lA-Dimethyl-^i2-PGE,
: LD50 82,9 mg/kg).
Die erfindungsgemäßen Verbindungen können als freie Säuren, in Form ihre? physiologisch unbederikfehcn
anorganischen oder organischen Salze oder als Ester von aliphatischen, cycfoalipbaiischcn oder araliphatischen
Alkoholen zur Anwendung kommen. Als Salze kommen beispielsweise h* Bctrach! Benz&ylamnionkim-, Tri-(.5
äihanolammonium oder Morpholinsalzc sowie Alkalisalze, als Ehtcr vorzugsivc&e die iisler von niedrigen aliphatischen

Alkohole, Dimethylsulfoxid oder Dimethylformamid, auch in Gegenwart von pharmakologisch unbedenklichen
Polymerträgern wie z.B. Polyvinylpyrrolidon zur Anwendung gelangen.
Als Zubereitungen kommen die üblichen galenischen Infusions- oder Irtjektionslösungen und "!tabletten,
sowie örtlich anwendbare Zubereitungen wie Cremes, Emulsionen, Suppositorien cder Aerosole iin Frage.
Die Verbindungen können für sich alieine oder zusammen mit anderen pharmakologischen Wirkungen wie
/. B. Diuretika oder Antidiabctika zur Anwendung gelangen.
Die Verbindungen der Formeln 111, V, Vl, VII, VHi, IX, X, XI sind wertvolle Zwischenprodukte fur die Synthese
der erfindungsgemäßcn Verbindungen der Formel I.
Synthese von 2-Oxa-3-oxo-6syn-formyl-7-anti-p-biphenyl-carboxy-cis-bicyclo[3,3,0]octan (III)
In einem 2 1 Vierhalskolben werden 1,34 leiner Lösung aus 21,3 g Cl2 in 1,5 1 abs. CCl4 unter Argon vorgelegt
und auf -100C abgekühlt, dazu werden 33,3 g Thioanisol tropfenweise zugegeben, wobei ein wei£li;r Niederschlag
ausfallt.
Nach beendeter Zugabe wird auf -200C abgekühlt und 30 Minuten gerührt. Inzwischen bereitet man eine
Lösung aus 3G g Lactonalkohol (II) in maximal 30Ö mi CH2Ci2 abs. Diese Lösung wird bei -20°C siitgig zügetropft
und anschließend 2-3 Stunden bei -200C gerührt. Danach werden 54,3 g Triäthylamin gelösüt in 50 ml
CI !,Cl; abs. innerhalb 1 Stunde langsam zugetropft, wobei gegen Ende des Zulropfens die Temperaturauf-5° C
ansteigen darf.
Dann wird das Reaktionsgemisch in eine eisgekühlte Lösung aus 600 ml 1 % HCl und 1,5 Jl Diisoprapyläther
gegossen. Der ausgefallene, weiße Niederschlag 24 g wird auf einer möglichst großen Nutsche abge saugt und
mit Äther gewaschen. Das Filtrat wird in einen Scheidetrichter gegeben und die organische Phase abgetrennt,
getrocknet und bei höchstens +15°C eingeengt, auf ein Volumen von ca. 750 ml. Die ausgefallen«1« Kristalle
werden nach guter Kühlung abgesaugt (5 g) und mit dem Filterrückstand vereinigt.
Ausbeute: 29 g weiße Kristalle (98%)

a) Synthese von Dimethyl-2-oxo-3,3-dimethyl-4-äthoxy-butyl-phosphonat (IVa)
18 g Dimelhylmethylphosphonat wurden in 100 ml Tetrahydrofuran unter Argon auf-700C abgekühlt. Unter
Rühren werden 100 ml einer 2 molaren n-Butyllithium Lösung in Hexan zugetropft. Nach 15 Minuten wurden
20 g l,l-Dimethyl-2-äthoxy-propion-säuremethylester in 75 ml Tetrahydrofuran bei -700C zugetnü'pft. Dann
wurde 2 Stunden gerührt. Mit 12 ml Eisessig wurde neutralisiert. Das Lösungsmittel im Vakuum eingeengt, der
Rückstand in Chloroform aufgenommen und mit Wasser gewaschen, die Chloroformphase getrocknet mit Mg/
SO4, eingeengt und der Rückstand im Vakuum destilliert.
Ausbeute: 10,5 g farbloses Öl IVa (37%)

