Die Erfindung betrifft den im Patentanspruch >o
gekennzeichneten Gegenstand. Die Verbindungen der Erfindung sind wertvolle Zwischenprodukte zur Herstellung
von öy-Dihydroxy-S.ey.e-tetrahydronaphthyloxyaminopropanolen,
die in der DE-OS 21 30 393 beschrieben sind.
Die Verbindungen der allgemeinen Formel I können ausgehend von 5,8-Dihydronaphthol der Formel II

hergestellt werden. Zu diesem Zweck wird das 5,8-Dihydronaphthol mit Essigsäureanhydrid in Gegenwart
einer organischen Base, wie Pyridin, zunächst in das entsprechende Acetat der Formel III

überführt.
Das Acetat der Formel III wird in Essigsäure gelöst und hierauf mit etwa 2 bis 4 Äquivalenten Silberacetat
und etwa 1 bis 2 Äquivalenten Jod versetzt. Das Gemisch wird auf eine Temperatur von etwa 80 bis
120° C während eines Zeitraums von etwa 1 bis 24 Stunden unter Stickstoff als Schutzgas erhitzt. Nach
Hydrolyse mit einer Base erhält man das trans-5,6,7,8-Tetrahydro-l,6,7-naphthalintriolder
Formel I-a

wird ebenfalls aus dem Acetat der Formel III hergestellt. In diesem Fall wird das Acetat in 92- bis 98prozentiger
Essigsäure, vorzugsweise 96prozentiger Essigsäure, gelöst und sodann auf die vorstehend beschriebene
Weise mit Silberacetat und Jod unter Stickstoff als Schutzgas behandelt Nach Hydrolyse mit einer Base
wird das cis-S.öy.e-Tetrahydro-l.ej-naphthalintriol I-b
erhalten.
Die Verbindungen der Formel I können nach dem in der DE-OS 21 30 393 beschriebenen Verfahren zu
öy-Dihydroxy-S.ej.S-tetrahydro-1 -[2,3-(epoxy)-propoxyl]-naphthalinen
und schließlich zu 6,7-Dihydroxy-5,6,7,8-tetrahydronaphthyloxyaminopropanolen der allgemeinen
Formel IV
Die trans-Verbindung der Formel I-a kann auch durch Umsetzung des 5,8-Dihydronaphthols der Formel II mit

umgesetzt werden, in der R einen unverzweigten oder verzweigten Alkylrest mit 1 bis 4 Kohlenstoffatomen
bedeutet. Die Verbindungen der allgemeinen Formel IV und ihre Salze mit Säuren sind wertvolle Arzneistoffe.
Insbesondere sind sie /J-Receptorenblocker, da sie die
Wirkung von Sympathomimetika an adrenergen /J-Receptoren hemmen. Wie alle bekannten /9-Receptoblokker
haben sich auch eine antihypertensive Wirkung. Die Verbindungen können oral oder parentereal in üblichen
Darreichungsformen gegeben werden.
Eine bevorzugte Verbindung ist das l-(tert.-Butylamino)-3-[(5,6,7,8-tetrahydro-cis-6,7-dihydroxy-1 -naphthyl)]-oxyl-propan-2-ol (Nadolol), das sich durch eine extrem lange biologische Halbwertszeit von 20 bis 24 Stunden auszeichnet. Dies ermöglicht eine Anwendungsweise von 1 χ täglich. Trotz der langen Halbwertszeit erfolgt bei normaler Nierenfunktion keine Kumulation. Nadolol steigert im Gegensatz zu anderen bekannten /J-Receptorenblockern die Nierendurchblutung.
Eine bevorzugte Verbindung ist das l-(tert.-Butylamino)-3-[(5,6,7,8-tetrahydro-cis-6,7-dihydroxy-1 -naphthyl)]-oxyl-propan-2-ol (Nadolol), das sich durch eine extrem lange biologische Halbwertszeit von 20 bis 24 Stunden auszeichnet. Einige Vergleichsversuche mit dem Standard /S-Receptorblocker Propranolol sind nachstehend wiedergegeben.
Sie zeigen die Überlegenheit einiger Verbindungen der allgemeinen Formel IV.

24-Stunden-Wirkung im Tierversuch
Die Wirkungsdauer von Nadolol wurde — im Vergleich zu Propranol — an Hunden bestimmt, wobei
die Hemmung der belastungsinduzierten Tachykardie als Parameter dieme. In einer cross-over Untersuchung
erhielten Hunde während 5 Tagen täglich eine Dosis von 0,5 mg/kg.

