DEUTSCHES
Die Erfindung betrifft ein Verfahren zur Herstellung von Aminosäureamiden und verwandten Verbindungen
aus leicht zugänglichen und daher billigen Ausgangsmaterialien auf einem besonders einfachen und billigen
Wege, und zwar vorzugsweise in einer einstufigen Reaktion. Bisher wurden Aminosäureamide und deren
Derivate durch Umsetzung von Aminen mit Aminosäureabkömmlingen, z. B. Aminosäureestern und Aminosäurehalogeniden,
dargestellt.
Die Erfindung beruht auf der Feststellung, daß Aminosäureamide und -thioamide durch Umsetzung von
Isonitrilen oder Isocyaniden mit Imoniumionen bequem und in guter Ausbeute zugänglich sind. Die meisten der
bekannten Reaktionen der Isonitrile stellen eine ionische Addition je eines elektrophilen und eines nucleophilen
Partners am »zweiwertigen« Kohlenstoffatom des Isonitrils gemäß dem folgenden Schema dar:

Es ist bekannt, daß einer Reihe von Kondensationsreaktionen, z. B. der Mannichkondensation, der Bettireaktion,
der Pictet-Spengler-Synthese, der Strecker-Synthese, und einigen speziellen Reaktionen, wie der
Robinsonschen Tropinon-Synthese, ein gemeinsamer Mechanismus zugrunde liegt, der darin besteht, daß ein
Imoniumion mit einem Kohlenstoffatom hoher nucleophiler Aktivität kondensiert.
Imoniumionen sind Reaktionsteilnehmer ausgeprägter elektrophiler Reaktivität und zeichnen sich dadurch aus,
daß sie im Gegensatz zu den meisten anderen elektrophilen Agenzien in relativ schwach saurem Medium
gebildet werden und reagieren können. Die Erfindung geht von der Überlegung aus, daß Imoniumionen infolgedessen
besonders günstige Reaktionspariner für Isonitrile sind. Sie reagieren als elektrophiler Partner (A®) unter
gleichzeitigem Einbau eines zur Verfügung stehenden nucleophilen Agens (B9) mit Isonitrilen gemäß obigem
Schema unter Bildung eines Primäradduktes, welchem die Struktur einer Imino-acyl-verbindung

zukommt. Diese labilen Primäraddukte stabilisieren sich Verfahren zur Herstellung
von Aminocarbonsäureamiden
und deren Abkömmlingen

durch Sekundärreaktionen, deren Art durch die Konstitution des Imoniumions und des nucleophilen Agens
bestimmt wird.
Auf Grund dieser Überlegung sieht die Erfindung ein Verfahren vor, bei dem man einen Aldehyd oder ein
Keton mit einem Amin bzw. die hieraus entstehende Schiffsche Base oder das entsprechende Enamin mit einem
Isonitril in Gegenwart von Wasser oder anderen nucleophilen Agenzien, wie Sulfiden, Aziden, Essigsäure oder
einem Amin, umsetzt. In der einfachsten Ausführungsform läßt man Isonitril, Carbonylverbindung und Amin
in Gegenwart einer Säure als Katalysator reagieren. In der Regel ist es zweckmäßig, die Reaktion unter Eiskühlung
durchzuführen. In vielen Fällen erweist es sich als zweckmäßig, die Reaktion in einem Gemisch aus
Wasser und einem organischen Lösungsmittel durchzuführen.
Das Verfahren nach der Erfindung verläuft nach folgendem Schema:

In diesen Gleichungen ' bedeutet R Wasserstoff, R1 aus Petroläther gereinigt werden kann. Schmelzpunkt

3,39 g Isobutylen-n-propylamin werden in 20 ecm 50 Lösungsmittel und die überschüssigen Komponenten im
Methanol gelöst und mit 2,70 g Cyclohexylisonitril Vakuum abgezogen. Die Rohausbeute an festem Produkt
versetzt. Unter Rühren und Eiskühlung werden nun ist 6,5 g. Das Produkt läßt sich durch Tieftemperatur-

4,98 g N-Cyclohexenylpiperidin werden in 20 ecm Vakuum eingeengt. Die Ausbeute an festem Rohprodukt

setzt. Unter Rühren und Eiskühlung werden nun 15 ecm werden 6,0 g Reinprodukt erhalten. Schmelzpunkt 148,5

werden in 40 ecm Methanol gelöst. Das Isonitril wird als letztes zugegeben. Nach 1 Stunde riecht
das Reaktionsgemisch nicht mehr nach Isonitril. Nach dem Stehen über Nacht wird das Lösungsmittel abgezogen.
Es bleibt ein kristalliner Rückstand zurück, der mit Benzol behandelt wird. Die filtrierte Lösung wird
eingeengt, der kristalline Rückstand schmilzt bei 120 bis 122° C und wiegt 6,3 g. Nach Umkristallisieren aus Benzol
steigt der Schmelzpunkt auf 145 bis 153°C an. Laut IR-Spektrum handelt es sich um das Amidin.

— CH2 — CS- NH — CH2 — C6H5
3,26 g Dimethylaminhydrochlorid werden in 2,50 ecm lOmolarer wäßriger Formaldehydlösung gelöst und mit
2 Tropfen 2 η-Natronlauge versetzt. 12,40 g Natriumthiosulfat (Pentahydrat) werden in möglichst wenig
Wasser gelöst und zugegeben. Unter Rühren wird die Lösung von 2,92 g Benzylisonitril in 30 ecm Tetrahydrofuran
eingetragen. Im Laufe der Zeit scheidet sich aus dem übersättigten System kristallines Natriumthiosulfat
aus, das durch Zugabe einiger Tropfen Wasser immer wieder in Lösung gebracht wird. Nach 3stündigem
Rühren bei Raumtemperatur wird das Reaktionsgemisch durch Zugabe von festem Kaliumcarbonat alkalisch gemacht.
Die Tetrahydrofuranschicht wird abgetrennt, die wäßrige Schicht wird noch zweimal mit je 15 ecm Tetrahydrofuran
ausgeschüttelt. Die vereinigten Tetrahydrofuranlösungen werden mit wasserfreiem Kaliumcarbonat
getrocknet und bei 150 bis 300 Torr eingeengt. Es bleiben 6,57 g eines gelben Öls zurück. Dieses Öl wird in 70 ecm
Benzol—Petroläther (1:1) gelöst. Bei O0C wird mit
Chlorwasserstoff gesättigt. Das ausgeschiedene Hydrochlorid wird abgesaugt. Die Ausbeute beträgt 5,03 g
(82°/0 der Theorie, Schmelzpunkt ab 160° C unter Zersetzung)
. Das Hydrochlorid wird durch Umkristallisieren aus Methanol gereinigt. Schmelzpunkt 170 bis 171° C
unter Zersetzung.
Claims (3)
1. Verfahren zur Herstellung von Aminocarbonsäureamiden und deren Abkömmlingen, dadurch ge
kennzeichnet, daß man einen Aldehyd oder ein Keton mit einem Arnin bzw. die hieraus entstehende
Schiffsche Base oder das entsprechende Enamin mit einem Isonitril in Gegenwart von Wasser oder Sulfiden,
Aziden, Essigsäure oder einem Amin umsetzt.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, daß man die Umsetzung in Gegenwart einer
Säure durchführt.
3. Verfahren nach Anspruch 1, dadurch gekennzeichnet, daß man die Reaktion in einem Gemisch aus
Wasser und einem organischen Lösungsmittel durchführt.