Zu einer Aufschlämmung von 0,67 g Natriumhydrid (80%ige Suspension in Öl) in 50 ml abs. 1,2 Dimethyloxyäthan
gibt man unter Argon innerhalb 15 Minuten 5 g Phosphonat IVa tropfenweise zu. Unter Entwicklung
von Wasserstoff tritt Lösung ein. Man rührt noch 40 Minuten und tropft dann 7,1 g Lactonaldehyd-(IIl) innerhalb
10 Minuten zu. Rührt noch 1 Stunde, neutralisiert mit Eisessig, klärt mit wenig Tierkohle, filtriert ab und engt
im Vakuum ein. Den Rückstand kristallisiert man aus iSU ml Isopropanol um. Hierbei wurden 5,1 g des
gewünschten Produkts erhalten.

Synthese von 2-Oxa-3-oxy-6-(3'-hydroxy-4',4'-dimethyl-6'-oxa-l'-octenyl)-7-(4"-biphenylcarbonyl)-
bicyclo[3,3,0]octan (Vl)
3,8 g der Verbindung Va wurden in 45 ml 1,2 Dimethoxyäthan gelöst, Bei 0°C wurden 30 ml einer 0,5 molaren 6o
Zinkborhydrid-Lösung (dargestellt wie folgt: 2,8 g Zinkchlorid in 45 ml 1,2 Dimethoxyäther,isuspendiert und
unter Kühlung und Rühren 1,52 g Natriumborhydrid zugegeben, 1/2 Stunde rühren und vom Ungelösten unter
Argon rasch abfiltriert) zugegeben. Bei Raumtemperatur wurde 2 1/2 Stunden gerührt. Dann wurde überschüssiges
Reagenz mit Eisessig bei 00C zersetzt. Mit Essigester-Wasser wurde das gesuchte Produkt extrahiert Die
organische Phase wurde mit Mg SO4 getrocknet, filtriert und im Vakuum eingeengt. 65

3,6 g der Verbindung VI wurden in 6(/ ml absolutem Methanol gelöst, bei Zimmertemperatur wurden dann
1,2 g feinst pulverisiertes Kaliumcarbonat zugegeben und unter Argon 2 1/2 Stunden gerührt. Dabei fiel p-Diphenylcarbonsäuremethylester
als kristalliner Niederschlag aus. Unter Eiskühlung wurde mit 1 N Salzsäure
auf pH 2 angesäuert, der p-Diphenylcarbonsäuremethylester pbgesaugt und das Filtrat mit Essigester-Wasser
* versetzt. Die organische Phase wurde nach dem Extrahieren abgetrennt, mit MgSO4 getrocknet und das
I Lösungsmittel im Vakuum entfernt. Die Ausbeute an der Verbindung VII betrug 1,86 g farbloses Ol (83 %).

Raumtemperatur gerührt, dann Essigester zugegeben und mit gesättigter Natriumbicarbonat-Lösung versetzt.
Die organische Phase wurde abgetrennt, mit Magnesiumsulfat getrocknet, das Lösungsmittel im Vakuum entfernt.
Der Rückstand (2,9 g farbloses Öl) wurde einer Saulcnchromatographie an Kicsclgcl (Merck) unterworfen.

(IX)
1,72 g der Verbindung VIII wurden in 25 ml Toluol gelöst, dann wurde auf -7O0C abgekühlt und innerhalb
von 3 Minuten unter einer Atmosphäre aus Argon 10 ml einer 1 M Lösung von Diisobutylaluminiumhydrid in
Toluol zugetropft. Zwei Stunden wurde bei -700C weitergerührt, dann mit 10 ml Methanol überschüssiges
Hydrierreagenz zersetzt. Mit Essigester und halb-gesättigter Natriumchlorid-Lösung wird das Reaktionsprodukt
extrahiert. Die organische Phase wird abgetrennt, mit Magnesiumsulfat getrocknet und das Lösungsmittel im ίο
Vakuum entfernt. Die Ausbeute an der Verbindung VIII betrug 1,66 g farbloses Öl (95%).

