Aus der Tabelle läßt sich ableeen, daß die e rf indungs gemäße -■
Verbindung I anabol lOmal wirksamer ist als Verbindung II
bzw. ^Omal wirksamer als Verbindung III· Des weiteren ist zu
erkennen-, daß sich das Samenblasengewicht im fall der erfin-
* . dungsgemäßen Verbindung I nicht erhöht. ·
Bekanntermaßen hat die Einführung einer 7-Methylgruppe einen
Einfluß auf die androgene und anabole Wirksamkeit von Steroiden
der Androstanreihe (Julius A. Vida "Androgens and Anabolic Agents"
^ Academic Press New York and London 1969)*
Es war zu erwarten, daß eine 7oc-Methylgruppe sowohl die erwünschte
anabole Wirkung als auch die unerwünschte androgene Hebenwirkung erhöhen würde. Es ist daher überraschend, daß die
erfindungsgemäßen Verbindungen in bezug auf ihre androgene
Wirksamkeit praktisch unwirksam sind, wodurch die Dissoziation zwischen anaboler und androgener Wirkung stark hervortritt·
Di© neuen Wirkstoffe sind bei allen Indikationen anwendbar,
bei denen eine Förderung des Eiweißaufbaues notwendig ist,
" wie z. B. bei Rekonvaleszenz," reduzierten» Allgemeinzustand,
* konsumierenden Erkrankungen , kachektischen Zuständen, Strahlen-
und Zytostatikatherapie, bei fortgeschrittenem Mamma- und
Genitalkarzinom als Zusatzbehandlung, bei Anämie zur Förderung
der Erythropoese, Langzeitbehandlung mit Kortikoiden, Osteoporose,
chronischen Leber- und Nierenerkrankungen, Hyperthyreose, Muskeldystrophie, diabetischer Retinopathie, bei
schlecht gedeihenden und dystrophisehen Säuglingen, bei

Lösungen in Ampullen zur intramuskulären Injektion genannt.
Für die orale Anwendung koimaen u. a. Tablett en 5, Sapp ein, Pillen,
Bragees, Granulate» Suspensionen und Lösungen laBetracht·
Sie Erfindung betrifft außerdem ein Verfahren zur Herstellung

Durch Chromatographie an Kieselgel und Umkristallisieren
aus Aceton/Hexan werden 260 mg la,2a-methylen-4-anärosten-3-on vom Schmelzpunkt 153-153»5° C
erhalten. ,

Eine Löaung von Methylmagneeiumjodid, hergestellt aus 3,5 g
Magnesiumspänen und 13 ml Methyljodid in 100 ml Äther» wird
mit 250 ml Tetrahydrofuran verdünnt. Man destilliert so lange
das Lösungsmittel ab, bis der Siedepunkt 62° C beträgt· Dann gibt man zn der auf Eaumtemperatur abgekühlten Suspension
unter starkem Hühren 750 mg Kupfer- (I) -chlorid-Lösung und
tropft sofort anschließend eine Lösung von 5 g 17ß~Acetoxyla,2a-methylen-4»6-androstadien-3-on
zu« Es wird noch 10 Hinuten nachgerührt und dann das Keaktionsgemisöh mit gesättigter
Ammoniumchloridlösung versetzt. Nach Ätherextraktion wird mit Essigsäureanhydrid in Eyridin nachacetyliert. Durch Chromatograph!
er en an Kieselgel mit 5-6,8 i» Aeeton/Hexaa. werden \,\ g
17ß-Acetoxy-7a-methyl-la,2a-methylen-5-androeten-3'-oa vom
Schmelzpunkt 121-122° C erhalten, das in Aceton mit 0,2 ml 70#iger Perchlorsäure zum 17ß-Aoetoxy-7«-aethyl-l«f2o>methyXen~
4-androsten-3~on vom Schmelzpunkt 155-156° C isomerisiert wird»
Ausbeute; 460 mg· »
= 14 500.

120 mg 17ß-Ac©tox3r--7a-nietliyl--li3t,2a~met]byleii-4«-aiidrosten~3'-on
werden in 1,5 ml Methylenchlorid und 3,5 ml Methanol mit 2,0
ml ln~Natriuinhydroxidlösung 1 Stunde bei Raumtemperatur unter
Stickstoff atmosphäre gerührt. Dann wird mit Essigsäure neutralisiert,
die Lösung im Vakuum weitgehend eingeengt, das Reaktionsprodukt mit Methylencnlorid extrahiert und die
Lösung mit Wasser gewaschen und getrocknet. Durch chromatographische Reinigung werden 70 mg 17ß-Hydroxy-7a'-methyl-»la,
2a-methylen-4-androsten-3-on als Schaum erhalten.

werden in 0,5 al Pyridin mit 0,3 ml Propionsäureanhydrid bei
Raumtemperatur gefiührt· Dann verdünnt man die Lösung mit
Methylenchlorid und schüttelt zur Abtrennung des Pyrldins mehr«=
male mit HCl (lsi)· Anschließend neutralisiert ma.n mit Natrium·
hydrogencarbonatlöBung und wäscht mit Wasser nach. Nach■ Chroma«
tDgraphieren an Kieselgel mit Aceton/Hexasi 'und Umkristallisieren aus'Aeeton/H©3tan werden 60 mg ITß-PsOplonylox^i'^

werden in 5»5 ml Capronsäureannydrid unter Zusatz einer
katalytischen Menge p-IoluolsSulfonsäure 24 Stunden bei Raumtemperatur
gerührt· Das Reaktionsgemisch wird mit Aceton verdünnt
und einer Wasserdampfdestillation unterzogen· Dann wird
das Reaktionsprodukt mit Methylenehlοrid extrahiert und das
Rohprodukt an Kieselgel mit Aceton/Hexan chromatographiert«
Man eluiert 45 mg 17ß-Hexanoyloxy-7a-metnyl-laf2«-methylen>4