Schlußfolgerungen
Zunächst hemmten beide Substanzen die belastungsinduzierte Tachykardie in etwa gleichem Ausmaß. Nach
24 Stunden jedoch zeigte Propranolol keinerlei Schutzwirkung mehr gegenüber dieser Streßbelastung.
Nadolol hingegen hemmte auch noch nach 24 Stunden den unerwünschten Pulsfrequenzanstieg um 82%.
Versuch 2
Direkte kardiodepressive Wirkung
Direkte kardiodepressive Wirkung
In einer Vergleichsuntersuchung von Nadolol gegen
Sotalol, Atenolol, Timolol, Acebutolol, Pindolol, Metroprolol, Alprenlol, Oxprenolol und Propranolol wurde
der Effekt dieser Sympathikolytika auf die myokardial Kontraktion am isolierten Meerscheinchenvorhofmuskel
untersucht. Unter Nadolol, Sotalol und Atenolol blieb die Kontraktionskraft voll erhalten. Eine 20prozentige
Verminderung der Kontraktionskraft erfolgte dagegen unter 100 bis 1000μg/ml Timolol und
Acebutolol; für dje anderen Betasympathikolytika Pindolol, Metroprolol, Butolol, Alprenolol, Oxprenolol
und Propranolol wurde ein 20prozentiger Abfall der Kontraktionskaft schon im Dosisbereich zwischen 10
und 100 μg/ml beobachtet.
Bestimmung der myokardialen Sicherheit
Atherosklerotische Kaninchen (nach 2% Cholesterindiät für 10 bis 14 Wochen) erhielten Dosen von
1 mg/kg/min. i.v. folgender Betasympathikolytika: Nadolol, Pindolol, Timolol, Atenolol, Oxprenolol, Alprenolol
und Propranolol. Es wurden folgende Parameter der myokardialen Kontraktilität untersucht: dp/dr (Kontraktionskraft),
Puls sowie linksventrikulärer Druck. Alle untersuchten Substanzen verursachten zunächst
die therapeutisch erwünschte Hemmung dieser Parameter als Resultat der Betasympathikolyse. Beim Weiterlaufen
der Infusion konnten die Effekte der verschiedenen Substanzen auf die myokardiale Kontraktilität
analysiert werden. Die MDso(also die toxische Dosis, die
die myokardiale Kontraktilität (dp/at) um 50% reduzierte)
sowie die ID5G (die »therapeutische« Dosis, die die Isoproterenol-induzierte Tachydardie um 50%
reduzierte) wurden für jede Substanz bestimmt und eine Meßzahl für die myokardiale Sicherheit (MD50/ID50)
errechnet.
Die Resultate zeigt Tabelle I.

Bestimmung der therapeutischen Dosis, die die Isoproterenol-induzierte Tachykardie um 50% reduzierte,
sowie der maximalen Dosis, die noch keine signifikante Verminderung der Kontraktilität des
Myokards hervorrief.
Die Versuche wurden am isolierten Meerschweinchenvorhofmusik durchgeführt. Die Ergebnisse sind in
Tabelle II durch die molare Konzentration angegeben,

Die Beispiele erläutern die Herstellung der Verbindungen der Erfindung.
-1,6,7-naphthalintriol
(a) 5,6,7,8-Tetrahydro-6,7-epoxy-1 -naphthol
25,0 g (etwa 0,12 Mol) m-Chlo-perbenzoesäure
werden innerhalb 10 Minuten zu einer eisgekühlten Lösung von 14,6 g (0,10 Mol) 5,8-Dihydro-l-naphthol in
225 mJ Äthylacetat gegeben. Das Napthol wurde gemäß der Arbeitsweise von Organic Syntheses, Coll. Vol. IV,
S. 887, hergestellt. Das Gemisch wird 16 Stunden bei Raumtemperatur stehengelassen und sodann in ein
gekühltes Gemisch aus 300 ml Diäthyläther und 300 ml lOprozentiger wäßrigen Natriumbicarbonatlösung eingerührt.
Nach 15 Minuten wird die organische Phase abgetrennt, mit Wasser und gesättigter Kochsalzlösung
gewaschen und getrocknet. Danach wird das Lösungsmittel abdestilliert. Es hinterbleibt ein Öl, das zweimal
mit jeweils 100 ml siedendem Hexan digeriert wird. Der Rückstand wird aus 150 ml eines 1 :1 Gemisches aus
Hexan und Äthylacetat umkristallisiert. Es werden 6,6 g (41% d.Th.) 5,6,7,8-Tetrahydro-6,7-epoxy-l-naphthol
vom F. 143 bis 146°C erhalten. Nach zweimaliger Umkristallisation einer kleinen Probe schmilzt das
Analysenpräparat bei 149,5 bis 155° C.