0,39 g Natriumhydrid (80%ige Suspension in Öl) wurde unter Argon mit 5 ml abs. Dimethylsulfoxyd versetzt
und I Stunde bei 60° C gerührt bis die Wasserstofientwicklung aufhört. Nach Abkühlung auf Raumtemperatur
wurden zu dieser Lösung tropfenweise 2,85 g 4-CarboxybutyItriphenyl-phosphoniumbromid (bei 12O0C im
Hochvakuum getrocknet) gelöst in 5 ml abs. Dimethylsulfoxyd zugetrorit. Dabei bildete sich unter intensiver
RotSirbung das für die Wittigreaktion benötigte Phosphorylid. Man rührte noch 30 Minuten bei 300C. Dann
wurden 1,66 g der Verbindung IX in 5 ml Dimethylsulfoxyd tropfenweise zugegeben. Es wurden 21/2 Stunden
bei Zimmertemperatur gerührt und anschließend auf Eiswasser gegeben, welches mit Diäthyläther überschichtcl
war. Man extrahiert die neutralen Substanzen und säuert die wäßrige Lösung mit 5%igerNatrh;mhydrogensullatlösung
unter Eiskühlung auf pH 2 an und extrahiert sofort mit Äther. Die Ätherlösung wird dann mit 0,5 N
Natronlauge extrahiert, die wäßrige alkalische Phase abgetrennt und erneut unter Eiskühlung angesäuert und
mit Äther extrahiert, die Ätherlösung wurde mit Magnesiumsulfat getrocknet, filtriert und im Vakuum eingeengt.
Die Ausbeute an der Verbindung IX betrug nach Säulenchromatographie an 250 g Kieselgel (LösungsmiUeisyslera:
Essigester-Essigsiiure 97SS : 2,5) L76 g, leicht gelbes Öl (90%).
Dünnschichtchromatogramm (Entwicklerlösung: Essigester-Essigsäure 97,5 : 2,5)
R1 = 0,56
Absorptionen im Infrarotspektrum (ohne Lösungsmittel):
3400 (OH-Bande), 2950, 1715 (Carbonyl-Bande), 1440, 1240, 1120, 1025, 970 cm"1.
Synthese von 9a-Oxo-llur, lSa-ditetrahydropyranyloxy-lo.lo-dimethyl-lS-oxa-S-cis-13-trans-prostadiensäure
(16,16-DimethyI-18-oxa-PGE2ir-l 1,15-bistetrahydropyranyläther) (XI)
0,85 g der Verbindung Xa werden in 30 ml Aceton gelöst. Bei -20°C bis -25° C werden unter Argon 2 ml
loncs Reagenz (2,1 g Chromsäure, 6 ml Wasser, 1,7 ml konz. Schwefelsäure) zugetropft. Man rührt 30 Minuten, o0
gibt dann 3 ml Isopropanol zu und rührt weitere 10 Minuten, um überschüssiges Oxydationsreagenz zu zerstören.
Es wurde dann mit 100 ml Methylenchlorid und 100 ml Wasser versetzt, ausgeschüttet, die Phasen
getrennt, der organische Extrakt mit MgSO4 getrocknet und das Lösungsmittel im Vakuum bei maximal +50C
eingeengt. Die Ausbeute an der Verbindung XI betrug 0,72 g fast farbloses, klares Öl (85%).

Absorptionen im Infrarotspektrum (NaCl-Platten):
2950, 1745 (Keton-Carbonyl), 1720 (Säure-Carbonyl), 1115, 1040, 970.
Synthese von 9-Oxo-ll a, lSar-dehydroxy-loJo-dimethyl-lS-oxa-S-cis-13-trans-prostadiensäure
(16,I6-DimethyI-18-oxa-PGE2ir) 15 S und 15 R Epimeren (I B)
0,72 g der Verbindung XIa wurden in 5 ml Tetrahydrofuran gelöst, 15 ml Essigsäure-Wasser im Verhältnis
2 : I zugegeben, und 4 Stunden bei 40° C gerührt. Dann wurde das Lösungsmittel im Vakuum unter mehrmali-
ger Zugabc von Benzol entfernt wobei die Temperatur +50C nicht übersteigen sollte. Die Ausbeute an der
Verbindung IB betrug, 0,6 g helles Öl. Nach Säulenchromatographic an Kicsclgcl-Mcrck (70-230 mesh) mit
Chloroform-Methanol = 22 :1 erhielt man in den Fraktionen (Einzclfraktion 2 ml)
S 130-170 104 mg 15REpimeres IB
u"d in den Fraktionen
170-280 102 mg 15 S Epimeres IB
Ausbeute: 0,206 g (41,5%)
Dünnschichtchromatogramm (Lösungsmittel: Essigsäureäthylester-Essigsäure = 97,5 :2,5) (Kieselgelplatten-Merck)

Die Spektren für das 15 R-und 15 S-Epimere von IB sind im Rahmen der üblichen Auflösung praktisch identisch.

