Verfahren zur Darstellung von Isatin und seinen im Benzolkern monomethylierten Derivaten.
Die Darstellung von Isatin durch Einwirkung von konzentrierter Schwefelsäure auf
Körper vom Typus
C6U5 NH- C-

ist von Sandmeyer (Ztschr. f. Farben und Textilchemie 2 [1903], S. 129; Geigy &Comp.
Patentschrift 113981, Kl. 12p) zuerst beschrieben.
Reißert (Ber. 37 [1904], S. 3725) untersuchte andere Derivate der Thiooxaminsäure
und fand, daß nur bei Oxanilsäurethiamid und Thiooxanilid mit konzentrierter Schwefelsäure in der Wärme Isatinbildung
stattfindet. Während aber die einseitig substituierten Derivate nur spurenweise Isatin
liefern, erhielt Reißert aus dem zweiseitig substituierten Monothiooxanilid
C.HKNH—CO-
C S-NHC6H6
bis 25 Prozent der theoretischen Ausbeute.
Die Versuche von Schultz, Rhode und
Herzog (J. pr. Ch. [2] 74 [1906], S. 74 u. S. 76) aus Hydrocyancarbodiphenylimid mittels konzentrierter
Schwefelsäure Isatin zu gewinnen, lieferten keine brauchbaren Ausbeuten.
Es wurde gefunden, daß sich Isatin und seine im Benzolkern monomethylierten Derivate
herstellen lassen, wenn man konzentrierte Schwefelsäure auf die substituierten Imidchloride
der Oxalsäure vom Typus

(wobei R ein Phenyl- oder ein o-, m-, p-Tolylrest ist) in der Wärme einwirken
läßt.
Derartige Imidchloride, die bisher noch nicht untersucht worden sind, können leicht
erhalten werden aus Diphenyl- bzw. Ditolyloxamid durch Kochen mit Phosphorpentachlorid
in Benzol oder Toluollösung.
Diphenyloxalimidchlorid ist von Wallach (Ber. 12 [1879], S. 1065; 13 [1880], S. 527
1. Abs.; 14 [1881], S. 740) dargestellt, aber
nicht im reinen Zustand isoliert und analysiert worden. Es schmilzt bei 1150, gelbe Blättchen
aus Ligroin.
Di-o-tolyloxalimidchlorid ist dargestellt und
analysiert von Bischoff (Ann: 279 [1894], S. 181 ff.) Schmp. 131 °, gelbe Nadeln aus
Benzol.
Di7m-tolyloxalimidchlorid Schmp. 720, gelbe
Prismen aus Gasolin.
Di-p-tolyloxalimidchlorid Schmp. 1070, gelbe
Blättchen aus Ligroin.
Die beiden letzterwähnten Imidchloride sind in der Literatur noch nicht beschrieben und
analysiert worden.
5 Gewichtsteile Di-o-tolyloxalimidchlorid
werden in kleinen Portionen in die fünffache Menge reiner konzentrierter Schwefelsäure,
welche auf dem Wasserbad erhitzt wird, eingetragen. Unter Aufschäumen entweicht Salzsäuregas.
Wenn eine Probe eine klare Lösung in Wasser gibt, die auch beim Alkalischmachen klar bleibt, läßt man das Gemisch
in Wasser einfließen. Die anfänglich klare Lösung trübt sich bald unter Ausscheidung
roter Kristallhaare von o-Methylisatin

welche bereits nach einmaligem Umkristallisieren aus Alkohol chemisch rein sind. Die
Ausbeute beträgt bis zu 50 Prozent der Theorie. Das o-Methylisatin ist unlöslich in Benzol,
Toluol, Äther, Ligroin, spurenweise löslich in Essigäther und Chloroform, sehr schwer löslich in Wasser, Alkohol, Methylalkohol,
Aceton, Amylalkohol, Eisessig, leicht löslich in Pyridin. Es sublimiert leicht in
langen roten Nadeln. Umkristallisiert aus Alkohol oder Wasser bildet es rote haarförmige
Kristalle vom Schmelzpunkt 2660. Es gibt die Indopheninreaktiori und zeigt
beim Lösen in Alkali den für Isatin charäkteristischeri
Farbumschlag. Vom Isatin unterscheidet es sich durch die Form der Kristalle
und durch seine viel geringere Löslichkeit in Wasser. 1 g o-Methylisatin braucht zur
Lösung etwa 1,5 1 siedendes Wasser, aus der es beim Abkühlen sich fast vollkommen und
sehr gut kristallisiert wieder abscheidet.
Sein Phenylhydrazon entsteht durch Versetzen einer wässerigen Lösung des o-Methylisatins
mit einer ■ essigsauren Lösung von Phenylhydrazin. Es kristallisiert aus Alkohol
mit etwas Eisessig in goldgelben Nadeln und schmilzt bei 2420. Es ist sehr schwer löslich
in den gewöhnlichen Lösungsmitteln, etwas leichter in Eisessig.
Das o-Methylisatoxim fällt aus der wässerigen
Lösung des o-Methylisatins aus, wenn man zu derselben eine freies Hydroxylamin
enthaltende wässerige Lösung zusetzt. Es ist löslich in Alkohol, Aceton, Essigäther
und Eisessig, sehr schwer löslich in Wasser, unlöslich in Benzol. Aus Alkohol umkristallisiert
bildet es gelbe Nadeln vom Schmelzpunkt 23 50.
Das o-Methylisatin ist nur einmal und ohne Beschreibung seiner Eigenschaften in 60-der
chemischen Literatur im Patent 105102, Kl. 12 (Friedlaender, Fortschritte der Teerfarb.
Fabr. V, S 395) erwähnt.
Beim Behandeln der anderen Imidchloride, welche sich vom Anilin, m- oder p-Toluidin
ableiten, scheidet sich das gewünschte Isatin, das sich in geringerer Ausbeute bildet, .nicht
sofort aus der wässerig-schwefelsauren Lösung' aus. In diesem Falle wird die Lösung in
der Wärme mit Ätzbaryt versetzt, bis man die Bildung des Bariumsalzes der Isatinsäure
an dem Farbumschlag violett in farblos erkannt hat. Aus der vom Bariumsalzschlamm
abfiltrierten Lösung wird das nicht gebundene Bariumhydroxyd mit Kohlensäure entfernt
und die Lösung stark eingedunstet. Säuert man darauf in der Kälte mit Salzsäure an,
so fallen Sulfosäuren des Anilins bzw. des m- oder p-Toluidins aus. Das fast farblose
Filtrat davon wird zum Sieden erhitzt, dabei geht seine Farbe in die tiefrote der
Isatinlösung über. Beim Abkühlen kristallisiert das betreffende Isatin (vgl. Ber. 38
[1905], S. 3552) aus.

- .. Patent-Anspruch:·Verfahren zur Darstellung von Isatin und seinen.; im Benzolkern tnonomethylierten Derivaten, darin bestehend, daß man Diphenyloxalimidchlorid bzw. o-, m- oder p-Ditolyloxalimidchlorid mit konzentrierter Schwefelsäure: auf dem Wasserbäde erwärmt und das betreffende Isatin entweder durch Eingießen der Schwefelsäurelösung in Wasser unmittelbar ausfällt oder nach Entfernen der Schwefelsäure mit Ätzbäryt oder Ätzkalk die Lösung eindunstet und dann mit Salzsäure das Isatin ausfällt.