(b) trans-5,6,7,8-Tetrahydro-1,6,7-naphthalintriol
Eine Lösung von 8,0 g (0,048 Mol) 5,6,7,8-Tetrahydro-6,7-epoxy-l-naphthol
in 100 ml Tetrahydrofuran wird auf 00C abgekühlt und mit 20 ml Wasser sowie 0,5 ml
70prozentigf:r Perchlorsäure versetzt. Nach 4 Stunden werden nochmals 1,5 ml Perchlorsäure zugegeben, und
die Lösung wird 16 Stunden bei Raumtemperatur gerührt. Danach wird die Lösung mit 100 ml Diäthyläther,
100 ml lOprozentiger wäßriger Natriumbicarbonatlösung und 100 ml gesättigter Kochsalzlösung
verdünnt. Die wäßrige Phase wird abgetrennt und mit 150 ml eines 1:1 Gemisches aus Diäthyläther und
Tetrahydrofuran ausgeschüttelt Die organischen Phasen werden vereinigt, mit gesättigter Kochsalzlösung
gewaschen, über Natriumsulfat getrocknet und eingedampft Es hinterbleibt ein öl, das beim Digerieren mit
Chloroform sich verfestigt. Die Umkristallisation 'liefert insgesamt 4,85 g eines Feststoffs, der nochmals aus
Äthylacetat umkristallisiert wird. Ey werden 3,84 g vom F. 179,5 bis 181,5°C erhalten. Nach zwei weiteren
Umkristallisationen einer kleinen Probe schmilzt das Analysenpräparat bei 183 bis 184° C.

-1,6,7-naphthalintriol
29,2g (0,2 Mol) 5,8-Dihydro-l-naphthol werden in 40 ml Essigsäureanhydrid und 100 ml Pyridin gelöst. Die
Lösung wird 16 Stunden stehengelassen. Danach wird das Lösungsmittel unter vermindertem Druck abdestilliert,
der Rückstand in Diäthyläther aufgenommen und mit 200 ml 5prozentiger Salzsäure, Wasser, 200 ml
lOprozentiger Natronlauge und gesättigter Kochsalzlösung gewaschen und getrocknet. Danach wird das
Lösungsmittel abdestilliert. Es hinterbleiben 34,2 g (90,5% d. Th.) des rohen Acetats, das in 900 ml
Essigsäure und 36 ml Wasser gelöst wird. Die Lösung wird mit 53,3 g (0,32 Mol) Silberacetat und sodann mit
40.6 g (0,16 Grammatom) Jod versetzt. Die Aufschlämmung wird 3 Stunden unter Stickstoff als Schutzgas auf
85 ± 10°C erhitzt und stark gerührt. Danach wird die Aufschlämmung abgekühlt und filtriert. Das Filtrat wird
unter vermindertem Druck eingedampft und der erhaltene Rückstand in 250 ml Methanol gelöst und auf
00C abgekühlt. Hierauf wird eine Lösung von 40 g Natriumhydroxid in 200 ml Wasser unter Stickstoff als
Schutzgas zugegeben und das Gemisch 17 Stunden gerührt. Die Hauptmenge des Methanols wird danach
unter vermindertem Druck abdestilliert. Der erhaltene feste Rückstand wird abfiltriert, in 150 ml Wasser gelöst
und mit 20 ml konzentrierter Salzsäure angesäuert. Beim Abkühlen erfolgt Kristallisation. Die Kristalle
werden abfiltriert und getrocknet. Es werden 16,5 g cis-S^y.e-Tetrahydro-l.ey-naphthalintriol vom F. 184,5
bis 187°C erhalten. Nach dreimaliger Umkristallisation aus wasserfreiem Äthanol schmilzt das Analysenpräparat
bei 188 bis 188,5° C.