1,4-2,7 Multiplen 12H (-CH2-, >CH-), 3,28 Singulett 2H (-(CH3)JC-CH2-O-),
3,46 Quartett 2 H (-OCH2CH3), 3,8-4,4 Multiplett 2H (>CH-OH),
5,25-5,75 Multiplett 4 H (olefinische H), 5,9-6,4 breites Singulett 3 H (2 x OH, 1 X COOH).

Durch H/D Austausch kann das Singual bei 5,9-6,4 ppm entfernt werden.

Claims (5)
- Patentansprüche:worin X einen gesättigten, verzweigten Alkylenrest mit 1 -5 C-Atomen und R die Methyl-, Äthyl-, Propyl-15 oder Isobutylgruppe bedeuten, sowie ihre physiologisch verträglichen Salze mit organischen und anorganischen Basen und ihre Alkylester mit 1—8 C-Atomen im Esterteil.
- 2. Verbindung der Formel I gemäß Anspruch 1 worin X die Gruppe - C(CH3J2 - CH2 - und R den VJiy lrcst darstellt.
- 3. Verwendung einer Verbindung der Formel I gemäß Anspruch 1 als cytoprotektives Mittel.20
- 4. Verfahren zur Herstellung von Verbindungen gemäß Anspruch 1, dadurch gekennzeichnet, daß man dieTetrahydropyranyt-Schutzgruppen in einer Verbindung der Füfffic! X!COOH X —O —RCXDworin X und R dieselbe Bedeutung haben wie in Formel I, durch saure Hydrolyse abspaltet und die erhaltene Verbindung der Formel I gegebenenfalls in ein physiologisch verträgliches Salz oder einen F.slcr überführt.
- 5. Verfahren zur Herstellung von Verbindungen gemäß Anspruch 1, dadurch gekennzeichnet, daß mana) den primären bicyclischen Alkohol der Formel IICH2OH(IDmit einem Oxydationsmittel in einem aprotis-cnen Lösungsmitte1 in inerter Atmosphäre zum Aldehyd der Formel IIICH== OCHI)oxydiert.b) den erhaltenen Aldehyd der Formel III mit einem Phosphonat der Formel IV CH3O O O\T I!P—CH2-C-X-O-R CH3OCHV) sworin X und R dieselbe Bedeutung wie in der Formel I haben, zu einem ungesättigten Keton der Formel V/ v_yX —O —R(V)=/ ίϊ O1520umsetztc) das erhaltene Keton der Formel V mit einem komplexen Metallhydrid zu dem Epimerengemisch der Alkohole der Formel VI/ V/Vc-όΧ—Ο —R3035worin X unJ R dieselbe Bedeutung wie in der Formel V haben, reduziertd) den erhaltenen Alkohol der Forme! VI als Epimerengemisch oder nach Trennung der Epimt,;en als reines S- oder R-Epimcres mit einem wasserfreien Alkali- oder Erdalkalicarbonat in alkoholischem Medium bei Raumtemperatur in ein Diol der Formel VII,Χ —Ο —R(νπ)45OHOHworin X und R dieselbe Bedeutung wie in Formel I haben, überführt,e) das erhaltene Diol der Formel VII durch sauer katalysierte Addition von 2,3-Dihydropyran in einem Di-Tctrahydropyranyläther der Formel VIIIO :Χ —Ο —R(Vffl)6065worin X und R die gleiche Bedeutung wie in der Formet I haben, überführtden erhaltenen Tetrahydropyranyläther der Formel VIII mit einem komplexen Aluminiumhydrid in einem aprotischen Lösungsmittel zu einem Lactol der Formel IXOHX-O — Rworin X und R die gleiche Bedeutung wie in Formel I haben, reduziertg) das erhaltene Lactol der Formel IX mit dem Ylid aus 4-Carboxy-buiyltriphenyIphosphoniumbromid in einer Lösung von Natriumhydrid im DimethylsulCoxyd in einer inerten Almosphiit c zu einer Säure der Formel X
